Abstract

ABSTRACTBarnyard grass (Echinochloa crus-galli (L.) P. Beauv.) is a major weed competitor to rice production in Indonesia. In order to develop integrated weed management program, a research to select competitive rice lines to E. crus-galli was conducted in a green house of Indonesian Centre of Agricultural Biotechnology and Genetic Resource Research and Development Bogor. The research design was split plot with four replications, E. crus-galli was designed as the main plot (rice without E. crus-galli compared  rice with four E. crus-galli per pot), and the sub-plots were 25 genotypes (23 lines, 1 tolerant variety and 1 sensitive variety). Level of tolerance was determined by the reduction percentage of grains weight, the number of productive tillers and dry matter weight. The results showed that rice competition with E. crus-galli reduced plant height, productive tiller numbers, filled spikelet numbers per panicle, dry matter weight and dry grain weight. Three lines, i.e., IR10L-155, IR10L-133 and BIO-R84-1 were classified as tolerant, 19 lines were moderate and 3 lines were sensitive to E. crus-galli competition.Keywords: Barnyard grass, competition, sensitive line, yield reduction

Abstract

ABSTRACTThe breeding of rainfed rice tolerant to drought can be accomplished using anther culture. The objectives of this research were to determine regeneration abilities of six F1 anther culture and its acclimatization ability. The experiment was arranged in completely randomized design with 14 replications. The treatments consisted of six F1 derived from crossing:  INPARI 18 x IR83140-B-11-B (G1), INPARI 18 x B12825E-TB-1-25 (G2), INPARI 18 x IR87705-14-11-B-SKI-12 (G3), INPARI 22 x IR83140-B-11-B (G4), Bio-R81 x O18b-1 (G5), Bio-R82-2 x O18b-1 (G6). Media for callus induction was based on N6 medium + 2.0 mg L-1 NAA + 0.5 mg L-1 kinetin + 1.0 mM putresin + 60 g L-1 sucrosa, media for regeneration was based on MS + 0.5 mg L-1 NAA + 2.0 mg L-1 kinetin + 1.0 mM  putresin, and media for rooting was based on  MS + 0.5 mg L-1 IBA + 30 g L-1 sucrosa. The result indicated that all six F1 had different ability in anther culture. Bio-R82-2 x O18-b1 (G6) and  Bio-R81 x O18-b1 (G5) F1 genotype had good response both of callus induction and plant regeneration. These two F1 genotypes also gave the highest ratio of green planlet production to number of anther inoculated (GP:AI) were 5.50% and 4.65%,  respectively. In this research, there were identified doubled haploid plants were developed from 4 F1 derived cross namely G2 (2 plants), G3 (4 plants),  G5 (21 plants), and G6 (26 plants).Keywords: Callus induction, doubled haploid, rice

Abstract

ABSTRACTThe use of probiotic bacteria as biocontrol agents is one of the methods of controlling anthracnose disease caused by Colletotrichum acutatum. The objective of this research was to suppress the infection of C. acutatum and increase chilli pepper seedling vigour. The research involved factorial experiments arranged in a completely randomized design with two factors. The first factor was seed coating involving six treatments, i.e., control, seed coating without bacteria, seed coating with Bacillus sp., Pseudomonas sp., Actinomycetes sp, and fungicide. The second factor was the seed storage period where six storage periods were experimented, i.e., 0, 1, 2, 3, 4, 5, and 6 months. The results showed that the coating treatment of Bacillus sp., Pseudomonas sp., and Actinomycetes sp. improved germination, growth rate and number of leaves and reduced the incidence of attacks and infection of hypocotyls at 5 month storage period.Keywords: Actinomycetes sp., Bacillus sp., Pseudomonas sp., seed coating, storage period

Abstract

ABSTRACTThe use of high quality seed is one of the key factors to improve productivity. Probiotic bacteria has been used to increase plant growth and to control pathogens. The objective of the research was to evaluate methods of chili seed production that yielded high physiological and pathological quality using probiotic fluorescent bacteria Pseudomonas (P24). The bacteria was expected to function as plant growth promoting bacteria as well as capable of controling seedborne pathogen Colletotrichum acutatum causes antrachnose. The experiment was conducted during March until October 2014 in Seed Health Laboratory, Plant Bacteriology Laboratory, and Leuwikopo experimental garden, IPB. The experiment was arranged in a split plot randomized complete block design with three replications. The main plots were untreated and inoculation of C. acutatum. The subplot was six treatments of fluorescent Pseudomonas (P24) application. The results showed that matriconditioning and spraying of fluorescent Pseudomonas (P24) on nursery decreased disease incidence significantly. Matriconditioning and spraying of fluorescent Pseudomonas (P24) on nursery and flowering phase increased number of healthy fruit and total weight of seeds per plant significantly. These applications also increased seed physiological  quality indicated by germination percentage (GP) 77.04%, growth rate (GR) 9.72% etmal-1, vigor index (VI) 29.74%, and seed health by suppresed C. acutatum infection up to 12.25%.Keywords: anthracnose, Colletotrichum acutatum, control, matriconditioning, spraying

Abstract

Knowledge of the magnitude of genetic variability, heritability and genetic advance in the selection of desirable characters could assist the plant breeders in ascertaining criteria to be used in the breeding programmes. Twenty three introgressed oil palm progenies were evaluated at the Specialized Centre for Oil Palm Research of Cameroon, from 2004 to 2014 to estimate performance, genetic variability, heritability and genetic advance of oil yield and some economic traits in terms to select new oil palm parent materials with the traits of interest. The results revealed high variability among oil palm population for all the characters. Moderate estimates of the phenotypic and genotypic coefficient of variations associated with high heritability and moderate genetic advance as percent of mean were obtained for characters of fresh fruit bunch, bunch number, oil yield, kernel to bunch, kernel to fruit, kernel yield and height increment. The results suggest the effectiveness of selection method for these traits and their improvement through their phenotypic performance. LM11087T x LM2749D and LM12960T x LM7409D were obtained as superior oil palm crossing parents with the potential production of 6.26 ton ha-1yr-1 of crude palm oil; they can be exploited in seed production and further breeding program.Keywords: genetic advance, heritability, introgressed progenies, oil yield, Phenotypic

Abstract

The objective of the study was to identify the combining ability of several introduced and local chilli genotypes and heterosis effects of their offsprings, as a result of the hybridisation process. This information will be used for production of hybrid varieties. The study was conducted from November 2006 to May 2007 at Tajur experimental station, Bogor, West Java, Indonesia. The fi ve parents with their ten F1 hybrids were evaluated in a randomized complete block design (RCBD) with three replications. The results showed that hybrid IPB C15 x IPB C19 had the highest fruit weight per plant, and hybrid IPB C8 x IPB C19 had the highest heterosis for this character. Hybrid IPB C8 x IPB C15 has highest number of fruits per plant and showed heterosis effects. Crosses among introduced and local genotypes resulted a high heterosis values. Additive variance of weight and number of fruits per plant were greater than the dominant variance. IPB C19 had the best general combining ability for fruit weight per plant character. Abstract

ABSTRACTThe fruit of ornamental chili functioned as ornamental as well as for consumption, and therefore it requires diversity of traits for selection process. Information on inheritance pattern of the traits are needed for effective selection. The ideotype of ornamental chili are shortened internode that form a bouquet of flowers, erect fruit orientation and contain anthocyanins for attractive appearance. The research was aimed to study qualitative characters inheritance associated with shortened internode, fruit orientation and anthocyanins content. This study used 6 population including female parent (P1) which has anthocyanin in flower parts, the male parent (P2) bird pepper which has the character of a shortened internode, F1, F1R, BCP1, BCP2, and F2. Data were analyzed with Chi-square test to determine the Mendelian ratio in the F2 population. The results showed that there were characters that was controlled by one gene or two genes. Shortened internode and erect fruit orientation were controlled by a single recessive gene with a ratio of 1:3. On the other hand the color of anthocyanin in the anther stem was controlled by one dominant gene with a ratio of 3:1. The characters controlled by two genes that were dominant and recessive epistasis was the color of anthocyanins in the anther with the ratio 13:3.Keywords: Mendelian ratio, antosianin, shortened internode

Abstract

Characters on hypocotyl dan cotyledon are very potential used for effective and efficient morphology marker for some crop plants. However, the information about the inheritance of qualitative and quantitative characters on hypocotyl and cotyledon of chili pepper was not available. The aim of this research was to determine the inheritance of qualitative and quantitative characters on hypocotyl and cotyledon of chili pepper. This research used purple chili pepper (P1 (IPB C20)), green chili pepper (P2 (IPB C2)), F1, F1R, BCP1, BCP2, and F2 populations. Analysis for qualitative characters used mendelian genetics analysis, whereas model genetic prediction for quantitative characters used join scaling test analysis. The result showed that hypocotyl and cotyledon color of chili pepper controlled by single gene. The gene controlling purple color was dominant than the gene controlling green color on hypocotyl, whereas  the gene controlling green color was dominant than the gene controlling purple color on cotyledon. The additive-dominant genetic model with influence of additive-additive and additive-dominant interaction was suitable for hypocotyl length. The additive-dominant genetic model with influence of additive-dominant and dominant-dominant interaction was suitable for hypocotyls diameter. The additive-dominant genetic model with influence of additive-additive and dominant-dominant interaction was suitable for cotyledon width and cotyledon length.  Keywords: genetic model, morfology marker, natural cross-pollination

Abstract

Pohpohan (Pilea trinervia Wight.) is potential indigenous vegetable to be developed as a commercial vegetables. The objectives of this research were to observe similarity and production potential of pohpohan landraces from several areas in West Java. Result based on cluster analysis at nine similarity scale, thirteen exploration pohpohan landraces were grouped into three clusters. Clusters I consisted of Warung Loa, Tugu Selatan, Palasari, Langensari, Kayu Ambon, Lebak Muncang, Situsari, Sukalilah and Lebaksiuh. Clusters II consisted of Curug Rendeng, Argalingga and Linggarjati and clusters III was Bobojong. Result from further test using Duncan Multiple Range Test (DMRT), Warung Loa is a leading landrace because showed the best result on plant height, primary branches number, leaf width, yield per plot and plant productivity. Abstract

Cassava is one of the important starch producer crops. Although Indonesia ranks as the third largest cassava producer country, Indonesia still imports cassava starch to meet the domestic demand in food and non-food industries. Gamma ray irradiation has been implemented to generate cassava mutants in order to support development of superior cassava variety, especially high yield and high starch content, especially high yield and high starch content. The objectives of this research were to evaluate the yield potential and the starch content of gamma irradiated 40 putative cassava mutants at M1V4 generation. This research was arranged in randomized complete block design with three replications and conducted at Cikabayan Experimental Field, IPB, Bogor, from May 2016 to March 2017. The starch content was measured using gravimetric methods. The results showed that five mutants, G2142, G3151, G2141, G1143, and G3111, had higher tuber weight per plant (6-7 kg per plant) compared to the background ‘Gajah’ genotype (4.7 kg per plant). ‘G2112’ mutant genotype showed higher starch content (23.86%) compared to the background ‘Gajah’ genotype (17.65%). The specific gravity (SG) was positively correlated with starch content (r = 0.905), indicated that the higher the SG, the higher the starch content of cassava.Keywords: gravimetric, mutant, specific gravity (SG), yield
