Abstract

Penelitian ini bertujuan untuk mengetahui efektifitas Implementasi pendekatan saintifik terhadap kemampuan kognitif sains anak usia 5-6 tahun di RA Annur Assalafy Tumpang Pacarkeling Kejayan Pasuruan. Dengan menggunakan pendekatan saintifik dengan pemahaman 5M ( mengamati, menanya, mengumpulkan informasi, menalar/mengasosiasi, membuat kesimpulan dan mengkomunikasikan) di harapkan proses pembelajaran di RA terutama dalam perkembangan kognitif sains bisa menarik perhatian anak sehingga minat anak dalam belajar sangat baik sehingga aspek perkembangan anak bisa tercapai sesuai yang diharapkan. Penelitian ini menggunakan penelitian kualitatif yang mendiskripsikan pemahaman peneliti dalam melaksanakan pendekatan saintifik pada proses pembelajaran terutama kognitif sains. Data tesebut di peroleh dengan cara wawancara, observasi, dan dokumentasi. Hasil penelitiannya adalah (1) Pemahaman awal peneliti tentang pendekatan kognitif sains sudah cukup baik. (2) perencanaan pembelajaran dalam pendekatan saintifik dalam menyususn rencana pelaksanaan pembelajaran harian (RPPH) dan media sudah mengacu dalam langkah-langkah yang sesuai karena peneliti sudah mendapatkan pelatian-pelatian di kampus sehingga dalam menyususn rencana pelaksanaan pembelajaran harian (RPPH) peneliti sudah cukup baik. (3) Pelaksanaan pembelajaran kognitif sains dengan menggunakan pendekatan sintifik di kegiatan pendahuluan, kegiatan inti, kegiatan penutup sudah cukup baik dengan pemahaman 5 M (mengamati, menanya, mengumpulkan informasi, menalar/mengasosiasi, membuat kesimpulan dan mengkomunikasikan). Pembelajaran berpusat pada anak. Anak lebih aktif dalam pembelajaran dibanding guru yang mengakibatkan keterampilan berfikir anak lebih tinggi.

Abstract

Objective: To investigate the phenotype and genotype of E. faecalis isolated from the root canal and saliva of primary endodontic patients with periapical lesions. Methods: Eighteen adult male and female individuals suffering from primary endodontic infection, either with or without periapical lesions, were involved in this study. Root canal scraping and saliva were collected from each subject and used for bacterial quantitation using a real-time polymerase chain reaction (RT-PCR). Enterococci were isolated using ChromAgar medium and then identified using both biochemical (Gram staining and catalase tests) and molecular biology (conventional PCR) methods. Gelatinase activity, polysaccharide capsul profile and mRNA ace expression level were determined using microbiological, biochemical and molecular biology approach, respectively. Genotype of E. faecalis was determined based on nucleotide sequence of ace and gelE genes analyzed using web-based 3730xl DNA Analyze software. Results: The results showed that except for its proportion, no significant difference was found in phenotypes (gelatinase activity and mRNA ace expression levels) and genotypes (polymorphism of Cps operon and variation of ace and gelE nucleotide sequences) of E. faecalis isolated from the root canal and saliva of primary endodontic patients had or had no periapical lesions. Conclusion: It can be concluded that E. faecalis proportion had a role in the occurrence of periapical lesions in the primary endodontic patients, but not gelatinase activity, mRNA ace expression level, Cps operon polymorphism or ace and gelE nucleotide sequence variations.

Abstract

Abstract Penelitian ini dilatarbelakangi oleh nilai agama dan moral anak usia dini (4-6 tahun) kurangnya kemampuan anak dalam nilai kemoralan dan agama anak usia dini. Rumusan masalah dari penelitian ini adalah bagaimana pola asuh orangtua terhadap perkembangan nilai agama moral anak usia dini di TK Wachid Hasyim Surabaya. Tujuan dari penelitian ini adalah menjawab rumusan masalah dan penelitian. Penelitian ini merupakan penelitian kualitatif melalui pendekatan fenomenologi dengan subyek penelitian yaitu anak usia 4-6 tahun di. Metode pengumpulan data menggunakan observasi, wawancara, dan dokumentasi. Data yang diperoleh diperiksa keabsahan datanya dengan triangulasi sumber, metode dan waktu. Data dianalisis dengan teknik analisis data model interaktif Miles and Huberman. Berdasarkan hasil penelitian menunjukan bahwa: 1) Pengembangan nilai agama dan moral di TK Wachid Hasyim Surabaya dengan pola asuh demokratis, dengan cara mengajarkan nilai kereligiusan, kemandirian, sopansantun, kemudian hal-hal yang baik melaluiproses pembiasaan. 2) Faktor pendukung pengembangan nilai agama dan moral pada anak usia dini yaitu (a) adanya relawan mahasiswa yang motivasi pada anak, (b) perhatian orangtua dalam memeberikan pembelajaran nilai agama dan moral pada anak. Sementara itu faktor penghambat pengembangan nilai agama dan moral pada anak usia dini adalah (a) kondisi lingkungan yang dapat memberikan pengaruh negatif kepada anak, baik itu dari perkataan, sikap, maupun cara berpakaian yang kurang baik dan sopan, (b) minimnya pemahaman orangtua terhadap pelajaran keagamaan, (c) minimnya orangtua memberikan contoh kepada anak, dikarenakan waktu orangtua bersama anak sangat sedikit.

Abstract

Infectious Bursal Disease (IBD) adalah penyakit virus yang bersifat akut dan infeksius serta menyerang pada unggas muda yang berumur kurang dari 4 bulan. Sejauh ini data molekuler virus IBD isolat Indonesia sangat minim, oleh karena itu penelitian ini bertujuan untuk mengidentifikasi dan mengarakterisasi gen VP-2 virus IBD yang telah ada di Indonesia. Sampel penelitian diperoleh dari kasus terdiagnosa IBD yang terjadi di peternakan ayam komersial broiler dan layer. Sampel Bursa dipersiapkan untuk dilakukan isolasi menggunakan telur ayam berembrio SPF. Membran korioalantois dipanen dan dilakukan identifikasi dengan metode RT-PCR dengan gen target VP-2. Hasil amplifikasi selanjutnya dilakukan pengurutan DNA. Data nukleotida hasil pengurutan DNA dianalisis dengan program MEGA 6, meliputi pesejajaran, prediksi asam amino, dan konstruksi pohon kekerabatan antara virus yang diteliti dengan beberapa virus yang telah dipublikasi di bank gen terutama virus IBD yang bersirkulasi di Indonesia. Hasil penelitian ini diperoleh data bahwa ayam yang terdiagnosis penyakit IBD dapat ditentukan penyebabnya sebagai virus IBD. Hasil analisis urutan penanda patogenisitas molekuler menunjukkan virus yang virulen. Analisis pohon kekerabatan 2 isolat IBD SR/Lay-WNO-DIY dan IBD Potrow/Lay-SLM-DIY termasuk dalam kelompok virus IBD tipe klasik, sedangkan lima virus lainnya, yaitu IBD Yanti/Lay-SLM-DIY; IBD Lampung/Bro/IL; IBD Fung/Lay-SLM-DIY-BF1, IBD Fung/Lay-SLM-DIY-BF2, dan IBD Fung/Lay-SLM-DIY-BF3 termasuk dalam kelompok vvIBD strain Indonesia.

Abstract

Resistance to benzimidazole at a population of gastrointestinal nematode, especially Haemonchus contortus is a widespread problem. Therefore, anthelmintic will not effective anymore. Benzimidazole resistance in nematode has been shown to be heritable and defined as an increase in frequency of individual worms able to drug tolerate. In Indonesia resistance to anthelmintic albendazole has been reported on sheep in district of Bogor, Kuningan, Kendal and province of Yogyakarta. Molecular mechanism of resistance to benzimidazole on nematode and fungi related to single gene which code ß tubulin. The aim of this research is to determine a genetics variability of isotype-1 ß tubulin genes from Indonesian strain of H. contortus which resistant to benzimidazole. Sequencing were carried out at a 520 bp fragment of the isotipe-1 ß tubulin gene of six H. contortus resistant strain from four sheep from SPTD Trijaya farm, district of Kuningan, West Java; UPTD Pelayanan Kesehatan Hewan farm, district of Bantul, Yogyakarta respectively. One H. contortus susceptible strains as a control come from farmer from Cicurug, district of Sukabumi, West Java. Results indicated that there are genetics variability of  fragment isotype-1 ß tubulin genes from H. contortus from UPTD Pelayanan Kesehatan Hewan farm, Bantul, Yogyakarta (H1, H2, and C2). There are no genetics variability in the fragment gene of H. contortus resistant strain from SPTD Trijaya farm, Kuningan, West Java (Kn1, Kn2 and K1). Abstract

Chrysomya bezziana is a fly causing myiasis in most livestock in Indonesia. To date, the genetic diversity of C. bezziana has been argued among researchers. The aim of this study was to analyse genetic diversity of C. bezziana population in Indonesia using mitochondrial cytochrome b gene (cyt b) as a marker. The preliminary study showed that the larvae instar III stadium (L3) was the most appropriate sample for molecular analysis and identification. Twenty-four L3 were collected from cattles, buffalos and horses in Bogor, Makassar and East Sumba. DNA samples were isolated from muscle tissue of the larvae. The fragment of cyt b gene (279 bp) was amplified using primer CB3FC-NINFA. The PCR product was subsequently purified, sequenced and analysed using PAUP version 4. The results showed that C. bezziana from Indonesia was different with those from Asia, basicaly for DNA sequences, however it had similar to those from Papua New Guinea. Two haplotypes were identified i.e. haplotype 6 (Makassar haplotype that similar to those from Papua New Guinea) and haplotype 7 (Bogor haplotype that similar to those from Makassar and East Sumba). Abstract

Periodontal disease, a common inflammatory oral disease involved periodontal tissues, has been linked with the evidence of some systemic disorders. Recently, periodontal disease has been suspected as a trigger of systemic disorders. Penetration of bacterial products, such as lipopolysaccharide (LPS) may reach into deeper periodontal tissues. Therefore there may affect systemic blood and cytokines production. Interleukin-1β (IL-1β) and Tumour Nuclear Factor-α (TNF-α) are known as pro-inflammatory cytokines. The production of systemic IL-1β and TNF-α of E. coli lipopolysaccharide-induced periodontitis model on rats was investigated in this research. Fifteen male Wistar rats, aged 6-8 weeks used for this study were divided into 3 groups. For group 1 and 2, silk ligature 3/0 were inserted in interdental area between upper right molar 1 and 2. First and second group received solution containing 10μg/ml and 1mg/ml E. coli lipopolysaccharide, respectively, mixedwith 2% carboxymethylcellulose (CMC) diluted in 100μl of phosphate buffer saline (PBS). The solution was topically applied on gingival tissues around the gingival sulcus, a single topical application of solution onceper 2 days for 14 days. Untreated subjects were used as negative control. On day 15, the blood was collected from vena orbitalis, and rats were sacrificed. The blood serum of each group was divided into 2 groups andcultured for 4 hours with or without 20μl of 100ng/ml of E. coli LPS. ELISA techniques were used to measure the cytokine productions of the supernatant. The data was analysed using Repeated Measure ANOVA. This study showed that there was a significant increase of IL-1β production on low dose of LPS compared to control and high dose of LPS groups (p<0.05). Whereas TNF-α not significantly showed increasing trend. The increasing trend of pro-inflammatory cytokine productions, such as IL-1β and TNF-α, on LPS-induced periodontitis model in this experiment supports the previous studies about the contribution of periodontal disease in the pathogenesis of systemic diseases.

Abstract

In office bleaching menggunakan hidrogen peroksida 40% sering memberikan efek samping berupa linu baik selama maupun setelah perawatan tersebut dilakukan. Penelitian ini dilakukan dengan tujuan untuk mengetahui pengaruh waktu aplikasi dan kandungan fluor desensitizing agent pada metode in office bleaching terhadap kandungan mineral gigi. Delapan gigi masing-masing dipotong menjadi 4 bagian, 6 potong digunakan untuk pemeriksaan XRD awal. Gigi dikeringkan dalam oven suhu 50° selama 30 menit diserbuk, diambil 1 mg untuk diperiksa kandungan mineralnya dengan goniometer. Dua puluh empat potong yang lain dibagi dalam 4 kelompok perlakuan. Kelompok I: gigi diaplikasi bahan bleaching 0,5-1 mm hidrogen peroksida 40% selama 1 jam, dicuci, dikeringkan, diikuti aplikasi 0,1 ml desensitizing agent tanpa fluor (CPP-ACP) selama 30 menit, dicuci, dikeringkan. Kelompok II : gigi diaplikasi CPP-ACP 30 menit, dicuci, dikeringkan, dibleaching menggunakan hidrogen peroksida 40% selama 1 jam. Selanjutnya gigi diaplikasi CPP-ACP 30 menit lagi, dicuci, dikeringkan. Kelompok III gigi dibleaching menggunakan hidrogen peroksida 40% selama 1 jam, dicuci, dikeringkan, diikuti aplikasi desensitizing agent yang mengandung fluor (CPP-ACFP) selama 30 menit, dicuci, dikeringkan. Kelompok IV: gigi diaplikasi CPP-ACFP 30 menit, dicuci, dikeringkan, dibleaching menggunakan hidrogen peroksida 40% selama 1 jam. Selanjutnya gigi diaplikasi CPP-ACFP selama 30 menit, dicuci dikeringkan. Sesudah perlakuan, semua gigi dilakukan pemeriksaan kandungan mineral gigi dengan prosedur yang sama. Uji Mann Whitney menunjukkan penurunan mineral paling sedikit terjadi pada kelompok IV (4500). Desensitizing agent mengandung F sebelum dan sesudah perlakuan in office bleaching menunjukkan penurunan mineral paling kecil. The Effectiveness of Desensitizing Agent with and without Fluorine in Office Bleaching Method to Tooth Mineral content. Tooth sensitivity arises during or after an in-office bleaching was performed is usually overcome by using desensitizing agent with or without fluor. So far, desensitizing method applied only reduces tooth sensitivity but it had not overcome demineralization problem yet. This study was aimed to determine the influence of an application process and fluoride containing desensitizing agent in tooth mineral lossEight teeth were divided into four parts. Six specimens were smoothened using agate mortar. One mg of powder was inserted into the sample holder and mounted on goniometer heads to examine mineral content before treatment using X-Ray Diffraction (XRD). Twenty-four specimens were classified into four groups. In group I, 0,1 ml desensitizing agent without fluoride (CPP-ACP) was applied for 30 minutes after an in-office bleaching using 40% Hydrogen Peroxide was performed, while in group II, CPP-ACP was applied before and after an in-office bleaching was performed. In group III 0,1 ml fluoride containing desensitizing agent (CPP-ACFP) was applied for 30 minutes after in-office bleaching using 40% Hydrogen Peroxide was performed, while in group IV fluoride containing desensitizing agent was applied before and after an in-office bleaching was performed. Finally, all of the specimens were measured in their after-treatment mineral contain. Mann Whitney test showed that the lowest mineral lost was detected in group IV (4500). Fluoride containing desensitizing agent applied before and after an in-office bleaching effectively reduced mineral loss.

Abstract

An indigenous strain of lactic acid bacterium (LAB) identified as Lactobacillus spp. Dad13 (Dad13), isolated from traditional fermented buffalo milk, was found to be potential as probiotic. The aim of this research was to study the effect of probiotic Dad13 on humoral immune response of Balb/C mice infected with Salmonella typhimurium. The specific objective was to find out the effect of different Dad13 consumption time (before and along with infection of S. typhimurium) on the humoral immune response of Balb/C mice. The experiment was conducted by in vivo trial on 20 males of Balb/C mice, age of 6-8 weeks, fed with AIN-93 standard diet. The mice were assigned into 4 groups. Each group received the following treatments, ie. Dad13 only, Dad13 before infection, Dad13 along with infection and Salmonella infection only. A volume of 100 µl Dad13 cell suspensions (1010  CFU/ml) were given by oral forced feeding daily for a week, at week 3 for group before infection and at week 4 for group of Dad13 only and Dad13 along with infection. Salmonella infection (109   CFU/ml) was given once orally at week 4 to all groups except group treated with Dad13 only. The humoral immune response of Balb/C mice was detected 2 weeks after infection by measuring the titers of IgG and IgA specific from serum and mucosal intestinal liquid samples using Enzyme-linked Immunosorbent Assay (ELISA) method. The result indicated that humoral immune response of Balb/C mice consuming Dad13 before and along with Salmonella infection were significantly different (p<0.05). Dad13 consumption along with Salmonella infection increased circulated IgG and IgA as well as secretory IgA. It can be concluded that Dad13 probiotic feeding along with infection increased humoral immune response more significantly compared to that before infection.

Abstract

The Enteropathogenic Escherichia coli (EPEC) is one of pathogenic strain of diarrheagenic E. coli group in children and infant that occurs in developing countries. The significant virulence factors in pathogenic EPEC are eaeA (E. coli attaching- effacing ), bfpA (bundle-forming pilus A) and espA (encoding secreted protein A) genes. The use of DNA probes to detect the virulence genes in E. coli in Indonesia is not common yet. In this experiment the gene fragments of eae, bfpA, and espA were used as probes to detect the EPEC among E. coli isolates from stool specimensin of diarrheic children attending Public Health Centers in Yogyakarta. The DNA samples were isolated from 49 diarrheagenic E. coli isolates. The DNA probes of eae, bfpA and espA were obtained by amplification of DNA fragment of EPEC O126 using PCR technique. Furthermore, those probes were used to identify the presence of those genes among E. coli isolates using hybridization technique. The results showed that 42 (85.7%) isolates were espA+,   25 isolates (51%) were eaeA+    (EPEC strains). Therefore among 25 isolates of EPEC, 20 isolates (80 %) among EPEC were bfpA+  (typical EPEC strains).
