Abstract

This study aim was to determine the best dimethyl sulfoxide (DMSO) concentration for native chicken’s(Gallus gallus domesticus) frozen semen. Semen was collected from three native chickens and then evaluatedmacroscopically and microscopically. Spermatozoa that used in this study had more than 75% spermmotility and more than 25 X 108 mL in sperm concentration. Semen was divided into three equal tubesand each of them diluted with Ringer lactate egg yolk (RLEY) extender consisted of 5, 7 and 9% of DMSO.Diluted semen was added into 0.25 mL of mini straw, then equilibrated at 5 °C for two hours. The semenwas then frozen placed in liquid nitrogen vapors for10 min. Finally, the semen was put into liquid nitrogenwith a temperature of – 196oC. Post thawing quality semen was assessed after 24 hours storage bythawing the straw in warm water (37 °C) for 30 seconds. Data were analyzed using one-way analysis ofvariance. The results demonstrated that the sperm motility and viability diluted with RLEY extenderwith 7% DMSO was better (p<0.05) than those in 5 or 9% DMSO. Semen quality decreased at every stepof freezing, especially between equilibration and thawing. Its concluded that 7% was the best DMSOconcentration for freezing of native chicken semen in Ringer lactate egg yolk extender compared to 5% and9%.

Abstract

The aim of the research was to identify the polymorphisms of HSP-70 gene of kampong, Arabic, andcommercial chickens. The study used whole blood samples taken from the branchial vein at the age of 12weeks from a group of chickens that were kept since DOC. Kampong chickens DOC were obtained byhatching the eggs that were collected from lowlands, medium and high altitude areas in Lombok island,while the Arabic chickens DOC were obtained from farmer. DOC of commercial chickens were purchased atthe poultry shop. The genotyping results using PCR-SSCP analysis showed that kampong and arabicchickens were polimorphic, while commercial chickens were  classified as monomorphic. Kampong chickenshad seven genotypes of HSP-70, namely AA, AB, AC, CC, AD, DD, and BC.  The AD genotypes had thehighest frequency, whereas in arabic chickens had six genotypes, namely AA, AB, AC, CC, AD and BC,which AC genotype had the highest frequencies.  However, the commercial chickens contain only onegenotype which was DD. There were three polymorphic sites found in amplificated area, namely AÆGmutation at site 617, AÆG mutation at site 628, GÆC mutation at site 646,  CÆT mutation at site 661,AÆG mutation at site 699, GÆC mutation at site 754, and AÆG mutation at site 792.  Mutations at sites628, 646 and 661 were considered as silent mutation. HSP-70 gene in kampung and arabic chickenpopulation in this study was in equilibrium. The value of Ho, He, and PIC of kampung and arabic chickenswere not different.

Abstract

Toll-like Receptor 4 (TLR4) gene is one of the genes that control resistance of chickens againstSalmonella sp. through non-specific immune response. These gene can be used as genetic markers inIndonesian local chickens, if known its diversity. Therefore this research was aimed at evaluating thegenetic polymorphism of TLR4 gene in several types of Indonesian lokal chickens (kampung, kampungpelungcrossbreed, sentul, and tolaki chicken) using PCR-RFLP. The total of samples were 136. There werethree stages of identification : extraction of DNA genom, PCR amplification of TLR4 gene (with size 220 bpon exon 2), and RFLP method using restriction enzym (MscI). The data were analyzed include frequency ofallele, frequency of genotype, heterozygosity values , and value of Polymorphic Informative Content (PIC).The result showed that TLR4|MscI was found polymorphic in all kind of chickens that was genotyped. Itwere found two alleles (A and G), and three genotypes (AA, AG, and GG). The value of x2 showed thatTLR4|MscI was in Hardy-Weinberg equilibrium. The value of Ho and He were 0,11-0,41 and 0,11-0,35. Thevalue of PIC (0,10-0,29) included in the category of low to moderately high. These results declared thatTLR4|MscI gave hope for used as genetic markers in resistance to Salmonella sp. infection in Indonesianlocal chickens.

Abstract

Bakteriosin adalah peptida dengan aktivitas antibakteri yang diproduksi oleh bakteri asam laktat dan digunakan sebagai pengawet alami. Penelitian sebelumnya menunjukkan bahwa Lactobacillus plantarum IIA-1A5 memproduksi bakteriosin yang diberi nama Plantarisin IIA-1A5 pada medium pertumbuhan yang dibuat dari whey yang diperkaya skim. Untuk aplikasi sebagai pengawet alami dan untuk memperbaiki masa simpan dan aktivitas anti mikrobanya, plantarisin perlu dienkapsulasi dan dikeringbekukan. Tujuan dari penelitian ini adalah untuk mengkarakterisasi dan mengevaluasi aktivitas antimikroba dari sediaan plantarisin IIA-1A5 yang terpurifikasi parsial dan terenkapsulasi kering beku. Ekstraksi dan purifikasi dari bakteriosin dimulai dengan presipitasi dengan ammonium sulfat, yang diikuti dengan dialysis, dan penukar kation kromatografi. Purifikasi parsial dari plantarisin kemudian dimikroenkapsulasi dengan maltodextrin kemudian dilanjutkan dengan proses kering beku. Berdasarkan pada SDS-PAGE, fraksi protein ke-7 (F7) dari plantarisin yang dipurifikasi parsial memiliki pita tunggal dan berat molekul sekitar 9,65 kDa. Konfirmasi lebih lanjut dengan menggunakan MALDI-TOF MS, ternyata pita tunggal tersebut terdiri dari 5 peptida yang diidentifikasi berbobot molekul masing-masing sebagai berikut 5,5, 7,80, 7,96, 9,09, dan 9,27 kDa. Plantarisin kering beku memiliki aktivitas antimikroba terhadap Staphylococcus  aureus tiga kali lipat dibandingkan dengan aktivitas antimikroba dari supernatan bebas sel, dan lebih tinggi dibandingkan dengan nisin, namun kurang bila dibandingkan dengan antibiotik ampisilin dan penisilin. Kesimpulannya, aktivitas antimikroba plantarisin kering beku dapat ditentukan dan lebih tinggi dibandingkan dengan nisin, ampisilin dan penisilin.Characterization of Plantarisin IIA-1A5 as Antimicrobial subtances and Evaluation of Acitivity of Freeze-dried Microencapsulated PreparationAbstractBacteriocins are peptides with antibacterial activity produced by lactic acid bacteria and used as natural preservatives. Previous studies showed that Lactobacillus plantarum IIA-1A5 produces bacteriocin named plantaricin IIA-1A5 in the medium consisting whey enriched with skim milk. For application as food preservatives and to improve its shelf-lie and activity, plantaricin was needed to be microencapsulated and freeze dried. The objective of this research was to characterize and evaluate the activity of partially purified freeze dried microencapsulated plantaricin IIA-1A5. Characterisation of partially purified plantaricin IIA-IA5 includes the identification of active fractions and molecular weight, evaluation of activity at different stage of purification and evaluation of antimicrobial activity of freeze dried microencapsulated plantaricin IIA-IA5. Extraction and prificafication of the bacteriocins started with precipitacion with ammonium sulfate, followed by dialysis, and cation exchange chromatography. The partial purified of plantaricin was then microencapsulated in maltodextrin followed by freeze drying. Based on SDS-PAGE, the protein fraction F7 of partially purified plantaricin had a single band and molecular weight about 9.65 kDa. Further analyses using MALDI-TOF, it revealed that five peptides were identified from one single band plantaricin with molecular weight 5.5, 7.80, 7.96, 9.09, and 9.27 kDa, respectively. The freeze dried plantaricin freeze showed antimicrobial activity against Staphylococcus aureus three times stringer as compared to the activity of cell free supernatant, and was higher than nicin, but less than antibiotic ampicilin and penicilin. As concusion, the activity of freeze dried plantaricin could be determined and had a higher value than nicin, ampicilin and penicilin.

Abstract

The development of the global genetic market has change in strategy, large-scale genetic companies, and artificial insemination has driven the industrialization of embryo market. As a result, the demand for embryo production is increasing. The purpose of this report is to briefly explain the current situation of the in vivo embryo production at the Livestock Embryo Centre (LEC), and review the development trends of global embryo transfer. In vivo embryo production data comes from secondary data for 2014-2020 from the LEC, while world transfer embryo development data comes from embryo transfer association. The average of ova (oocyte and embryo) ranged from 7-10 ova/flushing (averages 8 ova) with transferable embryos ranging from 3-5 embryos/flushing (average 4 embryo). Percentage of transferable embryo ranged from 36%-54% (averages 48%). Estimated numbers of ova and transferable embryos per flush in the world were 10.5 and 6.4 in 2019, respectively. In general, the average production of in vivo embryos and transferable embryos at LEC increases every year. Trend of world embryo production increases while in vivo embryo decreased accompanied by an increase in in vitro embryo production.

Abstract

The aims of this study were to identify the effects polymorphism of β-lactoglobulin gene  on milk yield and percentage of milk protein  and  fat  in  local sheep. A total number of 83  heads of  lactating ewes  raised under an extensive management at  Jonggol Animal Science Teaching and Research Unit (JASTRU) of  the Faculty of Anima Science, Bogor Agricultural  University were  studied.  Research  activities  were  carried  out  through  some  step involving blood collection. DNA isolation, DNA amplification and separation of DNA fragments by electrophoresis with silver staining method. By using polymerase chain reaction (P  R) then genotyped by single strand  conformation polymorphism technique (SSCP), it was success fully amplified a fragment length at 420 bp in β-lactoglobulin gene located on  the exon 7. The electrophoresis  pattern  revealed 5 types of β-lactoglobulin gene and designated as AB. C, D and E types. The proportion for  those respective 5 type from the  highest to  the lowest were for A (27.7%)C (16.87%), D (12.05%), (10.84%) and B (9.64%)  respectively. There were no  significant effect  polymorphism β-lactoglobulin gene on both milk yield and quality  in  I  cal  sheep at JASTRU.   Keywords : polymorphism. β-lactoglobulin in gene, PCR-SSCP and local sheep

Abstract

Karakterisasi sifat kualitatif dan kuantitatif diperlukan untuk menggali potensi sapi local sebagai informasi dasar untuk mengembangkan sumber daya genetic sapi tersebut.  Penelitian bertujuan untuk mengidentifikasi sifat kualitatif dan kuantitatif sapi Rancah di Kabupaten Ciamis.   Sampel  yang digunakan adalah 92 ekor sapi Rancah betina, umur > 3  tahun, masing masing 40 ekor dari kecamatan Tambaksari dan 52 ekor dari kecamatan Cijulang.  Parameter yang diukur adalah variasi warna bulu,  dan ukuran-ukuran tubuh pada kedua daerah penelitian.  Data  dianalisis secara deskriptif, perbedaan parameter bobot badan dan ukuran tubuh pada kedua populasi penelitian dianalisis dengan Uji T.  Hasil penelitian menunjukkan Warna bulu dominan sapi Rancah  adalah merah bata.  Bobot badan dan ukuran tubuh di Cijulang lebih besar dibandingkan  dengan Tambaksari, kecuali panjang dan lebar kepala sama.  Bobot badan dan ukuran-ukran tubuh sapi Rancah berada diantara sapi Bali dan PO.

Abstract

Penelitian ini bertujuan untuk menentukan jarak genetik antar subpopulasi kuda lokal di Sulawesi Utara dengan menggunakan analisis morfologi dan polimorfisme protein darah. Subpopulasi kuda local yang diamati meliputi 505 ekor kuda dewasa yang terdiri dari 310 kuda jantan (stallion) dan 193 kuda betina (mare) dengan umur 2-7 tahun. Analisis morfologi digunakan untuk menentukan ukuran tubuh dan bobot badan, sedangkan analisis polimorfisme protein darah dilakukan untuk menduga keragaman genetic berdasarkan polimorfisme protein Albumin (Alb), post-albumin (PAlb), transferin (Tf), post-transferin 1 (PTf-1), post-transferin 2 (PTf-2), alfa-hemoglobin(Hbα) dan beta-hemoglobin (Hbβ). Analisis morfologi menunjukkan bahwa rata-rata ukuran badan dan bobot tubuh subpopulasi kuda Tomohon lebih tinggi (p<0.05) dibandingkan dengan subpopulasi di 3 daerah lainnya (Manado, Minahasa selatan dan Minahasa). Jarak genetik terdekat (2,75) terdapat antara subpopulasi kuda Minahasa dan Minahasa Selatan, sedangakan jarak genetic terjauh (9,02) terdapat antara subpopulasi kuda Tomohon dan Manado. Hasil kedua analisis ini menunjukkan bahwa subpopulasi kuda Tomohon mempunyai hubungan kekerabatan yang jauh dengan ketiga subpopulasi kuda Sulawesi Utara lainnya.Kata kunci: jarak genetik, kuda, morfologi, polimorfisme protein darahDETERMINATION OF GENETIC DISTANCE ON NORTH SULAWESI NATIVE HORSES BASED ON ANALYSIS OF MORPHOLOGYAND BLOOD-PROTEIN POLYMORPHISMABSTRACTThis research aimed to determine genetic distance among subpopulations of North Sulawesi natice horses using analysis of morphology and blood protein polymorphism. The observed subpopulations of native horses included 505 mature horses 2-7 years old and consisted of 310 males (stallion) and 193 females (mare). Morphological analysis was used to determine the size and weight of body. Analysis of blood protein polymorphism was conducted to estimate genetic polymorphism of Albumin (Alb), Post Albumin (PAlb), Transferin (Tf), Post Transferin-1(PTf1), Post Transferin-2 (PTf2), alfa Hemoglobin (Hbα) and beta Hemoglobin (Hbβ). Morphological analysis showed that average size and weight of body in subpopulation of Tomohon horses were bigger (p<0,05) than 3 other ones (Manado, South Minahasa and Minahasa). The nearest genetic distance (viz. 2,75) was between Minahasa and South Minahasa horses, whereas the longest genetic distance was between Tomohon and Manado subpopulations. These analysis indicated that Tomohon horses bad far relationship to 3 other horse subpopulations ini North Sulawesi.

Abstract

The Thy1 gene is a member of the immunoglobulin superfamily, which is found in many different animals. This gene encodes a Thy1 protein (CD90) which measures 25–37 kDa. Although the function of the Thy1 gene is unknown, it is thought to play a crucial role in the immunological response, particularly in the activation of T cells. The goal of this study was to find a variation in the Thy1 gene in IPB D2  chickens that might be utilized as candidates for immunoresistance genetic markers. A total of 123 samples from IPB D2 chickens consisting of 38 samples from IPB D2 G0 (basic generation) chickens and 85 samples from IPB D2 G2 (second generation) chickens were used in this study. PCR methods and direct-DNA sequencing were used in this study. Data analysis in this study consisted of genotype frequency, allele frequency, observed heterozigosity, expected heterozygosity, and Hardy-Weinberg equilibrium. The results of the study  discovered two SNPs on the intron (g.2139 CT and g.2542 AG). Four haplotypes were created by combining two SNPs. The g.2139 CTand g.2542 AG were polymorphic, but not in the Hardy-Weinberg equilibrium.

Abstract

Cemani chicken is an Indonesian native chicken with black hyperpigmentation on feathers, skin, beak, comb, and flesh. Hyperpigmentation in chickens is called Fibromelanosis. Fibromelanosis in Cemani chickens is semi–dominant, producing two genotypes: homozygous (Fm/Fm) and heterozygous (Fm/fm+). Cemani chicken meat’s black color may indicate a higher mineral content than regular chicken meat. The study’s is aims are to detect genotype homozygous (Fm/Fm) and heterozygous (Fm/fm+) mutations and to determine the mineral content of Cemani chickens with homozygous (Fm/Fm) and heterozygous (Fm/fm+) genotypes. In the Cemani chicken population (n = 32), the Fm–specific allele genotype was detected using a Polymerase Chain Reaction–Restriction Fragment Length Polymorphism (PCR–RFLP) and the MluI restriction enzyme. The mineral contents of Cemani chicken tested were Fe, Zn, Mn, and Se. The results showed that homozygous Cemani chickens (Fm/Fm) had higher Fe and Zn mineral content. However, the homozygous (Fm/Fm) and heterozygous (Fm/fm+) Cemani chicken were not different in mineral content of Se and Mn. This study found that different genotypes of Cemani chicken had different mineral compositions. In the future, this analysis supports the selection of chicken strains with high antioxidant levels.
