Abstract

IPB D-1 chickens had good egg productivity. Chicken eggs are often used as functional ingredients in the form of powder because they have many advantages for the food industry. This research aims to produce egg yolk powder with good physicochemical, microbiological, and fatty acid qualities derived from IPB-D1 chicken eggs using various drying methods namely spray drying, oven drying, and vacuum drying. This study also aims to examine the functional properties of IPB-D1 chicken egg yolk powder by application to mayonnaise. It is compared with mayonnaise made from fresh egg yolks and commercial egg yolk powder. The experimental design used in this research was a randomized block design (RBD) based on the manufacturing period. The result showed that different drying methods for egg yolk powder can affect the emulsion activity, total plate count (TPC), aw, pH, water, ash, and fat content. The drying method did not affect the protein content and emulsion stability. Egg yolk powder with oven drying gave the best results for quality.

Abstract

Gelatin is extensively applied in various industries, including food, beverages, cosmetics, and pharmaceuticals. Although the determination of gelatin species is essential for religious, health, and consumer preference reasons, a standardized analysis method is absent. The challenge in identifying gelatin through DNA-based methods arises from the low DNA content and extensive DNA denaturation in the gelatin matrix. This study assessed the efficacy of two commercial DNA extraction kits, namely the Processed Food DNA Extraction (PF kit) and the DNeasy Mericon Food Kit (DM kit), for extracting DNA from porcine gelatin powder and commercial products derived from gelatin. Additionally, we evaluate the amplification of the extracted porcine DNA using real time polymerase chain reaction (RT-PCR) and droplet digital polymerase chain reaction (ddPCR) techniques. The PF extraction kit demonstrated successful DNA extraction from porcine gelatin powder and commercial samples of porcine gelatin-based candies with a higher concentration (32.24-286.07 ng/uL) and purity (A260/A280 ratio of 1.82-2.33) compared to the DM kit (3.95-7.30 ng/uL and an A260/A280 ratio of 1.29-2.45). RT-PCR and ddPCR analyses yielded positive results for porcine DNA from gelatin powder for both PF and DM kits, albeit with differing Cq values and copy numbers. The choice of DNA extraction kit significantly impacted the amplification results when analyzing commercial samples of porcine gelatin-based candies. Using RT-PCR, all samples yielded negative results with the DM kit, while the PF kit detected one positive result for porcine DNA. Improved outcomes were observed with more sensitive analysis methods such as ddPCR, where the DM kit identified one positive result for porcine DNA while the PF kit detected positive results for all tested candies.

Abstract

Kandungan asam lemak dipengaruhi oleh sejumlah gen dan di antaranya ialah selenoprotein W1 (SEPW1). Eksperimen ini bermaksud untuk mengetahui polimorfisme gen SEPW1 serta hubungannya terhadap kandungan asam lemak di domba. Sejumlah 102 ekor domba yang tersusun dari 20 domba ekor gemuk (DEG), 30 domba ekor tipis (DET), 9 domba komposit garut (DKG), 10 domba compass agrinak (DCA), 10 domba barbados cross (DBC), dan 23 domba jonggol (DJ) digunakan dalam penelitian ini. Analisis polimorfisme gen SEPW1 dicari dengan memanfaatkan Polymerase Chain Reaction-Restriction Fragment Lenght Polymorphism (PCR-RFLP), enzim BanI, serta produk amplifikasi berukuran 478 bp. Analisis Chi-square memberikan hasil bahwa populasi berada dalam keseimbangan Hardy-Weinberg, kecuali sampel domba jonggol. Polimorfisme gen SEPW1 teramati berada di semua sampel rumpun domba. Single nucleotide polymorphism (SNP) g.53617063 G>A gen SEPW1 berasosiasi (P<0,05) dengan asam laurat (C12:0), asam palmitat (C16:0), asam arakidat (C20:0), asam henekosilat (C21:0), asam miristoleinat (C14:1), asam oleat (C18:1n9c), asam linoelat (C18:2n6c) dan asam lemak total. Genotipe GG memiliki kandungan asam lemak tak jenuh yang paling tinggi dibandingkan genotipe lainnya. Gen SEPW1 memiliki kapasitas untuk dikembangkan sebagai penanda asam lemak di domba.

Abstract

Selenoprotein W1 (SEPW1) is a protein containing selenium that has an important role in muscle growth and development by protecting myoblasts from oxidative stress. This study was aim to identify the diversity of the SEPW1 gene and its association with mineral content in sheep. The samples used were 100 sheep consisting of 10 samples of compass agrinak sheep (CAS), 15 samples of Javanese fat-tailed sheep (JFT), 30 samples of Javanese thin-tailed sheep (JTT), 25 samples of garut composite sheep (GCS), and 20 samples of jonggol sheep (JS). Selected mineral contents were determined using the IK method LP-04.10-LT-1.0. Identification of SEPW1 gene polymorphism will be conducted using Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP). The size of the amplification product was 455 bp. Abstract

The Kerinci duck (Anas platyrhynchos), hailing from Jambi Province, represents one of the local duck breeds of Indonesia. This study aimed to ascertain the matrilineal origin of Kerinci ducks through analysis of the mitochondrial NADH Dehydrogenase subunit 2 (MT-ND2) gene. In this pursuit, forward sequences of the MT-ND2 gene (490 bp) were extracted from unsexed Kerinci ducks, totalling forty-eight (48) sequences, procured from blood samples. The findings demonstrated the identification of fifteen (15) distinct haplotypes within the MT-ND2 gene, with corresponding haplotype diversity (Hd) and nucleotide diversity (pi) values of 0.74 and 0.003, respectively. The resulting phylogenetic tree unveiled that Kerinci ducks exhibit two matrilineal origins: an Asian and an independent Kerinci lineage. Moreover, most Kerinci ducks were categorised within the H4 group (24 birds) of the Asian lineage. Nevertheless, this study also revealed the existence of an independent Kerinci lineage comprising eight (8) duck haplotypes. In conclusion, the analysis of the MT-ND2 gene underscored the genetic introgression of A. zonorhyncha and A. poecilorhyncha in Kerinci ducks.

Abstract

The weight of livestock is a crucial metric for evaluating management efficacy, informing policy decisions, and determining the market value of animals. In certain scenarios, conventional methods such as physical weighing and measurement calculations can prove challenging, including the absence of livestock health records or weighing equipment. This research aims to develop a predictive model for estimating the live weight of cattle through visual assessments and metadata, including age and pixel count, utilizing a combination of convolutional neural network (CNN) and artificial neural network (ANN) methodologies. A total of 223 data were obtained from a local farm before augmentation. The model's predictive capability was successfully demonstrated, with its performance quantified by an average mean absolute percentage error (MAPE) of 10% on test data. This study demonstrates that through the combination of CNN and ANN, as well as optimal parameter tuning, efficient prediction of cattle weight can be achieved.

Abstract

ABSTRAK. Pelepasan ayam IPB-D1 sebagai rumpun baru yang disebar di masyarakat maka sistem pemeliharaannya secara umum bersifat ekstensif dan pakan yang diberikan memanfaatkan pakan lokal. Tujuan penelitian ini adalah untuk mengetahui performa ayam IPB-D1 dengan perlakuan pemeliharaan P1 (sistem kandang Closed House dan penggunaan 100% pakan komersil) serta perlakuan P2 (sistem kandang umbaran dan pencampuran pakan lokal hingga 70%). Penelitian ini menggunakan ayam IPB-D1 berjumlah 503 ekor DOC yang dipelihara hingga umur 12 minggu. Performa ayam diukur berdasarkan bobot badan, laju pertumbuhan, konsumsi pakan, konversi pakan serta tingkat kematian ayam pada umur DOC-12 minggu. Data dianalisis secara deskriptif dengan menyajikan rataan, simpangan baku, koefisien keragaman dan analisis statistik menggunakan uji T. Hasil penelitian performa ayam IPB-D1 pada perlakuan P1 diperoleh bobot badan jantan 1378.2694.56 g dan betina 1178.33119.33 g, sedangkan pada perlakuan P2 diperoleh bobot badan jantan 1173.8260.1 g dan betina 957201.7 g. Laju pertumbuhan pada kedua perlakuan bersifat positif, serta konversi pakan pada perlakuan P1 (2.88) dan P2 (3.44) cukup baik. Kesimpulan penelitian ini adalah ayam IPB-D1 diperoleh performa yang sangat baik pada perlakuan P1 dan mampu beradaptasi pada perlakuan kandang sistem umbaran serta mampu memanfaatkan campuran pakan lokal yang baik pada perlakuan P2. (The growth performance of IPB-D1 chickens in different feed treatments and production systems) ABSTRACT. The release of the IPB-D1 chicken as a new breed that is spread in the community, the production system is generally extensive and the feed provided utilizes local feed. The purpose of this study was to determine the performance of IPB-D1 chicken with production treatment P1 (Closed House cage and use of 100% commercial feed) and P2 treatment (outdoor cage and use local feed mixing up to 70%). This study used 503 DOC IPB-D1 chickens which were maintained until 12 weeks of age. Chicken performance was measured based on body weight, growth rate, feed consumption, feed conversion and chicken mortality at 1-12 weeks. Data were statistically analyzed using the T test and descriptive analysis with means, standard deviations, and coefficient of diversity. The results of the study of the performance of IPB-D1 chickens in P1 treatment obtained male body weights 1378.26 94.56 g and females 1178.33 119.33 g, while in P2 treatment obtained male body weights 1173.8 260.1 g and females 957 201.7 g. Growth rates in both treatments were positive, and feed conversion was good in P1 treatments (2.88) and P2 treatments (3.44). The conclusion is that the IPB-D1 chicken obtained a very good performance in the P1 treatment and was able to adapt to the treatment of outdoor cages and was able to utilize the local feed mixture in the P2 treatment.

Abstract

Proteases are one of most important and abundant enzymes produced by the biotechnology industry, for scientific, physiological and industrial application and dominates of the whole enzyme market. Lactobacillus plantarum IIA-1A5 is an Indonesian lactic acid bacteria (LAB) isolated from beef Peranakan Ongole cattle. Preliminary analysis on its whole genome sequence indicated that this strain harbours some genes involved in protein degradation and might be promising to be further applied. This study aims to optimize the gene sequence of a lon-like protease of L. plantarum IIA-1A5 for heterologous expression system. The Lon-like gene expression system is made using genes that have been optimized first in silico. pET-28a(+), E. coli BL21(DE3), Nde1 and BamH1 were used in this study as a expression vector, a host and retriction enzyme, respectively. Molecular weight was validated using SDS-PAGE and expasy.org software. The results showed that optimization increased codon adaptation index value (CAI) and GC content to 0.97 and 56.57%, respectively, which were suitable for the E. coli expression system. The Lon-like IIA gene was successfully expressed in the cell cytoplasm by induction of 1 mM Isopropyl -D-1-thiogalactopyranoside (IPTG) at 37 C. As many as 88% of Lon-like IIA codons were distributed in the 91-100 quality group. Lon-like IIA was successfully expressed in a host cell induced with 1 mM IPTG at 37oC . IPTG induction was performed at the 3rd hour of incubation with OD600 0.59. In addition, Lon-like IIA molecular weight was detected approximately 43 kDa.

Abstract

The PPM1K (Protein Phosphatase, Mg2+/Mn2+ Dependent 1K) gene is assumed to associate with retail meat cuts. This study aims to determine the association of the PPM1K gene polymorphisms with retail meat cuts among various breeds of Indonesian sheep. The Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP) approach was used to determine the polymorphisms of the PPM1K gene in 130 Indonesian rams aged 10-12 months with body weights ranging from 20-25 kg. The association study between PPM1K and retail meat cuts was analyzed using General Linear Model (GLM). The research showed that the PPM1K gene was polymorphic, with three genotypes: AA (458 bp), GG (259 bp and 199 bp), and AG (458 bp, 259 bp, and 199 bp). The PPM1K gene polymorphisms were significantly (P<0.05) associated with breast cut, which is the breast muscle and bone. The GG genotype of the PPM1K gene had a higher value on breast lamb. This result implies that the PPM1K gene could be a potential candidate for marker-assisted selection for improving the lamb quality of Indonesian sheep.

Abstract

Standar kualitas semen yang berperan penting dalam keberhasilan inseminasi buatan (IB) sebagai upaya peningkatan populasi dan mutu ternak sapi di Indonesia terbukti tidak seutuhnya mampu menentukan tingkat fertilitas sapi pejantan di Indonesia. Studi terbaru membuktikan bahwa meskipun sudah memenuhi standar kualitas semen yang sudah ditetapkan, namun tiga dari enam ekor sapi pejantan di Indonesia masih tergolong fertilitas rendah (<60%). Potamine-1 (PRM1) merupakan molekul pada inti spermatozoa yang telah terbukti memiliki hubungan erat dengan fungsi fertilitas dan kualitas spermatozoa pada berbagai sapi pejantan di Indonesia. Hal tersebut mendasari usulan pengembangan uji berbasis molekuler PRM1 sebagai penentu tingkat fertilitas yang diharapkan dapat digunakan untuk proses seleksi dan kebijakan pengafkiran sapi pejantan unggul di Indonesia. Melalui policy brief ini diharapkan usulan pengembangan uji tersebut dapat lebih tersampaikan dengan segala solusi yang diberikan, termasuk terkait pendekatan uji berbasis molekuler yang paling tepat digunakan, kajian-kajian lanjutan, dan berbagai harapan dukungan dari segala pihak demi terwujudnya pengembangan uji PRM1.
