Abstract

The divorce rate in Indonesia continues to increase, as data shows that in 2021, there were 447,743 divorce cases, with 75.34% of them being divorce lawsuits filed by wives. This phenomenon is no exception in North Lore District, Poso Regency, where divorce lawsuits have increased significantly in the last three years. This study aims to analyze the main causes of the escalation of divorce lawsuits by wives against husbands in the area from the perspective of Maqashid Syari'ah. Data shows that domestic conflicts are often triggered by violations of the husband's rights and obligations, domestic violence, and unmanaged differences in character. In addition, the quality of marriage advice, which is still formal, contributes to the weak readiness of couples to face the challenges of marriage. Through a qualitative approach, this study found that the wife's increasing awareness of her rights is a significant factor in filing for divorce. This study emphasizes the importance of improving the quality of marriage advice based on Maqashid Syari'ah to prevent domestic conflict and maintain family integrity.

Abstract

Prostate cancer is a leading malignancy in men, ranking fourth globally and fifth in Indonesia (GLOBOCAN 2020). Conventional therapies, though available, are limited by high costs, side effects, and resistance, highlighting the need for accessible alternatives. Broccoli microgreens, rich in bioactive compounds, have shown potential in preventing and treating various cancers. This study hypothesized that bioactive compounds in broccoli microgreens interact with molecular targets involved in prostate cancer progression. To test this hypothesis, we employed a network pharmacology-based in silico approach to systematically explore these interactions and identify potential therapeutic mechanisms. Bioactive compounds in broccoli microgreens were identified using liquid chromatography-mass spectrometry (LC-MS) and analyzed via the PubChem database. The biological activities of these compounds were predicted using PASS Online, focusing on their capacity to modulate TP53 gene expression. Pharmacokinetic and toxicity evaluations were performed using ADMETLab 3.0 and Protox 3.0 to assess their safety and drug-like properties. Target proteins were identified through SwissTargetPrediction and GeneCards, while protein-protein interaction networks were constructed using STRING. The pharmacological network was visualized using Cytoscape to elucidate the molecular mechanisms of action. The analysis identified 528 relevant target proteins, with key roles attributed to SRC and EGFR, both critical in resistance to EGFR tyrosine kinase inhibitors and in regulating processes such as cell proliferation, apoptosis resistance, and metastatic potential. Through network pharmacology, bioactive compounds such as kaempferol and polydatin were identified as potential inhibitors of these targets, demonstrating their ability to modulate pathways essential to prostate cancer progression. In conclusion, broccoli microgreens contain bioactive compounds with potential pharmacological relevance for prostate cancer, particularly through their interaction with SRC and EGFR pathways, warranting further experimental validation.

Abstract

Marine-derived collagen peptides have been acknowledged for their therapeutic potential, especially in cancer therapy and inflammation management. The aim of this study was to investigate the molecular mechanisms that contribute to the anticancer, anti-inflammatory and antioxidant properties of yellowfin tuna collagen peptides (YFTCP) utilizing a network pharmacology approach. The YFTCP was extracted from the bones of yellowfin tuna (Thunnus albacares) and subsequently hydrolyzed with trypsin. Seventeen peptides were discovered using liquid chromatography in conjunction with high-resolution mass spectrometry (LC-HRMS). A network pharmacology method was utilized to investigate the interactions between the discovered peptides and their biological targets. Additionally, gene ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses were performed to identify pertinent biological pathways involved in the anticancer, antioxidant, and anti-inflammatory effects of these peptides. GO analysis revealed key associations between YFTCP and critical cancer- and inflammation-related genes encoding proteins such as CCND1, SRC, AKT1, IL-1β, TNF, and PPARG, which exhibited significant interactions. These proteins are essential for the regulation of the cell cycle, the development of tumors, and the response to inflammatory stimuli. The KEGG analysis also revealed that YFTCP was involved in a number of critical pathways, such as endocrine resistance, cancer pathways, Kaposi sarcoma-associated herpesvirus infection, proteoglycans in cancer, and human cytomegalovirus infection. These findings highlight the potential use of YFTCP as a multifaceted therapeutic agent, indicating their role in regulating important biological pathways associated with cancer development and inflammation. This study provides new valuable insights into the pharmacological properties of YFTCP, paving the way for future studies and drug development focused on these bioactive peptides.

Abstract

Abstract

Staphylococcus aureus, a Gram-positive bacterium, and Escherichia coli, a Gram-negative bacterium, are known to cause both enteric and systemic infections in humans. The improper use of antibiotics has often been associated with the emergence of antibiotic resistance. Purple chrysanthemum (Chrysanthemum morifolium) has shown significant potential as an alternative treatment, owing to its strong antioxidant, anti-inflammatory, and antibacterial properties. These properties are attributed to the presence of bioactive compounds such as polyphenols, flavones, essential oils, and other phytochemicals. This study aimed to identify secondary metabolites and evaluate the antibacterial activity of ethanol extracts from purple chrysanthemum (Chrysanthemum morifolium). The extraction process was conducted using Ultrasonic-Assisted Extraction (UAE), and the antibacterial activity was assessed using the disc diffusion method at extract concentrations of 12.5%, 25%, and 50%, with ciprofloxacin serving as the positive control. Phytochemical screening revealed the presence of alkaloids, flavonoids, tannins, phenolics, and steroids in the ethanol extract of purple chrysanthemum. Importantly, all ethanol extracts demonstrated antibacterial activity. The highest inhibition zones were observed at a 50% concentration, with S. aureus showing an inhibition zone of 12.35 ± 0.4 mm, while E. coli exhibited an inhibition zone of 15.08 ± 0.22 mm.
