Abstract

ABSTRACT This study aimed to analyze the genetic diversity of bacteria associated in bioflocs using 16S-rRNA polymerase chain reaction (PCR) with ARDRA technique. A total of 38 dominant bacterial isolates was obtained from bioflocs samples and of these isolates, 16S-rRNA gene was then isolated and amplified using PCR. The 16S-rRNA gene of the isolates was then cut using HaeIII (5’-GG↓CC) and HhaI (5’-GCG↓C) restriction enzymes resulting an ARDRA pattern which was further used as the binary data for the construction of phylogenetics tree that was used to estimate the group of bacteria. The result with HaeIII cut restriction enzyme from biofloc-associated bacteria gave 11 ARDRA patterns, while with the restriction enzyme HhaI gave eight ARDRA patterns. Phylogenetics of bacterial populations from biofloc-based cultivation system water consisted of at least 13 different bacterial species. Result of sequencing from two gene sample 16S-rRNA were identified as Microbacterium foliorumand and Pseudomonas putida. Keywords: bacterial diversity, ARDRA, biofloc, phylogeny  ABSTRAK Penelitian ini bertujuan untuk menganalisis keragaman genetika bakteri bioflok menggunakan metode polymerase chain reaction (PCR) 16S-rRNA dengan teknik ARDRA. Sebanyak 38 isolat bakteri dominan yang diperoleh diamplifikasi gen 16S-rRNAnya dengan PCR, kemudian dipotong dengan enzim restriksi HaeIII (5’-GG↓CC) dan HhaI (5’-GCG↓C). Pola ARDRA ini dijadikan data biner sebagai input untuk konstruksi pohon filogenetika yang dapat digunakan untuk memerkirakan jenis bakteri yang ada. Gen 16S-rRNA hasil PCR setelah dipotong dengan enzim restriksi HaeIII didapatkan 11 pola ARDRA, sedangkan dengan enzim restriksi HhaI menghasilkan delapan pola ARDRA. Berdasarkan pohon filogenetika, diketahui populasi bakteri pada air sistem budidaya bioflok sedikitnya terdiri atas 13 jenis bakteri. Berdasarkan sekuensing dari dua sampel gen 16S-rRNA teridentifikasi jenis bakteri Microbacterium foliorum dan Pseudomonas putida. Kata kunci: keragaman bakteri, ARDRA, bioflok, filogenetika

Abstract

ABSTRACT The aim of this study was to evaluate the supplementation of synbiotic with different doses of probiotics to prevent vibriosis in humpback grouper (Cromileptes altivelis). Grouper (3.0±0.48 g) fed by control feeds (without supplementation of synbiotic) which were K(-) and K(+), treatment feeds with supplementation of synbiotic with different doses of probiotic (probiotic 104 cfu/mL) 1% + prebiotic 2% (P1) v/w, (probiotic 106 cfu/mL) 1% + prebiotic 2% (P2) v/w, (probiotic 108 cfu/mL)1% + prebiotic 2% (P3) v/w) for 30 days. After a feeding trial period, there were observation of the bacterial counts in the fish intestine, the fish growth performance and immune response. Then all the grouper were challenged by Vibrio alginolyticus, except K (-). This study showed that survival, daily growth and food conversion ratio (FCR) of grouper in treatment P2 ((probiotic 106 cfu/mL) 1% + prebiotic 2%) and P3 (probiotic (108 cfu/mL) 1% + prebiotic 2%) were significantly better (P>0.05) than controls. The different doses of probiotic in synbiotic (probiotic 104 cfu/mL, probiotic 106 cfu/mL and probiotic 108 cfu/mL) provided better immune response than controls. Keywords: synbiotic, Vibrio alginolyticus, Bacillus sp., Cromileptes altivelis  ABSTRAK Tujuan penelitian ini adalah mengkaji pemberian sinbiotik dengan dosis probiotik berbeda untuk mencegah penyakit vibriosis pada ikan kerapu bebek (Cromileptes altivelis). Ikan kerapu dengan berat (3,0±0,48 g) diberikan pakan kontrol (tanpa penambahan sinbiotik) K(-) dan K(+), pakan perlakuan dengan penambahan sinbiotik dengan dosis probiotik berbeda: probiotik ((104 cfu/mL) 1% + prebiotik 2%) v/w (P1), ((probiotik 106 cfu/mL) 1% + prebiotik 2%) v/w (P2), dan ((probiotik 108 cfu/mL)1% + prebiotik 2%) v/w (P3) selama 30 hari. Setelah perlakuan pakan sinbiotik, dilakukan pengamatan terhadap jumlah total bakteri di usus, kinerja pertumbuhan dan respons imun. Kemudian ikan kerapu pada seluruh perlakuan, kecuali kontrol negatif (-) diberi uji tantang dengan Vibrio alginolyticus. Hasil penelitian menunjukkan bahwa sintasan, laju pertumbuhan harian (LPH), dan rasio konversi pakan (FCR) pada perlakuan P2 ((probiotik 106 cfu/mL) 1% + prebiotik 2%) dan  P3 (probiotik (108 cfu/mL) 1% + prebiotik 2%) secara signifikan lebih baik (P<0,05) bila dibandingkan dengan kontrol. Perlakuan sinbiotik dengan dosis probiotik berbeda (probiotik 104 cfu/mL, probiotik 106 cfu/mL, dan probiotik 108 cfu/mL) juga memberikan respons imun yang lebih baik dibandingkan dengan kontrol. Kata kunci : sinbiotik, Vibrio alginolyticus, Bacillus sp., Cromileptes altivelis

Abstract

ABSTRACT This study aimed to examine the effects of prebiotic, probiotic, and synbiotic in survival and immune response of white shrimp against co-infection of Vibrio harveyi and infectious myonecrosis virus (IMNV). The shrimps used had a body weigth of 2.04±0.20 g/individual, the shrimps were reared at a density of 20 shrimps in 60x30x35 cm3 sized aquarium. The study was conducted with five treatments consisting K(+) (without the addition of prebiotic, probiotic, and synbiotic with co-infection), K(-) (without the addition of prebiotic, probiotic and synbiotic, and without co-infection), P1 (the addition of prebiotic with co-infection), P2 (the addition of probiotic with co-infection), and P3 (the addition of synbiotic with co-infection). The results showed that the addition of prebiotic, probiotic and synbiotic could increase survival and immune response of white shrimp towards co-infection of Vibrio harveyi and IMNV. The best survival was obtained in probiotic treatment (79.17%), followed by prebiotic treatment (75%), synbiotic treatment (70.83%), while the positive control was only 50%. Keywords: white shrimp, prebiotic, probiotic, synbiotic, IMNV, Vibrio harveyi  ABSTRAK Penelitian ini bertujuan untuk menguji pengaruh pemberian prebiotik, probiotik, dan sinbiotik terhadap sintasan dan respons imun udang vaname dengan ko-infeksi Vibrio harveyi dan IMNV (infectious myonecrosis virus). Udang yang digunakan memiliki bobot 2,04±0,20 g/ekor, dipelihara sebanyak 20 ekor dalam akuarium berukuran 60x30x35 cm3. Penelitian dilakukan dengan lima perlakuan yaitu K(+) (tanpa pemberian prebiotik, probiotik, dan sinbiotik dengan ko-infeksi), K(-) (tanpa pemberian prebiotik, probiotik dan sinbiotik tanpa ko-infeksi), P1 (pemberian prebiotik dengan ko-infeksi), P2 (pemberian probiotik dengan ko-infeksi), dan P3 (pemberian sinbiotik dengan ko-infeksi). Hasil penelitian menunjukkan bahwa pemberian prebiotik, probiotik, dan sinbiotik mampu meningkatkan sintasan dan respons imun udang vaname terhadap ko-infeksi V. harveyi dan IMNV. Sintasan terbaik diperoleh pada perlakuan probiotik sebesar 79,17%, diikuti perlakuan prebiotik sebesar 75%, perlakuan sinbiotik sebesar 70,83%, sedangkan pada kontrol positif hanya mencapai 50%. Kata kunci: udang vaname, prebiotik, probiotik, sinbiotik, IMNV, Vibrio harveyi

Abstract

ABSTRACT The aim of this study was to test the effectiveness of a probiotic Bacillus for the prevention of motile aeromonad septicemia (MAS) disease caused by Aeromonas hydrophila in African catfish (Clarias gariepinus). The study consisted of the inhibition testing of A. hydrophila by Bacillus (in vitro) and the application of probiotic in African catfish (in vivo). The in vivo test, consisted of five treatments such as the addition of probiotic Bacillus P4I1 RifR, Bacillus P4I2 RifR, Bacillus P4I1 RifR + Bacillus P4I2 RifR (Kom), positive control (K+; only added with A. hydrophila) and negative control (K-; without probiotic nor A. hydrophila addition). African catfish (13.35±2.80 g) was maintained in 15 aquariums (40 L in volume) with 30 fishes each for 30 days. Probiotic bacteria was applied in water once a day, whereas pathogenic bacteria A. hydrophila RifR (103 cfu/mL) were added once in earlier treatment (except for the negative control). The result showed that the optimal concentration of Bacillus to inhibit A. hydrophila on in vitro test was 104 cfu/mL. In vivo test showed that the addition of probiotic in media of cultivation could reduce the number of A. hydrophila, improve immune response, and also increase the survival of African catfish compared to positive control. Penelitian terdiri atas pengujian penghambatan bakteri probiotik Bacillus terhadap A. hydrophila secara in vitro, dilanjutkan dengan aplikasi pada budidaya ikan lele dumbo (in vivo). Pada uji in vivo, penelitian terdiri atas lima perlakuan yaitu budidaya ikan lele dumbo dengan penambahan probiotik Bacillus P4I1 RifR, Bacillus P4I2 RifR, kombinasi probiotik Bacillus P4I1 RifR + Bacillus P4I2 RifR (Kom), kontrol positif (K+; hanya ditambahkan A. hydrophila) dan kontrol negatif (K-; tanpa pemberian probiotik dan A. hydrophila). Ikan lele dumbo (13,35±2,80 g) dipelihara pada akuarium volume 40 L dengan kepadatan 30 ekor/akuarium selama 30 hari. Bakteri probiotik ditambahkan pada media pemeliharaan ikan setiap hari, sedangkan bakteri patogen A. hydrophila RifR (103 cfu/ mL) diberikan sekali pada awal pemeliharaan (kecuali pada kontrol negatif). Hasil penelitian menunjukkan bahwa konsentrasi terbaik pada penghambatan in vitro adalah dengan penambahan Bacillus 104 cfu/mL. Hasil uji in vivo menunjukkan perlakuan penambahan probiotik pada media budidaya efektif dapat menekan jumlah bakteri A. hydrophila, memperbaiki respons imun, dan meningkatkan kelangsungan hidup ikan lele dumbo dibanding kontrol positif. Abstract

ABSTRACT The increasing demand of white shrimp Litopenaeus vannamei requires the application of intensive culture system. However, intensive culture system of white shrimp could increase the risk of disease outbreak. The application of sinbiotic may provide solution to the problem. This study was aimed to evaluate the effectiveness of the use of technical sinbiotic on the survival and growth of white shrimp in ponds. This study consisted of four treatments; treatment K (control), treatment A (probiotic 0.5% and prebiotic 1%), treatment B (probiotic 1% and prebiotic 2%), and treatment C (probiotic 2% and prebiotic 4%). The results showed that administration of sinbiotic had no significant differences on survival rate, growth rate, feed conversion ratio, size, and biomass of shrimp (P>0.05). However, based on analysis of business, sinbiotic A provided higher profits to the farmer (Rp10.230) compared to other symbiotic treatments and control. Keywords: Litopenaeus vannamei, sinbiotic, technical media ABSTRAK Permintaan terhadap udang vaname Litopenaeus vannamei yang semakin meningkat membuat sistem budidaya udang sebaiknya menggunakan sistem budidaya intensif. Namun demikian sistem intensif pada budidaya udang vaname dapat meningkatkan risiko timbulnya penyakit. Penggunaan sinbiotik diharapkan dapat memberikan solusi dalam mengatasi masalah tersebut. Penelitian ini bertujuan untuk menguji efektivitas penggunaan sinbiotik teknis terhadap sintasan, pertumbuhan, dan keuntungan usaha udang vaname yang dipelihara di tambak. Penelitian ini terdiri atas empat perlakuan, yaitu perlakuan K (kontrol), perlakuan A (probiotik 0,5% dan prebiotik 1%), perlakuan B (probiotik 1% dan prebiotik 2%), dan perlakuan C (probiotik 2% dan prebiotik 4%). Hasil penelitian menunjukkan bahwa pemberian sinbiotik tidak berbeda nyata dalam nilai sintasan, laju pertumbuhan, rasio konversi pakan, size, dan biomassa udang (P>0,05). Namun demikian, berdasarkan analisis usaha perlakuan A memberikan keuntungan lebih tinggi (Rp10.230) dibandingkan dengan perlakuan sinbiotik lainnya serta kontrol. Kata kunci: Litopenaeus vannamei, sinbiotik, media teknis

Abstract

ABSTRACTThis study was conducted to evaluate stress responses of transportation on red tilapia Oreochromis sp. which given feed containing chromium. Three isonitrogenous and isocaloric experimental feeds were prepared, these diets were control (without chromium), CrPic 1 mg/kg, and CrYst 2 mg/kg supplementation in feed, all group were arranged triplicate. Satiation feeding was done three times a day. After a 60-day feeding experiment, the experimental fishes were fasted and distributed in polyethylene bags (N=60 fish/bag) containing 3 L of water, subjected to condition of transport simulation for 13 hours. Survival rate, levels of plasma cortisol, blood glucose, superoxide dismutase (SOD), and malondialdehyde (MDA) enzyme were observed at before transportation, after transportation, one day, and two days after transportation. The result showed that chromium supplementation reduced the levels of plasma cortisol before and after transportation, one day, and two days after transportation. Also, it decreased blood glucose compared with control significantly before transportation and one day after transportation. The SOD enzyme concentration increased significantly after fish was fed with feed containing chromium for 30 days, while the MDA enzyme concentration increased significantly after two days of transportation. However, there was no significant difference in the survival of red tilapia between treatments. The best result was obtained in the treatment of fish which fed with feed containing chromium. CrPic supplementation 1 mg/kg and CrYst 2 mg/kg increased the body resistance in red tilapia by decreasing the negative effect of stress while transportation. Keywords: stress, transportation, red tilapia, chromium  ABSTRAKPenelitian dilakukan untuk mengevaluasi respons stres transportasi ikan nila merah Oreochromis sp. yang diberikan pakan yang mengandung kromium. Pada penelitian ini digunakan tiga jenis pakan, terdiri atas pakan tanpa suplementasi kromium (kontrol), pakan bersuplementasi kromium pikolinat (CrPic 1 mg/kg), dan kromium yeast (CrYst 2 mg/kg), semua perlakuan diulang sebanyak tiga ulangan. Pemberian pakan sebanyak tiga kali sehari dan dilakukan secara at satiation. Setelah 30 hari pemeliharaan, ikan uji dipuasakan dan didistribusikan dalam plastik polietilen (N=60 ekor ikan/kantong plastik) yang berisi 3 L air, dilakukan dengan simulasi transportasi selama 13 jam. Parameter yang diamati pada penelitian ini adalah kelangsungan hidup, kortisol, glukosa darah, enzim superoksida dismustase (SOD), dan malondialdehida (MDA) saat sebelum transportasi, sesaat setelah transportasi, sehari, dan dua hari setelah transportasi. Hasil yang didapatkan adalah suplementasi kromium menurunkan konsentrasi kortisol secara signifikan sebelum transportasi, sesaat, sehari, dan dua hari setelah transportasi. Suplementasi kromium menurunkan glukosa darah secara signifikan pada saat sebelum transportasi dan sehari setelah transportasi. Konsentrasi enzim SOD meningkat secara signifikan setelah pemberian pakan bersuplementasi kromium selama 30 hari, sedangkan konsentrasi enzim MDA meningkat secara signifikan setelah dua hari transportasi pada ikan yang diberi pakan bersuplementasi kromium. Namun, tidak ada perbedaan yang signifikan pada kelangsungan hidup ikan nila merah antarperlakuan. Hasil terbaik diperoleh pada perlakuan ikan dengan suplementasi kromium. Suplementasi 1 mg/kg CrPic dan 2 mg/kg CrYst dapat meningkatkan daya tahan tubuh pada budidaya ikan nila merah dengan menurunkan pengaruh negatif stres akibat transportasi. Kata kunci: stres, transportasi, nila merah, kromium

Abstract

ABSTRACTThe aim of this research was to determine optimum stocking density for growing-out of spiny lobster Panulirus sp. in controlled tanks that conducted for 30 days. The experimental spiny lobsters have the initial average weight of 130.39 ± 0.32 g and initial average total length of 140.70 ± 0.06 mm. This study used completely randomized design with three different stocking densities (KT10: 10 ind/m3; KT18: 18 ind/m3; and KT26: 26 ind/m3) and two replications. The parameters observed in this study included water quality (temperature, pH, salinity, dissolved oxygen, and total ammonia nitrogen), physiological responses (total haemocyte count, haemolymph glucose, and frequency of molt), and production performances, such as growth, specific growth rate, feed conversion ratio, and survival rate. The results showed that the spiny lobster could be reared in high stocking density in controlled tanks. Water quality during the study in each treatment was; temperature 26.56–28.65oC, salinity 29.7–33.6 g/L, pH 7.5–8.5, dissolved oxygen 6.15–6.58 mg/L, and total ammonia nitrogen 0.11–0.34 mg/L. The best stocking densities for spiny lobster was 18 ind/m3 (KT18) with 2.5‒3.5×106cells/mL total haemocyte counts, 24.6‒28.3 mg/dL haemolymph glucose, and 38.37 ± 3.20% frequency of molt. The final average body weight and length were 145.06 ± 0.42 g and 142.77 ± 0.19 mm, respectively. The survival rate reached 86.11 ± 3.92% with a specific growth rate 0.35 ± 0.01%/day, and feed conversion ratio 7.87 ± 0.31.Keywords: high stocking density, Panulirus sp., physiological responses, productivity.  ABSTRAKTujuan dari penelitian ini adalah untuk menentukan kepadatan terbaik dalam pembesaran lobster laut Panulirus sp. yang dipelihara dalam bak terkontrol selama 30 hari. Lobster laut yang digunakan pada awal penelitian memiliki bobot 130,39 ± 0,32 g dan panjang total 140,70 ± 0,06 mm. Penelitian dilakukan menggunakan rancangan acak lengkap dengan tiga perlakuan kepadatan berbeda, yaitu: 10 ekor/m3(KT10), 18 ekor/m3(KT18), dan 26 ekor/m3(KT26) dan dua ulangan. Parameter uji yang diamati dalam penelitian ini meliputi kualitas air (suhu, salinitas, pH, DO, dan TAN), respons fisiologis (total hemosit/THC, glukosa hemolim, dan frekuensi pergantian kulit), dan kinerja produksi meliputi pertumbuhan, laju pertumbuhan spesifik, rasio konversi pakan, dan tingkat kelangsungan hidup. Hasil penelitian menunjukkan bahwa lobster laut dapat dibesarkan dengan kepadatan tinggi dalam bak terkontrol. Pengukuran nilai kualitas air pada setiap perlakuan selama pemeliharaan adalah suhu berkisar 26,56–28,65oC, salinitas 29,7–33,6 g/L, pH 7,5–8,5, DO antara 6,15–6,58 mg/L dan TAN antara 0,11–034 mg/L. Perlakuan terbaik selama penelitian adalah dengan kepadatan 18 ekor/m3 (KT18) dengan nilai THC berkisar antara 2,5–3,5×106  sel/mL, glukosa hemolim 24,6–28,3 mg/dL,dan frekuensi pergantian kulit 38,37±3,20%. Bobot dan panjang lobster akhir rata-rata pada perlakuan tersebut masing-masing mencapai 145,06 ± 0,42 g, dan 142,77 ± 0,19 mm. Tingkat kelangsungan hidup mencapai 86,11 ± 3,92% dengan laju pertumbuhan spesifik 0,35 ± 0,01%/hari dan rasio konversi pakan selama penelitian adalah 7,87 ± 0,31.Kata kunci: padat pemeliharaan, Panulirus sp., produktivitas, respons fisiologis.

Abstract

ABSTRACT This study aimed to select lactic acid bacteria (LAB) as a potential probiotic that producing anti‒microbial compounds in order to treat motile aeromonads septicemia diseases caused by Aeromonas hydrophila on catfish Clarias sp. and evaluated its performance on gnotobiotic catfish. The in vitro assay was done to select several LAB isolates based on antagonistic activity against pathogenic bacteria. The selected isolate was tested in vivo to observe their ability to improve growth performances of catfish. The study was conducted with five treatments consists of K‒ (normal catfish without addition probiotic, without challenge test), K+ (normal catfish without addition of probiotic, with challenge test), Np (normal catfish with addition of probiotic and challenge test), G (gnoto catfish without addition of probiotic, with challenge test), and Gp (gnoto catfish with addition of probiotic and challenge test). The results showed that the addition of Pediococcus pentosaceus E2211 as selected probiotic could increase survival rate, specific growth rate, and immune response towards infection of A. hydrophila. The best survival rate after challenge test was obtained in Np and Gp treatments (88.46%), followed by G treatment (65.38%), while the K+ was only 53.84%. The conclusion of this study was P. pentosaceus E2211 potentially used as a probiotic candidate for normal and gnotobiotic catfish. The presence of normal microflora with P. pentosaceus E2211 in Np treatment showed the best probiotic performance with daily growth rate 3.28%, feed conversion ratio 1.79, and total intestinal bacteria reached 108 CFU/mL significantly different from other treatments (P<0.05).Keywords: Aeromonas hydrophila, catfish, LAB, probiotic, screening  ABSTRAKTujuan penelitian ini adalah menyeleksi bakteri asam laktat (BAL) sebagai probiotik potensial penghasil senyawa antimikrob guna menanggulangi penyakit motile aeromonad septicemia akibat Aeromonas hydrophila pada ikan lele Clarias sp. dan evaluasi kinerjanya pada ikan lele gnotobiotik. Pengujian in vitro dilakukan untuk menyeleksi beberapa isolat BAL sebagai kandidat probiotik berdasarkan aktivitas antagonis terhadap bakteri patogen. Isolat terpilih kemudian diuji in vivo untuk mengetahui kemampuannya dalam meningkatkan performa tumbuh ikan lele. Penelitian ini menggunakan rancangan acak lengkap dengan lima perlakuan, yaitu: K‒ (lele normal tanpa probiotik dan tanpa tanpa diuji tantang), K+ (lele normal tanpa probiotik dan diuji tantang), Np (lele normal diberi probiotik dan diuji tantang), G (lele gnoto tanpa probiotik dan diuji tantang), dan Gp (lele gnoto diberi probiotik dan diuji tantang). Hasil penelitian menunjukkan pemberian probiotik terpilih BAL Pediococcus pentosaceus E2211 mampu meningkatkan sintasan, laju pertumbuhan, dan respons imun ikan lele terhadap infeksi A. hydrophila. Sintasan terbaik pascauji tantang diperoleh pada perlakuan Np dan Gp yaitu sebesar 88,46%, diikuti perlakuan G sebesar 65,38%, sementara pada K+ hanya mencapai 53,84%. Kesimpulan dari penelitian ini ialah isolat BAL terpilih P. pentosaceus E2211 berpotensi sebagai kandidat probiotik untuk ikan lele normal dan lele gnotobiotik Clarias sp. Keberadaan mikroflora normal yang berasosiasi dengan P. pentosaceus E2211 pada perlakuan Np menunjukkan kinerja probiotik terbaik dengan nilai laju pertubuhan harian 3,28%, rasio konversi pakan 1,79 dan total bakteri usus mencapai 108 CFU/mL yang berbeda signifikan dibanding perlakuan lainnya (P<0,05).Kata kunci: A. hydrophila, BAL, ikan lele, probiotik, seleksi

Abstract

ABSTRACT This study aimed to evaluate the immune response and growth performance of white shrimp administered with prebiotic mannan oligosaccharides (MOS) with dosages of (0%, 0.2%, 0.4%, and 0.8% in diet) and used in the feeding trial. Shrimps (Litopenaeus vannamei) (the initial average weight was 3.416±0.064 g) were fed at satiation, three times a day. A completely randomized design was used  in the study. Shrimps were cultured at the stock density of 15 shrimps 40/L for each treatment in triplicates. After 30 days of the feeding trial, shrimp were challenged with white spot syndrome virus filtrate by intramuscular injection. The total gut bacteria, total haemocyte count (THC), phenoloxydase (PO), and respiratory burst (RB) activity were observed 4 times, before the experiment, day 30th before challenge test, day 32nd after challenge test, and day 36th the end of the experiment. The shrimp survival was observed at day 36th to evaluate the immune responses. The results showed that THC, PO activity, RB activity, growth performance, and shrimp survival administered with prebiotic 0.8% were significantly higher (P<0.05) than control. The administration of prebiotic with dose 0.8% was the best result and could effectively improve the immune responses and growth performance of whiteleg shrimp. Keywords: prebiotic, whiteleg shrimp, white spot disease  ABSTRAK Tujuan dari penelitian ini adalah untuk mengevaluasi respons imun dan performa pertumbuhan pada udang vaname yang diberi prebiotik mannan-oligosaccharides (MOS) dengan dosis berbeda (0%, 0,2%, 0,4%, dan 0,8%) pada pakan. Udang vaname (Litopenaeus vannamei) (dengan rata-rata bobot 3,41 ± 0,06 g) diberi pakan tiga kali sehari secara at satiation. Penelitian ini menggunakan rancangan acak lengkap. Udang dipelihara dengan kepadatan 15 ekor per 40/L pada setiap perlakuan dengan tiga kali pengulangan. Setelah 30 hari pemberian pakan, udang diuji tantang menggunakan white spot syndrome virus dengan diinjeksi secara intramuskular. Total bakteri usus, total haemocyte count (THC), aktivitas phenoloxydase (PO), dan aktivitas respiratory burst (RB) diamati 4 kali, yaitu sebelum perlakuan hari ke-30 sebelum uji tantang, hari ke-32 setelah uji tantang, dan hari ke-36 pada akhir penelitian. Hasil penelitian menunjukkan bahwa THC, aktivitas RB, aktivitas PO, performa pertumbuhan, dan kelangsungan hidup yang diberi prebiotik dengan dosis 0,8% lebih tinggi (P<0,05) jika dibandingkan dengan kontrol (dosis 0%). Pemberian prebiotik dengan dosis 0,8% merupakan hasil terbaik dan secara efektif mampu meningkatkan respons imun dan performa pertumbuhan pada udang vaname. Kata kunci: prebiotik, udang vaname, white spot disease

Abstract

ABSTRACTThis study aimed to produce microencapsulated probiotic Pseudoalteromonas piscicida (1Ub) and evaluate it with preb­iotic mannan-oligosaccharide (MOS) through the enrichment of Artemia sp., on bacterial population, growth performances, immune responses, and disease resistance of Pacific white shrimp larvae. Microencapsulation of probiotic was done by the freeze-drying method. The shrimp larvae were reared for 13 days and fed by the Artemia sp. enriched with microcapsule of probiotic 1Ub (10 g/L), prebiotic MOS (12 mg/L), synbiotic, and control without administration of microencapsulated probiotic and prebiotic, including negative (C-) and positive (C+) control. On the day 14, all of the experimental shrimp larvae except C- were challenged through immersion method with Vibrio harveyi MR5339 (107 CFU/mL). This study showed that the administration of microcapsule of probiotic 1Ub, prebiotic MOS, and synbiotic through the enrichment of Artemia sp. could increase the bacteria population, growth performances, immune responses, and disease resistance of Pacific white shrimp larvae. Moreover, synbiotic treatment demonstrated the best result compared to other treatments.Keywords: probiotic, prebiotic, synbiotic, Pacific white shrimp, microencapsulation ABSTRAKPenelitian ini bertujuan untuk membuat mikrokapsul probiotik Pseudoalteromonas piscicida (1Ub) dan mengevaluasinya dengan prebiotik mannan-oligosaccharides (MOS) melalui pengayaan Artemia sp. terhadap populasi bakteri, performa pertumbuhan, respons imun dan resistensi penyakit pada larva udang vaname. Mikroenkapsulasi probiotik dilakukan dengan metode freeze-drying. Larva udang dipelihara selama 13 hari dan diberi pakan Artemia sp. yang telah diperkaya dengan mikrokapsul probiotik 1Ub (10 g/L), prebiotik MOS (12 mg/L), sinbiotik, dan kontrol tanpa penambahan mikrokapsul probiotik dan prebiotik, termasuk kontrol negatif (C-) dan positif (C+). Pada hari ke-14, seluruh larva udang percobaan kecuali C- diuji tantang melalui metode perendaman dengan Vibrio harveyi MR5339 (107 CFU/mL). Hasil penelitian menunjukkan bahwa pemberian mikrokapsul probiotik 1Ub, prebiotik MOS, dan sinbiotik melalui pengayaan Artemia sp. dapat meningkatkan populasi bakteri, performa pertumbuhan, respons imun, dan resistensi penyakit pada larva udang vaname. Selain itu, perlakuan sinbiotik menunjukkan hasil terbaik dibandingkan perlakukan lainnya.Kata kunci : probiotik, prebiotik, sinbiotik, udang vaname, mikroenkapsulasi
