Abstract

Some food industries still use synthetic dyes. Synthetic dyes, if consumed, can harm your health. Turmeric can be used as a natural food coloring. This natural coloring is applied to sweet jelly. This natural coloring agent must be used in fresh juice. To extend the use time of the natural dyes turmeric-bay leaves, ginger-bay leaves, this research was carried out in a variety of storage conditions without using preservatives. The aim of the research was to determine the results of the content of secondary metabolite compounds in turmeric-laurel extract, curcuma-laurel extract, turmeric extract and curcuma extract, in making solid preparations using preservatives, namely citric acid, maltodextin and sucrose. This research uses experimental methods with Sari method is done on a mixture of turmeric-bay leaves and ginger-bay leaves using a water solvent using the distillation method. Determination of variations in storage conditions was carried out for 7 days and the effect of storage temperature. Color stability and intensity measurements were carried out using visible spectrophotometry and characterization methods. The results of this research were obtained. The absorbance intensity test on turmeric-bay leaves was carried out at a wavelength of 425.14 nm with a result of 0.442 (day 1), ginger-bay leaves had an absorbance of 0.490 ( day 1). In the color stability test, there was a change in color from day 3 at drying cupboard temperature, at refrigerator temperature and at room temperature, the color remained stable.

Abstract

Introduction: Single Cell Protein (SCP) is a term used for proteins produced from microbes such as bacteria, algae, yeasts, and fungi. One of the fungi known to produce PST is Aspergillus niger. Objective: The purpose of this study was to determine whether pineapple peel waste could produce single cell protein from A. niger culture and to determine differences in protein production with the addition of nutrients in the fermentation medium. Methods: This research method is an experimental method with the independent variables being MFKN1 medium (Pineapple Skin Fermentation Medium with the addition of KH2PO4 and sugar), and MFKN2 medium (Pineapple Skin Fermentation Medium with the addition of KH2PO4, (NH4)2SO4 and sugar); and the length of fermentation time used are days-0, 2, 4, and 6. The dependent variables of this study were analysis of protein content, dry weight of cells, glucose levels, pH and temperature. The data from this study were statistically analyzed using the two way Anova method. Results: The results of this study obtained the highest protein content in MFKN2 media, namely 0.80% (day-2); cell dry weight 0.473 g; glucose level 1.3406%; pH 4.6 and temperature 31 oC. Meanwhile, the highest protein content in MFKN1 media was 0.59% (day- 4); cell dry weight 0.346 g; glucose level 1.3406%; pH 3.7 and temperature 26.6 oC. Conclusion: The conclusion of this study is that pineapple peel waste can produce single cell protein from A. niger culture and there are differences in protein production results with the addition of nutrients, where protein content in MFKN2 medium is higher than protein content in MFKN1 medium.

Abstract

Bakteri Escherichia coli merupakan salah satu bakteri penyebab penyakit diare. Daun mangga harum manis (Mangifera indica L. var. harum manis) diketahui memiliki kandungan senyawa antibakteri. Dalam ukuran nanopartikel, luas kontak permukaan partikel menjadi lebih besar sehingga dapat meningkatkan aktivitas antibakteri. Tujuan penelitian ini untuk membandingkan aktivitas nanopartikel dengan ekstrak etanol daun mangga harum manis terhadap bakteri E. coli. Penelitian ini merupakan penelitian eksperimental dengan variabel bebas yaitu konsentrasi ekstrak etanol daun harum manis (EEDM 25%; EEDM 50%; dan EEDM 75%) dan konsentrasi nanopartikel ekstrak etanol daun mangga harum manis (NEDM 2,5%; NEDM 5%; dan NEDM 7,5%) dengan antibiotic pembanding yaitu tetracyclin.Variabel terikat yaitu aktivitas antibakteri ekstrak etanol dan nanopartikel ekstrak daun mangga harum manis terhadap bakteri E. coli. Karakterisasi nanopartikel ekstrak etanol menggunakan Particle Size Analyzer (PSA). Pengujian aktivitas antibakteri menggunakan metode difusi agar cakram Kirby Bauer. Hasil karakteristik ukuran nanopartikel ekstrak adalah 40,2 nm. Nilai Zone of Inhibition (ZOI) ekstrak etanol daun mangga harum manis adalah 12,4 mm (EEDM 25%); 14,1 mm (EEDM 50%); dan 16,1 mm (EEDM 75%). Nilai ZOI nanopartikel ekstrak etanol daun mangga harum manis adalah 13,8 mm (NEDM 2,5%); 14,5 mm (NEDM 5%); dan 18,6 mm (NEDM 7,5%). Kesimpulannya adalah ekstrak etanol daun mangga harum manis dapat dijadikan nanopartikel ekstrak dengan daya aktivitas antibakteri konsentrasi nanopartikel ekstrak 5% setara efektivitasnya dengan konsentrasi ekstrak etanol daun mangga harum manis 50% terhadap E. coli dengan kategori intermediate dibandingkan dengan Tetracyclin 30 μg, sehingga dapat dikatakan bahwa nanopartikel esktrak dapat memperkecil dosis suatu obat.

Abstract

Paper soap is soap in the form of thin sheets resembling paper. Rice bran from rice milling waste contains antibacterial compounds. The aim of this research was to formulate paper soap from rice bran ethanol extract and to determine its antibacterial activity. This research is a type of experimental research carried out in a laboratory. Includes steps such as taking and identifying samples, preparing and making ethanol extract of rice bran (Oryza sativa L.), and testing antibacterial activity against Escherichia coli using the Disc Disc method, where the inhibition zone of the ethanol extract of rice bran and the paper soap preparation is containing rice bran extract (Oryza sativa L.) was observed and measured as a result of the antibacterial activity test. The results of this research show that the ethanol extract of rice bran contains chemical compounds, namely alkaloids, flavonoids, saponins, tannins, triterpenoids/steroids and glycosides. Rice bran ethanol extract has characteristics in accordance with the 1989 MMI requirements. The results of determining the antibacterial activity of rice bran ethanol extract produce an inhibition zone against Escherechia coli bacteria ranging from 5mm - 12.5mm and Staphylococcus aureus ranging from 5.5 mm-16.67 mm. The results of the paper soap evaluation test, organoleptic examination, thickness examination, weight uniformity test, stability test, foam power test met the requirements and in the irritation tests F0, F1, F2 and F3 did not irritate the skin, in the hedonic test the most preferred formula was F1. The results of determining the antibacterial activity of paper soap produced an inhibition zone for Escherechia coli bacteria ranging from 8.16mm - 12.5mm and Staphylococcus aureus ranging from 15mm-14.83mm. From the research results it can be concluded that rice bran ethanol extract can be formulated into paper soap and has antibacterial activity.

Abstract

Human life always coexists with various types of microbes. One of them is kenaga leaves (Cananga odorata (Lam.) Hook. F. & Thomson), which is a plant that can be used as a traditional medicine, one of which can be used to treat itching on human skin and can inhibit bacteria, one of which is Staphylococcus epidermidis. This research aimed to determine the types of secondary metabolite compounds contained in ylang-ylang leaves (Cananga odorata (Lam.) Hook. F. & Thomson) and to test the antibacterial activity of the n-hexane and ethyl acetate fractions of ylang-ylang leaves (Cananga odorata (Lam.) Hook. F. & Thomson). This research method includes simplicia characterization and phytochemical screening on simplicia powder and 96% ethanol extract. Extraction was done by maceration with 96% ethanol solvent, then liquid-liquid fractionation with n-hexane and ethyl acetate solvents. The antibacterial activity test was carried out by measuring the diameter of the inhibition zone using the agar diffusion method. The results of the examination of the characteristics of ylang-ylang leaf Simplicia powder (Cananga odorata (Lam.) Hook. F. & Thomson) showed that the water content was 7.33%, the water-soluble essence content was 26.48%, the total ash was 3.52%, and acid insoluble ash content 0.18%. The results of phytochemical screening showed alkaloids, flavonoids, saponins, glycosides, tannins, and steroids. The activity test results on the n-hexane, ethyl acetate, and ethanol extract fractions had antibacterial activity. The n-hexane fraction gave strong results against Staphylococcus epidermidis bacteria at a 70 mg/ml concentration with an inhibitory zone diameter of 12.37 mm. The ethyl acetate fraction was strong against Staphylococcus epidermidis bacteria at a 70 mg/ml concentration with an inhibition zone diameter of 12.98 mm.

Abstract

Antioxidants are chemical compounds that, in specific amounts, can inhibit or slow down cell damage caused by free radicals. Antioxidants function to protect the body from free radical attacks. The more free radicals in the body, the more cells will be damaged. One plant that contains antioxidant compounds is casturi mango (Mangifera casturi Koesterm.). Casturi mango (Mangifera casturi Koesterm.) is a plant that contains secondary metabolite compounds such as saponins, tannins, triterpenoids, flavonoids, and phenolics which have potential as antioxidants. This research aimed to determine differences in secondary metabolite compounds and antioxidant activity of ethanol extract of casturi mango leaves (Mangifera casturi Koesterm.) growing in Gampong Drien Bungong, Pidie Jaya. The stages of this research include sample processing, making ethanol extract of kasturi mango leaves, characterization examination, phytochemical screening, and antioxidant activity test. The sample used was musk mango leaves. The ethanol extract of casturi mango leaves (Mangifera casturi Koesterm.) was extracted using maceration with 96% ethanol solvent. The antioxidant activity test was carried out using the DPPH (1-1-Diphenyl-2-Picrylhydrazyl) method using a UV-Vis spectrophotometer at a maximum wavelength of 516 nm and vitamin C as a comparison. Based on the results of phytochemical screening tests, there are differences in secondary metabolites in the ethanol extract of kasturi mango leaves in Gampong Drien Bungong, Pidie Jaya, namely alkaloids, flavonoids, tannins, saponins, steroids, and glycosides, and the results of antioxidant activity obtained an IC50 value of 9.06 ppm with the category very strong, equivalent to the antioxidant potential of vitamin C, having an IC50 value of 4.63 ppm in the robust category.

Abstract

Antioxidants are compounds that can delay, slow down, or inhibit the oxidation reaction of food or medicine. Antioxidants include electron-donor compounds that work by donating an electron to compounds that are free radicals so that radical activity is inhibited. One of the plants that can be used as a source of natural antioxidants is salam leaf (Syzygium polyanthum (Wight.) Walp). Salam leaves contain secondary metabolite compounds, including alkaloids, tannins, saponins, steroids, triterpenoids, and flavonoids, which act as antioxidants. The study aimed to determine whether the methanol extract of salam leaves (Syzygium polyanthum (Wight.) Walp.), which grows in Gampong Bunot, Pidie Jaya, contains secondary metabolites and has activity as an antioxidant. This research was conducted experimentally, where the sample used was salam leaves. The stages of this research included the processing of plant materials, preparation of methanol extract of salam leaves, characterization examination, phytochemical screening, and antioxidant activity test of methanol extract of salam leaves using the DPPH method. Salam leaf extract was prepared by maceration method using methanol solvent obtained and concentrated with a rotary evaporator, then the phytochemical screening test and the antioxidant activity test of the methanol extract of salam leaves were carried out using the DPPH method (1,1 diphenyl-2-picrylhidrazil) at a wavelength of 516 nm and vitamin C as a comparison. Based on the results of the phytochemical screening test, there were differences in secondary metabolites in the methanol extract of salam leaves grown in Gampong Bunot, Pidie Jaya, including alkaloids, flavonoids, saponins, tannins, steroids, and glycosides. The results of the antioxidant activity test of the methanol extract of salam leaves obtained the IC value50 of 25.68 µg/ml and the IC value50 Vitamin C of 4.537 µg/ml in the second category is very strong.

Abstract

Antioxidants are compounds that can ward off free radicals and prevent damage caused by free radicals. Free radicals are unstable and reactive molecules formed from unpaired electrons that can cause damage to the skin. To protect the body's skin from free radicals, it can be overcome by using an antioxidant compound. One is kedondong leaves, which contain polyphenolic compounds known to have antibacterial and antioxidant properties. This study aimed to conduct phytochemical screening and determine the antioxidant activity of ethanol extract of kedondong leaves with the DPPH method. This study was conducted experimentally, where the sample used was kedondong leaves. The stages of research include sample preparation, plant identification, making simplisia, phytochemical screening on kedondong leaf extract and simplisia, then antioxidant activity tests were carried out on kedondong leaf ethanol extract using the DPPH method (1,1-diphenyl -2-picrylhydrazil) at a maximum wavelength of 516 nm, as a positive control vitamin C was used. The research on phytochemical screening showed that ethanol extract of kedondong leaves contains secondary metabolite compounds, namely tannins, saponins, alkaloids, flavonoids, steroids, triterpenoids, and glycosides. In testing, the antioxidant activity of ethanol extract of kedondong leaves showed very strong antioxidant activity with an IC50 of 44.855 μg/ml, and vitamin C had very strong antioxidant activity with an IC50 of 4.44 μg/ml.

Abstract

Staphyococcus aureus bacteria is one of the bacteria that contaminates crispy chicken meat and can cause Extraordinary Events or Kejadian Luar Biasa (KLB) in Indonesia as a cause of diarrhea. This is due to lack of hygiene in processing, storing and serving crispy chicken. Poisoning can occur due to food contamination by S. aureus enterotoxin. The aim of the research was to analyze the prevalence and identify the presence of S. aureus bacteria found in crispy chicken meat in the Amplas area of Medan city using Rabbit Coagulase Plasma with EDTA. Method for identifying S. aureus using BPA (Baird Parker Agar Base) and Egg Yolk media. Specific identification testing using Gram staining, catalase test, and coagulase test using Rabbit Coagulase Plasma with EDTA. The value of S. aureus bacterial contamination using the TPC (Total Plate Count) method and analysis of the prevalence of S. aureus contamination. The results of research on roadside cart krispy chickens were found to be positive for S. aureus with characteristics of Gram positive, positive coagulase and positive catalase as well as contamination values above 1X102 CFU/ml exceeding the threshold so they do not meet SNI standards. Meanwhile, the local fast food crispy chicken value for the presence of S. aureus bacteria does not exceed the threshold of 1X102 CFU/ml so it meets SNI standards. The prevalence value of S. aureus contamination in roadside cart krispy chicken was 75.0% and local fast food was 8.3%.

Abstract

Dental and oral disease is a disease experienced by almost half of the world's population (3.58 billion people). Gum (periodontal) disease is the 11th most common disease in the world. One of the dental and oral diseases that has a fairly high prevalence in Indonesia is dental caries. The aim of this research was to evaluate the physical quality of toothpaste preparations with various concentrations (10%, 30% and 50%) and to determine the antibacterial activity of ethanol extract toothpaste preparations from butterfly pea flowers (Clitoria ternatea L.) against the growth of Streptococcus mutans bacteria.The stages of this research included processing plant materials, making ethanol extract of butterfly pea with ethanol 96% solvent, characterization examination, screening of phytochemicals, making herbal toothpaste preparations of butterfly pea extract, preparation evaluation, and testing the antibacterial activity of butterfly pea toothpaste against Streptococcus mutans using the well-diffusion method. Based on the research results, it can be seen that the antibacterial activity of the formula with the greatest zones of bacterial inhibition is F1 and F2, namely 21.85 mm and 21.15 mm with very strong inhibition criteria. The conclusion of this study is that herbal toothpaste with butterfly pea extract can prevent and treat dental caries caused by Streptococcus mutans bacteria.
