Abstract

The chalcone compound is a precursor of flavonoid and pyrazoline compounds. Chalcone has two aromatic rings (A and B) and one ?,?-unsaturated carbon atom. Chalcone compounds can be synthesized via the Claisen Schmidt condensation reaction. Chalcone synthesis uses the basic ingredients of alpha tetralone and benzaldehyde analogues. In this research, chalcone synthesis took place for 48 hours at room temperature. Synthetic materials using ethanol solvent and sodium hydroxide catalyst, as well as basic ingredients alpha tetralone, 3,4-dimethoxybenzaldehyde and 4-methoxybenzaldehyde. Characterization of synthesized compounds using spectroscopic methods, including 1H-NMR, 13C-NMR, Fourier Transform Infrared (FTIR) and mass spectroscopy. Characterization results using spectroscopic methods showed that the two chalcone analogue compounds were successfully synthesized with yields above 70%. Both chalcone compounds showed low activity against MCF-7 cancer cells. Molecular docking simulations showed that both chalcone analogues produced energy levels and intramolecular interactions that were no better than 4-hydroxytamoxifen. Keywords:          chalcone analogues, alpha tetralone, cytotoxic   Abstrak Senyawa kalkon merupakan salah satu prekursor dari senyawa flavonoid dan pirazolina. Kalkon mengandung dua cincin aromatis (A dan B) dan satu atom karbon ?,?-tak jenuh. Senyawa kalkon dapat disintesis melalui reaksi kondensasi Claisen Schmidt. Kalkon disintesis menggunakan bahan dasar alfa tetralone dan analog benzaldehida. Dalam penelitian ini senyawa kalkon disintesis selama 48 jam pada suhu ruang menggunakan pelarut etanol dan katalis natrium hidroksida serta bahan dasar alfa tetralone, 3,4-dimetoksibenzaldehida dan 4-metoksibenzaldehida.  Senyawa hasil sintesis dikarakterisasi menggunakan metode spektroskopi meliputi 1H-NMR, 13C-NMR, Fourier Transform Infrared (FTIR) dan spektroskopi massa. Karakterisasi menggunakan metode spektroskopi menunjukkan kedua senyawa analog kalkon berhasil disintesis dengan rendemen diatas 70%. Hasil pengujian sitotoksik menunjukkan bahwa kedua senyawa analog kalkon tidak aktif terhadap sel kanker payudara MCF-7. Simulasi penambatan molekuler menunjukkan bahwa kedua analog kalkon menghasilkan tingkat energi dan interaksi intramolekuler yang tidak lebih baik dibandingkan dengan 4-hidroksitamoksifen. Abstract

Rice washing water contains carbohydrates, vitamins, and minerals that serve as potential substrates for the growth of Lactic Acid Bacteria (LAB). This study aims to isolate and characterize LAB from the traditional East Kalimantan rice variety Beras Mayas. LAB isolation was performed using De Man Rogosa and Sharpe (MRS) Agar, followed by successive purification to obtain pure cultures. Identification of the isolates involved macroscopic and microscopic observations, Gram staining, and a series of biochemical assays. Three distinct LAB isolates, designated A, B, and C, were obtained. Isolates A and B exhibited circular colonies with yellowish-white pigmentation and undulate margins, suggesting affiliation with the genus Lactobacillus. Isolate C formed circular white colonies with entire margins, indicating a possible relationship with the genus Streptococcus. Biochemical testing revealed that all three isolates were negative for catalase, gelatin hydrolysis, motility, and indole production. In the Triple Sugar Iron Agar (TSIA) test, all isolates fermented glucose, lactose, and sucrose, producing yellow coloration in both the slant and butt portions of the medium. These results suggest that the isolates are homofermentative LAB.

Abstract

Rambutan (Nephelium lappaceum L.) seeds have long been utilized in traditional medicine due to their potential therapeutic properties. This study aimed to determine the secondary metabolite profile, toxicity, and antioxidant activity of the methanolic extract and ethyl acetate fraction of rambutan seeds to strengthen their potential application in traditional medicine. The powdered of rambutan seed was extracted with a maceration technique using methanol, followed by fractionation using n-hexane and ethyl acetate. The methanol extract and ethyl acetate fraction were tested for toxicity using the Brine Shrimp Lethality Test (BSLT) and for antioxidant activity using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay. Phytochemical screening was performed using standard qualitative method to identify secondary metabolites, which confirmed the presence of phenols, tannins, and flavonoids in both samples. Toxicity testing indicated LCâ‚…â‚€ values of 688.86 ppm for the methanol extract and 825.68 ppm for the ethyl acetate fraction. Antioxidant activity assessment revealed ICâ‚…â‚€ values of 328.61 ppm for the methanol extract and the higher activity for ethyl acetate fraction of 32.78 ppm. These results indicate that the ethyl acetate fraction of rambutan seeds may serve as a promising natural source of antioxidants for therapeutic development.

Abstract

The anti-inflammatory effect of fart plant leaf extract (Paederia scandens L.) on white rats (Rattus norvegicus) has been carried out. This research shows that the effective dose of the n-hexane fraction of P. scandens leaf extract provides the best anti-inflammatory effect on white rats' feet soles. The study used an artificial edema formation method and periodically measured the volume of white mice's legs using a plethysmometer. The doses used for the extract are 100mg/200g BW, 150mg/200g BW, and 200mg/200g BW. The research data was observed visually. The effective dose of P. scandens leaf extract, which has the best anti-inflammatory effect, is 150mg/200g BW.

Abstract

This study aims to determine the yields and free radical scavenging activity based on IC50 value of the extract and the fraction of Lagerstroemia speciosa Pers leaves. The leaves of L. speciosa were macerated using 96% methanol and then practioned using different kinds of solvent based on its polarity including n-hexane, ethyl acetate, and n-butanol. The result showed that the yield of methanol extract obtained from L. speciosa levaes was 5.45% of dried samples obtained, the fraction of n-hexane 19.12%, the fraction of ethyl acetate 4,7%, and the fraction of n-butanol 1.93%. Testing of antioxidant activity is performed with the 1,1-Diphenyl-2- Picryl Hydrazyl (DPPH) by spectrophotometry UV-Visible. Absorbanse measurement was conducted with spectrophotometry at the wave lenght of 516 nm. Data was analyzed with linear regresion to obtain value of IC50. The result of research showed that the extract and fractions of L. speciosa leaves have activity as an antioxidant with the IC50 value of methanolic extract L. speciosa leaves was 12.5 ppm, n-hexane fraction was 18.75 ppm, ethyl acetate fraction was 21.87 ppm, n-butanol was 8.37 ppm.

Abstract

Honey kelulut is derived from the kelulut bee (Trigona sp.), which is native to Kalimantan and belongs to the Trigona genus of stingless bees. The honey produced by the Trigona genus has a well-documented antibacterial property. Study objective accomplished. This text discusses the potent antibacterial action of kelulut honey. The antibacterial testing method employed is the well diffusion method, utilizing the bacterium Staphylococcus epidermidis as the test organism. An antibacterial test was conducted by measuring the diameter of the zone of inhibition against bacteria. The results were obtained from concentrations of 40%, 60%, 80%, and 100%. Then the resistance diameter was measured for each concentration. The most optimal outcomes were achieved in the antibacterial test using honey kelulut. The honey kelulut with an 80% concentration exhibited a medium resistance level of 10.2 mm, while the 100% concentration demonstrated a strong resistance level of 13.7 mm.

Abstract

Naphthalene derivative compounds named Eleutherol (1) and Eleutherol C (2) have been isolated from Bawang Tiwai Bulb Eleutherine bulbosa. We tested its cytotoxic activity against T47D breast cancer cells in vitro after isolating compounds 1 and 2 from the EtOH extract using conventional chromatography methods. Their chemical structures were elucidated based on spectroscopic analysis, including IR, HR-TOFMS, 1D, and 2D NMR, and by comparison to those related spectra previously reported. Compounds 1 and 2 were tested for their cytotoxic effects against T47D breast cancer cells and showed moderate cytotoxicity against T47D breast cancer cells with IC50 values of 117.15 and 80.21 µM, respectively, compared with cisplatin 24.07 µM.
