Abstract

Black nightshades [Solanum nigrum (L.)] is an indigenous vegetable originatied from tropical and subtropical regions. In Indonesia, black naightshades widely spread in Java, especially West Java. Black nightshades has a bitter taste caused by the content of tannins. The objective of this study was to identify and analyze the morphological and tannin content of 20 black nightshades accessions. The research was started with germplasm exploration in November 2015-February 2016, followed by planting and analysis until September 2016 at a laboratory of the Center for Tropical Horticultural Studies (PKHT) IPB. The experiment was arranged in a single-factor randomized complete block design with three replications. The results from cluster analysis based on morphological variability showed that the accessions of black nightshades can be grouped into two groups. The accessions had different fruit tannin contents, and some accessions can be selected for breeding material.Keywords: group analysis, indigenous vegetable, qualitative character, quantitative character

Abstract

The information on birds eye chili (Capsicum frutescens L.) resistance to melon aphids (Aphis gossypii) is rarely reported. Objectives of this research were to evaluate genetic variability of 19 genotypes bird chili and identify the potential genotypes resistant to A. gossypii. This research consisted of two experiments. The first experiment was conducted in April-December 2015 at Leuwikopo Experimental Field, Faculty of Agriculture IPB. Genetic materials used were 19 genotypes of bird chili from Plant Breeding Laboratory IPB which were planted from seedling stage until harvesting stage. The second experiment was conducted in May 2017-May 2018 at Faculty of Agriculture, IPB Darmaga Campus. Material used were 19 genotypes of bird chili at seedling stage, and melon aphids which were collected from Leuwikopo Experimental Field IPB. Melon aphid infestation method used no-choice test. The results showed the genetic variability of bird chili had more than 0.30 coefficient of dissimilarity. Genetic variability for canopy width, anther length, fruit weight, and fruit length were classified to broad, but total fruit weight per plant was classified to narrow. There were six genotypes clasified resistant to melon aphid, i.e., C332, C343, C333, C334, C290, C346. The potential genotypes for breeding program of bird chili resistant to melon aphid were C343 and C334.Keywords: Aphis gossypii, Capsicum frutescens, cluster, resistant variety

Abstract

Genetic information plays an important role in plant selection. This research was aimed to obtain genetic information on pattern inheritance of yield and yield component characters of two crosses population of birds chili. The research used biparental mating design consisted of 40 plants of parents population (P1 and P2), 40 plants of first generation (F1) and reciprocal (F1R) populations, 100 plants of backcross populations (BCP1 and BCP2), and 300 plants of second generation population (F2). Scaling test and joint scaling test indicated the presence of gene interaction (epistasis) in two crosses population. Estimates gene effect also showed that fruit weight character in crosses population of IPB C145 x IPB C174 was controlled by duplication epistasis, while yield per plant character in crosses population of IPB C145 x IPB C291 was controlled by complementary epistasis. All characters of both crosses populations had high broad sense heritability, but the characters in crosses population of IPB C145 x IPB C174 had medium to high narrow sense heritability, while the characters in crosses population of IPB C145 x IPB C291 had low to medium narrow sense heritability. Genetic difference between parents seemed to affect gene action of a character, in which yield per plant, number of fruits per plant, fruit weight, and fruit length characters in crosses population of IPB C145 x IPB C291 with small genetic difference were more controlled by the action of non additive gene than crosses population of IPB C145 x IPB C291 with large genetic difference.Keywords: epistasis, gene action, genetic difference, heritability, yield

Abstract

Improvement of chilli’s quantitative traits requires several stages of breeding program, one of these is selection stage. The objective of this study was to obtain the information concerning the means and ranges of some quantitative characters, inbreeding depression, heritability estimates, and response to selection from six generations derived from crossing two parents of chili peppers. Materials used were IPB C160 (female), IPB C291 (male) as parents, F2, F3, F4, F5, and F6 derived from crossing of these parents with a total number of 630 plants. Results from this study showed that the mean valuesbetween the generations F2 to F6 showed an increase in the observed character except of harvesting time. The highest value of inbreeding depression for harvest time, weight per fruit, and fruit length was found in F5 generation. The highest value of inbreeding depression for fruit weight per plant, number of fruit per plant, and fruit weight was found in F6 generation, and for fruit diameter was found in F4 generation. The narrow-sense heritability estimates were classified into low to moderate criteria. Abstract

Polyploidy induction in Phalaenopsis amabilis is one of the methods to increase plant genetic diversity. A highly meristematic condition of protocorm induced by cytokinines may improve the success rate of polyploidy induction. This experiment was aimed to study the effect of Benzylaminopurine (BAP), Thidiazuron (TDZ) and coconut water (CW) in a half concentration of Murashige & Skoog (MS) medium before colchicine treatment on growth, multiplication and the success rate of polyploidy induction on Phalaenopsis amabilis protocorm. The experiment used completely randomized  design with one factor, medium treatment (15% of CW); 0.51 mg L-1 BAP; 0.50 mg L-1 TDZ; 15% CW + 0.51 mg L-1 BAP; 15% CW + 0.50 mg L-1 TDZ). Each treatment consisted of three replications with five culture bottles of protocorm for each replication and ten protocorms per bottle. The protocorms were incubated for eight weeks in the medium before treated with 50 mg L-1 of colchicine for ten days. The results showed that 15% CW, 15% CW + 0.51 mg L-1 BAP or 0.5 mg L-1 TDZ gave the highest percentage (100%) of alive protocorm. The highest percentage of protocorm multiplication were resulted from 15% CW+0.51 mg L-1 BAP (65.3%) and 15% CW + 0.5 mg L-1 TDZ (62.7%). The best treatment to promote formation of leaves and roots from protocorm was 15% CW. The percentage of polyploid plantlets of 3% was found in plantlet from 15% CW + 0.50 mg L-1 TDZ media treatment while from other treatments could not be detected.Keywords: BAP, chromosome, coconut water, plantlet, TDZ

Abstract

Breeding improved yardlong bean varieties is not only aimed for yield, but also for enhancing nutritional content such as anthocyanin and carotenoids concentrations. The objectives of this research were to elucidate the anthocyanin and carotenoids concentrations as well as yield components in several yard long bean genotypes. This research was conducted at the Leuwikopo Experimental Station and Post Harvest Laboratory of Department of Agronomy and Horticulture IPB from November 2017 to January 2018. The experiment was arranged in a single-factor randomized complete block design with 4 replicates, involving 15 yardlong bean genotypes. The results showed that Fagiola 2 had relatively high anthocyanin and carotenoids contents. Peleton had higher yield than Kinaya 1, Pangeran Anvi, Parade, and Fagiola 2. Anthocyanin content was not correlated with yield components, whereas carotenoids content was positively correlated with the length of pod stalks, length of pods, weight per pod, chlorophyll a, chlorophyll b, and total chlorophyll. Pod weights per plant were positively correlated with the number of pods per plant (r = 0.81, P<0.01), and can be predicted by a linear model involving number of pods per plant and days to harvest (R2 = 0.75, P<0.01).Keywords: biofortification, chlorophyll, correlation, stepwise regression, Vigna unguiculata var. sesquipedalis

Abstract

The fruit thinning treatment is known to be able to improve seed quality. This research aimed to increase the yield component and seed quality on two okra varieties (Naila and Zahira) through fruit thinning. This experiment was conducted in May until September 2018 at the Leuwikopo IPB Field, Postharvest Laboratory, and Seed Testing Laboratory, Department of Agronomy and Horticulture, Faculty of Agriculture, Bogor Agricultural University. This experiment conducted using the two-factors nested design, which is a fruit thinning factor as subplots consisting of five levels and okra varieties consisting of two levels. The results showed that the fruit thinning factor influenced the parameters of yield components (fruit length and weight per fruit) and 1,000 seeds weight. The variety factor influenced most of the yield component parameters and seed quality in okra plants. The thinning treatment of the first two fruits and elimination of one fruit interval resulted in the highest physical quality of seed based on 1,000 seeds weight (67.99 g). Abstract

Varietas cabai yang stabil dan berdaya hasil tinggi diperlukan untuk peningkatan produksi cabai nasional. Penelitian ini bertujuan untuk mengevaluasi stabilitas hasil 16 genotipe cabai rawit (Capsicum fructescens L.) dengan menggunakan analisis stabilitas parametrik (Francis-Kannenberg, Wricke, Finlay-Wilkinson, AMMI dan GGE). Penelitian ini dilaksanakan pada bulan Agustus 2018 sampai September 2019 pada empat lokasi, yaitu Kolaka, Palembang, Aceh dan Bogor. Penelitian ini menggunakan rancangan kelompok lengkap teracak dengan tiga ulangan. Genotipe cabai rawit yang dievaluasi adalah F5285290-237-6-1, F6285290-6-10-1-1, F5285290-290-2-1, F5285290-290-9-1, F5285290-290-9-3, F5321290-40-2-1, F5285290-123-6-15, F6321290-252-10-8-4, F5285290-38-6-3, F6321290-252-10-8-23, F6321290-252-10-8-75, Bonita IPB, Inul (varietas lokal), Cakra putih (varietas komersial), Taruna (varietas komersial), dan Tobasco. Interaksi genotipe x lingkungan berpengaruh nyata terhadap hasil. genotipe F6321290-252-10-8-4 diidentifikasi sebagai genotipe stabil berdasarkan beberapa metode stabilitas parametrik, namun genotipe tersebut tidak memiliki rata-rata hasil tertinggi. Sedangkan analisis GGE mengidentifikasi Bonita IPB sebagai genotipe yang stabil dengan daya hasil tertinggi. Kata kunci: AMMI, analisis stabilitas, cabai rawit, varietas

Abstract

Kecipir (Psophocarpus tetragonolobus (L.) DC) dijuluki sebagai kedelai daerah tropis dengan potensi kandungan gizi tinggi. Kurangnya informasi mengenai komponen keragaman dan heritabilitas dalam pemuliaan kecipir menjadi salah satu penyebab belum berkembangnya program pemuliaan kecipir di Indonesia. Penelitian ini bertujuan untuk memperoleh informasi ragam, heritabilitas dan hubungan antar karakter kecipir. Penelitian dilakukan di tiga lingkungan yaitu dua lingkungan di Bogor, dan satu lingkungan di Palembang, pada bulan Januari 2019 sampai dengan Juni 2020. Pengujian dilakukan terhadap 11 genotipe kecipir dan 11 karakter pengamatan. Hasil menunjukkan karakter umur berbunga dan jumlah biji per polong memiliki keragaman genetik yang luas. Nilai heritabilitas tinggi ditunjukkan oleh karakter jumlah biji per polong (89.48%), umur berbunga (83.32%), panjang polong muda (70.97%) dan bobot 100 biji (68.63%). Hasil analisis lintas menunjukkan karakter produktivitas polong muda (1.07), bobot biji per plot (0.81), umur berbunga (0.29), panjang polong muda (0.17), lebar polong muda (0.12), bobot biji per tanaman (0.09), dan bobot 100 biji (0.01) memiliki pengaruh langsung positif terhadap produktivitas biji kecipir. Umur berbunga, jumlah biji per polong dan bobot biji per tanaman merupakan karakter yang tepat menjadi kriteria seleksi dalam merakit kecipir berproduksi tinggi. Abstract

Konsumsi kentang di Indonesia terus meningkat namun belum sepenuhnya diperoleh dari dalam negeri. Hal ini disebabkan produktivitas kentang nasional masih rendah akibat kultivar impor yang tidak adaptif. Perakitan kultivar yang berpotensi hasil tinggi dan tahan penyakit layu bakteri merupakan upaya untuk meningkatkan produksi kentang. Penelitian ini bertujuan untuk mengetahui tingkat ketahanan penyakit layu bakteri secara in vitro dan potensi hasil genotipe kentang IPB. Penelitian dilakukan di laboratorium Pusat Kajian Hortikultura Tropika (PKHT) IPB untuk uji ketahanan penyakit dan di lahan pertanaman kentang milik petani di Pangalengan, Kabupaten Bandung untuk uji daya hasil pada bulan Januari sampai Juni 2018. Sebanyak tujuh genotipe uji yang terdiri dari terdiri dari PKHT 4, PKHT 6, PKHT 9, PKHT 10, PKHT 12, Intan, dan Medians, serta dua pembanding yaitu Atlantik dan Granola digunakan dalam penelitian ini. Uji ketahanan penyakit layu menggunakan metode in vitro dengan cara menginokulasi plantlet dengan metode gunting pucuk dilanjutkan dengan mengamati periode inkubasi, tingkat kejadian penyakit dan derajat ketahanan. Adapun uji daya hasil dilakukan dengan cara membandingkan pertumbuhan tanaman dan produksi umbi di lapangan. Hasil uji ketahanan menunjukkan genotipe PKHT 4 dan PKHT 9 mempunyai tingkat ketahanan, yaitu agak tahan, yang lebih baik dibandingkan Atlantik dan Granola sebagai kultivar pembanding. Uji daya hasil menunjukkan terdapat dua genotipe yang memiliki potensi hasil tinggi yaitu PKHT 6 (38.91 ton ha-1) PKHT 10 (28.75 ton ha-1) yang melebihi Atlantik dan Granola sebagai pembanding.
