Abstract

Microgels have been recognized as promising scaffolds for diabetic wound care due to their favorable properties in promoting wound healing. This study aims to synthesize a microgel using sulfated carboxymethyl cellulose (SCMC) derived from Lindur fruit (Bruguiera gymnorrhiza) for potential application in diabetic ulcer treatment. The synthesis was conducted through a double-crosslinking reaction, and the resulting microgel was characterized using infrared (IR) spectroscopy, scanning electron microscopy (SEM), and in vivo analysis in male mice (Mus musculus). The results demonstrated that the yield of Ca-alginate isolate ranged from 18.67% to 19.46%, with a purity of 19.5%33.1%, viscosity of 31.1931.46 cP, and stable pH at 7. The Ca-alginate isolate exhibited antioxidant activity at 50 ppm with an inhibition percentage of 22.3%. The modification of SCMC was successfully achieved, as confirmed by the presence of characteristic absorption peaks at 1220 cm (asymmetric SO stretching) and 870 cm (symmetric COS stretching vibrations), indicating successful sulfate group incorporation. The microgel swelling ratio was recorded at 660% for microgel A, 700% for microgel B, and 720% for microgel C, with microgel C exhibiting the highest swelling capacity. In vivo wound healing assessment revealed that microgel C facilitated more rapid wound closure, with wound sizes reducing progressively from 0.9 cm on day 3 to 0.5 cm on day 5, 0.3 cm on day 7, and 0.1 cm on day 14. These findings suggest that SCMC-based microgel with alginate derived from Lindur fruit holds significant potential as an innovative formulation for diabetic wound management

Abstract

Sungkai (Peronema canescens Jack) is known to contain flavonoid compounds with antioxidant properties that may contribute to blood glucose reduction. Cinnamon (Cinnamomum burmanii (Nees & T. Nees) Blume) also contains key antidiabetic compounds, including methylhydroxy chalcone, cinnamaldehyde, and proanthocyanidins. The combination of multiple medicinal plants is believed to produce a synergistic effect, potentially enhancing antidiabetic activity. This study aimed to evaluate the effects of a combination of sungkai and cinnamon leaf extracts on body weight and blood glucose levels in alloxan-induced mice (Mus musculus). The extracts were obtained through maceration and subjected to phytochemical screening. The antidiabetic test was conducted using seven treatment groups, with observations recorded on days 3, 5, and 7 following extract administration. Phytochemical analysis revealed that the combined extract contained alkaloids, flavonoids, tannins, saponins, steroids, and triterpenoids. The treatment groups (C+, P1–P5) showed a lower percentage of body weight loss compared to the negative control group; however, no statistically significant differences were observed. In contrast, the P5 group (250 mg sungkai extract + 100 mg cinnamon extract) demonstrated the highest percentage reduction in blood glucose levels, reaching 43.97% on day 5, significantly outperforming other treatment groups. These findings suggest that the combination of sungkai and cinnamon leaf extracts has promising potential as an effective antidiabetic agent. Abstract

Peronema canescens Jack (commonly known as sungkai) is an endemic medicinal plant native to Sumatra and Kalimantan, known for its broad spectrum of pharmacological activities, including antioxidant, antibacterial, antidiabetic, antihyperuricemic, and anti-inflammatory effects. These properties are largely attributed to its high content of bioactive phenolic and flavonoid compounds. Conventional extraction techniques typically utilize organic solvents such as ethanol or methanol, which raise concerns regarding toxicity, cost, and environmental sustainability. This study explores a green extraction strategy employing Natural Deep Eutectic Solvents (NADES), formulated from sodium acetate and glycerol at varying molar ratios, to optimize the recovery of antioxidant compounds from sungkai leaves. The extraction process involved maceration for 2 × 24 hours. FTIR analysis identified key functional group absorptions (O–H, C–H, C=C), with distinct spectral shifts and intensity variations, indicating interactions between NADES components and bioactive molecules. Among the four tested formulations, NADES 4 (sodium acetate:glycerol, 1:3) yielded the highest total phenolic and flavonoid contents, and exhibited the strongest antioxidant activity, with an IC50 value of 32.30 ppm—classified as very strong and comparable to ascorbic acid. These results demonstrate that the compositional adjustment of NADES significantly influences solvent polarity and extraction efficiency, underscoring their promise as sustainable alternatives for the extraction of natural antioxidants.

Abstract

Antibacterial compounds are bioactive substances capable of inhibiting bacterial growth by disrupting the metabolism and cellular processes of pathogenic microorganisms. Natural plant-derived compounds have been widely explored for their antibacterial properties, with star fruit (Averrhoa bilimbi L.) and betel leaf (Piper betle) recognized for their potent antimicrobial effects. This study aimed to evaluate the antibacterial activity of a combination of star fruit and betel leaf extracts against Escherichia coli and Staphylococcus aureus, two clinically significant bacterial pathogens. The extraction process was performed using hydro-extraction at different temperatures (40°C, 50°C, 60°C, and 90°C) to determine the optimal conditions for bioactive compound yield. Antibacterial activity was assessed using the disc diffusion method, measuring inhibition zones to indicate bacterial susceptibility. The results demonstrated that the optimal inhibitory effect occurred at 50°C, producing an inhibition zone of 19.75 mm for Staphylococcus aureus and 11.75 mm for Escherichia coli. These findings suggest that temperature plays a critical role in maximizing the antibacterial potential of plant extracts. The study highlights the potential application of star fruit and betel leaf extracts as natural antibacterial agents, particularly against Gram-positive and Gram-negative bacteria. Further research is recommended to explore the mechanism of action, phytochemical composition, and potential synergy of these extracts in antimicrobial formulations.

Abstract

Pluchea indica L. is a plant have some secondary metabolite compounds namely flavonoids, alkaloids, tannins, saponins, terpenoids, and phenols that have antioxidants activity. The purpose of this study was to compare the content of antioxidant compounds analyzed using LC-MS and determine the relationship of antioxidant activity through the DPPH test on P. indica leaves extracted by maceration and soxhlation. The extracts from maceration and soxhlation of P. indica leaves were analyzed using LC-MS to determine the composition of the compounds contained therein then continued with the antioxidant activity test using the DPPH method. In the antioxidant activity test using DPPH, each extract was made 3 variations of concentration, namely 10 ppm, 50 ppm, and 100 ppm, and measured based on the absorbance of DPPH at a wavelength of 515 nm using a UV-vis spectrometer. The results of analysis using LC-MS on macerated extracts of P. indica leaves are 112 compounds consisting of flavonoids 72.4841%, phenols 9.00625%, terpenoids 6.11827%, alkaloids 6.99075%, tannins 2.2256%, and saponins 2.04458%. The Soxhlet extraction contained 116 compounds consisting of flavonoids 67.14345%, phenols 8.95854%, terpenoids 13.82165%, alkaloids 5.35637%, tannins 2.53305%, and saponins 0.2969%. The results of the antioxidant activity test using DPPH on maceration extracts have an IC50 value of 76.7576 and soxhlation 62.580 ppm. Both maceration and soxhlation extracts of P. indica leaves contain compounds that have high antioxidant potential and have good antioxidant activity, namely in the strong antioxidant category. However, the soxhlation extract has more compounds and a lower IC50 value. Thus, the more antioxidant compounds contained in an extract, the lower the IC50 value so that the better the antioxidant activity.

Abstract

The Sungkai plant (Peronema canescens Jack) is a widely recognized medicinal plant in Indonesia, with its leaves recently gaining attention for their potential health benefits. This study explores the microencapsulation of ethanol extract from Sungkai leaves using three different coating materials—maltodextrin, inulin, and Arabic gum—at varying concentrations. The aim of this study was to identify the optimal microencapsulation formulation using these materials. Microencapsulation was performed using the extrusion method, and the best formulation was characterized by evaluating its physicochemical properties, morphology, and infrared (IR) spectrum. Antioxidant activity was measured using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay. The results showed that microencapsulant formulation A1 exhibited superior physicochemical properties compared to other formulations. Scanning electron microscopy (SEM) analysis of sample A1 revealed a smooth surface with a slightly rounded shape and minimal wall folds or cracks, suggesting good stability. Fourier-transform infrared (FTIR) analysis confirmed effective encapsulation of the ethanol extract. While the crude extract demonstrated the highest antioxidant activity, microencapsulation slightly reduced this activity. Among the microencapsulated samples, formulation A1 (using Arabic gum) retained the most antioxidant potential. In conclusion, formulation A1, utilizing Arabic gum as the coating material, was found to be the optimal microencapsulation formulation for the ethanol extract of Sungkai leaves.
