Abstract

Mercury contamination in Lebong Regency has caused serious impacts on public health and environmental quality. Bacteria living in contaminated environments can adapt and develop resistance to mercury through the presence of the merA gene, which plays an important role in converting mercury into a less toxic form. This study aimed to identify and characterize the merA gene in Pseudomonas putida. Molecular identification was carried out using 16S rDNA gene analysis and the polymerase chain reaction (PCR) technique. The obtained DNA sequences were further analyzed to determine gene similarity and phylogenetic relationships. The results showed that the bacterial isolate shared 99% homology with Pseudomonas putida strain CRSD9. The merA gene was 820 bp in length and encoded a mercury reductase enzyme consisting of 310 amino acids. This gene showed 97.78% homology with previously reported mercury reductase genes and contained two important domains, namely Pyridine Nucleotide-Disulfide Oxidoreductase (Pyr Redox) and FAD-linked domains. These findings indicate that Pseudomonas putida has strong potential as a bioremediation agent for reducing mercury pollution in the environment.

Abstract

Buah potong (rujak) merupakan salah satu makanan siap saji yang banyak dikonsumsi masyarakat, namun memiliki risiko tinggi terhadap kontaminasi mikrobiologi akibat proses pengolahan dan penyimpanan yang kurang higienis. Keberadaan bakteri Coliform dan Escherichia coli pada pangan dapat menjadi indikator rendahnya sanitasi serta berpotensi menimbulkan gangguan kesehatan. Penelitian ini bertujuan untuk menganalisis cemaran bakteri Coliform dan Escherichia coli pada buah potong (rujak) yang dijual di kawasan Pantai Panjang Kota Bengkulu serta membandingkan efektivitas metode Compact Dry EC (KIT) dan metode Most Probable Number (MPN). Penelitian menggunakan metode deskriptif dengan pengambilan sampel secara purposive sampling terhadap 10 pedagang rujak. Analisis dilakukan menggunakan metode KIT untuk mendeteksi jumlah koloni bakteri dan metode MPN melalui uji penduga, uji penguat, serta uji pelengkap. Hasil penelitian menunjukkan bahwa 5 dari 10 sampel positif terkontaminasi Escherichia coli. Metode KIT menunjukkan jumlah bakteri tertinggi sebesar 800 CFU/mL, sedangkan metode MPN menunjukkan nilai tertinggi ≥1898/100 mL. Uji pelengkap pada media EMBA menunjukkan terbentuknya koloni hijau metalik yang mengindikasikan adanya Escherichia coli. Berdasarkan hasil tersebut, sebagian sampel buah potong (rujak) di Pantai Panjang Kota Bengkulu belum memenuhi standar keamanan pangan secara mikrobiologis.

Abstract

Various crafts may be made with resin materials, such as earrings, bracelets, rings, and souvenirs. The availability of souvenirs may prove an attractive feature for tourists visiting any given area. Enggano Island a tourist destination in Bengkulu Province, lacks a distinctive range of souvenirs representative of the region. Thus, a training program was conducted for the local community, focusing on creating souvenirs utilizing resin and various materials surrounding Enggano Island. The program aimed to increase community skills in making souvenirs and strengthen the community’s entrepreneurial spirit. The program started with a presentation and followed by a souvenir-making practice. Evaluation result, the participants (100%) already knew the tools and materials needed to make resin souvenirs, they knew how to make a good quality of resin souvenirs, and they could identify substitute materials that could be obtained surrounding Enggano Island. Youth participants already knew how to get tools and materials to make resin souvenirs and market them through e-commerce. Additionally, the program has improved participants' knowledge and skills on how to make resin souvenirs, using materials found around Enggano Island.Bahan resin dapat dipergunakan untuk membuat berbagai macam kerajinan seperti anting, gelang, cincin, dan berbagai macam suvenir. Ketersediaan suvenir dapat menjadi salah satu daya tarik bagi wisatawan yang berkunjung di suatu daerah. Pulau Enggano sebagai salah satu wilayah tujuan wisata di Provinsi Bengkulu belum memiliki suvenir khas wilayah tersebut. Oleh karena itu kami melakukan pelatihan pembuatan suvenir kepada masyarakat dengan memanfaatkan resin dan berbagai bahan isian yang dapat diperoleh dari alam sekitar Enggano. Tujuan dari kegiatan ini untuk meningkatkan keterampilan masyarakat dalam membuat suvenir berbahan resin serta mendorong semangat wirausaha masyarakat. Pelaksanaan kegiatan diawali dengan pemaparan materi, kemudian dilanjutkan dengan praktik pembuatan suvenir, serta evaluasi pelaksanaan pelatihan. Hasil evaluasi menunjukkan bahwa 100% peserta telah mengetahui alat dan bahan yang dibutuhkan serta cara pembuatan suvenir berbahan resin, peserta telah mampu membuat suvenir berbahan resin dengan hasil yang cukup baik, dapat mengidentifikasi pilihan alternatif bahan yang dapat diperoleh di sekitar pulau Enggano. Sebagian peserta terutama dari kelompok anak muda dapat mengetahui cara mendapatkan alat dan bahan yang diperlukan, dan mengetahui cara pemasaran melalui e-commerce. Kegiatan ini juga telah meningkatkan pengetahuan dan keterampilan peserta mengenai cara pembuatan suvenir berbahan resin dengan memanfaatkan bahan-bahan yang ada di sekitar Pulau Enggano.

Abstract

Patikan Kebo (Euphorbia hirta L.) is a plant that lives scattered in the yard, has potential as an antibacterial, one way to extract plants is by isolating endophytic bacteria. Endophytes are bacteria found in plant tissue. The purpose of this study was to isolate and identify the morphology of endophytic bacteria from  plant Patikan Kebo (Euphorbia hirta L.). The method used is the Total Plate Count (TPC) method by taking samples, isolating bacteria, pure culture, morphological identification, and Gram staining. Endophytic bacteria were obtained by isolating parts of the plant Patikan Kebo (E. hirta L.), namely the roots of patikan kebo (PRE), Patikan kebo stem endophyte (PSE) and Patikan kebo leaves endophyte (PLE). The results of the study found 22 isolates, endophytic bacterial isolates have diverse morphological characteristics including appearance, elevation, edges and color. There are 20 isolates of endophytic bacteria that belong to the Gram positive group and 2 isolates of endophytic bacteria that belong to the Gram negative group. It can be concluded that endophytic bacterial isolates from Patikan Kebo (Euphorbia hirta L.) plants have different morphologies

Abstract

The increasing intensification of dragon fruit (Hylocereus sp.) cultivation is followed by an increase in disease attacks that have the potential to reduce plant productivity. This study aims to isolate, identify, and test the pathogenicity of bacteria that cause dragon fruit stem rot in Pematang Donok Village, Kepahiang, Bengkulu. Bacterial isolation was carried out from disease-infected stem tissue, followed by isolation purification. Bacterial identification was carried out based on colony morphology and Gram staining, while pathogenicity and hemolysis tests were carried out on potatoes and blood agar media, respectively. The data were analyzed descriptively qualitatively through observation of colony morphological characteristics and Gram staining, as well as pathogenicity tests on potatoes and hemolysis tests on blood agar. The isolation results obtained two bacterial isolates, namely isolates BBNK1 and BBNK2 with different colony morphology characteristics. Both isolates are Gram-positive bacteria with bacillus and coccus cell shapes. Pathogenicity tests on potatoes showed that both isolates were able to cause soft rot symptoms such as tissue softening, discoloration, and mucus formation compared to the control. Hemolysis tests showed that only isolate BBNK1 showed hemolytic activity with the formation of a clear zone around the isolate. The findings of this study offer foundational data to support future research on molecular identification and the development of more environmentally friendly and sustainable disease management strategies.

Abstract

The bacterium Salmonella typhii ATCC 13313 causes typhoid fever, which remains a medical problem in Indonesia. To prevent bacterial resistance, the misuse of antibiotics may lead to the use of alternative antibacterial agents derived from natural sources. The dadap serep leaf, or Erythrina lithosperma Miq, is a plant with potential as an antibacterial agent because it contains secondary metabolites such as alkaloids, flavonoids, triterpenoids, and phenols. The objective of this study was to determine the antibacterial properties of a 96% ethanol extract from dadap serep leaves that affect the growth of Salmonella typhii ATCC 13313. The disk diffusion method was used in this study. The maceration method with 96% ethanol was used to prepare the dadap serep leaf extract. Concentrations of 20%, 40%, 60%, 80%, and 100% were tested. Chloramphenicol was used as a positive control, and DMSO was used as a negative control. After 24-hour incubation at 37°C, inhibition zones around the disk paper indicated antibacterial activity. The results of the study show that the ethanol extract of Erythrina lithosperma Miq. leaves has a yield of 13.86% and is capable of inhibiting the growth of Salmonella typhii ATCC 13313. The higher the concentration of the extract, the greater the inhibitory effect it produces. Therefore, the ethanol extract of Erythrina lithosperma Miq. leaves may serve as a natural antibacterial agent against Salmonella typhii ATCC 13313.

Abstract

Liquid palm sugar is a natural sweetener widely used in traditional beverages, particularly by coconut ice vendors in the Pantai Panjang area of Bengkulu City. The processing method, which is still carried out traditionally with inadequate sanitation, has the potential to cause contamination by Coliform and Escherichia coli bacteria. This study aims to analyze the presence of Coliform and Escherichia coli bacteria in liquid palm sugar using the KIT (Compact Dry EC) method and the Most Probable Number (MPN) method. This study is descriptive in nature, with a sample consisting of 10 liquid palm sugar samples collected from coconut ice vendors in Pantai Panjang, Bengkulu City. Testing was conducted using the KIT method as an initial screening and followed by the MPN method as a confirmatory test. The results showed that 2 out of 10 samples tested positive for Escherichia coli contamination based on the KIT method. Further testing using the MPN method revealed an index value of ≥1898 MPN/100 mL in one sample, while the other sample showed a value of 6 MPN/100 mL at the enrichment stage. A supplementary test using EMBA medium revealed the formation of metallic green colonies, indicating the presence of Escherichia coli. Based on these results, some of the liquid palm sugar samples did not meet food safety standards as defined by SNI, posing a potential health risk to consumers.

Abstract

Barumanis Village, located in Bermani Ulu District, Rejang Lebong Regency, Bengkulu Province, has considerable horticultural agricultural potential, particularly in the production of fruits and vegetables. Intensive agricultural activities generate large amounts of organic waste that have not been optimally managed, leading to environmental pollution, unpleasant odors, and the proliferation of disease vectors. One environmentally friendly and economically valuable alternative for managing organic waste is the cultivation of Black Soldier Fly (BSF) larvae (Hermetia illucens). This community service program aimed to enhance the knowledge and practical skills of local residents in utilizing organic waste as a substrate for BSF larvae cultivation, thereby supporting the development of community-based creative economic activities. The program was implemented in three stages: (1) educational outreach on the characteristics, benefits, and cultivation techniques of BSF larvae; (2) demonstrations and hands-on training in organic waste management and BSF cultivation; and (3) evaluation and monitoring using pre-test and post-test instruments. The results demonstrated a significant improvement in participants' understanding of the benefits of BSF larvae, cultivation techniques, and their potential application as a sustainable solution for organic waste management and an alternative source of household income. Evaluation results indicated that participants who initially had limited knowledge of BSF cultivation showed marked improvement, as reflected by higher post-test scores compared to pre-test scores. This program is expected to encourage the implementation of sustainable organic waste management practices while improving community welfare through the development of BSF cultivation enterprises based on local resources.
