Abstract

ABSTRACTCommercial varieties of chili is still susceptible to anthracnose disease. Anthracnose is one of diseases which caused yield loss about 10-80% in rainy season and 2-35% in dry season. One of the disease control method is to develop new varieties resistant to anthracnose through gamma-ray irradiation. This research was aimed to determine radiosensitivity level and heritability of chili which generated by mutation inductions of three genotypes: IPB C2, IPB C10, and IPB C15. Seed of the three genotypes were irradiated by gamma rays at dosages of 100, 200, 300, 400, 500, 600, 700, 800, 900 and 1,000 Gy. The LD50 in IPB C2, IPB C10, and IPB C15 were 317.9, 591.4, and 538.8 Gy, respectively. The character of IPB C2 mutants ranged between very susceptible to resistant against anthracnose disease, IPB C10 tended to very susceptible to highly resistant, and genotype IPB C15 tended to susceptible to highly resistant. Genotype IPB C2 had high level in heritability for disease resistance character, while medium level in heritability value was obtained on genotype IPB C10 and IPB C15.Keywords: genotype, LD50, mutant, mutation, seed

Abstract

ABSTRACTIdentification of fruit and seed characters when they reach physiological maturity is very important to get high qualiy chili seed. The purpose of the research was to study the fruit characters and to determine seed physiologycal maturity of six chili genotipes (Capsicum annuum L.) i.e. Anies 1, Anies 2, Seloka 1, Seloka 2, Seloka 3 and SSP. Characterization was assayed at 32, 35, 38, 41, 44 DAA. Chili seed was taken from two groups of branches, lower branches (2nd-5th branches) and upper branches (6-7th branches). The result revealed that seed physiological maturity was achieved at 38-44 DAA, that was indicated by changes of the fruit color from brown to red (Anies 1) and dark green to red on five others genotipe; maximum seed dry weight, seed viability and vigor. When the seed reached physiological maturity, SSP’s had the longest fruit but fruit diameter and weight, seed number and seed weight per fruit was the lowest. However, SSP seed rendement was the highest. Flowers of lower branches developed faster, their fruits size, seed dry weight, and seed number was significantly higher than the upper branches. Nevertheles the seed viability and vigor was not different between lower and upper branches.Keywords: fruit color, seed dry weight, seed viability, seed vigor

Abstract

ABSTRACTPhalaenopsis amabilis (L.) Blume and Phalaenopsis amboinensis J.J. Smith (diploid) are important in Phalaenopsis breeding. Polyploid species are needed for crossing with polyploid hybrid varities of Phalaenopsis. The objectives of this study were to obtain effective concentration of colchicine to induce polyploidy and to produce polyploid plantlets of P. amabilis and P. amboinensis. Experiment was arranged in randomized complete block design with one factor, the colchicine concentration. Protocorms of P. amabilis and P. amboinensis were immersed in half strength of Murashige-Skoog (1/2 MS) liquid media added with colchicine (0; 0.5; 5; 25; 50, and 75 mg L-1) for 10 days. The results showed that higher concentration of colchicine on both species did not have significant effect on the survival of the plantlets at 24 weeks after treatment. The average number of leaves and roots of colchicine treated planlets from both species were less than the control plantlets. Immersing protocorm in colchicine at concentration of 50 mg L-1 for 10 days was effective in inducing polyploid plantlets of P. amabilis and P. amboinensis with the frequency of 33.3% and 40%, respectively. Polyploid plantlet has larger stomata size and lower stomata density than the diploid ones.Keywords: chromosome number, colchicine, polyploid, protocorm, stomatal density, stomatal size

Abstract

Selection is very important activity in plant breeding program. The purpose of this study was to obtain genetic information, heritability and expected genetic advance, and to compare the genetic advance expectations with the selection advance. The research was conducted from September 2013 to November 2015 at Research Station of Leuwikopo, Dramaga, Bogor. Distribution of F2 population data in this study was broader than that of F3 populations. Broad sense heritability estimated by F3 populations showed in agreement to that estimated by F2 population. Character of fruit weight, fruit length, pedicel length and yield had positive value of selection advance appropriate to the genetic advance expectations in F2 populations. The value of heritability estimate and genetic advance were high indicating that the phenotypic variances were controlled by action of additive genes.Keywords: genetic advance, heritability, selection advance

Abstract

ABSTRACTKenikir (Cosmos caudatus Kunth.) is an Indonesian indigenous vegetable which is potential to be developed. Exploration conducted in Bogor, Sukabumi, Bandung, Bandung Barat, Subang, Garut, Majalengka, Kuningan, and Tasikmalaya had succesfully collected 20 accessions of Cosmos sp. The objectives of this study were  to observe similarity and estimate the yield of Cosmos accessions from West Java. Clusters analysis grouped 20 Cosmos accessions into three clusters. Cluster I consisted of Pendeuy, Saribakti, Karang agung, Sindangbarang, Langensari, Perbawati, Sudajaya girang, Karang tengah, Argalingga, Warnasari, Sukaresmi, Ciwidey, Jalan cagak, Lebaksiuh, Tugu selatan, Ciwarak, Linggarjati and Babakan accessions. Cluster II and III consisted only one accession each which were Ciaruteun and Dramaga, respectively. Seven accessions from different sub cluster were evaluated to estimate the yield. A significant variability was found among the accessions. Results showed that those accessions were significantly different on plant height, stem girth, numbers of primary branches, number of leaf, leaves width, leaves length, days to flowering and yield. The results showed that Dramaga and Ciaruteun acessions were recomended to be developed because of its high yield. Keywords: characterization, cluster analysis, exploration

Abstract

Induction of polyploid gametes is one of useful plant polyploidization methods. Some of its benefits are to obtain triploid and tetraploid progenies at the same time by cross and self pollination. Previous research showed that some morphological characters which could be the indications of polyploidy plantlets before the analysis of chromosome number. Colchicine treatment on flower bud of diploid Phalaenopsis amabilis was conducted to determine the effect of colchicine on flower bud development, plantlets morphology and growth, and potential of polyploidy induction based on plantlets morphology. Colchicine concentrations in the experiment were 0, 50, 500, 1,000, and 2,000 mg L-1, with three days duration of treatments with aluminium foil wraps on flower buds. The results showed that high colchicine concentrations (2,000 mg L-1) inhibited flowers blooming of treated flower buds. Based on morphological characters, plantlets were classified into normal and putative polyploid plantlets. Putative polyploid plantlets from colchicine with the concentration of 50, 500, and 1,000 mg L-1 were 71.2, 86.4, and 100.0% respectively.Keywords: colchicine concentration, morphological characters, normal plantlets, putative polyploidy, reproductive organ

Abstract

ABSTRAK Development of black rice is quite rapid in Indonesia. The objective of this study was to obtain doubled haploid black rice lines with good agronomic characters through anther culture. Materials used in this experiment were 6 F1 derived from black rice landrace (Melik) and two cultivated varieties (Inpari 13 and Fatmawati): Melik/Inpari13//Inpari13, Melik / Inpari13// Melik, Melik/Fatmawati //Fatmawati///Fatmawati, Melik/ Fatmawati//Fatmawati// /Fatmawati, Melik/Fatmawati, and Melik/Fatmawati//Melik. The experiment was performed using a completely randomized design consisting of 12 replications. An experimental unit was a petri dish containing ± 150 anthers. Results of the anther culture varied widely between crosses. Crosses Melik/Inpari13//Melik, Melik/Fatmawati//Melik, and Melik/Inpari13 //Inpari13 showed the best response on callus induction and plant regeneration compared to the other crosses. Melik / Inpari13 // Melik produced  the highest number of acclimatized plants, the highest number and percentage of doubled haploid: 63 plants, 42 plants and 91.3% respectively. The highest number of doubled haploid having black aleuron layer were obtained by Melik/Inpari13//Melik and Melik/Fatmawati//Melik.Keywords: anther culture, black rice, doubled haploid, regeneration

Abstract

ABSTRACTCurcuma aeruginosa Roxb., popularly known as “temu ireng”, is considered as a potential source of medicinal plant for pharmacological activities. However, varieties of C. aeruginosa are still limited in Indonesia so it needs more accessions for improvement and development of new varieties. Rhizome colors, phytochemical contents and extract yield from 20 promising lines of C. aeruginosa were investigated by qualitative method for rhizome colors and phytochemical contents, and maceration method using 70% ethanol for yield extract. Similarity analysis was used for cluster analysis based on rhizome colors, phytochemical contents and yield extract. Blue was the color characterization of rhizome C. aeruginosa. The extract yield for 20 promising lines of C. aeruginosa varied from 7.92 to 19.71%, with KN and BH promising lines having the lowest and highest value, respectively. All promising lines of C. aeruginosa contain saponin and triterpenoid. Based on similarity analysis, all promising lines could be divided into 3 clusters. Cluster I consisted of 14 promising lines i.e. WG, SH, KA, GD, BH, KP, NW, PW, MB, PR, PT, KN, MD, and PK. Cluster II consisted of 4 promising lines i.e. LC, CB, KL, and GK. Abstract

Eggplants (Solanum melongena L.) are consumed worldwide in a variety of colors, shapes and sizes, thus vegetable plant breeders place great interest in developing high yielding genotypes in response to the population growth and the need to increase vegetable production. This research was aimed to determine the proper character as selection character for breeding high yielding eggplants by using estimated heritability value, coefficient of genetic diversity, characters correlation and path analysis. The experiment was conducted during 2014 to August 2015 at Cikabayan Experimental Field, Tajur Experimental Field and Pasir Sarongge Experimental Field, IPB, West Java. A total of 12 characters was observed in the 25 genotypes. Eleven characters has a high estimated heritability value and high coefficient of genetic diversity value. Those characters were time of flowering, time of harvest, fruit length, fruit weight, fruit diameter, plant height, stem length, fruit apical scar length, calyx length, the number of fruits per plant and fruit weight per plant. Results of correlation and path analysis showed that fruit length, fruit weight, fruit diameter, stem diameter, calyx length and number of fruits per plant have a direct positive effect on fruit weight per plant. Fruit weight, fruit diameter and number of fruits per plant are suitable characters in determining the selection criteria for developing high productivity eggplant.Keywords: path analysis, correlation, heritability, genetic diversity coefficient

Abstract

The objective of this study was to identify the stability of seven hybrid chili pepper genotypes that have been developed at Genetics and Plant Breeding Laboratory, Department of Agronomy and Horticulture IPB. The study used eight yield stability analyses and Additive Main Effect Multiplicative Interaction (AMMI) methods. The design was randomized complete block design with three replications as blocks using the genotypes of IPB CH1, IPB CH2, IPB CH3, IPB CH5, IPB CH25, IPB CH28, IPB CH50, and fi ve commercial varieties, i.e. Adipati, Biola, Gada, Hot Beauty and Imperial. These genotypes were planted at six different locations at Ciherang, Leuwikopo, Tajur, Subang, Rembang and Boyolali. IPB CH28, IPB CH25, IPB CH1 and IPB CH2 were more stable cultivars than IPB CH3, IPB CH5, IPB CH50, Adipati and Biola, which had 10, 9, 8, and 6 out of all 10 stability statistics used, respectively. IPB CH28 and IPB CH25 being the most stable cultivars. IPB CH3 was the best genotype compared to the checks based on pair wise GxE interaction test. Based on post predictive success, the AMMI2 model was able to explain 85.51% of the interaction-infl uenced variation. The stable genotypes in six locations were IPB CH1, IPB CH2, IPB CH25, IPB CH28, and IPB CH50. IPB CH3 genotype was locally adapted for Subang.
