Abstract

Spiny lobster is a member of crustaceans that has high economic value. The demand for export markets in Asian, European and American countries as well as locally, is quite high with an expensive selling price. However, there are obstacles that often occur in spiny lobster rearing cultivation, namely infectious diseases that will cause high mortality. The purpose of this study was to investigate infectious diseases in rearing spiny lobster Panulirus spp. that were kept in floating net cages at Tanjung Putus, Lampung. The research method involved sampling five spiny lobsters in floating net cages, observation of clinical symptoms, bacterial isolation, identification of bacteria, detection of Milky Hemolymph Disease of Spiny Lobster (MHD-SL) through polymerase chain reaction (PCR) analysis and histopathology. The sampling location was at PT. Saibatin Perikanan Indonesia. The bacteria Vibrio parahaemolyticus and Vibrio alginolyticus were detected. Five lobsters were infected by MHD-SL, characterized by milky white hemolymph. All five lobster samples were positively infected with Milky Hemolymph Disease of Spiny Lobster (MHD-SL) through PCR analysis. Histopathological observations showed pathological microanatomical changes in the lobster hepatopancreas tissue, indicating that the tissue changes, in the form of encapsulation and infiltration were due to Rickettsia-like bacteria (RLB) infection. The management of spiny lobster stocking density, maintenance biosecurity, and increased lobster immunity must be implemented in order to prevent MHD-SL disease in floating net cages. Permintaan pasar ekspor pada negara Asia, Eropa, dan Amerika maupun lokal cukup tinggi dengan nilai jual yang mahal. Akan tetapi, terdapat kendala yang sering terjadi pada budidaya pembesaran lobster yaitu infeksi penyakit yang akan menyebabkan tingginya mortalitas Tujuan penelitian ini adalah untuk menginvestigasi penyakit infeksi pada pembesaran lobster Panulirus spp. yang dipelihara di keramba jaring apung Tanjung Putus, Lampung. Metode penelitian ini meliputi pengambilan sampel lobster sebanyak lima ekor dalam karamba jarring apung (KJA), pengamatan gejala klinis, isolasi bakteri, identifikasi bakteri, deteksi Milky Hemolymph Disease of Spiny Lobster (MHD-SL) melalui analisis Polymerase Chain Reaction (PCR), dan histopatologi. Lokasi pengambilan sampel di PT. Saibatin Perikanan Indonesia, Lampung. Terdapat bakteri Vibrio parahaemolyticus dan V. alginolyticus. Lima ekor lobster terinfeksi oleh MHD-SL yang ditandai dengan hemolim berwarna putih susu. Lima sampel lobster positif terinfeksi penyakit Milky Hemolymph Disease of Spiny Lobster (MHD-SL) melalui analisis PCR. Hasil pengamatan histopatologi terdapat perubahan mikroanatomi patologi pada jaringan hepatopankreas lobster menunjukkan bahwa adanya perubahan jaringan berupa enkapsulasi dan infiltrasi karena adanya infeksi Rickettsia-like bacteria (RLB). Pengelolaan kepadatan penebaran lobster, biosekuritas pemeliharaan, dan peningkatan imunitas lobster harus ditingkatkan untuk mencegah penyakit MHD-SL di KJA. Abstract

This study aimed to evaluate the inhibition of V. parahaemolyticus infection in the whiteleg shrimp (Penaeus vannamei) reared in different volumes of biofloc system. Post-larval shrimp with an average body weight of 0.28 ± 0.01 g were reared in 15 aquariums with working volume of 33.3 L and stocking density of 3 shrimp per liter. The shrimp were reared for 21 days in a biofloc system (C:N ratio of 10) with biofloc volume of 5, 10, and 15 mL/L, and challenged with V. parahaemolyticus at a density of 103 CFU/mL initially. The positive control treatment involved shrimp reared without biofloc and challenged, while the negative control treatment involved shrimp reared without biofloc and without challenged. The shrimp was fed with commercial feed while the protein content of 39-40% four times a day. The results showed that the presumptive Vibrio count (PVC) and the population of V. parahaemolyticus in the biofloc treatments were significantly lower than the positive control (p<0.05). Furthermore, the biofloc treatment with a volume of 15 mL/L demonstrated the best results compared to other treatments in decreasing PVC and V. parahaemolyticus population as evidenced by the immune response, survival rate, and growth performance of whiteleg shrimp. Keywords: biofloc, immune response, Penaeus vannamei, Vibrio parahaemolyticus ABSTRAK Penelitian ini bertujuan untuk mengevaluasi penghambatan infeksi V. parahaemolyticus pada udang vaname (Penaeus vannamei) yang dipelihara dalam sistem bioflok dengan volume yang berbeda. Post-larvae udang dengan bobot tubuh rata-rata 0,28 ± 0,01 g dipelihara dalam 15 akuarium dengan volume kerja 33.3 L dan padat tebar 3 udang per liter. Udang dipelihara selama 21 hari dalam sistem bioflok (rasio C:N 10) dengan volume bioflok 5, 10, dan 15 mL/L, dan diuji tantang dengan V. parahaemolyticus pada kepadatan awal 103 CFU/mL. Perlakuan kontrol positif melibatkan udang yang dipelihara tanpa bioflok dan diuji tantang, sementara perlakuan kontrol negatif melibatkan udang yang dipelihara tanpa bioflok dan tanpa diuji tantang. Udang diberi pakan komersial dengan kandungan protein 39-40% empat kali sehari. Hasilnya menunjukkan bahwa presumptive Vibrio count (PVC) dan populasi V. parahaemolyticus dalam perlakuan bioflok signifikan lebih rendah daripada kontrol positif (p<0,05). Selain itu, perlakuan bioflok dengan volume 15 mL/L menunjukkan hasil terbaik dibandingkan dengan perlakuan lain dalam menurunkan PVC dan populasi V. parahaemolyticus sebagaimana dibuktikan oleh respons imun, tingkat kelangsungan hidup, dan kinerja pertumbuhan udang vaname. Abstract

Vibrio parahaemolyticus infection is a major threat to Pacific white shrimp aquaculture, causing significant shrimp mortality. Bacteriophages (phages) provide a promising antibiotic alternative due to their specificity towards specific bacteria. This research includes essential steps isolating phages from shrimp pond water and characterizing them, including plaque morphology, spot tests, phage density evaluations, host range assessments, and in vitro studies targeting V. parahaemolyticus. In-vitro tests encompassed six treatments, each with three replicates: negative control (K-), positive control (K+), antibiotic control, phage density of 107 (FB7), phage density of 108 (FB8), and phage density of FB109 (FB9). Phages were successfully isolated from shrimp pond water in Banyuwangi, indicated by plaque formation on double-layer agar media. These phages exhibited morphological characteristics, featuring small, round plaques (d= 0.02-0.5 ± 0.08 cm) with flat, wavy edges and clear plaque. Phage plaques were amplified with an average density of 3.5×109 PFU/mL. Host range tests revealed that only V. parahaemolyticus Situbondo was susceptible to the isolated phages, while five other bacterial strains were not. In bacterial inhibition tests, treatments with different phage densities significantly outperformed the negative control, media control, and antibiotic control. However, the antibiotic control performed better than phage density treatments (0.61 ± 0.01). All phage density treatments effectively reduced V. parahaemolyticus density compared to the negative control, with the most favorable results observed in the FB9 treatment at a density of 109 PFU/mL. Penggunaan bakteriofage (fage) dapat menjadi alternatif solusi untuk menggantikan antibiotik, karena fage bersifat spesifik pada bakteri tertentu. Penelitian ini meliputi beberapa tahapan, yaitu isolasi fage dari beberapa sumber air tambak, karakterisasi fage meliputi morfologi plak fage, spot test, dan uji kisaran inang, serta uji in vitro penghambatan V. parahaemolyticus oleh fage. Uji in vitro dilakukan dengan enam perlakuan dan masing masing tiga ulangan yaitu kontrol negatif (K-), kontrol positif (K+), kontrol antibiotik, kepadatan fage 107 (FB7), kepadatan fage 108 (FB8), dan kepadatan fage 109 (FB9). Penelitian ini berhasil mengisolasi fage yang bersumber dari air tambak udang di Banyuwangi ditandai dengan terbentuknya plak pada media double layer agar, dengan ciri morfologisnya berbentuk titik-titik dan bulat besar (d= 0,02 ± 0,08 cm), tepian rata dan bergelombang serta memiliki warna plak yang bening. Plak fage dapat diperbanyak dengan densitas fage rata-rata 3,5×109 PFU/mL. Untuk uji kisaran inang menggunakan 6 jenis bakteri dengan sumber dan strain yang berbeda, menunjukkan bahwa hanya bakteri V. parahaemolyticus Situbondo yang dapat lisis oleh fage hasil isolasi, sedangkan lima jenis bakteri lainnya tidak. Pada daya hambat bakteri, perlakuan dengan penambahan pada setiap densitas fage berbeda nyata dibandingkan dengan kontrol negatif, kontrol media dan kontrol antibiotik, walaupun kontrol antibiotik lebih baik dari perlakuan densitas fage (0,61 ± 0,01). Semua perlakuan kepadatan fage mampu mengurangi kepadatan V. parahaemolyticus dibandingkan dengan perlakuan kontrol negatif. Hasil terbaik terdapat pada perlakuan FB9 yaitu 109 PFU/mL. Abstract

This study evaluated the efficacy of Eurycoma longifolia, Curcuma zedoaria, and Allium sativum in improving the immune response and resistance of whiteleg shrimp to prevent Vibrio parahaemolyticus infection. The study consisted of two phases, an in vitro phase to determine the compounds contained in three medicinal plants as antibacterials, followed by an in vivo phase to evaluate the effect of the medical plant extract on immune response and robustness against V. parahaemolyticus. The results from the first phase revealed that bioactive compounds present in E. longifolia were more varied and had higher concentrations with a lower bactericidal value when compared to those found in C. zedoaria or A. sativum. In the second phase of the experiment, the medicinal plant extract was added to the feed with a dose that was determined according to the first phase results. The treatments tested in the second phase were 1.6% E. longifolia extract dietary addition (EL16), 6.4% C. zedoaria extract dietary addition (CZ64), 6.4% A. sativum extract dietary addition (AS64) and phytobiotics mixture of 1:1:1 (C1) dietary addition, as well as no phytobiotic for negative control treatment and positive control. The results from the second stage demonstrated that dietary phytobiotic extract addition enhances the immunological responses and improves the shrimp survival against V. parahaemolyticus challenge compared to the control group. In conclusion, E. longifolia, C. zedoaria, and A. sativum showed different bioactive compound profiles, which affect their efficacy against V. parahaemolyticus, with EL16 showing higher efficacy. Penelitian ini terdiri dari dua tahap: tahap in vitro untuk menentukan senyawa yang terkandung dalam tiga tanaman obat sebagai antibakteri, diikuti oleh tahap in vivo untuk mengevaluasi efek ekstrak tanaman obat terhadap respon imun dan ketahanan terhadap V. parahaemolyticus. Hasil dari tahap pertama mengungkapkan bahwa senyawa bioaktif yang ada dalam E. longifolia lebih bervariasi dan memiliki konsentrasi yang lebih tinggi dengan nilai bakterisida yang lebih rendah jika dibandingkan dengan yang ditemukan di C. zedoaria atau A. sativum. Pada percobaan tahap kedua, ekstrak tanaman obat ditambahkan ke pakan dengan dosis yang ditentukan sesuai dengan hasil tahap pertama. Perlakuan yang diuji pada tahap kedua adalah penambahan 1,6% ekstrak E. longifolia dalam pakan (EL16), penambahan 6,4% ekstrak C. zedoaria dalam pakan (CZ64), penambahan 6,4% ekstrak A. sativum dalam pakan (AS64) dan campuran fitobiotik 1:1:1 (C1) dalam pakan, serta tanpa fitobiotik untuk perlakuan kontrol negatif dan kontrol positif. Hasil dari tahap kedua menunjukkan bahwa penambahan ekstrak fitobiotik dalam pakan meningkatkan respons imunologi dan memperbaiki kelangsungan hidup udang terhadap tantangan V. parahaemolyticus dibandingkan dengan kelompok kontrol. Sebagai kesimpulan, E. longifolia, C. zedoaria, dan A. sativum menunjukkan profil senyawa bioaktif yang berbeda, yang mempengaruhi efikasinya terhadap V. parahaemolyticus, dengan EL16 menunjukkan efikasi yang lebih tinggi. Abstract

Vibrio harveyi is one of the common bacteria that cause disease in vaname shrimp. The aim of this study is to determine the efficacy of bacterial metabolites Proteus myxofaciens JB 20B as an antibiofilm agent in the prevention and treatment of V. harveyi infected vaname shrimp (Litopenaeus vannamei). The experimental design included eight treatments and three replicates: K- (negative control), K+ (positive control), PCA and PGA (antibiotic control 50 g/100ml for preventive and treatment), PCE (preventive extract 0.1 ml/Kg feed), PCS (preventive supernatant 20 ml/Kg feed), PGE (treatment extract 0.1 ml/Kg feed), and PGS (treatment supernatant 20 ml/Kg feed). On day 15th, vaname shrimp were intramuscularly infected with 106 CFU/mL of V. harveyi. Examining clinical symptoms, viewing histology, assessing total hemocyte count, phagocytosis activity, phenoloxidase activity, respiratory burst, and growth performance. According to the findings of this study, the treatment of bacterial metabolites P. myxofaciens JB 20B (PCE, PCS, PGE, and PGS) is superior to the positive control (K+) treatment in terms of reducing shrimp hepatopancreas necrosis, increasing the value of total hemocyte count, phagocytosis activity, phenoloxidase activity, respiratory burst, and growth performance of vaname shrimp through feeding in both prevention and treatment. The use of P. myxofaciens JB 20B bacterial metabolites to control V. harveyi infection in vaname shrimp yielded the best results in the supernatant treatment. Tujuan dari penelitian ini adalah untuk mengetahui efektivitas metabolit bakteri Proteus myxofaciens JB 20B sebagai agen antibiofilm dalam pencegahan dan pengobatan udang vaname (Litopenaeus vannamei) yang terinfeksi V. harveyi. Rancangan penelitian ini terdiri dari delapan perlakuan dan tiga kali ulangan: K- (kontrol negatif), K+ (kontrol positif), PCA dan PGA (kontrol antibiotik 50 g/100ml untuk pencegahan dan pengobatan), PCE (ekstrak pencegahan 0,1 ml/Kg pakan), PCS (supernatan pencegahan 20 ml/Kg pakan), PGE (ekstrak pengobatan 0,1 ml/Kg pakan), dan PGS (supernatan pengobatan 20 ml/Kg pakan). Pada hari ke-15, udang vaname diinfeksi secara intramuskular dengan 106 CFU/mL V. harveyi. Memeriksa gejala klinis, melihat histologi, menilai jumlah hemosit total, aktivitas fagositosis, aktivitas phenoloxidase, respiratory burst, dan performa pertumbuhan. Berdasarkan hasil penelitian ini, perlakuan metabolit bakteri P. myxofaciens JB 20B (PCE, PCS, PGE, dan PGS) lebih unggul dibandingkan dengan perlakuan kontrol positif (K+) dalam hal mengurangi nekrosis hepatopankreas udang, meningkatkan nilai jumlah hemosit total, aktivitas fagositosis, aktivitas phenoloxidase, respiratory burst, dan performa pertumbuhan udang vaname melalui pemberian pakan pasca infeksi baik pada pencegahan maupun pada pengobatan. Penggunaan metabolit bakteri P. myxofaciens JB 20B untuk mengendalikan infeksi V. harveyi pada udang vaname memberikan hasil terbaik pada perlakuan supernatan. Abstract

Forest onion potentially acts as an immunostimulant, enhancing the immune response of tilapia to pathogen infection. This study aimed to evaluate the effectiveness of forest onion simplisia on growth performance, immune response, and resistance of tilapia to Streptococcus agalactiae infection. The research was divided into in vitro and in vivo tests. In vitro testing was conducted to determine the dose of forest onion extract inhibiting the growth of S. agalactiae with seven treatments, namely positive control, negative control, addition of forest onion extract at 0.625, 1.25, 2.5, 5, and 10 mg/mL. In vivo testing to evaluate the administration of forest onion simplisia through feed in enhancing growth performance, immune response, and resistance of tilapia to S. agalactiae. Tilapia with an average weight of 7.57 ± 0.26 g were maintained in glass aquariums sized 60×30×30 cm3 with 15 units at a density of 15 fish/aquarium for 30 days. Experimental treatments included maintaining tilapia with the addition of forest onion simplisia at 12.5, 25, and 50 g/kg feed, negative and positive controls. The results showed that the addition of 12.5 g/kg simplisia significantly increased growth which was significantly different from the other treatments, while the immune response and resistance to S. agalactiae in all simplisia treatments showed values that were not different, but significantly different from the control treatment. The conclusion is supplementation of forest onion simplisia at a dose of 12.5 g/kg effectively enhances growth performance, immune response, and resistance of tilapia to S. agalactiae infection. Keywords: forest onion, immunostimulant, Nile tilapia, Streptococcus agalactiae ABSTRAK Bawang hutan berpotensi sebagai imunostimulan yang dapat meningkatkan respons imun ikan terhadap infeksi patogen. Penelitian ini bertujuan mengevaluasi efektivitas simplisia bawang hutan terhadap kinerja pertumbuhan, respons imun dan resistansi ikan nila terhadap infeksi Streptococcus agalactiae. Penelitian terbagi menjadi pengujian in vitro dan in vivo. Pengujian in vitro dilakukan untuk menentukan dosis ekstrak bawang hutan dalam menghambat pertumbuhan bakteri S. agalactiae dengan delapan perlakuan yaitu kontrol positif, kontrol negatif, penambahan ekstrak bawang hutan 0,625,1.25, 2,5, 5, dan 10 mg/mL. Adapun pengujian in vivo untuk mengevaluasi pemberian simplisia bawang hutan dalam meningkatkan kinerja pertumbuhan, respons imun, dan resistansi ikan nila terhadap S. agalactiae. Ikan nila berukuran 7,57 ± 0,26 g dipelihara pada akuarium berukuran 60×30×30 cm3 sebanyak 15 unit dengan kepadatan 15 ekor/akuarium selama 30 hari. Perlakuan uji meliputi pemeliharaan ikan nila dengan penambahan simplisia bawang hutan 12.5, 25, dan 50 g/kg pakan, kontrol negatif dan positif. Hasil penelitian menunjukkan penambahan simplisia 12,5 g/kg signifikan meningkatkan pertumbuhan yang berbeda nyata dengan perlakuan lainnya, adapun respons imun dan resistansi terhadap S. agalactiae pada semua perlakuan simplisia menunjukkan nilai yang tidak berbeda, akan tetapi berbeda nyata dengan perlakuan kontrol. Kesimpulan penelitian ini adalah suplementasi simplisia bawang hutan 12,5 g/kg efektif meningkatkan kinerja pertumbuhan, respons imun, dan resistansi ikan nila terhadap S. agalactiae. Abstract

Vannamei shrimp is one of the most economically valuable aquaculture commodities in Indonesia. One of the pathogenic bacteria that is often found in vannamei shrimp farming is Vibrio parahaemolyticus. This study aimed to analyze the effectivity of Lactobacillus paracasei probiotics and paraprobiotics through feed with different cell densities on growth, and immune responses infected with V. parahaemolyticus. Vannamei shrimp of size 0.63 ± 0.01 were reared in containers with a stocking density of 15 shrimp per container and supplemented feed for 30 days. The research design consisted of six treatments, each with three replicates, namely (K-) feeding without supplements and injected with phosphate-buffered saline (PBS), (K+) feeding without supplements and infected with V. parahaemolyticus (104 CFU/mL), (PRI) feeding with 1% (v/w) probiotic L. paracasei with cell density of 106 CFU/mL, (PRII) 1% (v/w) probiotic L. paracasei cell density 109 CFU/mL, (PAI) 1% (v/w) paraprobiotic L. paracasei cell density 106 CFU/mL, (PAII) 1% (v/w) paraprobiotic L. paracasei cell density 109 CFU/mL. All treatments, except K-, were infected with V. parahaemolyticus (104 CFU/mL). Vannamei shrimp rearing was continued post the challenge test with V. parahaemolyticus which was conducted up to six days post injection. The results showed that feeding both probiotic and paraprobiotic L. paracasei through feed has improved growth, immune response, protein fat retention, and digestive enzyme activity of vannamei shrimp better than those of control. As the recommendation for the disease control of V. parahaemolyticus is feed supplementation with 1% (v/w) probiotic L. paracasei with cell density of 109 CFU/mL. Salah satu bakteri patogen yang sering ditemukan dalam budidaya udang vaname ialah bakteri Vibrio parahaemolyticus. Penelitian ini bertujuan untuk menganalisis pemberian probiotik dan paraprobiotik Lactobacillus paracasei melalui pakan dengan kepadatan sel berbeda terhadap pertumbuhan, dan respons imunitas yang diinfeksi V. parahaemolyticus. Udang vaname dengan ukuran 0.63 ± 0.01 gr dipelihara di dalam kontainer dengan padat tebar 15 ekor per wadah dan pemberian pakan bersuplemen selama 30 hari. Rancangan penelitian terdiri dari enam perlakuan, tiga ulangan, yaitu (K-) pemberian pakan tanpa suplemen dan diinjeksi PBS, (K+) pemberian pakan tanpa suplemen dan diinfeksi V. parahaemolyticus (104 CFU/mL), (PRI) pemberian pakan dengan probiotik L. paracasei kepadatan sel 106 CFU/mL dosis 1% (v/w), (PRII) probiotik L. paracasei kepadatan sel 109 CFU/mL dosis 1% (v/w), (PAI) paraprobiotik L. paracasei kepadatan sel 106 CFU/mL dosis 1% (v/w), (PAII) paraprobiotik L. paracasei kepadatan sel 109 CFU/mL dosis 1% (v/w), dan masing-masing diinfeksi V. parahaemolyticus (104 CFU/mL). Pemeliharaan udang vaname dilanjutkan setelah uji tantang dengan V. parahaemolyticus yang dilakukan hingga enam hari pasca injeksi. Hasil penelitian menunjukkan bahwa pemberian probiotik maupun paraprobiotik L. paracasei melalui pakan telah meningkatkan pertumbuhan, respons imun, retensi lemak protein, dan aktivitas enzim pencernaan udang vaname lebih baik dibandingkan kontrol. Untuk pengendalian V. parahaemolyticus diperoleh hasil terbaik dengan aplikasi probiotik L. paracasei dosis 1% (v/w) dengan kepadatan sel 109 CFU/mL. Abstract

Penyakit bakterial pada kegiatan budidaya ikan sudah menjadi masalah yang sering dihadapi pembudidaya. Salah satu jenis bakteri yang menyebabkan penyakit pada ikan lele (Clarias sp.) adalah bakteri Aeromonas hydrophila. Tujuan penelitian ini untuk mengevaluasi efek pemberian probiotik B. megaterium PTB 1.4 dan P. pentosaceus E2211 terhadap respons imun dan kelangsungan hidup ikan lele selama uji tantang dengan bakteri A. hyrophila. Probiotik yang digunakan adalah kombinasi 0,5% B. megaterium PTB 1.4 dan 0,5%  P. pentosaceus E2211. Pemberian probiotik dilakukan pada hari ke-0 hingga hari ke-30 melalui pakan. Uji tantang dilakukan pada hari ke-30 hingga hari ke-40. Hasil penelitian menunjukkan bahwa pemberian probiotik dapat berpengaruh terhadap respons imun dan kelangsungan hidup ikan lele dalam menekan infeksi A. hydrophila. Total eritrosit, hemoglobin, hematokrit, total leukosit, aktivitas fagositik dan aktivitas lisozim signifikan (P<0,05) teramati meningkat pada hari ke-35 dan menurun pada hari ke-40. Kelangsungan hidup ikan lele berhasil dipertahankan pada 87,62±4,83%. Total bakteri A. hydrophila meningkat pada hari ke-35 dan menurun pada hari ke-40. Pemberian probiotik B. megaterium PTB 1.4 dan P. pentosaceus E2211 pada dosis 0,5% dapat menekan pertumbuhan bakteri A. hydrophila dan meningkatkan status kesehatan ikan lele.

Abstract

Catfish (Clarias sp.) aquaculture in Kediri Regency, East Java, faces challenges due to the limited availability of high-quality broodstock and the lack of mastery in catfish spawning technologies, such as broodstock selection and the use of natural materials and probiotics to accelerate gonad maturation. These issues formed the basis for implementing a community empowerment program titled "Empowering Fish Farmers through Reproductive Process Education to Improve Catfish Seed Productivity in Kediri Regency." The program aimed to educate fish farmers on proper catfish breeding techniques, from broodstock selection to the use of probiotics and pine fruit extract to enhance gonad maturation and broodstock rematuration, through outreach activities. The community service program included public outreach to fish farming communities and discussions on program sustainability with the Kediri Regency Fisheries Department. Key stakeholders involved in the program included IPB University, the Fisheries Department of Kediri Regency, fisheries extension officers (Ministry of Marine Affairs and Fisheries), and catfish farmer groups in the region. The outreach began with a pre-test and concluded with a post-test to evaluate the program’s effectiveness. The materials covered included broodstock selection, application of pine fruit extract and probiotics for accelerating gonad maturation and rematuration. The community responded positively, as evidenced by an increase in average test scores from 78.42 (pre-test) to 92.63 (post-test), indicating that participants had gained a general understanding of catfish farming techniques and, through the outreach, comprehended the technology needed to produce high-quality broodstock.

Abstract

One of the common diseases affecting the Pacific white shrimp (Penaeus vannamei) culture is vibriosis, caused by infection with Vibrio species, including Vibrio parahaemolyticus. Certain strains of V. parahaemolyticus that carry the PirA and PirB toxins are responsible for causing acute hepatopancreatic necrosis disease (AHPND). This study aimed to assess the pathogenicity of V. parahaemolyticus in P. vannamei using bacterial isolates from different sources. A challenge test was conducted using P. vannamei with an average weight of 0.9±0.1 g, exposed to bacterial concentrations of 104 CFU/mL, as determined by lethal concentration 50% (LC50). The experiment followed a completely randomized design with three treatments and three replicates: a control group (K) of uninfected shrimp, shrimp infected with V. parahaemolyticus from Tasikmalaya, West Java (Vp-1), and shrimp infected with V. parahaemolyticus from Situbondo, East Java (Vp-2). Parameters observed included the confirmation of V. parahaemolyticus and AHPND via polymerase chain reaction (PCR), mortality rate, clinical symptoms, bacterial load, and immune response indicators, including total haemocyte count, phagocytic activity, respiratory burst, phenoloxidase activity, and histopathological examination of the hepatopancreas and intestines. Both Vp-1 and Vp-2 isolates were identified as V. parahaemolyticus AHPND strains, infecting shrimp with clinical signs such as pale hepatopancreas, empty intestines, and lethargic movement. Tissue damage, including extensive necrosis in the hepatopancreas and intestines, was observed, leading to mortality rates of 73.33-81.67% with an average time to death ranging from 24.28 to 65.44 hours postinfection.
