Abstract

ABSTRACT Ornamental fish is non consumption fish which is an important source of Indonesian foreign exchange. The objective of this study is to analyze the productivity of bronze corydoras Corydoras aeneus ornamental fish through increased stocking density with biofloc technology. The average weight of the experimental corydoras was 0.61 ‒0.72 g with 2.32‒2.40 cm standard length. This study used a randomized design method with biofloc technology treatment in 3000, 4500, and 6000 fish/m2 stocking densities. The results showed that the daily length and weight-growth rate among treatments were not significantly different (P>0.05), while survival rate and the number of fish production on all treatments were significantly different (P<0.05). The water quality during the rearing period, such as temperature, pH, alkalinity, ammonia, nitrite, and nitrate, were in a tolerable range for corydoras culture. The total suspended solids tended to be higher due to higher stocking density. The best productivity using biofloc technology obtained from 6000 fish/m2 stocking density. Keywords: Biofloc technology, Corydoras aeneus, growth rate, stocking density, survival rate. ABSTRAK Ikan hias merupakan produk perikanan non konsumsi yang menjadi sumber devisa Indonesia yang cukup penting. Penelitian ini bertujuan untuk menganalisis produktivitas ikan hias koridoras melalui peningkatan padat tebar dengan teknologi bioflok. Ikan yang digunakan adalah ikan hias koridoras (Corydoras aeneus) berbobot 0,61‒0,72 g dan panjang baku 2,32‒2,40 cm. Penelitian ini menggunakan rancangan acak lengkap dengan perlakuan teknologi bioflok pada padat tebar 3000, 4500, dan 6000 ekor/m2. Hasil penelitian menunjukkan bahwa laju pertumbuhan harian panjang dan bobot antar perlakuan tidak berbeda nyata (P>0,05), sedangkan kelangsungan hidup dan jumlah produksi ikan pada semua perlakuan berbeda nyata (P<0,05). Nilai kualitas air selama pemeliharaan yakni suhu, pH, alkalinitas, amonia, nitrit, dan nitrat yang berada pada kisaran yang cukup baik untuk budidaya ikan. Total padatan tersuspensi cenderung tinggi akibat dari semakin tinggi padat tebar. Produktivitas terbaik pada budidaya ikan koridoras dengan teknologi bioflok adalah pada padat tebar 6000 ekor/m2. Kata kunci:  Corydoras aeneus, kelangsungan hidup, padat tebar, pertumbuhan, teknologi bioflok

Abstract

ABSTRACT White spot disease caused by White Spot Syndrome Virus (WSSV) is the most serious viral disease and has a major impact on the decline in production of white shrimp farm. Improving the immune response through the application of prebiotic, probiotic and synbiotic is expected to be one of the environmentally friendly alternatives to prevent the disease. This study aimed to evaluate the effect of administrating Baccillus sp. NP5 probiotic, honey prebiotic and a combination both (synbiotic) in enhancing immune response and resistence of white shrimp to WSSV infection. This study consisted of five treatments and three replications, namely positive control (feeding without probiotics, prebiotics, and synbiotics then challenged with WSSV), negative control (feeding without probiotics, prebiotics, synbiotics, then injected with PBS), pro (feeding with the addition of Bacillus sp. NP5 probiotics then challenged against WSSV), pre (feeding with the addition of honey prebiotics then challenged against WSSV), and sin (feeding with the addition of synbiotics then challenged against WSSV). White shrimp with an average weight of 1.8±0.06 gram/shrimp were reared at a density of 15 shrimps per aquarium (60 cm x 30 cm x 30 cm and water height of 20 cm) for eight weeks, then challenge against WSSV at lethal dose (LD50) dose as much as 0.1 ml per shrimp. The results showed that the immune response and resistence of white shrimp after treatments of probiotics, prebiotics, and synbiotics were better than those in controls with the optimal found in prebiotics treatment. NP5, honey, immune response, white shrimp, WSSV ABSTRAK White spot disease yang disebabkan oleh infeksi white spot syndrome virus (WSSV) merupakan penyakit viral paling serius dan berdampak besar terhadap penurunan produksi budidaya udang vaname. Perbaikan respons imun melalui aplikasi probiotik, prebiotik dan sinbiotik dapat menjadi salah satu alternatif ramah lingkungan untuk pencegahan serangan penyakit tersebut. Penelitian ini bertujuan untuk mengevaluasi pengaruh pemberian probiotik Bacillus sp. NP5, prebiotik madu dan gabungan keduanya (sinbiotik) dalam meningkatkan respons imun dan resistansi udang vaname terhadap infeksi WSSV. Penelitian ini terdiri dari lima perlakuan dan tiga ulangan yaitu kontrol positif (pemberian pakan tanpa probiotik, prebiotik, dan sinbiotik kemudian diuji tantang dengan WSSV), kontrol negatif (pemberian pakan tanpa probiotik, prebiotik, dan sinbiotik kemudian diinjeksi PBS), pro (pemberian pakan dengan penambahan probiotik Bacillus sp. NP5 kemudian diuji tantang dengan WSSV), pre (pemberian pakan dengan penambahan prebiotik madu kemudian diuji tantang WSSV), dan sin (pemberian pakan dengan penambahan sinbiotik kemudian diuji tantang dengan WSSV). Udang vaname dengan bobot rata-rata 1.8±0.06 gram/ekor dipelihara dengan kepadatan 15 ekor per akuarium (60 cm x 30 cm x 30 cm) selama delapan minggu, kemudian diuji tantang dengan WSSV sebanyak 0.1 ml per ekor pada dosis LD50. Hasil penelitian menunjukkan bahwa respons imun dan resistansi udang vaname setelah pemberian probiotik, prebiotik dan sinbiotik lebih baik (P<0.05) dibandingkan kontrol dengan hasil optimal pada perlakuan prebiotik. Abstract

ABSTRAK Cairan fermentasi daun mangrove Avicennia marina mengandung mikroorganisme, nutrient, dan metabolit sekunder. Penelitian ini bertujuan untuk mengidentifikasi bakteri dan senyawa dalam cairan fermentasi daun mangrove A. marina dan mengukur daya hambatnya terhadap bakteri patogen Stenotrophomonas maltophilia penyebab penyakit ice-ice pada rumput laut. Hasil analisis molekuler dengan target gen 16S rRNA menunjukkan bahwa bakteri dalam cairan fermentasi terdiri atas delapan jenis Bacillus, yaitu Bacillus subtilis MSAR-01, Bacillus megaterium MSAR-02, Bacillus firmus MSAR-03, Bacillus thuringiensis MSAR-04, Bacillus subterranerus MSAR-05, Bacillus vietnamensis MSAR-06, Bacillus sp. MSAR-07, Bacillus circulans MSAR-08, dengan daya hambat terbaik ditunjukkan oleh B. subtilis MSAR-01, B. vietnamensis MSAR-06, dan Bacillus sp. MSAR-07. Pemberian asam laktat, bakteriosin, cairan fermentasi total, dan supernatan sebanyak 15 mL menghasilkan daya hambat terhadap bakteri S. maltophilia lebih baik daripada menggunakan salah satu atau kombinasi beberapa jenis bakteri isolat. Daya hambat cairan fermentasi dan supernatan yang diperkaya bakteri tunggal lebih baik daripada pengayaan kombinasi bakteri. Kata kunci: Avicennia marina, fermentasi, ice-ice, mangrove

Abstract

ABSTRACT Motile aeromonad septicaemia (MAS) in catfish can be done by improving the immune responses through probiotic administration. Co-administering probiotics producing digestive enzymes are expected to have an impact on fish growth. This study was aimed to evaluate the effectivity of probiotics B. megaterium PTB 1.4 and P. pentosaceus E2211 to improve the growth performance and immune response of catfish infected by Aeromonas hydrophila. Catfish with the initial body weight of 7.36 ± 0.21 g were reared in a pond. This study was conducted with five treatments, i. e. K- (without probiotic addition and A. hydrophila injection), K+ (no probiotic addition with A. hydrophila injection), Bm (B. megaterium PTB 1.4 addition and A. hydrophila injection), Pp (P. pentosaceus E2211 addition and A. hydrophila injection), and Bm+Pp (B. megaterium PTB 1.4 addition and P. pentosaceus E2211 and A. hydrophila injection). The study results showed that Bm, Pp, and Bm+Pp treatments were able to improve the growth performance of catfish including digestive enzyme activity, daily growth rate, feed conversion ratio, and final biomass with the best results was Bm+Pp treatment. The immune response of catfish before and after the challenge test showed better survival rate, higher total leukocytes, phagocytic activity, respiratory burst, and lysozyme activity on Bm, Pp, and Bm+Pp treatment (P<0.05) than the control treatment. In conclusion, the combination of probiotics B. megaterium PTB 1.4 and P. pentosaceus E2211 in feed synergistically improved the growth performance and immune response on catfish against A. hydrophila infection. Pemberian bersama probiotik penghasil enzim pencernaan diharapkan dapat berdampak pada pertumbuhan ikan. Penelitian ini bertujuan mengevaluasi efektivitas pemberian probiotik B. megaterium PTB 1.4 dan P. pentosaceus E2211 terhadap kinerja pertumbuhan dan respons imun ikan lele terhadap infeksi A. hydrophila. Penelitian ini menggunakan rancangan acak lengkap dengan lima perlakuan, yaitu: K- (tanpa probiotik dan tanpa diinjeksi A. hydrophila), K+ (tanpa probiotik dan diinjeksi A. hydrophila), Bm (diberi B. megaterium PTB 1.4 dan diinjeksi A. hydrophila), Pp (diberi P. pentosaceus E2211 dan diinjeksi A. hydrophila), dan Bm+Pp (diberi B. megaterium PTB 1.4 dan P. pentosaceus E2211 dan diinjeksi A. hydrophila). Hasil penelitian menunjukkan perlakuan Bm, Pp, dan Bm+Pp mampu meningkatkan pertumbuhan ikan lele. Aktivitas enzim pencernaan, laju pertumbuhan harian, rasio konversi pakan, dan biomassa panen dengan hasil terbaik yaitu perlakuan Bm+Pp. Respons imun ikan lele dengan kombinasi probiotik B. megaterium PTB 1.4 dan P. pentosaceus E2211 mampu meningkatkan sintasan, total leukosit, aktivitas fagositosis, respiratory burst, dan aktivitas lisozim, baik pada sebelum dan setelah uji tantang. Pemberian kombinasi probiotik B. megaterium PTB 1.4 dan P. pentosaceus E2211 pada pakan mampu bekerja sinergis dalam meningkatkan kinerja pertumbuhan dan respons imun ikan lele terhadap infeksi A. hydrophila. Abstract

ABSTRACT The intensification of catfish culture requires a regular and sustainable good seeds quality. Microbial-based aquaculture with probiotic application is expected to improve the reproductive performance of catfish. The aim of this study was to evaluate the reproductive performance of catfish given probiotics Bacillus sp. NP5 with different concentrations through feed. This study used a Completely Randomized Block Design (CRBD) with the administration of probiotic Bacillus sp. NP5 at 2% concentration of 108 CFU/mL and 1010 CFU/mL in feed given and control (without probiotics), then repeated four times. Catfish broodstocks with an average size of 955 ± 261.27 g were kept in a net sized 1 m×1 m×1 m with seven broodstocks per net. Parameters observed included the Gonadosomatic Index (GSI), fecundity, number of matured eggs, frequency distribution of egg diameter, fertilization rate, and hatching rate. The result showed that the GSI value of female catfish broodstock was significantly different in sixth week as the highest value was obtained from 1010 CFU/mL probiotics with 5.19%. Moreover, the fecundity of all treatments showed significantly different results (P<0.05) as the highest value was on the concentration of 1010 CFU/mL Bacillus sp. NP5 with 137,123 ± 32,635 eggs/broodstock. This study concludes that the administration of probiotics Bacillus sp. NP5 with 1010 CFU/mL in feed given to catfish broodstock increases the fecundity (137,123 eggs/broodstock), GSI (5.19%), number of matured eggs percentage (64.50%), and hatching rate (82.53%). Keywords: Bacillus sp. NP5, catfish, reproductive performance ABSTRAK Intensifikasi budidaya ikan lele menuntut penyediaan benih yang berkualitas secara teratur dan berkelanjutan. Budidaya berbasis mikroba diharapkan dapat meningkatkan kinerja reproduksi ikan lele. Tujuan penelitian ini adalah mengevaluasi performa reproduksi ikan lele yang diberi probiotik Bacillus sp. NP5 dengan konsentrasi berbeda melalui pakan. Penelitian ini menggunakan rancangan acak kelompok lengkap (RAKL) dengan pemberian probiotik Bacillus sp. NP5 pada konsentrasi 108 CFU/mL dan 1010 CFU/mL sebanyak 2% dari pakan yang diberikan serta kontrol (tanpa pemberian probiotik), masing-masing dengan empat ulangan. Induk ikan lele dengan ukuran 955 ± 261.27 g dipelihara pada jaring hapa ukuran 1 m × 1 m × 1 m sebanyak tujuh ekor per jaring hapa. Parameter yang diamati meliputi indeks kematangan gonad (IKG), fekunditas, jumlah telur matang, sebaran frekuensi diameter telur, derajat pembuahan, dan derajat penetasan. Hasil penelitian menunjukkan bahwa nilai IKG induk betina ikan lele berbeda nyata pada minggu keenam dengan nilai tertinggi diperoleh pada perlakuan probiotik 1010 CFU/mL yaitu sebesar 5.19%. Selain itu, fekunditas semua perlakuan menunjukkan hasil yang berbeda nyata (P<0.05) dengan nilai tertinggi pada perlakuan pemberian probiotik Bacillus sp. NP5 dengan konsentrasi 1010 CFU/mL sebesar 137,123 ± 32,635 telur/induk. Kesimpulan, pemberian probiotik Bacillus sp. NP5 dengan konsentrasi 1010 CFU/mL melalui pakan kepada induk ikan lele mampu meningkatkan nilai fekunditas 137,123 telur/induk, GSI 5,19%, persentase jumlah telur matang 64,50% dan derajat penetasan telur 82,53%. Abstract

ABSTRACT This study was aimed to evaluate the effectiveness of Bacillus sp. NP5 paraprobiotic administration through commercial feed on growth performance, immune response, and resistance of Nile tilapia against Streptococcus agalactiae infection. Bacillus sp. NP5 paraprobiotic was produced through heat-inactivation at 95°C for 1 h, then performed a viability test on tryptic soy agar (TSA) media and incubated for 24 hours. Paraprobiotics could be used whether the bacteria did not grow on the TSA media. This study used a completely randomized design, containing three treatments with five replications, i.e. 1% (v/w) probiotic addition, 1% (v/w) paraprobiotic addition, and no addition of probiotic or paraprobiotic (control). The experimental fish were reared for 30 days. On day 31 of rearing, fish were challenged with S. agalactiae (107 CFU/mL) through intraperitoneal injection route, while the negative control was injected with PBS. This study results significantly improved growth performances and immune responses (P<0.05), compared to control after 30 days of probiotic and paraprobiotic Bacillus sp. NP5 administration. After challenge test, increased immune responses in probiotic and paraprobiotic of Bacillus sp. NP5 treatment had higher survival rates (P<0.05) than positive control. The administration of Bacillus sp. NP5 probiotic and paraprobiotic through commercial feed were effective in increasing growth performance, immune response, and resistance of Nile tilapia against S. agalactiae infection. NP5 melalui pakan dalam meningkatkan kinerja pertumbuhan, respons imun, dan resistansi ikan nila terhadap infeksi Streptococcus agalactiae. Proses pembuatan bakteri paraprobiotik yaitu Bacillus sp. NP5 diinaktivasi panas pada suhu 95°C selama 1 jam, dilanjutkan dengan pengujian viabilitas dengan menyebarkannya pada media tryptic soy agar kemudian diinkubasi selama 24 jam. Jika bakteri tidak tumbuh, maka paraprobiotik dapat digunakan. Penelitian ini menggunakan rancangan acak lengkap dengan tiga perlakuan dan lima ulangan, yaitu penambahan probiotik 1% (v/w), penambahan paraprobiotik 1% (v/w), dan tanpa penambahan probiotik atau paraprobiotik (kontrol). Ikan perlakuan dipelihara selama 30 hari dan pada hari ke-31, ikan diuji tantang dengan S. agalactiae (107 CFU/mL) melalui injeksi intraperitoneal, sementara perlakuan kontrol negatif diinjeksi dengan PBS. Hasil penelitian setelah 30 hari pemberian probiotik dan paraprobiotik Bacillus sp. NP5 menunjukkan kinerja pertumbuhan dan respons imun yang meningkat signifikan (P<0.05) dibandingkan dengan kontrol. Pascauji tantang, peningkatan respons imun pada perlakuan probiotik dan paraprobiotik Bacillus sp. NP5 menunjukkan tingkat kelangsungan hidup yang lebih tinggi (P<0.05) dibandingkan kontrol positif. Pemberian probiotik dan paraprobiotik Bacillus sp. NP5 melalui pakan dapat meningkatkan kinerja pertumbuhan, respons imun, dan resistansi ikan nila terhadap infeksi Streptococcus agalactiae. Abstract

An effort to increase the production of juvenile catfish is limited by the high mortality rate, especially caused by cannibalism. The hormonal treatment has been conducted as an effort to control cannibalism. This study used completely randomized factorial design, consisted of six treatments and three replications. There were two factors examined in this study, the first factor was different doses of 17β‒estradiol hormone (0, 30, and 60 mg estradiol‒17β/kg) incorporated in the feed, and the second factor was stocking density (150 and 300 fish/m2). Juvenile catfish with the size of 4.0 ± 0.1 cm were reared for 30 days in the 84 L aquarium. The results showed that hormone treatment could reduce cannibalism rate, type-I and II cannibalism compared to control (P<0.05). The results showed that the lowest of mortality was obtained in the treatment B (17β‒estradiol administration of 30 mg/kg, at stocking density of 150 fish/m2; P<0.05). The lowest cortisol level was found in treatment F, and the highest estradiol level was found in treatment F (17β‒estradiol administration of 60 mg/kg, at stocking density of 300 fish/m2; P<0.05). The lowest of blood glucose level was found in treatment B (P<0.05). The highest specific weight growth was found in the stocking density of 150 fish/m2 (P<0.05). The results of this study indicated that administration of 17β‒estradiol in feed could reduce the level of cannibalism in African catfish juvenile. Keywords: cannibalism, Juvenile, catfish, estradiol‒17β, cortisol ABSTRAK Upaya untuk meningkatkan produksi benih ikan lele dibatasi oleh tingginya angka kematian, terutama yang disebabkan oleh kanibalisme. Pendekatan hormonal telah dilakukan sebagai upaya pengendalian kanibalisme. Penelitian ini menggunakan rancangan acak lengkap faktorial, terdiri dari enam perlakuan dan tiga ulangan. Ada dua faktor yang diteliti dalam penelitian ini, faktor pertama adalah dosis hormon 17β‒estradiol yang berbeda (0, 30, dan 60 mg/kg) yang diberikan melalui pakan, dan faktor kedua adalah padat tebar (150 dan 300 ekor/m2). Benih ikan lele berukuran 4.0 ± 0.1 cm dipelihara selama 30 hari di akuarium (84 L). Hasil penelitian menunjukkan bahwa perlakuan hormon dapat menurunkan tingkat kanibalisme tipe‒I dan II dibandingkan kontrol (P<0.05). Hasil penelitian menunjukkan bahwa mortalitas terendah diperoleh pada perlakuan B (pemberian 17β‒estradiol 30 mg/kg, pada padat tebar 150 ekor/m2; P<0.05). Kadar kortisol terendah ditemukan pada perlakuan F, dan kadar estradiol tertinggi ditemukan pada perlakuan F (pemberian 17β‒estradiol 60 mg/kg, pada padat tebar 300 ekor/m2; P<0.05). Kadar glukosa darah terendah ditemukan pada perlakuan B (P<0.05). Pertumbuhan bobot spesifik tertinggi ditemukan pada padat tebar 150 ekor/m2 (P<0.05). Hasil penelitian ini menunjukkan bahwa pemberian hormon 17β‒estradiol dengan dosis 30 mg/kg pakan, dan padat tebar 150 ekor/m2 dapat menurunkan tingkat kanibalisme pada benih ikan lele di Afrika. Abstract

This study aimed to evaluate the effectivity of microencapsulated synbiotic (MS), Bacillus sp. NP5 and mannan oligosaccharide (MOS) dietary in different feeding frequencies in Pacific white shrimp culture field experiment. The MS was administered as a feed supplementation to enhance the immunity for prevention against co-infection with WSSV (White Spot Syndrome Virus) and Vibrio harveyi. The synbiotic was microencapsulated by the spray dryer method. Shrimps were reared in the floating net cages in the pond. Treatments included the administration of MS at different frequencies i.e, daily (A), twice a week (B), once a week (C), and without MS supplementation (consisted of negative and positive controls) with a feeding rate of 6% of shrimp biomass (5 times a day). During the challenge trial, shrimps were removed and further reared in plastic tanks, for 7 days. The shrimps (except negative control treatment) were intramuscularly injected by WSSV filtrate at the infective dosage of 10-4 copies.ml-1. Twenty four hours after WSSV injection the shrimps were immersed in the water-containing cells suspension of V. harveyi at the cell’s population dosage of 106 CFU.ml-1. Immune responses were observed for 7 days after experimental infection. The shrimps that have been treated with daily MS supplementation (A) showed better immune responses i.e., total haemocyte counts, phenoloxidase, respiratory burst, and the lower pathogenic cells abundance in the intestine compared to other treatments groups.

Abstract

The aim of the study is to test the effectiveness of adding various paraprobiotic doses of Bacillus sp. NP5 through feed in improving the immune responses and resistance of tilapia juvenile (Oreochromis niloticus) to infection of Streptococcus agalactiae. Bacillus sp. NP5 paraprobiotic was produced through heat-inactivation at 95°C for 1 h. This study used a completely randomized design, which consisted of four treatments with three repetitions. The test feed was enriched with 1% paraprobiotic with a density of 108, 109, and 1010 CFU ml-1. The tilapia juvenile (10.29 ± 0.22 g) were reared for 30 days and on day 31, the fish was challenge tested with S. agalactiae (107 CFU ml-1) injected by intraperitoneal route unless the negative control was injected with PBS. The results of this study showed significantly improved growth performances and immune responses (P<0.05), compared to the control group after 30 days of paraprobiotic Bacillus sp. NP5 administered. The increased of immune responses in paraprobiotic of Bacillus sp. NP5 showed a higher survival rate (P<0.05), than positive control after the challenge tested with S. agalactiae. Administration of paraprobiotic Bacillus sp. NP5 through the commercial feed able to improve the growth performances, immune responses, and resistance of nile tilapia against S. agalactiae infection with the best result at dose of 1010 CFU ml­-1.

Abstract

Vibrio alginolyticus which becomes the main cause of vibriosis disease in grouper culture and causes great economic loss in Asian aquaculture industries. This study investigated the antibacterial activity of clove extract and the effect of adding clove powder to the diet on survival cantang grouper infected with V. alginolyticus. The clove extraction used a maceration method. Two dose levels of clove powder were used, namely 10 and 15 g kg−1. The control treatments without clove powder supplementation contained positive control (CP) and negative control treatment (CN). The results found that the clove extract contained five major compounds. The highest compound was phenol, 2-methoxy-4-(2-propenyl)-Eugenol (64.07%). Phytochemical analysis of clove extract contained phenolics, flavonoids, and tannins at (28.53 ± 0.00) mg/g, (0.38 ± 0.00) mg/g, and (0.15 ± 0.00) %, respectively. The diameter of the clove extract inhibition zone was significantly different (P < 0.05) in all treatments. The Scanning Electron Microscopy (SEM) result presents that the clove extract can alter the V. alginolyticus cell morphology. The dietary supplementation of clove powder improves the survival rate significantly higher (P<0.05) post-challenge test. The conclusion of this research is that clove extract has antibacterial activity that can inhibit growth and cause cell morphological damage to V. alginolyticus. The application of clove powder at a dose of 15g kg-1 was able to improve the survival value which was a higher post-challenge test.
