Abstract

Abstract: Mercury-resistant bacteria can be found in the oral cavity, especially in dental plaques which are exposed to mercury in amalgam. Albeit, not all mercury resistant bacteria must be resistant to antibiotics. This study aimed to determine the level of the resistance of Escherichia coli in mercury and to determine whether the mercury-resistant bacteria have become resistant to the antibiotic ciprofloxacin. This study used a descriptive exploratory method with samples of Escherichia coli bacteria, mercury, and antibiotics that were available in the Laboratory of Pharmaceutical Microbiology University of Sam Ratulangi Manado. The bacterium E. coli were grown in four concentrations of mercury. The results showed that E. coli was resistant to mercury. However, in three repetitions of antibiotic it was found that E. coli was still sensitive to ciprofloxacin. Based on the results, it is advisable to do a similar study in groups using the same antibiotic but with different treatments.Keywords: mercury, bacteria, resistant, ciprofloxacin antibioticAbstrak: Bakteri resisten merkuri bisa ditemukan di dalam rongga mulut pada plak gigi yang terpapar merkuri (amalgam). Bakteri yang resisten merkuri tidak harus resisten terhadap antibiotic. Penelitian ini bertujuan untuk mengetahui tingkat resisensi bakteri Escherichia coli (E.coli) terhadap merkuri dan untuk mengetahui apakah bakteri yang resisten merkuri ini juga resisten terhadap antibiotik siprofloksasin. Penelitian ini menggunakan metode deskriptif eksploratif dengan sampel E.coli, merkuri, dan antibiotik yang tersedia di Laboratorium Mikrobiologi Farmasi Universitas Sam Ratulangi Manado. Hasil penelitian memperlihatkan bahwa E. coli yang ditanam pada empat konsentrasi merkuri resisten terhadap merkuri. Dari hasil uji resisten antibiotik dengan tiga kali pengulangan didapatkan bahwa E. coli yang resisten merkuri ini masih sensitif terhadap siprofloksasin. Berdasarkan hasil penelitian disarankan untuk dilakukan penelitian yang serupa secara berkelompok dengan menggunakan antibiotik yang sama tetapi dengan perlakuan yang berbeda.Kata kunci: merkuri, bakteri, resisten, antibiotik siprofloksasin

Abstract

Abstract: Betel fruit contains saponins, tannins, alkaloids, flavonoids, and steroids which are antibacterial compounds. Alkaloids are one of the secondary metabolites that are found in nature and have physiological activity. This study aims to determine the presence of alkaloid content in betel fruit and to determine the inhibitory activity of betel fruit extract against Staphylococcus epidermidis bacteria. This study is using the disc method in the antibacterial test, TLC method, color reaction and UV-Vis spectrophotometry to determine the presence of alkaloids in betel fruit. The results showed that betel extract contained alkaloids, as evidenced by the presence of orange stains on TLC with chloroform: methanol (1: 4) eluent sprayed with Dragendrof reagents. UV-Vis spectrophotometer analysis results, the alkaloids are at a maximum wavelength of 282 nm.The results of the antibacterial activity test of betel extract against Staphylococcus epidermidis shows that the concentration of 10% and 20% had a strong antibacterial activity with an average inhibition zone of 12.8 ± 1.40 mm and 15.03 ± 0.723 mm and a concentration of 40% had Antibacterial activity is very strong with an average inhibition zone of 21.53 ± 1.530 mm. In conclusion, betel fruit extract contains alkaloid compounds and has antibacterial activity with strong to very strong categories.Keywords: Betel Fruit (Piper betle L), Alkaloids, Antibacterial, Staphylococcus epidermidis.  Abstrak: Buah sirih memiliki kandungan saponin, tanin, alkaloid, flavonoid, dan steroid yang merupakan senyawa antibakteri. Alkaloid merupakan salah satu metabolit sekunder yang banyak ditemukan di alam dan mempunyai aktivitas fisiologis. Penelitian ini bertujuan untuk mengetahui adanya kandungan alkaloid pada buah sirih dan untuk mengetahui adanya aktivitas daya hambat dari ekstrak buah sirih terhadap bakteri Staphylococcus epidermidis. Penelitian ini menggunakan metode cakram pada uji antibakteri, metode KLT, reaksi warna dan Spektrofotometri UV-Vis untuk mengetahui adanya alkaloid pada buah sirih. Hasil penelitian menunjukkan bahwa ekstrak buah sirih memiliki kandungan alkaloid, terbukti dengan adanya noda berwarna jingga pada KLT dengan eluen kloroform:methanol (1:4) yang  disemprotkan pereaksi Dragendrof. Hasil analisis Spektrofotometer UV-Vis, alkaloid tersebut berada pada panjang gelombang maksimum 282 nm. Hasil uji aktivitas antibakteri ekstrak buah sirih terhadap bakteri Staphylococcus epidermidis diperoleh bahwa konsentrasi 10% dan 20% memiliki aktivitas antibakteri kuat dengan zona hambat rata-rata sebesar 12,8±1,40 mm dan 15,03±0,723 mm serta konsentrasi 40% memiliki aktivitas antibakteri sangat kuat dengan zona hambat rata-rata sebesar 21,53±1,530 mm. Sebagai kesimpulan ekstrak buah sirih mengandung senyawa alkaloid dan memiliki aktifitas antibakteri dengan kategori kuat sampai sangat kuat.Kata kunci : Buah Sirih (Piper betle L), Alkaloid, Antibakteri, Staphylococcus epidermidis

Abstract

Abstract: Betel leaf (Piper betle L) contains alkaloid compounds and has antibacterial and antiseptic functions. The purpose of this research was to isolated alkaloid compounds also to tested the effectiveness of inhibition of the betel leaf extract against the Staphylococcus epidermidis bacterial growth. Simplicia betel leaf was extracted used 96% of ethanol solvent. Antibacterial activity tested of betel leaf extract with a concentration variation of 25%, 20%, 15%, 10% and 5% using the liquid dilution method. The results of identification of alkaloid isolates used UV-Vis Spectrophotometry method can be known alkaloid compounds contained in betel leaves include alkaloids with indole base framework that was absorption at 262 nm and 274 nm wavelengths. The results of the effectiveness of betel leaf on Bacteria Staphylococcus epidermidis used the liquid dilution method showed that betel leaf extract with Optical density values at concentrations of 25%, 20%, 15% and 10% before and after incubation decreased respectively by -0.347, -0.304, -0.192 and -0.104, while at a concentration of 5% there was increase in Optical density values of 0.162. From the results of the research, it can be concluded that the ethanol extract of betel leaf contains an alkaloid with an indole base framework and was inhibitory activity against Staphylococcus epidermidis bacteria with a MIC at a concentration of 10%.Keywords: betel leaf, Staphylococcus epidermidis, alkaloid compound, liquid dilution.  Abstrak: Daun Sirih (Piper betle L) mengandung senyawa alkaloid dan memiliki fungsi sebagai antibakteri dan antiseptik. Penelitian ini bertujuan untuk mengisolasi senyawa alkaloid dan menguji efektivitas penghambatan dari ekstrak daun sirih hijau terhadap pertumbuhan bakteri Staphylococcus epidermidis. Simplisia daun sirih hijau diekstraksi dengan menggunakan pelarut etanol 96%. Uji aktivitas antibakteri ekstrak daun sirih dengan variasi konsentrasi 25%, 20%, 15%, 10% dan 5% menggunakan metode dilusi cair. Hasil identifikasi terhadap isolat alkaloid menggunakan metode Spektrofotometri UV-Vis dapat diketahui senyawa alkaloid yang terkandung dalam daun sirih termasuk alkaloid dengan kerangka dasar Indol yang mempunyai serapan pada panjang gelombang 262 nm dan 274 nm. Hasil uji efektivitas daun sirih terhadap bakteri Staphylococcus epidermidis dengan metode dilusi cair menunjukan bahwa ekstrak daun sirih dengan nilai densitas optik pada konsentrasi 25%, 20%, 15% dan 10% sebelum dan sesudah inkubasi mengalami penurunan berturut-turut sebesar -0.347, -0.304, -0.192 dan -0.104, sedangkan pada konsentrasi 5% mengalami kenaikan nilai densitas optik sebesar 0.162. Dari hasil penelitian yang diperoleh, dapat disimpulkan bahwa ekstrak etanol daun sirih mengandung alkaloid dengan kerangka dasar indol dan memiliki aktivitas penghambatan terhadap bakteri Staphylococcus epidermidis dengan KHM pada konsentrasi 10%.Kata Kunci: daun sirih, Staphylococcus epidermidis, senyawa alkaloid, dilusi cair.

Abstract

ABSTRACTPalm horse weevil contains medicinal properties that are useful in the process of woundhealing. The purpose of this study is to to examine formulate an ointment of palm horse(Ipomoea pes-caprae (L) Sweet) weevil extract and to test the ointment to heal open woundon the skin of rabbits. This study is experimental descriptive in the laboratory. Palm horseweevil extracts used in the manufactured of ointment formulation 10%, 15 % and 20 %. Theresults showed that the palm horse weevil formulation meets the ointment test requirementaccording, making ointment formulation using cassava tuber extract used as 5 rabbits.Divided into 5 treatment gropus, namely, test the ointment treatment group, negative controlgroup (basic ointment) and positive control group (chloramphenicol ointment). Observed thehealing effects for 7 days. The results showed that from day-1 to day-7 observations burnssuffered diminution in diameter. It was concluded that concentration of 10 % cassava extractontment has healing effect and increasing the concentration of 15 % and 20 % containedshowed more effective and speed up the healing process.Keywords : Extract leaf palm horse , ointment, wound healing, rabbit.

Abstract

This study aimed at finding out the effectivity of Salacca (Salacca zalacca [Gaertn]Voss) rind extract on blood sugar decrease of white male wistar (Rattus norvegicus L.)induced with sucrose. Completely randomized design method was used in this experiment.The subject of the experiment were 15 white male wistar which were divided into 3 (three)groups namely negative control group, treatment group with the dose of 150 mg extract/kgbody weight, and positive control group using glibenclamide 0.45 mg/kg body weight. Dataobtained from the examination of the fasting blood sugar were 30 minutes after inductionwith sucrose and at minute 15, 30, 60, and 120 after treatment was given. The data isprocessed using ANOVA and LSD test to compare between the negative control group,treatment, and a positive control. The analysis revealed a significant difference betweennegative control and treatment, negative control and positive control, but did not show asignificant difference between treatment groups with the positive control. The conclusion ofthis study is Salacca rind extract has an effect on blood sugar levels decrease of white malewistar induced with sucrose.Keywords: Salacca zalacca (Gaertn.) Abstract

The aims of this research was to determine the effect of ethanol extract paku sisik nagaleaves against lipid peroxidation in male wistar liver induced by CCl4. Method of thisresearch used laboratory experiment using 9 white male wistar were divided into 3 treatmentgroups. All groups were given treatment during ten days. The first group (negative control)was given distilled water ad libitum, the second group was given ethanol extract of paku sisiknaga leaves 97.02 mg/kg body weight per oral and the third group (positive control) wasgiven Curcuma 45 mg/kg body weight per oral. On the tenth day, after two hours oftreatment all groups were given CCl4 1mL/kg body weight intraperitonial, eighteen hoursafter that rats in all groups were dissected and taken his liver. Then rat liver analyzed byTBARS method (Thiobarbiturate Acid Reactive Subtance) to measure the levels of MDA(Malondialdehyde) which is an indicator of lipid peroxidation in the rat liver. Data wereanalyzed by SPSS ver.20, significant difference between treatments was tested by one-wayANOVA. The results showed a significant difference with 95% confidence level (α = 0.05)with a mean value of MDA 3,828 μM negative control group, group of ethanol extract ofpaku sisik naga leaf was 2,666 μM and 2,211 μM for positive control group. Ethanol extractof paku sisik naga leaves with dose 97.02 mg/kg body weight can prevent the process of lipidperoxidation in rat liver.Keywords: Drymoglossum piloselloides L. Abstract

This paper reports the identification and determination of benzoate preservative level intomato sauce on the market in Manado. Extraction of the benzoate with diethylether anddetermination of the preservative (benzoic acid) was determined using UV-Visspectrophotometer. Qualitatively, it was found that the sauce samples contained benzoate.Quantitatively, it was found that four samples of tomato sauce used benzoate as apreservative. two of them used benzoic above the threshold set uot in Decree No. Permenkes.722/Menkes/Per/IX/88.Keywords: benzoate, ketchup, Manado, spectrophotometer UV- Vis.

Abstract

ABSTRACTThis research aims to determine the phytochemical compounds and testing the analgesiceffects of ethanol extract of kucing-kucingan plant (Acalypha indica Linn.). Phytochemicaltesting conducted according to Harbone methods and analgesic activity of rats reaction timeis determined based on the thermal response after giving the ethanol extract of A indica L.This study used 20 female Wistar rats were divided into a treatment group was given 0,035mg / kg extract of leaves, stems and roots, negative control receive aquadest and positivecontrol using aspirin. Reaction time data were analyzed using One Way ANOVA followed byDuncan's test multipe Range Test. The results showed that the ethanol extract of A.indica L.contains alkaloids and saponins in the extract while the third type of flavonoid and tanninfound only in leaf extracts. Testing the analgesic activity of ethanol extract A. indica L.showed that the leaf extract gave the highest reaction time of 3.16 seconds, and stem and rootextracts followed by 1.72 seconds and 2.85 seconds. Abstract

ABSTRACTIodine deficiency disorder can be prevented by consumption of salt which contains iodine into the body. Based on Indonesian National Standard (SNI), concentration of Potassium iodide(KIO3) were 30-80 ppm. The objectives of this research were to identify and determineconcentration of potassium iodide in salt which marketed in Bitung region. Samples were 9branded salts and 1 sample unbranded. Identification was done by Potassium cation test andIodide anion test. Determination of Potassium iodide were done using spectrophotometerUV-Vis. The results shows that 9 samples contains Potassium iodide (Branded salts), and 1sample did not contains Potassium iodide (Unbranded salt). Abstract

ABSTRACTLeaves of Langsat parasite (Dendrophthoe petandra (L.) Miq.) contains flavonoids. Theobjective of this research was to examine hepatoprotector effect of leave extract from langsatparasite on white male rats induced with CCl4. Treatment was conducted for 8 days to 13 rats,which group I (P1) were administered with EDBL 35mg/KgBB,group II (P2) wereadministered with EDBL 70mg/KgBB,group III (K+) were administered withCurcuma81mg/KgBB orally. Group IV (K-) and V (KP) were administered with aquadestad libitum.At 8th day, all groups except (KP) were induced with CCl4 1mL/KgBB intraperitoneally.Eighteen hours after inducing time, all rats in every group were sacrafied to take its liver.Rats liver were analyzed using TBARS (Thiobarbituric Acid Reactive Subtance) method tocalculate MDA (Malondialdehid) level that were the end product of lipid peroxidation in ratsliver.MDA content of P1 were 2,715μM, P2 were 2,080μM, K+ were 1,708μM, and K- were3,413μM.
