Abstract

This study aims to improve the ability of children science Group B RA RoudlotulMujtahidin Jrambe Village District Dlanggu Mojokerto through Sentra BahanAlam.Penelitian using quantitative research methods with the type of research isexperimental research. This research uses Pre-Experimental Designs research design usingOne-Group Pre-Test-Post-Test Designs.The value of improvement of the Science Ability of the 26 students who have done theapplication of learning Material Sentra Nature average score or value increased 13.50. TestStatistics above Asymp value. Sig (2-tailed) is 0, Because the value of Asymp. Abstract

Male Aedes aegypti mosquito has been considered to not have any important role in transmitting dengue virus (DENV). The purpose of this study is to prove that male Ae. aegypti mosquito does have an important role in transmitting DENV 3 through venereal transmission with their potency of its polygamy behavior. The data collection was done using colonization method and intrathoracal injection. The presence of DENV 3 on male and female mosquitoes was proven by RT-PCR method (profile of DNA band specifically on 511 bp) and serotyping PCR (290 bp). The data were analyzed using univariate analysis followed by bivariate analysis with parametric test ANOVA. The result of the study demonstrated that male Ae. aegypti mosquitoes do have an important role in transmitting DENV 3 through venereal transmission with the potency of their polygamy behavior. There was no significant difference between the polygamy behaviors of Ae. aegypti male mosquito infected by DENV and the non-infectious Ae. aegypti male mosquito.

Abstract

Poultry dust exposure may increase workers’ health risks, particularly in the form of respiratory allergic reactions. This study aimed to identify mold content of the dust and to analyze the association between dust exposure, IgE level, history of allergy, and symptoms of allergy. This study used cross sectional design with total samples of 33 workers. The data were analyzed using chi-square test and multivariate logistic regression. This study found 93.33% growth of Aspergillus sp., 69.7% work duration > 3 years, 84.8% high IgE levels, 18.2% history of allergy, and 69.7% symptoms of allergy. Dust exposure and history of allergy did not show significant correlation with symptoms of allergy while IgE levels had significant correlation with p-value of 0.036. Workers with high IgE-level were 15.986 times more likely to have symptoms of allergy (p-value 0.028). Aspergillus sp. as dust allergen potentially increased IgE levels and might become the base for facilitation of early and independent preventive and promotive efforts of workers’ health.

Abstract

Aedes aegypti mosquito population could be controlled by using lethal ovitrap. The addition of hay infusions as a attractant greatly enhance Aedes aegypti eggs trapped, and papaya leaf juice may inhibit Aedes aegypti eggs evolve to larvae or a larvae to pupae stage. This study was conducted to find out the chemical compounds in hay infusion and papaya leaf juice. We used phytochemical test using UV-Vis Spectrophotometry, Thin Layer Chromatography, and High Performance Liquid Chromatoraphy (HPLC) method. The results showed that hay infusion contains 12,75 mg/L ammonium and <1,20 ppm (µg/mL) lactic acid and papaya leaf juice contains 0,25% alkaloid, 0,14% flavonoid, 0,30% saponin, ≤68 mg/L steroid and 11,34% tannin, but negative terpenoid. We concluded that hay infusion and papaya leaf juice contains chemical compounds that could be use as attractant and bioinsecticide to Aedes aegypti, respectively.

Abstract

Dianella nemorosa Lam. (Liliaceae) is a medicinal plant traditionally used in Papua for treatment of cancer, injury, fracture, inflammation and also for antiseptic. The aim of this study was to evaluate antipriliferative activities of methanol extract of D. nemorosa leaves against HCT+116 (colorectal cell line), C2C12 (adherent mouse myoblast cell line) and 293A (Primary embryonal human kidney transformed with human adenovirus type 5 DNA). Powder of leaves was extracted using methanol. Antiproliverative activities were determined by cell proliferation reagents WST-1 assay for 1 h, 2h and 4h after 72h incubation. The result showed that methanol extract of D.nemorosa leaves showed remarkable antiproliferatiove activities against HTC-116 cell line with IC50 values of 199.31 μg/ml (1h), 197.87 μg/ml (2h) and 161.12 μg/ml (4h). The activities against C2C12 cell line resulted in the IC50 values of 405.51 μg/ml (1h), 435.12 μg/ml (2h) and 394.38 μg/ml (4h), while the IC50 values for 293A cell line were 580.81 μg/ml (1h), 442.21 μg/ml (2h) and 366.74 μg/ml (4h), respectively. Those results indicated that methanol extract of D.nemorosa leaves posses potential antiproliferative activities against HCT-116, C2C12 and 293A cell line. Further study is necessary to investigate the inhibitory mechanism of methanol extract of D. nemorosa leaves on HCT-116, C2C12 and 293A cell lines.Keywords : Dianella nemorosa Lam, cancer cell, HCT-116, C2C12, 293A

Abstract

Demam tifoid adalah penyakit endemis yang disebabkan oleh strain bakteri S. typhi, bakteri tersebut dapat dikelompokkan berdasarkan perbedaan mikromorfologi, morfologi koloni, penggunaan sumber karbon, produksi enzim, kemampuan mendegradasi makromolekul. Oleh karena itu penelitian ini bertujuan melakukan klasifikasi numerik-fenetik 4 starin S. typhi asal Jawa Tengah dan 2 strain asal DIY dengan 2 strain acuan S. typhi NCTC 786 dan BLKS berdasarkan karakterisasi fenotipik dengan analisis hubungan similaritas yang didasarkan atas analisis Simple Mattching Coefficient (SSM) serta algoritme UPGMA (unweighted pair group methode with averages) yang kemudian dipresentasikan dalam bentuk dendogram. Hasilnya dapat dikelompokkan menjadi empat klaster, klaster pertama beranggotakan 3 strain berasal dari Jawa Tengah dan 1 strain acuan BLK Semarang (similaritas 100%). Klaster kedua beranggotakan satu strain acuan S. typhi NCTC 786 (similaritas 98,6%) dengan klaster pertama. Klaster ketiga beranggotakan 1 strain MDA dari Jawa Tengah (similaritas 96,8%) dengan klaster pertama dan kedua. Klaster keempat beranggotakan 2 strain S. typhi asal DIY yang memiliki (similaritas 96,4%) dengan ketiga klaster lainnya.

Abstract

Penelitian ini bertujuan mengetahui jumlah sel Purkinje cerebellum anak mencit umur 21 hari (pascasapih) setelah induksi Ochratoksin A selama periode organogenesis. Tiga puluh ekor mencit bunting dibagi secara acak menjadi 5 kelompok perlakuan dengan masing-masing 6 ulangan. Ochratoksin A dilarutkan dalam Sodium Bicarbonat, diberikan secara oral pada saat kebuntingan hari ke 7 sampai hari ke -14. Dosis perlakuan Ochratoksin A adalah 0,5 ; 1,0; 1,5 mg/kg bb dan sebagai kontrol tidak diberi perlakuan, serta kontrol placebo diberi perlakuan pelarut Sodium Bicarbonat. Induk mencit dipelihara sampai melahirkan. Pada umur ke 21 hari (pascasapih), anak mencit dikorbankan dan diambil bagian otaknya. Otak mencit selanjutnya dipreparasi dengan metode parafin dan pewarnaan menggunakan pewarnaan Haematoksilin Eosin. Data jumlah sel Purkinje dianalisis dengan Anava Satu Arah dan dilanjutkan dengan uji DMRT untuk mengetahui beda nyata antar perlakuan. Hasil penelitian menunjukkan bahwa Ochratoksin A yang diberikan pada mencit bunting selama periode organogenesis menyebabkan terhambatnya pertumbuhan jumlah sel Purkinje mencit perlakuan yang ditandai dengan semakin menurunnya jumlah sel Purkinje dibandingkan dengan kontrol dan kontrol placebo.

Abstract

Telah dilakukan percobaan untuk menentukan tahapan palatogenesis pada mencit (Mus musculus L.) yang rentan terhadap efek polutan 2,3,7,8-Tetraklorodibenzo-p-dioksin (TCDD). Percobaan dirancang mengikuti Rancangan Acak Lengkap dengan pola faktorial (4X3). Empat puluh delapan ekor mencit bunting dicekok TCDD dengan dosis 0 (kontrol), 5, 10, atau 20 μg/kg bb. Perlakuan diberikan pada hari kebuntingan (Hk) 9−10, 11−12, atau 13−14. Mencit kontrol dicekok pelarut saja (98,5% minyak wijen + 1,5% DMSO). Pada Hk 18 mencit dibius lalu dibunuh dengan teknik cervical dislocation, persentase fetus cleft palate (cp) dihitung, derajat penutupan palatum diberi skor, preparat dengan ketebalan 6 µm dibuat, dan mikrostruktur kraniofasial diamati. Hasil menunjukkan, pemberian TCDD antara hari ke 9 dan 12 menginduksi cacat cp, dengan kecenderungan hasil tertinggi pada pemberian Hk 910. Perlakuan TCDD dosis 10 atau 20 μg/kg bb pada Hk 910 menghasilkan fetus cacat cp >90%. Persentase fetus cp tetap tinggi pada pemberian Hk 1112, khususnya pada kelompok dosis 20 μg/kg bb (87,3%). TCDD dosis terendah (5 μg/kg bb) menginduksi cp dominan bercelah sempit, menunjukkan adanya hambatan pada tahap fusi. Dosis 10 dan 20 μg/kg bb menginduksi cp bercelah sedang atau lebar, mengisyaratkan terjadi hambatan pada tahap inisiasi atau elevasi. Disimpulkan, seluruh tahapan palatogenesis rentan terhadap efek TCDD, namun tahap paling rentan adalah tahap fusi palatum.

Abstract

The periodontopathogens Porphyromonas gingivalis may directly or indirectly attack the fetus by releasing toxins into the blood stream that could reach the placental and influence fetal development . This study aimed to determine the effect of periodontal infection with P. gingivalis on fetal development in pregnant rat model. Female rats were infected with live-P. gingivalis at concentration of 109colony forming unit/ml into subgingival sulcus before and/or during pregnancy. Group I: without P. gingivalis infection ; group II: P. gingivalis infection before and during pregnancy; group III: P. gingivalisinfection before pregnancy; and group IV: P. gingivalis infection during pregnancy. They were sacrified on gestational day 14 and 20. Fetuses were evaluated for weight and crown-tail length. The results showed that the dams infected with P.gingivalis significantly decrease the mean of fetal weights, lengths and placental weights compared with the control group (p<0.05). The percentages of fetal growth restriction at the time of sacrifice were 6.66, 100, 72.97 and 87.09% growthrestricted fetuses in group I, II, III, and IV, respectively. When weights of growth- restricted fetuses of the treated groups were compared with the control group there were significant differences (p<0.05). P. gingivalis was detected by API ZYM system in the blood of umbillical cord from the treated groups. In conclusion, in pregnant rat models, periodontal infection with P. gingivalis affects fetal development. The maternal P. gingivalis infection on periodontal tissue can result in P. gingivalis dissemination to umbillical cord and induction of FGR.
