Abstract

Myostatin plays a role in inhibiting skeletal muscle growth in vertebrates. This study aimed to investigate the full sequence of the myostatin gene in fast-growing and slow-growing chickens. Fast- and slow-growing chicken models were produced from F2 Kampung x broiler. The full sequence of the myostatin gene was identified using 24 pairs of primers covering about 8,000 bp. mRNA expression analysis of muscle tissue was performed to examine whether the expression levels of myostatin are affected by chicken lines, sex, or muscle type. The results showed 170 mutations in fast- and slow-growing chickens. One hundred and sixty-one of them are novel mutations. A total of five and twenty-two alleles were specific alleles found only in the fast-growing and slow-growing groups of chickens, respectively. There were no differences in amino acids and gene expression levels of myostatin between the fast- and slow-growing chickens. In summary, the results of this study showed that specific alleles for the fast-growing or slow-growing chicken groups were found, suggesting that these specific alleles potentially be used as genetic markers for muscle growth in chickens.

Abstract

One of gene which involves on mediation an antioxidant cascade in beta cells as an intracellular target of excess fat intake is Kinesin Family 12 (KIF12). This study aims to identify the diversity of SNP g. 9617965 C>T KIF12 and association with carcass characteristic and meat quality. The total sample used were 102 ram DNA samples consisted of 10 barbados cross sheep (BCS), 52 javanese thin tail (JTT), 15 jonggol sheep (JS), 15 garut sheep (GS), and 10 compass agrinak sheep (CAS). Identification of the diversity of KIF12|BfaI was conducted using the Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP) method. The results showed that the amplification product was 696 bp. The diversity of the KIF12 gene was found three genotypes namely CC, CT, and TT. The allele frequency in all types of sheep was in Hardy-Weinberg equilibrium. A SNP of the KIF12 was associated (P<0.05) with characteristic of carcass, including length of carcass. The association of KIF12 with meat quality was significant (P<0.05) too associated with pH and water holding capasity (WHC). It could be concluded that KIF12 gene is a potential candidate gene for carcass characteristic and meat quality traits in sheep

Abstract

This study aimed to evaluate nutrient requirements and zinc supplementation for IPB D-2 pre-layer chicken line candidates on performance, blood profile, and blood malondialdehyde (MDA). This study used 200 of 16 weeks IPB-D2 chickens line candidates reared until 19 weeks of age (pre-layer phase). This study used a completely randomized design (CRD) with 2 treatments and 10 replications. The treatment rations were formulated to fill the chicken nutrient requirements according to Lohman (2020) without the addition of Zinc and with the addition of 120 ppm Zinc. The data were analyzed using the T test. The results showed that the ration supplemented with 120 ppm zinc did not significantly affect performance, but it was able to increase body weight uniformity. The 120-ppm Zinc supplementation significantly (p<0.05) decreased erythrocytes, however it tended to lower blood MDA. In conclusion, 120 ppm Zinc supplementation in the ration is sufficient to meet the nutrient requirement of IPB-D2 chicken line candidates in the pre-layer phase. Abstract

A candidate marker that influences the flavor and odor of Indonesian sheep that is the Potassium Channel Tetramerization Domain Containing 2 (KCTD2) gene. This study aims to examine the polymorphism and association between the KCTD2 gene and lamb flavor and odor in Indonesian local sheeps. This study used DNA taken from the longissimus dorsi (LD) muscle of 100 rams including 75 Javanese Thin-Tail sheep (JTTS), 10 Javanese Fat-Tail Sheep (JFTS), and 15 Jonggol Sheep (JS), with ranged in weight from 20 to 35 kg and were 10 to 12 months old. The PCR-RFLP technique and GLM test analysis were used to identify polymorphisms and association of KCTD2 gene. The restriction enzyme BafI digests the PCR-RFLP product of the KCTD2 gene. The results showed that the KCTD2 gene was polymorphic (CC and CT). KCTD2 gene analysis showed a significant (P<0.05) association with 3-Methylphenol (MP). This research provide information regarding the role of the KCTD2 gene in lamb flavor and odor, especially in 3-Methylphenol (MP) compounds, and explain KCTD2 as a functional gene in the selection of premium sheep with low flavor and odor in lamb meat.

Abstract

The Maleo Senkawor bird (Macrocephalon maleo) is endemic and widely distributed in Sulawesi. Maleo Senkawor bird is monomorphic so it is difficult to distinguish between male and female individuals. This study aimed to determine the kinship relationship among M. maleo in various geographic areas in Sulawesi through the use of the gene marker Cytochrome-b (Cyt-b), determine the sex ratio, and examine the reliability of the 2550F/2718R primer. Fifteen (feather and eggshell) samples from North Sulawesi, Southeast Sulawesi, and Central Sulawesi were collected. Total DNA isolation was performed using the Dneasy® Blood and Tissue kit (Cat. No. 69504) following a Qiagen protocol with modification. PCR amplification (35 cycles) used a forward primer MMCytb_F (5'- GAAAATCCCACCCCCTACTA-3'), a reverse primer MMCytb_R (5'- GTTGGCTACGAGGAGTCAGA-3') and a primer for sexing used 2550F/2718R. Analysis was performed on the length of 903 bp Cyt-b mtDNA gene sequences of M. maleo along with their amino acid sequences (301 AA) and Chromo Helicase DNA-Binding (CHD) gene sequences. Based on the genetic distance of the Kimura 2-Parameter model and p-distance, the interpopulation of M. maleo in Sulawesi (the population of Southeast Sulawesi was separated from the population of Central Sulawesi and North Sulawesi) was 0.002 (0.2%) – 0.003 (0.3%). Each population has formed a different haplotype. Primer 2550F/2718R was able to amplify the CHD gene and could distinguish the sex identification. Male individuals are more dominant than female individuals with a sex ratio of 6,5:1

Abstract

The objective of this experiment was to determine the addition of sodium dodecyl sulfate (SDS) to Ringer Lactate-Egg Yolk (RL-EY) extender on pelung chicken semen preservation. Semen was collected three times a week from three pelung chickens. Collected semen was evaluated macroscopically and microscopically. Only ejaculates of at least 70% sperm motility was then divided into three equal microtubes. Each of them diluted with RL-EY with ratio 90%:10%, then added with 0.00, 0.025, and 0.05% SDS respectively. The liquid semen then stored at 5 °C for 72 hours. Sperm motility and viability were observed every 12 hours. The addition of 0.025% SDS showed higher spermatozoa motility and viability (72.08±1.44% and 80.82±1.30%) which were significantly higher (P0.05) than 0.00 and 0.05% SDS addition at 24 hours of storage. There was no differences on the spermatozoa motility and viability between 0.00 and 0.05% SDS addition. The decrease of spermatozoa motility and viability was observed in 0.025% SDS addition (4.17±0.56% and 4.65±0.59%, respectively) that significantly lower compared to 0% and 0.05% SDS addition at 24 hours of storage. In conclusion, 0.025% SDS addition in a Ringer Lactate -Egg Yolk extender maintained pelung spermatozoa motility and viability better than 0.00 and 5% SDS addition.

Abstract

This study aimed to investigate the effectivity of goat milk fermentation as tyrosinase inhibitory with Lactobacillus rhamnosus TW 2. The examination of fresh milk contained the density, protein, and fat content. The culture of Lactobacillus rhamnosus TW 2 acid lactid bacteria starter was added with the concentration of 3, 4, and 5%, inoculated in pasteurized goat milk, then incubated at 37° C for 24 hours. Fermented milk were extracted by centrifugation, then supernatant was collected and was used for inhibition of tyrosinase enzymes activity on L-tyrosin and L-dopa substrate. The result showed that the density, fat content, and protein content of Etawah crossbred goat milk are 1.028, 3.73, and 5.45%, respectively. Re-identification of lactic acid bacteria showed similar morphology, physiology, and bio-chemistry with the isolated lactic acid bacteria. The growth curve of TW 2 was observed in 12 hours. The 5 % of Lactobacillus rhamnosus TW 2 was the best concentration to inhibit tyrosinase activity in L-Tyrosin substrate. In conclusion, fermentation of goat milk using Lactobacillus rhamnosus TW 2 at concentration of 5% as starter is effective to inhibit tyrosinase activity significantly.Key words: goat milk, fermentation, tyrosinase inhibitory

Abstract

The aim of this study was to examine the preservation of sentul crossbreed chicken semen in ringer lactate egg yolk diluent supplemented with various monosaccharide. Semen was collected from three roosters using massage method. Immediately after collection, the semen was evaluated macroscopically and microscopically. Semen with more than 70% motility was divided into four tubes. Each of them diluted with ringer lactate egg yolk glucose (RLEYG), ringer lactate egg yolk fructose (RLEYF), ringer lactate egg yolk xylose (RLEYX) and ringer lactate egg yolk mannose (RLEYM). Semen was stored in refrigerator (5o C) for sixty hours and evaluated every twelve hours for spermatozoa motility and viability. Results showed that no significant difference (P0.05) among diluents used on spermatozoa quality parameters after dilution and during preservation. Semen quality decrease during storage and at sixty hours of storage, the motility and viability of spermatozoa ranging from 48.33±2.56 to 55.42±2.26% and 58.59±2.87 to 64.83±2.42%, respectively. This research conclude that glucose, fructose, xylose and mannose can be used as energy source for roosters semen during preservation.

Abstract

The apolipoprotein A-V (APOA5) gene is a candidate gene that plays an important role in carcass characteristics and lamb quality. This study aimed to association and polymorphism of the APOA5 gene with carcass characteristics and lamb quality in sheep. The breed of sheep were used 100 sheep consisting of Thin Tailed Sheep (TTS) (n=55), Compass Agrinac Sheep (CAS) (n=10), Garut Sheep (GS) (n=18), Garut Composite Sheep (GCS) (n=8), and Barbados cross sheep (BCS) (n=9). Identification of APOA5 gene polymorphism was performed using PCR-RFLP method and association study was performed using GLM analysis. The results showed that the APOA5 (g.26929941 C>T) gene polymorphism was polymorphic in TTS, CAS, GS, and GCS, while BCS was monomorphic. The APOA5 gene has three genotypes (CC, CT, and TT). Association showed that the APOA5 was a significantly (P<0.05) associated with carcass percentage and carcass length. The polymorphism of the APOA5 (g.26929941 C>T) was also significant (P<0.05) associated with lamb quality of the pH dan tenderness. The TT genotype showed greater carcass length and pH than the CC and CT genotypes. The CT genotype showed a greater percentage of carcass and tenderness than the CC and TT genotypes. The APOA5 gene helps improve sheep carcass characteristics and lamb quality.

Abstract

On the freezing semen chicken cryoprotectants required to overcome the damage of spermatozoa due to cold shock. This study aims to get the best concentrations of cryoprotectants glycerol concentration of 5%, 7% and 9% in freezing Sentul chicken semen. The semen used in this study came from three chickens Sentul and be repeated nine times. Semen was collected by  messase methods for three times a week. Semen was evaluated macroscopic and microscopic. Furthermore spermatozoa diluted with egg yolk and the addition of three concentrations of cryoprotectants glycerol (5%, 7% and 9%). Semen diluted 0:25 ml is packed into straw. Then equilibrated at a temperature of 5°C for two hours. After equilibration to evaluate the motility and viability of spermatozoa. Furthermore, frozen in liquid nitrogen vapor for 10 minutes. Frozen semen is then stored in liquid nitrogen containers with temperature -196°C. After 24 hours, semen is thawed at 37°C for 30 seconds. The results showed that the percentage of sperm motility and viability of frozen semen cock Sentul using glycerol cryoprotectants 5% better P (0.05) compared with the use of glycerol 7% and 9%. The use of glycerol 5% at this stage of equilibration and storage can reduce the damage of spermatozoa in the semen of chicken Sentul. Neither glycerol 5% could increase recovery rate after thawing
