Abstract

Mekanisme kadmium menginduksi toksisitas pada hepar yaitu dengan menimbulkan stres oksidatif. Stres oksidatif menghambat enzim-enzim yang berperan dalam proses metabolisme glukosa. Penelitian ini bertujuan untuk menganalisis perbedaan kadar glukosa pada homogenat hepar. Penelitian ini merupakan penelitian eksperimental laboratorik yang dilakukan pada 4 kelompok: kelompok P0 yang tidak dipajankan Cd, kelompok P1 dipajankan Cd 0,03 mg/L, kelompok P2 dipajankan Cd 0,3 mg/L, dan  kelompok P3 dipajankan Cd 3 mg/L. Hasil penelitian didapatkan rata-rata kadar glukosa pada kelompok P0 sebesar 21.667 μM, kelompok P1 sebesar 33.278 μM, kelompok P2 sebesar 69.889 μM, dankelompok P3 sebesar 150.667 μM. Melalui uji statistik Kruskal-Wallis didapatkan perbedaan yang bermakna p=0,000 (p<0,05) dan uji pos hoc Mann-Whitney juga menunjukkan perbedaan yang bermakna pada semua kelompok. Dapat disimpulkan bahwa semakin tinggi konsentrasi pemajanan Cd terhadap homogenat hepar maka semakin tinggi pula peningkatan kadar glukosa pada hepar tikus.

Abstract

Hiperglikemia merupakan suatu keadaan meningkatnya kadar glukosa darah melebihi normal. Keadaan hiperglikemia ini memiliki peran terhadap komplikasi diabetes mellitus. Meskipun secara umum osteoporosis tidak digolongkan sebagai komplikasi diabetes, pada pasien diabetes tipe 1 dan 2 terjadi peningkatan resiko terjadinya osteoporosis. Penelitian ini bertujuan untuk mengetahui korelasi antara kadar glukosa darah terhadap kadar kalsium tulang pada tikus putih. Penelitian ini merupakan penelitian eksperimental murni dengan posttes-only with control group design, menggunakan Rancangan Acak Sederhana, terdiri atas 10 kelompok yang masing-masing kelompok terdiri atas 5 ekor tikus putih (Rattus norvegicus), yaitu 1 kelompok kontrol dan 9 kelompok perlakuan yang diinduksi streptozotocin dengan dosis 50 mg/kgBB, yang setiap kelompok dilakukan pengukuran kadar glukosa dan kalsium setiap 3 hari sekali. Kadar kalsium tulang diukur dengan metode permanganometri. Data yang diperoleh diuji korelasi Pearson dengan tingkat kepercayaan 99%. Berdasarkan penelitian dapat disimpulkan bahwa keadaan peningkatan kadar glukosa darah dapat menyebabkan penurunan kadar kalsium tulang.

Abstract

Ethinyl estradiol is an estrogen derivative used as contraceptive or hormone replacement therapy to maintain woman?s sexual function. The aim of this study was to examine ethinyl estradiol antioxidative activity and its potency to inhibit protein oxidation by glycocylation reaction. This study was an in vitro assay and its antioxidative activity by H2O2 scavenging, ?OH scavenging, metal chelating were measured. Activity as inhibitor of Advanced Oxidation Protein Products (AOPP) was carried out by using glycocylation reaction. Three groups (n=6), i.e P0 control (3 ml of serum), P1 (3 ml of serum + glucose 500 mM), and P2 (3 ml of serum + glucose 500 mM + ethinyl estradiol 0,15gr/100mL) were used in this study. The result of this study showed that H2O2 scavenging, ?OH scavenging, metal chelating were 48,889%, 15,139%, 11,538% respectively. For AOPP inhibition, P0 and P1 was different significantly (p 0,05), P0 and P2 was not different significantly (p 0,05), and P1 and P2 was different significantly (p 0,05). It was suggested that antioxidative mechanism from ethinyl estradiol could inhibit AOPP formation generated by glycocylation reaction.

Abstract

Maillard reaction or glycosylation reaction is a reaction between amine group and aldehyde group from glucose to form dicarbonyl compound and Advanced Glycation End Products (AGEs). The formation of AGEs have a role in diabetic complications. This work was conducted study the tyrosine degradation reaction model and the formation of AGEs caused by Maillard reaction. The research design was an experimental study with pre and posttest control group design with followed up posttest in 48 hours to 20 days. Two solutions, A and B were used; solution A was a mixture of BSA 30% + buffer phosphate pH 7 + aquadest, and solution B was BSA 30%+ buffer phosphate pH 7+ aquadest+ glucose. The degradation of tyrosine was measured by its absorbance at l=470 nm and the absorbance of AGEs formation at l=340 nm. Model of AGEs formation in solution A followed the equation of Y=0,0289X ? 0,01847 with correlation coefficient R2=0,9767, where as that in solution B followed equation of Y=0,6552X ? 1,5267 with correlation coefficient R2=0,9463. The tyrosine degradation model in solution A followed equation of Y=-0,1232X + 2,7166 with correlations coefficient R2=0,9175 where as that in solution B followed equation of Y=-0,03552X + 0,7762 with coefficient of correlation R2=0,9175. Negative value was observed in correlation between AGEs absorbance with tyrosine in solution A and B reflecting the formation of AGEs followed by tyrosine degradation

Abstract

The present study was undertaken to evaluate the effect of a topical application of ethanolic extracts of Mauli banana stem on buccal mucosa wounds in rats (Rattus novergicus). Three sets of experiments with 3 groups male rats each consisted of 9 animals were used for studying wound oxidative stress status. Group 1 (P0) as the negative control was left to heal spontaneously; group 2 (P1) as the postive control were treated with alocair topically; and group 3 (P2) as treatment group were treated with ethanloic extracts of Mauli banana stem topically, respectively, 24h after wound creation for 3 days. The oxidative stress status was evaluated by monitoring the SOD, CAT activity, MDA and CC levels. The effect of ethanolic extracts of Mauli banana stem on oxidative stress status revealed significant increased in SOD activity, decreased in MDA levels, and no significant change both in CAT activity and CC levels compared to negative control. These results showed that The ethanolic extracts of Mauli banana stem might affect the oxidative stress status during wound healing process. Abstract

AbstrakPestisida merupakan bahan kimia yang digunakan untuk mengendalikan serangga, jamur, gulma dan lain-lain. Penggunaan pestisida yang tak terkendali menyebabkan keracunan dan berdampak pada ganguan metabolisme. Untuk membuktikan hal tersebut telah diteliti hubungan antara kadar enzim asetilkolinesterase dengan kadar glukosa darah petani yang terpajan pestisida. Rancangan penelitian ini bersifat analitik, dengan pendekatan cross sectional. Subjek penelitian adalah seluruh anggota kelompok tani Kurnia Makmur di Kecamatan Landasan Ulin Utara Banjarbaru dengan menggunakan jumlah sampel minimal sebesar 60 orang. Pemeriksaan sampel kadar enzim asetilkolinesterase dalam darah dan glukosa dalam darah dilakukan dengan bekerjasama dengan Labkesda Kabupaten Banjar dengan No. 001/LKA/C/VII/2016 s/d 60/LKA/C/VII/2016. Data dikumpulkan dari Dinas Kesehatan Banjarbaru tahun 2016. Analisis bivariat digunakan untuk menjelaskan hubungan dua variabel yaitu antara kadar kolinesterase dengan kadar glukosa darah dengan menggunakan uji Spearman dengan derajat kepercayaan 95%. Hasil penelitian didapatkan rerata kadar asetilkolinesterase pada petani berjenis kelamin laki-laki dan perempuan melebihi batas normal (L= 4,6-11,5 U/L; P=3,9-10,8 U/L). Hal ini menandakan bahwa sebanyak 63,34% petani mengalami keracunan pestisida. Sementara itu, rerata kadar glukosa darah petani masih tergolong normal (Gula darah sewaktu „T 125 mg/dL) meskipun pada petani berjenis kelamin wanita ditemukan sebanyak 5% mengalami hiperglikemia. Hasil uji Spearman didapat r =0,049 (p=0,768; p>0,05) yang berarti tidak ada hubungan yang bermakna antara kadar asetilkolinesterase dengan kadar glukosa darah petani yang terpajan pestisida.Kata-kata kunci: Pestisida, kolinesterase, glukosa, diabetesAbstractPesticides are chemicals used to control insects, fungi, weeds and others. Uncontrolled use of pesticides causes poisoning and affects metabolic disorders. To prove the above has been investigated the relationship between the levels of enzyme acetylcholineserase with blood glucose levels of farmers exposed to pesticides. The design of this study is analytic, with cross sectional approach. Research subjects were all members of Kurnia Makmur farmer group in Kecamatan Landasan Ulin Utara Banjarbaru by using minimum sample amount of 60 people. Examination of samples of cholinesterase enzyme levels in blood and glucose in blood was done in collaboration with Labkesda Kabupaten Banjar with no. 001 / LKA / C / VII / 2016 s / d 60 / LKA / C / VII / 2016. Data was collected from Banjarbaru Health Office in 2016. Bivariate analysis was used to explain the relationship between two variables, ie, between cholinesterase and blood glucose level using Spearman test with 95% confidence degree. The result showed that the level of acetylcholinesterase in male and female farmers exceeded the normal limit (L = 4.6-11.5 U / L; P = 3.9-10.8 U / L). This indicates that as many as 63.34% of farmers are poisoned by pesticides. Meanwhile, the average blood glucose level of farmers is still normal (blood sugar at „T 125 mg / dL) although the female-female farmers found as many as 5% had hyperglycemia. Spearman test result obtained r = 0.049 (p = 0.768; p> 0.05) meaning there is no significant relation between level of acetylcholinesterase with blood glucose level of farmer exposed by pesticide.Keyword: pesticide, acetylcholinesterase, glucose, diabettes

Abstract

Pesticide sale circumstances in Paser District through measuring cholinesterase enzymes by the laboratory of the Paser District on the blood of pesticide traders found 37.5% experienced organopospat group pesticide toxicity. This research aim to analyze risk factors related to the level of pesticide toxicity in pesticide traders namely age, nutritional status, educational level, length of work, work period and use of personal protective equipment. The study design used was observational with a cross sectional approach. The number of samples is 42 people taken by simple random sampling. Measurement data using observation and examination sheetcholinesterase levels in the blood of respondents using Tintometer tool brands inScienPro Pest 100. The data were analyzed using Chi-square and multiple logistic regression. Pesticide traders who experienced pesticide toxicity were 42.9%. Result of Chi-Square test shows three variables that have a significant relationship with the incidence of pesticide toxicity. Risk factors for pesticide toxicity were age (p=0.021; OR=6.250; 95% CI=1.511-25.860), length of work (p=0.038; OR=6.769; 95% CI=1.268-36.139) and work period (p=0.043; OR=4.900; 95% CI=1.238-19.399). Multiple logistic regression showed that the most dominant factor associated with the incidence of pesticide toxicity namely the length of work and work period. Risk factors associated with the incidence of pesticide toxicity are age, length of work and work period where is the last two are the dominant factors.

Abstract

The results of monitoring and examination of farmers in Barito Utara District by examining the cholinesterase enzyme by the Regional Health Laboratory and the District Health Office of Barito Utara in the blood of farmers found 71.19% experienced organopathic pesticide poisoning. This study aims to analyze the risk factors associated with the level of pesticide poisoning in rice farmers in Teweh Selatan Subdistrict, Barito Utara District, namely mixing dosage, spraying time, spraying frequency, spraying duration, working period, wind direction, and use of personal protective equipment. The research design used was observational with cross sectional approach. The number of samples is 55 people taken by simple random sampling. Rice farmers who experienced pesticide poisoning by 56.4%. Chi-Square Test showed four variables had a significant relationship with the incidence of pesticide poisoning and were a risk factor for the occurrence of pesticide poisoning namely mixing dose (p=0.003 OR=6.909; 95% CI=2.020-23.627), frequency of spraying (p=0.004; OR=6,300; 95% CI=1.913-20.749), working period (p=0.020; OR=4.416; 95% CI=1.402-13.906) and wind direction (p=0.007; OR=5.714; 95% CI=1.756-18.591). Multiple logistic regression test showed that the most dominant factor related to the incidence of pesticide poisoning was the frequency of spraying. Risk factors associated with the incidence of pesticide poisoning are mixing dosage, frequency of spraying, working period and wind direction.

Abstract

Brotowali (Tinospora crispa [L] miers), bitter melon (Momordica charantia), and madagascar periwinkle (Catharanthus rosesus [L.] G. Don) are plants that have been used for some disease treatments. The mechanism of action of these plants to cure some diseases corellate with the balance of oxidant and antioxidant. To prove that statement, this study was conducted to evaluate the of antioxidant potential activity in these plants. The method used was antioxi¬dant trial activity with modification of DNP method. The result showed that Madagascar periwinkle (Catharanthus rosesus [L.] G. Don) infusion has the highest antioxidant activity, that was 90, 27%.Tanaman brotowali (Tinospora crispa [L] miers), pare {Momordica charantia), dan tapak dara {Catharanthus rosesus [L.] G. Don) adalah beberapa tanaman yang telah banyak dipergunakan untuk mengobati berbagai macam penyakit. Mekanisme tanaman tersebut dalam mengobati penyakit diduga terkait dengan keseimbangan antara oksidan dan antioksidan. Untuk membuktikan hal tersebut, maka dilakukan penelitian untuk mengetahui potensi aktivitas antioksidan tanaman brotowali, pare, dan tapak dara. Metode yang dipakai berupa pengujian aktivitas antioksidan dengan metode DNP yang dimodifikasi. Hasil penelitian menunjukkan bahwa infus daun tanaman tapak dara {Catharanthus rosesus [L.] G. Don) mempunyai aktivitas antioksidan paling baik, yaitu 90,27 %.

Abstract

Lidah buaya (Aloe vera) mengandung enzim antioksidan yang dapat menghambat kerja dari mediator inflamasi dan penghilang rasa sakit. Pada penelitian ini akan diukur aktivitas enzim antioksidan askorbat dependent peroksidase dan katalase ekstrak air Aloe vera serta potensinya sebagai antiinflamasi pada tikus yang mengalami luka terkontaminasi. Penelitian ini menggunakan rancangan post test only control group dengan simple random sampling pada 36 ekor tikus yang terbagi atas 2 kelompok, yaitu kelompok kontrol (P0)  dan perlakuan (P1) merupakan kelompok tikus dengan luka terkontaminasi yang diberikan balutan dengan menggunakan ekstrak air lidah buaya 0,2 mg/g BB. Hasil penelitian menunjukkan bahwa aktivitas enzim antioksidan askorbat dependent peroksidase dan katalase masing-masing 37,8 menit-1 dan 3,145 menit-1 dan rata-rata penurunan intensitas warna kemerahan dari eritema pada kelompok yang diberi ekstrak air lidah buaya (Aloe vera) lebih cepat daripada kelompok kontrol. Diismpulkan bahwa ekstrak air lidah buaya berpotensi sebagai antiinflamasi pada model tikus luka terkontaminasi. Aloe vera contained an antioxidant enzyme that can inhibit the work of the mediators of inflammation and pain. With this research, however, will be measured the antioxidant enzyme activity of ascorbic dependent peroxidase and catalase on water extract Aloe vera and its potential as an anti-inflammatory on rat model wound contaminated. This research uses the post test only control group with simple random sampling techniques, with 36 rats were divided into two groups, namely the control and treatment groups. The control group (P0) is a control group and treatment group (P1)  is a group of mice with wounds contaminated given the wrap by using water extracts of Aloe vera 0.2 mg/g BB. In this study it was concluded that the antioxidant enzyme activity of activity of ascorbic dependent and catalase each 37.8 seconds-1 minute-1 and 3,145. In addition, the decrease in intensity of redness of erythema on the group that was given a water extract of Aloe Vera (Aloe vera) is faster than the control group. It can be concluded that the water extract of Aloe vera as anti-inflammatory potential in a mouse model of contaminated wounds.
