Abstract

The research of nitric oxide scavenging by hemoglobin and blood pressure in preeclampsia had been done. Aims of this research were to measure methemoglobin level as an index nitric oxide and its correlation to blood pressure. Sixty pregnant were women involved in this study. Thirty subject were normal pregnancy and preeclampsia. Preeclampsia group were consist of 18 mild preeclampsia and 12 severe preeclampsia. The methods, to measure methemoglobin based on Evelyn and Malloy methods with spectrophotometer. Meanwhile, Blood pressure is measured with auscultation methods. The result showed that methemoglobin level, systolic and diastolic pressure preeclampsia higher than normal significantly. There is a positive correlation between methemoglobin level and blood pressure in mild preeclampsia. In severe preeclampsia, there is negative correlation between methemoglobin level and blood pressure. Beside that, correlation between methemoglobin level and sistolic pressure has positive correlation and negative correlation found between methemoglobin level and diastolic pressure in normal pregnancy. This study conclude that nitrite oxide scavenging activity by hemoglobin have correlation to bloodpressure change in normal pregnancy and preeclampsia.Telah dilakukan penelitian tentang aktivitas scavenging mtrit oksida oleh hemoglobin dan tekanan darah. Penelitian ini bertujuan untuk mengukur kadar methemoglobin sebagai parameter nitrit oksida dan korelasinya dengan tekanan darah. Sebanyak 60 orang wanita hamil terlibat dalam penelitian ini. Pada subjek tersebut, 30 orang tergolong hamil normal dan 30 orang didignosis preeklampsia. Kelompok preeklampsia terdiri atas 18 orang preeklampsia ringan dan 12 orang preeklampsia berat. Metode pengukuran kadar methemoglobin digunakan metode Evelyn dan Malloy dan pengukuran tekanan darah dengan metode auskultasi. Hasil penelitian menunjukkan bahwa kadar methemoglobin, tekanan sistolik dan diastolik pada preeklampsia (ringan dan berat) lebih tinggi bermakna daripada tanpa preeklampsia. Terdapat korelasi positif antara kadar methemoglobin dengan tekanan sistolik dan diastolik preeklampsia ringan. Pada preeklampsia berat korelasi kadar methemoglobin dan tekanan sistolik dan diastolik bernilai negatif. Pada kehamilan normal korelasi kadar methemoglobin bernilai positif dengan tekanan sistolik, sedangkan terhadap tekanan diastolik, berkorelasi negatif. Kesimpulanpenelitiani ini adalah aktivitas scavengingmint oksid olehhemoglobin mempunyai korelasi terhadap perubahan tekanan darah pada kehamilan normal dan preeklampsia.

Abstract

The three-dimensional (3D) structure of proteins is necessary to understand the properties and functions of proteins. Determining protein structure by laboratory equipment is quite complicated and expensive. An alternative method to predict the 3D structure of proteins in the in silico method. One of the in silico methods is homology modeling. Homology modeling is done using the SWISS-MODEL server. Proteins that will be modeled in the 3D structure are proteins that do not yet have a structure in the RCSB PDB database. Protein sequences were obtained from the UniProt database with code A0A0B6VWS2. The results showed that there were two models selected, namely model-1 with the PDB code template 1q0e and model-2 with the PDB code template 3gtv. The results of sequence alignment and model visualization show that model-1 and model-2 are identical. The evaluation and assessment of model-1 on the Ramachandran Plot have a Favored area of ??97.36%, a MolProbity score of 0.79, and a QMEAN value is 1.13. Model-1 is a good 3D protein structure model.

Abstract

Cadmium (Cd) and mercury (Hg) are toxic metals that affect human organs function, including liver and kidney. This toxic activity is because the heavy metal could induce oxidative stress and interfere antioxidant activities, including catalase (CAT). The present study was aims to evaluate the effect of Cd and Hg to liver and kidney CAT kinetic parameters in vitro. In this experiment, liver and kidney were taken from male rats (Rattus novergicus). Sample the homogenized and divided into three groups with; T0 served as control which contains liver or kidney homogenate + H2O2, T1 which contains liver or kidney homogenate + H2O2 + 0.03 mg/L CdSO4; and T2 which contains liver or kidney homogenate + H2O2 + 1 mg/L Hg. Solutions then incubated at 37ºC for 1 hour and then was prepared for CAT activity measurement. The CAT activity was measured using spectrophotometer at 240 nm. For measuring the kinetic parameters, different concentration of H2O2 were used. The kinetics parameters (Km and Vmax) were calculated using Lineweaver-Burk plot. The results shows that Cd and Hg could decrease the affinity of CAT-H2O2 complex which expressed by the higher Km and Vmax values. Also from the results, Cd has better activity to decreased the affinity of CAT-H2O2 complex than Hg. From this results, it can be concluded that Cd and Hg treatments could inhibit CAT activity in liver and kidney in vitro.

Abstract

Corona Virus Disease (COVID-19) is becoming a global pandemic. Indonesia, especially South Kalimantan had recorded increasing cases with a high fatality rate of 3.7%. Information about factors related to outcomes based on clinical and laboratory features in Indonesia is still limited. Identification of the risk is crucial to determine optimal management and reducing mortality. This retrospective study enrolled 455 adults COVID-19 patients, and data were extracted from medical records of Ulin General Hospital Banjarmasin. The latter is COVID-19 referral hospital in South Kalimantan between March-November 2020. Demographic data, comorbidities, and laboratory were all collected. Data were compared between survivors and non-survivors. Fisher’s exact test and chi-square were used to compare categorical variables. The Mann_Whitney U test was used to compare continuous variables. Analysis was continued by multivariate logistic regression then receiver operating characteristic (ROC) curve to determine cut-off value. The multivariate analysis showed that number of comorbidities [odds ratio (OR) 1,339 (95% confidence interval (CI): 1,064-1,685, P = 0,013) was a significant risk factor to the outcome. In laboratory, lactate dehydrogenase (LDH) [OR: 1.001, 95% CI: 1,000-1.002, P = 0.001], Ferritin (OR 1.000, CI: 1,000-1.001, P = 0.013), APTT (OR: 1.045, CI: 1.010-1.082, P = 0.012), and D-dimer (OR: 1.188, CI: 1.064 - 1.327, P = 0.002) were significant predictor factors but only LDH, ferritin, and D- dimer were obtained good AUC 0.731, 0.715, and 0.705, respectively. The cut of the value of LDH was 656.5 U/L, ferritin was 672.18 ng/ml, and D-dimer was 2.28 mg/L. Sensitivity and specificity were 66.7% and 68,0% for LDH, 83,2% and 56,3% for ferritin, and 62,8 and 70,8% for D-dimer. From this research, we revealed that the number of comorbidities was a risk factor for death. Elevated LDH, ferritin, and D-dimer could be good predictive factors for poor outcomes, thereby considering the accelerating management of COVID-19 patients.

Abstract

Fever is increased  temperature  regulation  of  the  body.  In  the  process  is according  indirect  which  increased  of  free  radicals,  as  anion  superoxide  (•O2) and  will  trigger  oxidative  stress  happened.  Oxidative stress will effect in endothelial damaged. A celluler marker of damage the endothelium is increased number of Circulating  Endothelial  Cells (CEC).  The  aim  of  this  research  is  to valuated the influence of watery plant kalakai extract (Stenochlaena palustris)to number  of  Circulating  Endothelial  Cells  in  Marmota  caligata had  been  fever  and to calculated the average of CEC. The research is true experimental study, with Posttest-Only with Control Group Design, with 2 control group and 5 treatments group  of  each  4  Marmota  caligata. The CEC is   measured  by  Hladovec  method. Data  was  analyzed  by  using  Kruskal-Wallis  test  with confidence  rate  at  95  %. The  analyzed  results  got  p  =  0.001  (p  <  0.05)  means there  be  a  significant different  between  treatment  group.  From the  result, can  be  conclude  that  the present of watery  plant kalakai  extract is decreasing CEC in plasma of Marmota caligatafever induced.Key words: Fever, Circulating Endothelial Cells, Stenochlaena palustris

Abstract

Background: Posyandu cadre performance refers to the role of cadres in posyandu activities as inthe Posyandu cadre handbook, explaining the role of posyandu cadres is divided into three stages,namely before posyandu opening days, posyandu opening days and after posyandu opening days. Manystudies report different results on the performance of health cadres related to knowledge and incentives.The purpose of this paper is to analyze the relationship between knowledge and incentives with theperformance of posyandu cadres.Method: The research method used is meta-analysis to assess how big the effect of the knowledge andincentive variables will be on the performance of posyandu cadres.Result: Based on the results of the study of research articles, the combined P-value of 0,000 on theknowledge variable, and the combined p-value of 0,039 on the incentive variable.Conclusion: There is a relationship between knowledge and incentive variables on the performance ofposyandu cadres.

Abstract

Abstract : Interleukin-10 as pro-inflammatory cytokines have a role to protect the damage of severe malaria. Kelakai contain bioactive compounds that have anti-inflammatory activity. The purpose of this study was to determine the potential of the extract kelakai on levels of IL-10 in mice BALB / s infected by P. berghei ANKA. This study is true experimental studies with posttest-only with Control Group Design. The treatment group was divided into 8 groups. Two groups received kelakai extract per oral 10 mg/kgBW and 100 mg/kgBW. Four groups received kelakai extract per oral 10 mg/kgBW and 100 mg/kgBW 3 hours after infection and when the parasitaemia reaches 15-20%. Negative controls do not receive the extract kelakai and parasitic infections. Positive controls get a parasitic infection. Treatment was given for 4 days. Blood samples were taken 24 hours after the last treatment. Levels of IL-10 were measured by sandwich ELISA method. Data were analyzed with Games Howell test, with a confidence level of 95%. There were no significant differences between treatment groups. Extract kelakai Giving oral dose of 10 mg/KgBW and 100 mg/KgBW not significantly increase the levels of interleukin-10. Keywords: Stenochlaena palustris (Burm.f) bedd; Interleukin-10 Abstrak : Interleukin-10 sebagai Sitokin pro-inflamasi memiliki peran melindungi kerusakan pada malaria berat. Kelakai mengandung senyawa bioaktif yang mempunyai aktifitas anti-inflamasi. Tujuan penelitian ini adalah untuk mengetahui potensi ekstrak kelakai terhadap kadar IL-10 pada mencit BALB/s yang diinfeksi P. berghei ANKA. Penelitian ini merupakan studi eksperimental murni dengan Posttest-only with Control Group Design. Kelompok perlakuan dibagi menjadi 8 kelompok. Dua kelompok mendapat ekstrak kelakai per oral 10 mg/kg berat badan dan 100 mg/kg berat badan. Empat kelompok mendapat ekstrak kelakai per oral 10 mg/kg berat badan dan 100 mg/kg berat badan 3 jam setelah infeksi dan pada saat parasitemia mencapai 15-20%. Kontrol negatif tidak mendapat ekstrak kelakai dan infeksi parasit. Kontrol positif mendapat infeksi parasit. Perlakuan diberikan selama 4 hari. Sampel darah diambil 24 jam setelah perlakuan terakhir. Kadar Il-10 diukur dengan ELISA metode sandwich. Data dianalisa dengan tes Games Howell, dengan tingkat kepercayaan 95%. Tidak didapatkan perbedaan bermakna antar kelompok perlakuan. Pemberian Ekstrak kelakai secara oral dosis 10 mg/Kg berat badan dan 100 mg/Kg berat badan tidak signifikan meningkatkan kadar Interleukin-10. Abstract

Background: Mengkudu is a traditional plant with active compound that has antioxidant activity. Diabetes mellitus complication has causative link with AGEs formation that involves free radical. Objective: To know antioxidant activity of mengkudu and the role to inhibit AGEs formation. Methods: This experiment used pre and post test control group design. Antioxidant activity was deter-mined'by reacting the mengkudu juice with 2,4 dinitrophenylhydrazin (DNPH) and analyzed with spectro-photometer a.=390 nm. AGEs absorbancy was measured every 48 hours until 20 days with spectropho-tometer ?=340 nm. Dicarbonyl level was determined with DNPH method that was developed by Uchida and modified by Sadikin.Result: Antioxidant activity of 0.25 g/ml mengkudu juice (Morinda citrifolia) was 125.46±27.79%.Beside that, mengkudu juice can inhibit advanced glycation end products (AGEs) formation.Conclusion: Mengkudu juice can decrease AGEs formation and has antioxidant activity. Key words: antioxidant - mengkudu juice - AGEs - glycosilation

Abstract

The research of an antioxidant activity of Daun Dewa leaf boiled (Gynura pseudochina) and the role as inhibitor of advanced gly cation end products (AGEs) formation in vitro had been done. Aims of this research are to measure an antioxidant activity by Daun Dewa leaf boiled and the role as inhibitor of AGEs formation. The methods, to measured an antioxidant activity by Daun Dewa leaf boiled is reacted with 2,4-dinitrophenylhydrazin (DNPH) and ana¬lyzed with spectrophotometry methods with 1—390 nm. Meanwhile, AGEs com¬pounds is measure every 48 hours during 20 days of research and analyzed with spectrophotometry methods with 1=340 nm Content of dicarbonyl com¬pounds is measured with DNPH methods that developed by Uchida and modi¬fied by Sadikin. The result, showed that an antioxidant activity of Daun Dewa leaf boiled at 25% concentration is 66,34 ± 3,72 %. On the other hand, is able to block rate of AGEs formation.Telah dilakukan penelitian aktifitas antioksidan rebusan daun tanaman Daun Dewa dan perannya sebagai penghambat pembentukan AGEs. Penelitian ini bertujuan menentukan aktifitas antioksidan rebusan daun tanaman Daun Dewa dan perannya sebagai penghambat pembentukan AGEs. Untuk penentuan aktivitas antioksidan, rebusan dauan tanaman Daun Dewa direaksikan dengan 2,4-dinitrophenylhydrazin (DNPH) dan dianalisis secara spektrofotmetri pada 1 = 390nm. Senyawa AGEs diukur setiap 48 jam selama 20 hari dengan menggunakan spektrofotometri pada 1 = 340 nm. Kadar senyawa dikarbonil ditentukan dengan cara metoda DNPH yang dikembangkan Uchida dan dimodifikasi oleh Sadikin. Hasil penelitian menunjukkan bahwa aktifitas antioksidan rebusan daun tanaman Daun Dewa konsentrasi 25% adalah 66,34 ± 3,72%. Selain itu, daun Dewa juga dapat menghambat pembentukan AGEs.
