Abstract

?-mangostin (?-MG) is a metabolite of xanthones isolated from fruit of Garcinia mangostana Linn pericarps. Skin cancer is one of the most common malignancies affecting many people. The development of skin cancer is mainly due to intermittent or long-term exposure to ultraviolet (UV) radiation resulting in reduced cell-mediated immune response, production of reactive oxygen species (ROS), and DNA changes. Increased levels of ROS can induce the production of matrix metalloproteinases (MMPs). ?-MG showing potential as an anti-metastatic agent with reduced the expression of MMP-1. This study aims to determine the affinity and stability profiles of ?-MG as anti-skin cancer by applying the In-Silico method. Molecular simulation of ?-MG was successfully docked to the MMP-1. ?-MG showed stable results after 100 ns molecular dynamics simulation based on the root mean square deviation (RMSD) and the root mean squared fluctuation (RMSF). The binding energies of the xanthone derivatives were calculated using the MM/PBSA method.

Abstract

HIV/AIDS merupakan penyakit menular yang hingga kini masih menjadi masalah kesehatan masyarakat global dan nasional. Kelompok usia remaja, khususnya siswa Sekolah Menengah Kejuruan (SMK), berada pada fase perkembangan yang rentan terhadap perilaku berisiko akibat pengetahuan yang terbatas dan pengaruh lingkungan sosial. Di sisi lain, stigma dan diskriminasi terhadap Orang dengan HIV/AIDS (ODHA) masih menjadi hambatan serius dalam upaya pencegahan dan penanganan kasus. Kegiatan ini bertujuan untuk meningkatkan literasi kesehatan siswa mengenai HIV/AIDS serta membangun sikap empatik dan inklusif terhadap ODHA di SMK Kesehatan Mandonga. Kegiatan dilaksanakan melalui metode ceramah, diskusi, tanya jawab, serta pembagian leaflet edukatif. Hasil kegiatan menunjukkan adanya peningkatan pemahaman dan kesadaran siswa mengenai HIV/AIDS, yang ditunjukkan melalui partisipasi aktif dalam diskusi dan kemampuan menjelaskan kembali materi dengan benar. Kesimpulan dari kegiatan ini adalah peningkatan literasi kesehatan remaja dan pembentukan perilaku hidup sehat dalam membangun generasi muda yang sehat, peduli, dan bebas stigma terhadap HIV/AIDS di lingkungan sekolah.

Abstract

Penggunaan obat yang tidak rasional masih menjadi permasalahan di kalangan remaja, termasuk siswa sekolah kejuruan kesehatan, yang mulai melakukan swamedikasi tanpa pemahaman memadai. Kurangnya literasi obat berpotensi menimbulkan efek samping, kesalahan dosis hingga penyalahgunaan obat. Program DAGUSIBU (Dapatkan, Gunakan, Simpan dan Buang Obat) sebagai materi utama edukasi disampaikan melalui integrasi teknologi digital. Tujuan kegiatan ini adalah membentuk siswa yang tidak hanya memiliki pemahaman yang benar tentang penggunaan obat, tetapi juga meningkatkan keterampilan digital sehingga mampu beradaptasi dengan perkembangan teknologi di bidang kesehatan. Metode pelaksanaan dilakukan dalam tiga sesi: (1) pemaparan materi Dagusibu melalui video edukasi yang dikembangkan menggunakan Canva, LARAS, dan ChatGPT; (2) sesi praktik dan diskusi interaktif mengenai cara mendapatkan, menggunakan, menyimpan, dan membuang obat secara benar; serta (3) sesi tanya jawab. Hasil kegiatan menunjukkan peningkatan antusiasme dan pemahaman siswa terkait prinsip DAGUSIBU. Media digital terbukti membantu siswa menyerap materi dengan lebih baik melalui visualisasi dan alur penyampaian yang lebih menarik dibandingkan metode konvensional. Kesimpulannya, pemanfaatan media digital dalam edukasi DAGUSIBU efektif meningkatkan literasi obat siswa SMK Kesehatan Mandonga.

Abstract

Forest honey is a natural product derived from the nectar of wild plants and is known to contain various bioactive compounds, such as flavonoids and phenolics, which contribute to its antioxidant and anti-inflammatory potential. Buton Regency in Southeast Sulawesi is recognized as a producer of forest honey; however, scientific studies on its biological activities remain limited. This study aimed to evaluate the free radical scavenging activity and anti-inflammatory effects of Buton forest honey through both in vitro and in vivo approaches. Antioxidant activity was assessed using DPPH and ABTS assays, while anti-inflammatory evaluation involved inhibition of BSA protein denaturation and a mouse ear edema model induced by xylene. The honey exhibited IC₅₀ values of 71.69 ± 2.73 mg/L (DPPH) and 49.23 ± 4.24 mg/L (ABTS). Its protein denaturation inhibition showed an IC₅₀ of 14.17 ± 3.18 mg/L. In vivo testing demonstrated that high-dose honey significantly reduced ear thickness in mice, with a percentage of inhibition comparable to diclofenac sodium. Statistical analysis revealed significant differences (p < 0.05) across most parameters. These findings indicate that Buton forest honey holds promise as a natural antioxidant and anti-inflammatory agent.

Abstract

Antibiotik merupakan obat yang digunakan untuk mengatasi penyakit infeksi yang disebabkan oleh bakteri. Namun, di masyarakat masih sering terjadi penyalahgunaan antibiotik seperti pemakaian tanpa indikasi medis yang sesuai, sehingga berpotensi meningkatkan kasus resistensi. Pengabdian ini bertujuan untuk meningkatkan pemahaman masyarakat mengenai penggunaan antibiotik secara rasional. Kegiatan dilaksanakan di Kelurahan Gunung Jati, Provinsi Sulawesi Tenggara, melalui metode sosialisasi disertai pretest dan posttest. Data dianalisis berdasarkan nilai jawaban benar (skor 1) dan salah (skor 0). Hasil pretest menunjukkan tingkat pemahaman masyarakat tergolong rendah, yaitu sebesar 72,7% dengan 15 responden (68,2%) berada pada kategori kurang. Setelah dilakukan sosialisasi, terjadi peningkatan pemahaman yang signifikan, ditunjukkan oleh hasil posttest sebesar 95,5%, dengan 14 responden (63,7%) berada pada kategori baik. Peningkatan ini mencerminkan efektivitas metode sosialisasi yang diterapkan kepada masyarakat. Hasil ini juga mengindikasikan bahwa edukasi langsung kepada masyarakat mampu meningkatkan kesadaran dan pengetahuan terkait penggunaan antibiotik yang tepat.

Abstract

Pogostemon cablin (Patchouli), a medicinal plant traditionally used in Indonesia, is gaining scientific attention for its potential pharmacological activities. This study aimed to investigate the anti-inflammatory and anticancer effects of P. cablin ethanolic leaf extract specifically sourced from North Konawe, Southeast Sulawesi. To evaluate its anti-inflammatory activity, two in vitro models were used: the protein denaturation inhibition assay, simulating inflammatory protein response, and the human red blood cell (HRBC) membrane stabilization assay, which mimics lysosomal membrane stability in inflamed tissues. The extract showed significant, dose-dependent inhibition of protein denaturation, with an IC50 value of 62.98 µg/mL. In the HRBC assay, the extract demonstrated membrane stabilization activity with a maximum inhibition of 64.24% at the highest tested concentration (100 µg/mL). The cytotoxic potential was assessed using the MTT assay on MCF-7 hormone-responsive breast cancer cells. The extract exhibited potent anti-proliferative activity, with an IC50 value of 91.56 ± 1.31 µg/mL, indicating its effectiveness in inhibiting breast cancer cell growth. These findings highlight P. cablin from North Konawe as a promising natural source of anti-inflammatory and anticancer agents, with the potential to contribute to the development of plant-based therapeutics. However, this study is limited to in vitro analyses; further investigations are needed to isolate active compounds and confirm efficacy through in vivo and mechanistic studies.

Abstract

Background: Hypertension in the elderly increases the risk of stroke, heart disease, and kidney failure, while adherence to pharmacological therapy remains a major issue requiring attention. Objective: This study aimed to analyze factors associated with antihypertensive medication adherence among elderly patients in the working area of the BLUD UPTD Benu-Benua Community Health Center. Methods: A cross-sectional design was employed involving a total of 67 hypertensive elderly respondents, determined using the Slovin formula (margin of error 10%) from a total population of 207 elderly individuals and selected through random sampling. Data were collected from November to December 2024 using a structured questionnaire that had been tested for validity and reliability (α = 0.685–0.922). Data analysis was conducted bivariately using the Chi-Square test with continuity correction and multivariately using binary logistic regression. Results: The results showed that medication adherence was higher among elderly individuals with good knowledge (76.9%) compared to those with poor knowledge (29.3%; p = 0.000), positive attitude (80.6%) compared to negative attitude (19.4%; p = 0.000), good motivation (63.6%) compared to low motivation (32.4%; p = 0.020), good family support (61.0%) compared to poor support (26.9%; p = 0.014), and good health worker support (64.1%) compared to poor support (25.0%; p = 0.004). Multivariate analysis showed that only the attitude variable significantly influenced medication adherence (p = 0.009 < 0.05; OR = 7.881; 95% CI = 1.678–37.021). Elderly individuals with positive attitudes were 7.881 times more likely to adhere to medication compared to those with negative attitudes. Attitude was the most dominant factor influencing antihypertensive medication adherence. Conclusion: These findings highlight the importance of educational interventions and family support to improve treatment adherence among elderly individuals with hypertension.

Abstract

Resistensi antibiotik merupakan masalah kesehatan global yang terus meningkat akibat penggunaan antibiotik yang tidak rasional di masyarakat. Kurangnya pengetahuan mengenai indikasi, dosis, dan durasi penggunaan antibiotik menjadi salah satu faktor utama terjadinya resistensi. Kegiatan pengabdian kepada masyarakat ini bertujuan untuk meningkatkan pengetahuan dan kesadaran masyarakat tentang penggunaan antibiotik yang tepat melalui edukasi DAGUSIBU (Dapatkan, Gunakan, Simpan, dan Buang obat dengan benar). Kegiatan dilaksanakan secara luring pada 20 November 2025 di Kelurahan Puday, Kecamatan Abeli, Kota Kendari, Sulawesi Tenggara, bekerja sama dengan Puskesmas Abeli dan mahasiswa Program Studi Profesi Apoteker Universitas Halu Oleo. Metode pelaksanaan meliputi penyuluhan interaktif, pembagian leaflet edukatif, sesi tanya jawab, serta evaluasi menggunakan pre-test dan post-test. Peserta kegiatan berjumlah 39 orang masyarakat. Hasil evaluasi menunjukkan adanya peningkatan pengetahuan yang signifikan setelah pemberian edukasi, khususnya terkait pemahaman indikasi penggunaan antibiotik, kewajiban menghabiskan antibiotik, serta bahaya penggunaan antibiotik tanpa resep dokter. Kegiatan ini juga didukung dengan pemeriksaan kesehatan gratis untuk meningkatkan partisipasi masyarakat. Kesimpulan dari kegiatan ini adalah bahwa edukasi DAGUSIBU efektif dalam meningkatkan pengetahuan dan kesadaran masyarakat mengenai penggunaan antibiotik yang rasional, sehingga berpotensi menurunkan risiko terjadinya resistensi antibiotik di masyarakat.

Abstract

Pneumonia is one of the leading causes of morbidity and mortality worldwide. The irrational use of antibiotics in pneumonia management can reduce therapeutic effectiveness and increase the risk of antimicrobial resistance. This study aimed to evaluate the rationality of antibiotic use in pneumonia patients at Hospital X using the Gyssens method. This research employed a descriptive observational design with a retrospective approach, utilizing medical record data of adult inpatients diagnosed with pneumonia during the period of January–December 2024. A total of 86 patient records meeting the inclusion criteria were analyzed. Evaluation was conducted using the Gyssens method by comparing the appropriateness of therapy with the Guidelines for the Management of Adult Pneumonia issued by the Indonesian Ministry of Health (Regulation No. 28 of 2021) and the Infectious Diseases Society of America/American Thoracic Society (IDSA/ATS, 2023) guidelines. The results showed that the most frequently used antibiotics were Ceftriaxone (34.88%), Azithromycin (32.56%), and Levofloxacin (32.56%). The most common duration of therapy was five days (53.9%). Based on the Gyssens categories, rational antibiotic use (category 0) was found in 57 cases (66.28%), while the remaining cases were categorized as I (6.98%), IIa (11.63%), IIIa (9.30%), and IIIb (5.81%). These findings indicate that most antibiotic use at Hospital X was rational; however, continuous monitoring and periodic evaluation through the Antimicrobial Resistance Control Program (PPRA) are still necessary to optimize therapeutic effectiveness and prevent antibiotic resistance.

Abstract

Forest honey is a natural product produced by wild bees that gather nectar from various types of flowers, creating a unique chemical composition influenced by the biodiversity of the surrounding environment. This honey contains bioactive compounds such as phenolics, flavonoids, terpenoids, and volatile substances that contribute to its pharmacological properties, particularly as an anti-inflammatory agent. This study aimed to identify bioactive compounds in forest honey from East Kolaka using Gas Chromatography-Mass Spectrometry (GC-MS) and to evaluate its anti-inflammatory activity through in vitro assays, including red blood cell (RBC) membrane stabilization, protein denaturation inhibition, and nitric oxide (NO) production inhibition. GC-MS analysis revealed several volatile compounds with antioxidant, antimicrobial, and anti-inflammatory properties. The RBC membrane stabilization assay showed increased membrane stability as honey concentration increased, reaching 92.55% at 100 mg/L compared to 97.00% for sodium diclofenac. In the protein denaturation assay, East Kolaka forest honey had an IC?? value of 12.70?±?0.02?mg/L, categorized as moderate, while the NO inhibition assay recorded an IC?? of 2.97?±?1.34?mg/L, indicating strong activity, approaching that of vitamin C (1.07?±?0.39?mg/L). Statistical tests showed significant differences (p < 0.05) in all tested parameters compared to positive controls. These results support the potential of East Kolaka forest honey as a natural anti-inflammatory agent through mechanisms involving membrane stabilization, protein protection, and nitric oxide suppression.
