Abstract

Karakteristik gen-laktoglobulin dan hubungannya dengan sifat produksi susu pada 42 sapi Hissar dianalisis menggunakan teknik PCR-RFLP. Fragmen DNA-produk CPR dan hasil pemotongan menggunakan enzim restriksi Hae III dipisahkan menggunakan elektroforesis gel poliarkrilamid (PAGE) kemudian dideteksi dengan pewarnaan perak. Jumlah allel gen-laktoglobulin yang terdeteksi ada dua macam yaitu alel-A dan B dengan frekuensi masing-masing 0.19 dan 0.81. Uji signifikansi Chi-Square menunjukkan bahwa distribusi genotip gen-laktoglobulin dalam populasi sapi Hissar di Moyo Hilir Sumbawa-NTB berada dalam ketidakseimbangan dimana frekuensi genotip BB 69.05%; AB 23.81% dan AA 7.14%. Tingkat produksi susu memiliki korelasi yang kuat dengan genotip gen -laktoglobulin. Rata-rata produksi susu sapi Hissar dengan genotip AA, AB dan BB berturut-turut: 4.30 lt/hr; 3.11 lt/hr, dan 2.17 lt/hr.

Abstract

Survey of stingless bee diversity, nesting preferences, and forage plants was conducted in West Halmahera across 134 collection sites. This research was aimed to determine species diversity, nesting preference and habitat, and dominant forage plants. There were three species found, the most common species being Tetragonula clypearis (Friese), followed by T. sapiens (Cockerell), and last T. biroi (Friese). Based on the morphology characters of each species, the key identification was provided. The most colonies were found in public houses (80.39%), followed by plantations (13.73%), and the community forest (5.88%), respectively.Most colonies nested in stone cavities, parts of the houses, wooden materials, tree trunks, logs, tree roots, bamboo, and sometimes iron cavities. The forage plants consist of forage plantation, crops, fruits, vegetables, ornamental flowers, wild plants and shrubs. The three species found were new record in West Halmahera. Bees lived in various hollow places that were safe for their colony. Bees made use of a variety of flowering plants and secrete resins around the nest site.

Abstract

The decrease in fertility is considered to be the main cause of reproductive loss in dairy cattle and beef industry. Many candidate genes that play an important role in fertility and embryonic development. The purpose of this study was to detect genetic variations of the growth hormone gene family (GH|MspI, GH|AluI, GHR| AluI, GHRH|HaeIII and Pit-1|HinfI) and its association with superovulation response, ovulation, fertilization and transferable embryos rate. A total of 45 blood samples taken from cows that have been superovulated Angus, Brahman, HF, Limousin and Simmental. DNA was extracted with phenol-chloroform protocol followed by polymerase chain reaction technique (PCR) using specific primers for GH, GHR, GHRH and Pit-1 gene. PCR product was cut with restriction enzyme MspI, AluI, HaeIII and HinfI and electrophoresed on agarose gel and stained with ethidium bromide (EtBr). Superovulation is done by injecting a totally of 20 ml FSH for 4 days. Injecting the prostaglandin hormone (PGF2α) was performed on the eleventh day of CIDR implantation. Artificial insemination (AI) performed two or three days after the injection of PGF2α and Flushing was done on the seventh day after the AI.  The results showed that the Angus, Limousin, Brahman and Simental GH loci diversity of GH|MspI, GH|AluI, GHR|AluI, GHRH|HaeIII and Pit-1|HinfI was not associated with superovulation response, ovulation, fertilization and transferable embryo rate. In HF dairy cattle, genotype on Pit-1|HinfI AA has higher percentage of superovulation response (P < 0.05) when compared to AB genotype, but did not differ to BB genotype. Dairy cattle HF AA genotype also had higher ovulation rate (P < 0.05) when compared to AB and BB genotypes, but AB and BB have the same ovulation rate. Abstract

Information on the estimation of genetic distances and differentiation among sheep breeds are needed in crossing and conservation programs. This research aims to study of utilizing behaviour characteristic variables to differentiate and estimate genetic distance between the sheep breeds.  The study was conducted at Cilebut and Bogor Animal House of Indonesian Research Institute for Animal Production.  Five sheep breeds used were Barbados Black Belly Cross (BC), Garut Composite (KG), Garut Local (LG), Sumatera Composite (KS) and St. Croix Cross (SC), with total sample of 50 heads.  A total of 10 variables of behavior traits were observed in this study. Analysis of variances and significance tests were applied to compare between sheep breeds and performed for all of behavior traits using PROC GLM of SAS Program ver. 9.0. PROC CANDISC was used for canonical discriminant analyses, the hierarchical clustering was performed using the PROC CLUSTER by Average Linkage method (Unweighted Pair-Group Method Using Arithmetic Averages, UPGMA), and the dendogram for the five sheep breeds was described using PROC TREE.  The differentiator variables for the behavior traits were standing and feeding duration.  The canonical plotting based on behavioral characteristics could differentiate BC, KS and LG (with KG and SC) sheeps. Estimation of genetic distance based on the behavior traits is less accurate for grouping of sheep breeds.

Abstract

Diacylglycerol acyltransferase 1 (DGAT1) gene is one of the major genes that has an important role in milk fat synthesis. This research was aimed at to identifying genetic polymorphism of the DGAT1 gene by PCR-RFLP method and its association to milk fatty acid components.  Animals studied were Holstein Friesian (HF) cattle from BBPTU Baturraden (123 cows) and BPPT SP Cikole (36 cows). The length of PCR product of the DGAT1gene was 411 bp. Genotyping resulted in two types of alleles, namely K (411 bp) and A (203 and 208 bp); and two genotypes, namely KK (411 bp) and AK (203, 208 and 411 bp). For both locations, genotype frequency of AK (0.75) was higher than KK (0.25). The allele frequency of K (0.64) was higher than A (0.36). Heterozygosity of HF cattles at both locations was relatively high (Ho>He). The DGAT1 gene of the observed HF cattle was polymorphic. Result showed that there was an association between the DGAT1 polymorphism with unsaturated fatty acids especially in nervonat acid. The AK cows had a significant effect on unsaturated fatty acid content of which having a higher nervonat content (0.05%) (P<0.05) than that of the KK cows (0.03%). From the results, it is concluded that the DGAT1 gene can be functioned as a marker of selection for milk fatty acids.

Abstract

Avian Beta Defensin 2 (AvBD2) gene, which is located in chromosome 3, plays an important role in the immune system of the chicken by inhibiting the development of microorganisms such as bacteria that infect body tissues. Defensins are produced through epithelial cells immediately after tissue injury or infection, which then processes the maturation of dendritic cells to initiate an immune response in the lymph nodes. The purpose of this study was to discover the polymorphism of the AvBD2 gene in IPB-D1 chickens. PCR and direct-DNA sequencing methods were used to identify the diversity of intron 1, exon 2, and intron 2 AvDB2 genes in 47 chickens. Genotype and allele frequency, and heterozygosity calculations were carried out to obtain information of the AvBD2 gene polymorphism. A total of 10 single nucleotide polymorphisms were found in the AvBD2 gene located in intron 1 (g.4843T>A, g.4853G>A, and g.4859T>C), exon 2 (g.4881A>G, g.4889G>A, and g.5002C>T), and intron 2 (g.5075C>T, g.5111T>G, g.5116G>T, and g.5177G>T). All SNPs are polymorphic. The g.5002C>T mutation causes changes in the amino acid Ala to Val which has the potential to be a candidate for characterizing disease resistance in IPB-D1 chickens.

Abstract

Protein content in milk is an important indicator of milk. Accordingly, genetic improvement to produce Holstein Friesian (HF) dairy cattle is important. The objective of this study was to evaluate the genetic variant of milk protein genes and its effect on milk component traits of Holstein Friesian (HF). A total of 100 HF were used in this study. The HF cattle used have physiological status in the lactation period 1 up to 3 and lactation change of 1 up to 12 months. Genotype variants of milk protein genes were identified using Real Time-Polymerase Chain Reaction method.  Analysis of milk component was carried out covering the component of protein, fat, lactose, and solid non-fat (SNF) by using a milk quality measuring device (Lactoscan). Genotyping of cattle blood samples consisted of DNA extraction, genes amplification using the RT-PCR method. The result showed that protein milk was significantly affected (p<0.05) by the genetic variants of CSN1S1-192 and CSN2-67 genes. Fat milk was significantly affected (p<0.05) by the genetic variants of CSN1S1-192 and CSN3 genes.  Meanwhile, solid non-fat milk was significantly affected (p<0.05) by the genetic variants of CSN-BMC9215, CSN-BMC6334, CSN1S1-14618, CSN2_67, and CSN3 genes. Lactose milk was significantly affected (p<0.05) by the genetic variants of CSN-BMC9215 and CSN2-67 genes. It was concluded that genetic variants of the milk protein genes have an association with the component of cow's milk (protein, fat, solid non-fat, and lactose).

Abstract

Sound analysis has been carried out in various activities including identification and differentiation of species as well as the preparation of the taxonomy of some animals’ species because of several advantages, including no need to capture or too close to the subject observed. Analysis of voice used to differentiate and to estimate of breeds’ sheep genetic distance has not been reported. This research was conducted to study the character of a few breeds’ sheep sound and likely to be used as a predictor of genetic distance between breeds of sheep. The study was conducted in the Animal House at Indonesian Research Institute for Animal Production, Bogor.  A total of 20 head adult of five sheeps (St. Croix cross / SC, Barbados Black Belly cross / BC, Local Garut/LG, Composite Garut / KG and Composites Sumatra / KS) used in this study. Call sound recorded using a digital voice recorder. Sound analysis performed by Raven Software Pro 1.3 for Windows to count as many as 24 variables sound. Analysis of variance of each variable sound was performed using PROC GLM of SAS software Ver. 9.0. It used PROC CANDISC for canonical discriminant analysis and then PROC TREE to build a dendogram. The results showed that there were variations in amplitude, energy, power and frequency variables among the five breeds of sheep. By plotting canonical, LG, KS and BC sheep were from a different group. It was concluded that the sound characteristics variables which can be used as a differentiator breeds of sheep were the third quartile frequency, center frequency, maximum frequency and the first quartile time. Dendogram showed that KG sheep was in the less accurate group. Genetic distance estimation method using voice characteristic data may be applied on sheep. Abstract

The aim of this research is to know the influence of the number of follicles produced per ovary on the quality of oocytes and the day length of developing blastocytes by in vitro fertilization (IVF) at 7-,8-, and 9-days. Material of the research is ovary of Fries Holland dairy cattle gathered from slaughterhouse. A number of 138 ovaries are grouped into four based on the number of follicles per ovary, including group I (≤5 follicles), group II (6-10 follicles), group III (11-20 follicles), and group IV (≥20 follicles). The result showed that the number of follicles per ovary are not significantly (P>0.05) influencing both on oocyte quality and the days length of forming blastocytes at 7-, 8-, and 9-days. Number of oocytes for grade A-B (percentage of the number of oocytes IVF per the number of oocytes obtained per ovary) for group I - IV consecutively is 83.5%, 91.3%, 97.1%, and 91.2%. The number of developing blastocytes (percentage the number of blastocytes per the number of oocytes) for group I - IV at 7-days consecutively is 5.19%, 5.56%, 5.45%, and 6.03%; at 8-days is 15.58%, 12.94 %, 13.64%, and 11.64%; at 9-days is 1818%, 8.96%, 8.18%, and 10.35%.   Key words: Oocytes, IVF, blastocyte

Abstract

The bone morphogenetic protein 15 (BMP15) gene or commonly called FecX (fecundity chromosome X) is a gene that controls the prolific properties. This study aims to identify the mutation of BMP15 gene and to analyze its polymorphism in Boer, Kacang, and Boerka goats. The total of 50 female goat bloods were identified using PCR-Sequencing method, 17 Boer, 16 Kacang and 17 Boerka respectively. BMP15 gene amplification resulted fragment with the length of 141 bp. Genotyping of BMP15 gene produced three genotypes. The result showed that BMP15 gene found two polymorphic SNP were analyzed by genotype frequency, allele frequency, heterozygosis and equilibrium of genotype in all population was detected by the Hardy-Weinberg equilibrium test (χ2). Sequence analysis results of BMP15 gene showed that there were two mutation between adenine (A) and guanine (G) bases and determination of genotype BMP15 gene produces three genotypes showed that there were GG, GA and AA. In conclusion, there was found the mutation of BMP15 gene in Boer, Kacang and Boerka goats and genetic polymorphism were identified using PCR-Sequencing method.
