Abstract

The aims of this research was to determine the effect of photodynamic therapy (PDT) on the healing of incision wounds in rat skin (Rattus norvegicus) that infected with MRSA bacteria by evaluating and give a score (0 – 4) at wound healing factors such as epithelialization, angiogenesis, inflammatory cells, fibroblast cells and collagen density. This study used 25 white rats divided randomly into 5 groups with 5 replications, namely P0 = MRSA infection wound + Povidone Iodine, P1 = MRSA infection wound + Silver Sulfadiazine, P2 = MRSA infection wound + 5-minute PDT irradiation, P3 = MRSA infection wound + 10- minute PDT irradiation and P4 = MRSA infection wound + 15-minute PDT irradiation. Histopathological observations and scoring showed epithelialization and collagen density in groups P2, P3 and P4 increased significantly compared to groups P0 and P1 (p<0.05). The histopathological observations and scoring of angiogenesis, inflammatory cells, fibroblast cells of P0 and P1 groups had a higher score than the treatment group with photodynamic therapy P2, P3 and P4 (p<0.05). This condition showed that in the P0 and P1 groups are still in the inflammatory phase, while in the P2, P3 and P4 groups have entered the remodeling phase which is characterized by high epithelialization scores and collagen density. Giving photodynamic therapy to wounds infected with MRSA can accelerate the wound healing process.

Abstract

Pasteurella multocida type B:2 is a bacteria which causing the disease was called Septicemia Epizooticae in ruminants, expecially cattle and buffalo. The aims of this research to determine the encoding gene SodC of Pastuerella multocida type B:2. This isolate of Pasteurella multocida type B:2 was obtained from the BBVet Denpasar, Bali that originally from East Nusa Tenggara (NTT) Indonesia. The sample was grown on Blood agar (BA). Separated colonies were grossly observed identified by macroscopicly (transparent, round and sweet-smelling) and microscopicly identified (bipolar and Gram negative), Biochemical test included Triple Sugar Iron Agar (TSIA), Urease, Simmnons Citrate Agar (SCA), Sulphid Indol Motility (SIM), Sugars Test (Sucrose, Manitol, Maltose, Glucose and Lactose). This Isolate had been test using Polymerase Chain Reaction (PCR), the result of PCR test if Pasteurella multocida had band in 235bp. This research was continued with Sequencing test to reveal the nucleotides sequence of the encoding SodC gene in Pasteurella multocida and also the homology from Genbank or NCBI. Result of Blasting in NCBI revealed that this encoding gene had similar nucleotides sequence with Pasteurella multocida strain 4407 (97%), Chinese 9N Pasteurella multocida strain (98%), Chinese Pasteurella multocida strain BS168 (97%), Pasteurella multocida strain EB168 China (97 %), Pasteurella multocida subsp. multocida strain CIRMBP-0884 France (97%), and Pasteurella multocida subsp. multocida strain RCAD0259 China (96%).

Abstract

Avian influenza virus (AIV) subtype H9N2 is one of the infectious agents that threatens laying poultry farms, because it has an impact on drastically reducing production in the population. The aim of this study was to isolate and analyze phylogenetically the partial gene encoding the surface proteins of the AIV subtype H9N2 from laying hens in East Java. A total of 30 suspected AIV subtypes of H9N2 were taken from laying hens which had decreased production by up to 70% in three sub districts each in Kediri, Blitar and Tulung Agung regency, in East Java Province. The virus was isolated in embryonated chicken eggs and then followed by a Hemagglutination (HA) test. Detection of the presence of H9 and N2 genes was carried out through Reverse Transcriptase polymerase Chain Reaction (RT-PCR) and continued with partial gene sequencing of the two surface proteins. Data analysis was processed using BioEdit version 7.2.5 and Mega 7.0. The results show that only one sample, namely code B2.2 from Blitar regency, is an AIV subtype of H9N2. The virus in this study belong to clade h9.4.2.5 of the AIV subtype H9N2. The conclusion of this study is that the VAI subtype H9N2 was successfully isolated from laying hens in East Java and successfully identified phylogenetically.

Abstract

This study aims to early detect Salmonella spp. and identify serotypes in commercial chicken eggs from layer chicken farms and traditional markets in Bali. Early detection study of Salmonella spp. was carried out by conventional bacteriological methods, while serotype identification by duplex Polymerase Chain Reaction (d-PCR) test against the invA gene from Salmonella spp. and the sefA gene from Salmonella enteritidis. Egg samples in this study were taken from 10 layer chicken farms in Bali which included districts of Bangli, Gianyar, Tabanan and Karangasem. Egg samples from traditional markets were taken from 18 traditional markets from the districts of Bangli, Gianyar, Tabanan, Karangasem, Badung, and Denpasar City. Samples were eggshells and egg whites. Analysis of positive results from Salmonella spp. described descriptively. The results showed that eggshells and white eggs from all of the layer chicken farms are negative contaminated with Salmonella spp. (0%). In eggshell samples taken from the traditional markets of Taman Bali and Tulikup from the districts of Bangli and Gianyar, positive with Salmonella spp. (11,1%) by conventional bacteriological tests. In the duplex Polymerase Chain Reaction test, S. enteritidis serotypes were identified. The finding contamination of Salmonella enteritidis in commercial chicken eggs from traditional markets require periodically detection to prevent the occurrence of salmonellosis due to consumption of contaminated chicken eggs in traditional markets in Bali.
