Abstract

Red palm oil (RPO) has various bioactive and nutritional components with high potential to be developed as a source of nutraceuticals in addition to its potential as a functional food. The fermentation technology is a processing process that affects metabolite activity, increasing the durability of a product. Staphylococcus epidermidis is one of the bacteria that is generally used in the sugar and oil fermentation process. RPO fermentation by Staphylococcus epidermidis is supposed to increase nutraceutical value by adding probiotic properties to RPO products. Moreover, the instability of bioactive compounds in RPO needs to be protected with coating technology microencapsulation. So, this study aims to formulate a microencapsulation procedure for fermented RPO using Staphylococcus epidermidis ATCC 12228 as a potential nutraceutical. RPO was fermented using S. epidermidis with three variations of MRS media and RPO comparison, then microencapsulated using emulsification and extrusion methods. The physicochemical properties and bioactivity of the product, microcapsule, were then analyzed. Our data shows that RPO fermentation was able to increase the chemical components. RPO fermentation produced more 1,2-Benzenedicarboxylic acid (2-Ethylhexyl) ester. Furthermore, microencapsulation of fermented RPO has better yield, efficiency, and solubility in water than non-fermented RPO microencapsulation. Moreover, our work also shows that the microencapsulation process increased RPO stability.

Abstract

Sungkai (Peronema canescens Jack) and Cinnamon (Cinnamomum burmannii) are herbal plants which are known to have a high content of secondary metabolite compounds as a source of natural antioxidants which are able to inhibit free radical compounds in the body and convert these free radical compounds into non-radical compounds. The greater the free radical scavenger activity in the product, the better the SPF value will be. However, the use of the extract as an active substance in cosmetic dosage forms has a low pharmaceutical elegance, so the extract is formulated using the SNEDDS (Self Nano-Emulsifying Drug Delivery System) technique. Nanoemulsion able to increase the solubility of natural ingredients by eliminating variability in absorption, permeability, and the bioavailability of drugs or bioactive compounds from natural ingredient extracts by maximizing drug penetration into the body and minimizing side effects. The aim of this research is to characterize a nanoemulsion preparation from a combination of sungkai leaves extract and cinnamon in order to obtain the optimal formula for a nanoemulsion preparation as an antioxidant product and skin protection factor agent. This research began with making extracts using the maceration method using 96% ethanol solvent, followed by making nanoemulsions using the spontaneous method. The extract concentration used was 200 mg. Observation of physical properties includes centrifugation test, organoleptic test, percent transmittance, solubility, pH, emulsion type, particle size, polydispersity index, antioxidant activity, SPF value of nanoemulsion preparation and irritation test. Analysis was carried out based on descriptive test parameters. Based on the research results, nanoemulsion preparations were made with Tween 80 surfactant, PEG 400 cosurfactant, VCO oil phase, and aqua deion water phase. So that the optimum formula nanoemulsion preparation (F1) was obtained with an active substance concentration of 100 mg sungkai extract and 100 mg cinnamon extract that met the requirements as a transparent, stable, visual nanoemulsion preparation product, with a transmittance value of 98%, a skin pH range of 6.14, having emulsion type (o/w) with a nanoemulsion particle size of 35.0 nm, PI of 0.479 and has a very strong IC50 antioxidant activity of 20.29 ppm with an SPF value of 7.4 in extra protection.

Abstract

Diabetes is a common and rapidly spreading disease that affects 6.6% of the world’s population. Metformin hydrochloride (HCl) is an effective oral pharmacological treatment for type 2 diabetes mellitus (T2DM) patients because of its capability to reduce glucose levels and the risk of hypoglycaemia. However, gastrointestinal sensitivities to this drug can cause diarrhoea, nausea, vomiting and stomach ache. An alternative is controlled release technology by coating metformin with carboxymethyl cellulose (CMC) and zinc-metal-organic framework (Zn-MOF) through the encapsulation method. Therefore, this study aimed to coat metformin formulations using CMC in a Zn-MOF to control release into the body and minimise the side effects related to doses. The results showed that CMC/metformin@Zn-MOF encapsulation had a controlled blood sugar lowering effect and stable healing effectiveness even at a low concentration of 1:10 of local metformin. In vitro, the released concentrations from local metformin tablets and CMC/Metformin@Zn-MOF were 199.50 ppm of 500 ppm and 42.182 ppm of 50 ppm, with release percentages of 39.90% and 84.364%, respectively. This study found that metformin masking using CMC and Zn-MOF maximally produced a modified release formula along the intestine by reducing the concentration to increase tolerability in lowering blood sugar effectively for T2DM patients with minimal side effects.
