Abstract

Congenital heart disease (CHD) represents nearly one-third of congenital birth defects annually, with ventricular septal defect (VSD) being the most common type. The aim of this study was to explore the role of specific GATA binding protein 6 gene (GATA6) mutations as a potential etiological factor in the development of VSD through an in silico approach. Data were collected from the human gene databases: DisGeNET and GeneCards, with protein-protein interaction networks constructed via STRING and Cytoscape. Gene ontology and pathway enrichment analyses were conducted using DAVID, with data analysis in R with significance set at FDR p<0.05. Target single nucleotide polymorphisms (SNPs) of GATA6 were obtained from NCBI dbSNP, and non-synonymous single nucleotide polymorphism (nsSNP) effects were predicted using SIFT, PolyPhen-2, I-Mutant 2.0, Fathmm, MutPred 2.0, SNP&GO, and PON-P2. Conserved regions of GATA6 were analyzed using ConSurf, with functional classification, variant conservation, and stability changes evaluated in Google Colab. Multiple sequence alignment was performed using ClustalW. Mutation modeling and molecular dynamics analysis, using GROMACS, revealed that among 87 intersecting genes, 16 proteins were interconnected with GATA6, showing a centrality value of 0.4378. Gene ontology analysis highlighted atrioventricular canal development, protein-DNA complexes, and transcription factor regulation as key processes for cardiac development, especially in the ventricular septum. NsSNP and molecular dynamics analyses identified rs387906818 and rs387906820 as having the highest pathogenic potential for VSD due to amino acid structural changes.

Abstract

This study aims to identify the misconceptions of 10th-grade students of a Senior High School (SMA N 1) Ciawi Bogor on chemical bonding by using a two-tier diagnostic test consisting of nine questions. The research method that was carried out was quantitative research with adescriptive approach. The participants of this study were classes X-2, X-4, and X-8 of SMAN 1 Ciawi in the academic year 2023/2024, with a total sample of 96 students taken from three classes using purposive sampling. Moderate misconceptions concerning ionic bonding have been discovered in the responses provided by the students for questions 1 and 2, with percentages of 37.50 and 34.38%, respectively. In Question 3, which covered lattice and intermolecular forces on silicon carbide, misconceptions among students were found to be 72.92%, indicating a high level of misconception. The percentage of students experiencing misconceptions about ionic bonding in question 4 (58.33%) was considered moderate. Students' misconceptions about bonding and intermolecular forces on magnesium oxide were determined to be relatively low, at 30.21%. Students who experienced misconceptions about the conductivity of graphite (Question 6) were 43.75%, which can be categorized as moderate misconceptions. In Question 7 about sulfur, student misconceptions were observed at 15.63%, but there were 47.92% who did not have prior knowledge about sulfur as a simple molecular compound. When asked about the differences in intermolecular forces on water and hydrogen sulfide, the percentage of students who had misconceptions was observed at 50.00%, then on the concept of lattice and intermolecular forces on macromolecules at 25.00%. These results are also influenced by students' prior knowledge of macromolecules, which is quite limited. Teachers and educators can employ some potential futuristic instructional methods to help students understand chemical bonding more efficiently, such as implementing VR and AR simulations, integration of gamification and game-based learning, andvirtual field trips and interactive demonstrations.

Abstract

The law of guarantees recognizes two types of guarantees, namely general guarantees and special guarantees, while special guarantees can still be distinguished into material guarantees and guarantees of people who guarantee debts. General guarantees and debt guarantees do not fully provide certainty regarding debt repayment, because the creditor does not have priority rights so that the creditor's position remains as a concurrent creditor against other creditors. Fiduciary guarantees are conventional products that are determined to provide protection for creditors, especially if the debtor defaults. Fiduciary guarantees that have not been able to be applied comprehensively to all financial institution systems in Indonesia, need to be conducted several studies, especially in the perspective of Islam and Law No. 42 of 1999 which uses the sharia financial system. The focus of the study that is used as the formulation of the problem is how is the law of fiduciary guarantees in the perspective of Islamic law and Law No. 42 of 1999 concerning fiduciary guarantees in Indonesia. In this study, normative law is used, or it can be called library research. The results of the study show that the practice of implementing fiduciary in Indonesia has a very important position and must be fulfilled by financing service providers. Meanwhile, according to Islamic views, there is no fiduciary guarantee in the aspect of Islamic law, but matters concerning guarantees can be equated with rahn. Thus, in Islamic Financial Institutions, rahn is guided as a guarantee system in lending activities in accordance with Islamic law, this is based on the word of Allah QS. Al-Baqarah verse 283 and Law Number 21 of 2008 concerning Islamic Banking. Guarantees of property are known in law, namely (1) guarantees in the form of pawns regulated in Articles 1150-1160 of the Civil Code, (2) mortgages regulated in Articles 1162-1178 of the Civil Code, (3) mortgage rights as regulated in Law No. 4 of 1996, and (4) fiduciary guarantee, which is regulated in Law No. 42 of 1999 concerning fiduciary guarantee. Its implementation has not gone well, still experiencing obstacles: (a) Problems around the basis of trust where the submission of guarantees based on such trust is referred to as fiduciare eigendom overdracht, (b) The existence of intent as an unlawful act by one party, and (c) Related to Personal Guarantees where the recipient of the fiduciary does not directly own the object that is the fiduciary guarantee submitted by the fiduciary giver, so that the fiduciary guarantee is a guarantee theory.
