Abstract

ABSTRACTAcrylamide has been classified as a cancer causing compound or potentially carcinogenic in humans. Acrylamide is produced from foods that contain high carbohydrates with temperatures over 1200C in processing. One of foods that are popular and potentially produce acrylamide compounds was fried potato. This study aims to determine the levels of acrylamide contained in french fries using UV-Vis Spectrophotometry method. Samples were obtained from 3 fast food restaurants in the City of Manado. In this study the analysis of acrylamide compounds from 3 samples were carried out at 267 nm wavelength using an aquadest blank with 2 repetitions. The estimated result of three samples are K samples of 0.69 µg/g, M of 0.58 µg/g, and T samples of 0.67 µg/g. The three samples did not contain acrylamide because the levels obtained were still below the detection limit (LOD) of 1.54 µg/g. Keywords: Acrylamide, French Fries, UV-Vis Spectrophotometry, Fast Food Restaurants in Manado. ABSTRAKAkrilamida telah diklasifikasikan sebagai senyawa yang menyebabkan kanker atau berpotensi sebagai karsinogenik pada manusia. Akrilamida dihasilkan dari makanan yang mengandung karbohidrat tinggi dengan suhu lebih dari 1200C pada pengolahannya. Makanan yang banyak digemari serta berpotensi menghasilkan senyawa akrilamida salah satunya adalah kentang goreng. Penelitian ini bertujuan untuk menetapkan kadar akrilamida yang terkandung dalam kentang goreng dengan menggunakan metode Spektrofotometri UV-Vis. Sampel diperoleh dari 3 restoran cepat saji di Kota Manado.  Pada penelitian ini analisis senyawa akrilamida dari 3 sampel dilakukan pada panjang gelombang 267 nm menggunakan blanko aquadest dengan 2 kali pengulangan. Hasil perhitungan dari ketiga sampel adalah sampel K sebesar 0,69 µg/g, M sebesar 0,58 µg/g, dan sampel T sebesar 0,67 µg/g. Ketiga sampel tidak mengandung akrilamida karena kadar yang didapat masih berada di bawah batas deteksi (LOD) yang didapat yaitu 1,54 µg/g. Kata Kunci:   Akrilamida, Kentang Goreng, Spektrofotometri UV-Vis,  Restoran cepat saji di Manado

Abstract

Rhodamin B is a synthetic dye that was banned for use in foods and is certified as a hazardousmaterial according to Minister of Health of Indonesian Republic No. 722/Menkes/Per/IX/1988because it can cause liver, kidney, and spleen damage, and followed by anatomical changes in theform of organ enlargement. Rhodamin B dye is often used for a food and beverage products. Thestudy was conducted to identify Rhodamin B on cakalang fufu in the market of Manado city.Cakalang fufu samples were taken from four markets in the city of Manado, namely : the Paal 2market, Bahu market, Karombasan market and Bersehati 45 Market. Samples were extracted by twomethods, the method of extraction followed by purification and extraction method using wool yarnand absorption followed by identification using thin layer chromatography (TLC) and detected withUV light 254 nm and 366 nm. Rhodamin B content is read using a UV-Vis spectrophotometry. Theresult showed that out of eight samples examined with two tests, there are four samples containingRhodamin B with higher levels of the average value of Rhodamin B in samples, that is from themerchant two of Bahu Market is 0,054 ug/ml, at merchant two Karombasan market is 0.184 ug/ml,34merchant one at Bersehati 45 Market is 0.103 ug/ml and merchant two at Bersehati 45 is 0.514 ug/ml.Based on these results, some of cakalang fufu circulating in Manado City Markets are not safe forconsumption.Keywords: Rhodamin B, Cakalang fufu, TLC and UV-Vis spectrophotometry

Abstract

ABSTRACTJackfruit leaves is a plant that work as medicine on healing process of skin diseases,especially in wounds. The purpose of this study is to determine the quality of ointment fromethanolic extract of Jackfruit leaves at the concentration on 5%, 10% and 15% and the effectof the open wound healing in rabbits. Type of this research is laboratory experimental. In thisresearch maceration was done as the extraction method to got the extract of Jackfruit leaves.The ointment of ethanolic extract from jackfruit leaves made within 3 concentrations of 5%,10% and 15%. On the testing ointment using organoleptic test, homogenity test and pH test.The effectiveness assay to heal the open wounds using 5 groups, Betadine ointment (positivecontrol), basic ointment (negative control), the ointment of ethanolic extract from Jackfruitleaves 5%, 10%, and 15% of the 3 rabbits with a 1.5 cm was done and observed until 7 days.The data were processed statistically using One Way ANOVA (Analysis Of Variant) andfollowed by LSD (Least Significant Different). Results of this study showed that the ointmentof ethanolic extract from Jackfruit leaves on the concentration of 5%, 10% and 15% producequalified ointment of organoleptic test, homogenity test and pH test, also give the effect ofopen wound healing in rabbits and the most effective cure is the ointment of ethanolic extractfrom Jackfruit leaves 15%.Keywords: ethanolic extract of Jackfruit leaves, ointment, wound, rabbit.

Abstract

ABSTRACT Algae have long been used for the treatment of various types of diseases. Ulva lactuca is one of the algae, which contains bioactive substances as antimicrobial, antifungal, and antioxidant. This study aims to determine the microorganisms growth inhibitory activity from Ulva lactuca algae obtained from the of Lembeh Strait waters in City of Bitung against microorganisms Escherichia coli, Staphylococcus aureus and Candida albicans. Ulva lactuca algae was extracted using maceration method with ethanol solvent and fractionated with methanol, n-hexan and chloroform solvents. Testing of antimicrobial activity using agar diffusion method. The result showed that extracts and fractions of Ulva lactuca algae did not have antimicrobial activity against the microorganisms Escherichia coli, Staphylococcus aureus and Candida albicans. The chemical composition of algae is influenced by season, geographical distribution, as well as environmental factors such as temperature, water, salinity, light, nutrition, and mineral availability.Keywords: Ulva lactuca, Antimicrobials, Maseration, Fractions, Agar DiffusionABSTRAK Alga telah lama digunakan untuk keperluan pengobatan berbagai jenis penyakit.  Ulva lactuca merupakan salah satu alga yang memiliki kandungan zat bioaktif sebagai antimikroba. Penelitian ini bertujuan untuk mengetahui adanya aktivitas penghambat pertumbuhan mikroorganisme dari alga Ulva lactuca yang diperoleh dari perairan Selat Lembeh kota Bitung terhadap mikroba Escherichia coli, Staphylococcus aureus, dan Candida albicans. Alga Ulva lactuca diekstraksi menggunakan metode maserasi dengan pelarut etanol dan difraksinasi dengan pelarut methanol, n-hexan, dan kloroform. Pengujian aktivitas antimikroba menggunakan metode difusi agar. Hasil penelitian menunjukkan ekstrak dan fraksi dari alga Ulva lactuca tidak memiliki aktivitas antimikroba terhadap Escherichia coli, Staphylococcus aureus, dan Candida albicans. Komposisi kimia alga dipengaruhi oleh musim, distribusi geografis, serta faktor lingkungan seperti suhu, air, salinitasi, cahaya, nutrisi, dan ketersediaan mineral.Kata kunci : Alga Ulva lactuca, Antimikroba, Maserasi, Fraksi, Difusi Agar

Abstract

ABSTRACTLactic Acid Bacteria (LAB) are a group of Gram-positive bacteria that produce lactic acid as the major metabolic end product. They are cocci or rods, nonsporulating and are anaerobic or facultative anaerobes bacteria. Most LABs are probiotics that are known to have good benefits to health, such as inhibiting some pathogens. This study was aimed to examine the potential probiotics properties of LAB isolates from Romaine lettuce fermentation which is included  antibacterial activity of isolates. The isolates are spread on MRS agar  supplemented with 1% of CaCO3 and then purified by using streak method to obtain pure isolates. The results showed that there are 4 isolates from Romaine lettuce fermentation which have the potential to inhibits some pathogens.Key words: Lactic acid bacteria, fermentation, Romain lettuce, potential probiotic ABSTRAKBakteri Asam Laktat (BAL) merupakan sekelompok bakteri Gram positif yang menghasilkan asam laktat sebagai produk akhir metabolisme. Berbentuk kokus atau batang, tidak memiliki spora dan bersifat anaerob atau fakultatif anaerob. Sebagian besar BAL merupakan probiotik yang diketahui memiliki manfaat baik bagi kesehatan, seperti memiliki aktivitas antibakteri. Penelitian ini bertujuan untuk menguji kemampuan isolat BAL dari hasil fermentasi selada Romain sebagai probiotik potensial yang mencakup aktivitas antibakteri isolat. Isolat bakteri ditumbuhkan pada media MRS agar  yang ditambahkan 1% CaCO3 kemudian dimurnikan menggunakan metode gores (streak) sehingga diperoleh isolat murni yang kemudian diuji. Hasil penelitian menunjukkan bahwa ke empat isolat yang diperoleh dari hasil fermentasi selada romain yang berpotensi memiliki aktivitas penghambatan bakteri patogenKata kunci: Bakteri asam laktat, fermentasi, selada Romain, probiotik potensial

Abstract

Abelmoschus manihot L. has been used traditionally as folks medicine plant and it containflavonoid and steroid that has health benefit. The objectives of this research were todetermine anti obesity and hypolipidemic activity of flavonoid and steroid extract fromAbelmoschus manihot L. on white male Wistar induced with high content of protein and lipidfood. Anti obesity test were evaluated by the decreasing of rats weight, whereashypolipidemic test were evaluated by the decreasing of total cholesterol levelsphotometrically using Liebermann-Burchard method. The rats were divided into four groups,namely the negative control (distilled water), extract treatment groups (flavonoids andsteroids), and a positive control group (simvastatin). The percentations of Weight reducingand cholesterol level were analyzed by ANOVA, if there were significant in groups, itcontinued with Duncan Multiple Range Test. The results showed that treatment group possesanti obesity activity, whereas flavonoid extract decrease 7.85% body weight of rats andsteroids extract were 13.70% of body weight. Flavonoids showed hypolipidemic effects bydecrease rats cholesterol levels by 86.45% and steroid extract decrease cholesterol levels by72.53%.Keywords : Abelmoschus manihot L., flavonoid extract, steroid extract, anti obesity,hypolipidemic

Abstract

Isolation and identification flavonoid compounds in beluntas leaf (Pluchea indica L.) have beenconducted. Isolation was carried out by using 96% p.a ethanol solvent of maceration extraction for 7days and partition using n-heksana solvent to obtain concentrated beluntas leaf extract. The extractwas purified by thin layer chromatography using eluent n-butanol : asetic acid : water (BAA) (4:1:5).Isolate 3, result from thin layer chromatography contain flavonoid compound. Ultra Violet - Visiblespectra showed that the flavonoid compound was flavonol, with characteristic wavelengths from 250to 280 nm for band II and 350-385 nm for band I.Keywords: beluntas, flavonoid, Thin Layer Chromatography, UV-Vis Spectrophotometer

Abstract

ABSTRACTVitamin C is one of the nutrients that role as antioxidant and effectively overcome the freeradicals that can damage cells or tissue. Fruit is a good source of vitamin C, such as themango fruit. This study aimed to determine the levels of vitamin C in the mango fruit usingUV-Vis spectrophotometry method and Iodimetry method, and compare the results of bothmethods. Samples were identified as ripe mango. In this study, the levels of vitamin C wereanalyzed by using UV-Vis spectrophotometry and Iodimetry. The results of the analysis ofvitamin C in the mango dodol by using UV-Vis spectrophotometry is 15.88 g/100g, whereasthe content of vitamin C by using Iodimetry was 3.5 g/100g. From these results, showed thelevels of vitamin c by using the spektrofotometry uv-vis method is higher than the iodimetrymethod.Keywords: Vitamin C, mango, UV-Vis Spectrophotometry, Iodimetry.

Abstract

The objective of this research was to determine the species or genus of mercury resistant bacteria culture from mercury polluted waters. Microbiology identification of culture S3.2.2 was done with morphology, physiology and biochemistry test. Molecular analysis was done by amplifying 16S rRNA gene using PCR method. Universal Bact- F1 forward/Uni-B1 Reverse was using as primer. Amplification result was electrophorize with agarose 1,5%, furthermore sequencing was done against 16SrRNA gene. The result shows that culture S3.2.2 were negative grams bacteria, rod shaped, nonmotil, catalase (+), citrat (+), H2S (-), fermentase (+), indol (+) and lisin (+). Blast analysis against nucleotide sequence of 16S rRNA gene possesses 99% similarities with Klebsiella pneumoniae.
