Abstract

Western blotting is an important technique used in cell and molecular biology. Using the western blot, researchers can identify specific proteins from the complex mixture of proteins extracted from cells. This technique uses three elements to accomplish this task: (1) separation by size, (2) transfer to a solid support, and (3) marking target proteins using appropriate primary and secondary antibodies to visualize. This paper will attempt to explain the techniques and theory behind western blot, and offer several ways to solve the problem

Abstract

A B S T R A C TTrials in pre-clinical trials are very complex multidisciplinary studies. Extrapolatingdata from animals to humans requires information from many fields of science. Frompre-clinical trials, more detailed information and data on efficacy and safety wereobtained, especially at doses equivalent to doses in humans, and the presence orabsence of cumulative effects and whether these effects could return to normal(reversible) after the administration of the test material was stopped. This test canbe used to predict the negative impact on humans if he is exposed to the materialfor a long time.

Abstract

Regardless of the method, it is essential to induce unconsciousness as rapidly as possible if euthanasia is to be aesthetically and scientifically successful. Criteria that have been considered in recommending the methods of euthanasia include: time required to produce unconsciousness, time required to produce death, purposes, research results and compliance with the AVMA Guidelines on Euthanasia. There are special considerations for euthanizing rodent embryos, fetuses and neonates.

Abstract

Recognition of pain depends upon intact pathways from pain receptors to the thalamus and cerebral cortex, as well as functional cerebral cortex and subcortical structures. Thus any means that renders the cerebral cortex nonfunctional, such as hypoxia or drug depression, prevents pain. When this happens, stimuli that evoke motor nerve reflexes that may be painful to the conscious animal are not painful in the unconscious animal. Equally painful stimuli administered to animals chemically paralyzed by curare or succinylcholine will not evoke a motor reflex simply because of paralysis, but will cause pain because of the conscious state. Hence, it is possible that unconscious animals may feel no pain but respond to certain stimuli, and paralyzed animals may feel pain but cannot respond.

Abstract

Mice are usually caught and lifted by the tail. The tail should be grasped abouttwo-thirds of the way down. With this simple method of holding, they may betransferred to another cage or a balance, identified or sexed; but such restraint isnot sufficient for treatment and close examination. For more effective control, themouse may be held by the tail and placed on a table or other surface, preferablyone that the mouse can grasp, and the loose skin over the neck and shouldersgrasped with thumb and fingers.

Abstract

Although significant progress has been made in cancer treatment in recent decades, the effectiveness of modern therapeutic approaches is often limited by their toxic effects on other organs. In addition, many people in the world have limited or unequal access to cancer treatment services. Therefore, utilizing information from traditional medicine systems to identify alternative methods to prevent and control cancer. The use of traditional medicine can lead to the discovery of new bioactive compounds and drugs that are available, safe and affordable. In Islamic medicine, cancer is known as a disease with many treatment options. Traditional Islamic medicine suggests several strategies for cancer control and treatment. Surgical manipulation, venesection, dietary adjustment, and use of natural medicines including solid, semisolid, and liquid dosage forms by oral and external routes of administration are among these methods. This literature study will presents cancer from the perspective of traditional Islamic medicine, its aetiology and management.

Abstract

Sambiloto (Andrographis paniculata) is one of the most common plants in Indonesia. Sambiloto contains quite varied secondary metabolites, where this plant is rich in flavonoids, alkaloids, terpenes and glycosides. This study aims to assess the effect of Sambiloto (Andrographis paniculata) leaf extract on blood sugar levels and the expression of GLUT4 protein in muscle tissue which indicates the potential of the test extract's ability to improve blood glucose intake to cells so that it can maintain blood sugar regulation. The process of extracting sambiloto is carried out by maceration in which 500 grams of simplicia are macerated with 96% ethanol for 72 hours. After 1 week of adaptation, the mice were randomly divided into the following six groups, each containing 5 animals: Normal control group, diabetes group (negative control), diabetes + metformin group (Met; 45 mg/kg), Diabetes + ES group (50 mg/kg), diabetes + ES group (100 mg/kg) and diabetes + ES group (200 mg/kg). The treatment with sambiloto extract was able to reduce blood sugar levels significantly, were at the ES 100 and 200 mg/kg BW doses it was able to reduce blood sugar levels to reach the target below 200 mg/kg BW. The dominant flavonoids in Sambiloto leaf extract are believed to be responsible for the effect of blood glucose regulation. In conclusion, Sambiloto extract affects lowering blood sugar levels in diabetes mellitus white rats by increasing glucose intake in cells and tissues.

Abstract

Diabetes mellitus is a chronic condition that disturbs the body's blood sugar regulation. Bay leaves contain entirely various secondary metabolites, and this plant is rich in flavonoids, alkaloids, terpenes, and glycosides. This study aims to assess the effect of bay leaf extract (Syzygium polyanthum) on blood sugar levels and the expression of GLUT4 protein in muscle tissue. A total of 30 white rats (Rattus norvegicus) Wistar strains were obtained from the Eureka Research Laboratory (Palembang, Indonesia) weighing between 200 – and 250 grams. Bay leaf Simplicia was obtained from the Tawangmangu Herbal Research Center, Karanganyar, Indonesia. After 1 week of adaptation, the mice were randomly divided into the following six groups, each containing 5 animals: Normal control group, diabetes group (negative control), diabetes + metformin group (Met; 45 mg / kg), Diabetes + BLE (75 mg/kg), diabetes + BLE group (150 mg / kg) and diabetes + BLE group (300 mg/kg). Alloxan-induced white rats showed a very significant increase in blood sugar levels, where the use of the drug metformin was able to reduce blood sugar levels significantly even though they had not reached the target blood glucose target of less than 200 mg / dL. The treatment with bay leaf extract was able to reduce blood sugar levels significantly. The administration of metformin drugs or bay leaf extract showed the ability to increase the level of GLUT4 protein. In conclusion, bay leaf extract affects reducing blood sugar levels in diabetes mellitus white rats by increasing glucose intake in cells and tissues.

Abstract

Cholesterol is an important precursor that plays a role in the production and regulation of various sterol group compounds, especially steroid hormone compounds, androgen hormone compounds, cortisol compounds, and estrogen compounds. This herb is known to have the effect of improving glucose regulation by increasing glucose intake into cells and tissues. Its ability to improve cell and tissue metabolism is mediated by the content of secondary metabolite compounds. This ability is believed to have the potential to improve cholesterol regulation. Bay leaf extract is effective in lowering cholesterol levels through regulation of Acetyl CoA production.
