Abstract

The purpose of study was to determine kinetic parameters and inhibitory effect of Mg2+ ions on pectinase. Activity test performed at pH 4,5 temperature 55oC for 50' with 10 mm concentrations of each ion 0-10mm. Galacturonic acid content, as product of pectin substrate hydrolysis, was used as basis for determining activity and analyzed by visible spectrophotometry. At concentrations 2 and 4 mm Mg2+ act as an activator, but at concentrations 6, 8, and 10 mM as inhibitors. The value of KM with and without inhibitors is almost same, that is 0.3145% and 0.3105% but value of Vm from both are different, that is 80,645 ?g.ml-1.minute-1 and 62,112 ?g.ml-1.minute-1. The conclusion that type of inhibitory pectinase with Mg2+ is a non-competitive inhibition. The inhibition constant value (Ki) is 26,84.

Abstract

The Covid-19 pandemic caused the effects of learning activities from home. The phenomenon requires improvisation as a quick response so that the teaching and learning process continues to run and maintains learning motivation, especially in the scope of college. Activities that are assumed to be able to maintain motivation online is to provide tasks in the form of projects or Project Based Learning (PBL) which in this context does not mean eliminating face-to-face activities offline, but in the sense of providing sustainable projects in one semester and staying connected to lecture materials. This PBL is combined with Collaborative Learning (CL) in order to conduct a continuous project review. This research was conducted in Ma Chung University's Information Systems study program for Programming Language courses. The results of this implementation are evaluated by conducting FGD (Focus Group Discussion) in two stages. In the first stage there are some shortcomings that are then corrected by adding the obligation to upload the project to Github and intensify the involvement of lecturers. So in the second stage, FGD results show improved results. The results of the study are generic prescriptions, namely: the implementation of PBL accompanied by a mature plan and preparation, improvisation of PBL accompanied by the application of CL accompanied by the role of lecturers as facilitators need to be evaluated gradually, and the implementation of online collaboration can be applied because students can quickly adapt online. Abstract

One form of efficiency that can be done by palm oil mill companies so that production targets are achieved is to reduce the number of oil losses or loss of oil in the process of making their products, in this case, are CPO and PKO. This is because the lower the amount of oil losses, the higher the CPO and PKO that will be produced. This study aims to analyze oil losses in the processing of Palm Kernel Oil or PKO with the seven tools method. Data collection techniques in this study used case studies based on literature studies and field studies. Data were analyzed descriptively qualitatively. The conclusion of this research is the analysis of oil losses in the process of making palm kernel oil using check sheets and histograms, the data obtained shows high oil losses in the company PT. XYZ with an average of 8.92% for 4 months, so it is necessary to make improvements that at improving the quality of good palm kernel oil (PKO). Analysis of PKO oil losses at PT. XYZ is also performed using a cause-and-effect diagram. This aims to identify the causes of oil loss which include raw materials, human resources (HR), machinery, and the environment in order to fulfill the oil loss standard designed by the company, which is less than 8.00%, and also prevent high oil losses. which may happen again in the future.
