Abstract

The digestive system of sea urchins functions to process organic material from food into bioactive compounds. . However, the direct exploitation of bioactive compounds from sea urchins may trigger ecological imbalances. These bioactive compounds can also be produced through interactions between sea urchins and their diverse associated bacteria within the body. Therefore, alternative methods are needed to obtain bioactive compounds without disrupting the aquatic ecosystem. This study aims to explore bioactive compounds from the associated bacteria of sea urchins (Stomopneustes variolaris Lamarck and Tripneustes ventricosus Lamarck) originating from Kaur Regency, Bengkulu. A total of 30 bacterial isolates were successfully obtained using Zobell Marine Agar (ZMA) medium. Potential isolates were identified through morphological observation, Gram staining, and biochemical tests. Antibacterial activity assays showed that 12 isolates had potential as antibacterial producers based on tests against Bacillus subtilis, Escherichia coli, and Staphylococcus aureus. Two isolates, TVL 6 and TVL 11, were able to inhibit all test bacteria. Additionally, assays using pellets and supernatants showed that all 12 isolates exhibited antibacterial activity, with 5 isolates showing activity in the supernatant. Isolates TVL 6 and TVL 11 were proven effective in inhibiting all test bacteria in both pellet and supernatant forms, with the highest inhibition observed against Staphylococcus aureus. These results indicate that the associated bacteria in the digestive tract of sea urchins have potential as candidates for antibacterial compound producers.   Sistem pencernaan bulu babi berfungsi untuk mengolah bahan organik dari makanan menjadi senyawa bioaktif. Namun, eksploitasi senyawa bioaktif dari bulu babi secara langsung dapat memicu ketidakseimbangan ekosistem. Senyawa bioaktif ini juga dapat dihasilkan melalui interaksi antara bulu babi dengan bakteri asosiasi yang beragam di dalam tubuhnya. Oleh karena itu, diperlukan metode alternatif untuk memperoleh senyawa bioaktif tanpa merusak keseimbangan ekosistem perairan. Penelitian ini bertujuan untuk mengeksplorasi senyawa bioaktif dari bakteri asosiasi bulu babi (Stomopneustes variolaris Lamarck dan Tripneustes ventricosus Lamarck) asal Kabupaten Kaur, Bengkulu. Sebanyak 30 isolat bakteri berhasil diisolasi menggunakan media Zobell Marine Agar (ZMA). Identifikasi isolat potensial dilakukan melalui pengamatan morfologi, pewarnaan Gram, dan uji biokimia. Uji aktivitas antibakteri menunjukkan bahwa 12 isolat memiliki potensi sebagai penghasil antibakteri berdasarkan pengujian terhadap kultur uji Bacillus subtilis, Escherichia coli, dan Staphylococcus aureus. Dua isolat, TVL 6 dan TVL 11, mampu menghambat semua bakteri uji tersebut. Selain itu, pengujian menggunakan pelet dan supernatan menunjukkan bahwa 12 isolat mampu menghasilkan aktivitas antibakteri, dengan 5 isolat menunjukkan aktivitas pada supernatan. Isolat TVL 6 dan TVL 11 terbukti efektif menghambat semua bakteri uji baik pada pelet maupun supernatan, dengan penghambatan tertinggi terhadap Stapylococcus aureus. Hasil ini mengindikasikan bahwa bakteri asosiasi dalam saluran cerna bulu babi memiliki potensi sebagai kandidat penghasil antibakteri.

Abstract

The bark of the traditional plant, Archidendron bubalinum, known locally as kabau, exhibits promising qualities as a natural antibacterial agent. This research was conducted with two main objectives: (1) to evaluate the antibacterial efficacy of kabau fruit peel extract against two common pathogenic bacteria, Staphylococcus aureus and Escherichia coli, and (2) to design, validate, and assess the effectiveness of a research-based Microbiology E-Module in fostering students' critical analytical skills. The antibacterial capacity was tested using the paper disc diffusion method across a range of extract concentrations (5%, 10%, 20%, 40%, and 80%), using both pure water (aquades) and 2% dimethyl sulfoxide as carriers. Statistical analysis (one-way ANOVA, p = 0.020 < 0.05) indicated a notable difference in the zone of inhibition diameter across the various concentrations. The most potent antibacterial effect was observed at the 80% concentration with the aquades solvent, yielding inhibition zones of 3.97 mm against S. aureus and 8.42 mm against E. coli. These empirical results served as the core content for the innovative E-Module. The developed E-Module attained a high level of validity, achieving a CVR score of 1.00 from 5 expert validators, thus confirming its readiness for use. Subsequent evaluation during instructional application revealed a marked elevation in students' critical thinking abilities. Students in the Biology Department at FMIPA, Universitas Bengkulu, showed an initial mean pre-test score of 41.67, which surged to 88.00 in the post-test. This change resulted in an N-gain score of 0.79, positioning the improvement in the "high" category. Therefore, A. bubalinum extract is confirmed to hold antibacterial potential, and the E-Module is proven to be an effective educational tool for enhancing students' critical thinking competence.

Abstract

Limbah organik berupa sayuran dan buah-buahan di Desa Sukasari, Kabupaten  Kepahiang, Kota Bengkulu yang melimpah dapat dijadikan sebagai produk Eco-enzyme sebagai bahan alternatif pembuatan Bio-handsanitizer dan Biofertilizer. Dari hasil kegiatan pengabdian kepada masyarakat mengenai aplikasi pemanfaatan ecoenzyme dari limbah organik sebagai bahan alternatif pembuatan Bio-handsanitizer dan Bio-fertilizer, para kelompok tani  telah mengetahui dan memahami cara pembuatan dan manfaat ecoenzyme. Hal ini menunjukkan kegiatan pengabdian sukses dan mencapai sasaran. Sosialisasi dan demonstrasi pelatihan yang dilakukan memberikan hasil persentase yang baik ketika pemberian umpan balik kepada peserta. Maka dari itu kegiatan pengabdian ini diharapkan untuk dapat memberi solusi terhadap kasus  pengolahan serta penanganan limbah organik melalui pembuatan produk ecoenzyme sebagai bentuk pengolahan limbah organik, seperti limbah sisa buah dan sayur dan diharapkan dapat memberi manfaat berupa wawasan untuk dimanfaatkan dalam realisasi produk ecoenzyme di Desa Suka Sari, Kecamatan Kabawetan, Kabupaten Kepahiang. Sehingga kedepannya kelompok tani dapat mengelola limbah organik secara tepat sehingga menghasilkan produk bermanfaat seperti biohandsanitizer dan biofertilizer dengan nilai jual yang tinggi dan diharapkan bisa mengurangi penggunaan pupuk kimia.

Abstract

Trans Malakoni Village, located on Enggano Island, Bengkulu Province, faces significant challenges in providing access to clean, potable water. Limited access to adequate freshwater sources and water quality that frequently fails to meet health standards are major obstacles to the community's sustainable livelihood. This research aims to explore the potential and implementation of household-scale reverse osmosis (RO) technology as an innovative solution for producing clean, potable water in Trans Malakoni Village. The research methodology involves raw water quality analysis, the design of a household-scale RO system suitable for local conditions, and an evaluation of system performance and community acceptance. Initial results indicate that existing water sources in Trans Malakoni Village, although limited, can be effectively treated using RO technology to achieve established drinking water quality standards. The proposed household-scale RO system design emphasizes energy efficiency, operational ease, and affordable maintenance costs, thereby enabling sustainable adoption by households. The implementation of RO technology is expected not only to enhance access to clean water but also to contribute to improved public health, self-reliance in water resource management, and sustainable living in Malakoni Village. This study recommends further research on system optimization and community education programs to maximize the positive impact of RO technology in other remote island regions.

Abstract

Background: The peppermint plant (Mentha piperita L.) is a type of medicinal plant that is widely found in temperate regions, especially Europe, North America and Africa. This plant contains essential oils, menthol, and menthone, menthafuran, sesquiterpenes, triterpenes, flavonoids, carotenoids, tannins and various minerals are also widely contained in peppermint leaf oil (M. piperita L.). The purpose of this study was to isolate and determine the morphological diversity of endophytic bacteria from peppermint plants (M. piperita L.). Methodology: The method used in this study is to take isolation samples, pure culture, characterization and gram staining to determine whether it includes gram-negative or positive bacteria. The results of the study of 12 cultures of endophytic bacterial isolates from plants (M. piperita L.) were taken 10 isolates from each different culture media. Findings: The results of morphological observations of bacterial colonies show diverse characteristics in terms of surface, appearan, ceelevation, edges, and colour. Gram Staining Test of Endophytic Bacterial Isolates from the roots, leaves, and stems of mint plants against endophytic bacteria showed that there were gram-positive and gram-negative bacterial isolates. Then the results of bacterial cell shape morphology have a diversity of Cocobacil, Coccus, and Bacil forms so it can be concluded that endophytic bacterial isolates from peppermint plants (M. piperita L.) have varied morphological diversity. Contribution: Data on the morphological diversity of endophytic bacteria from M. piperita L. provides important basic data for further research on the potential of M. piperita L. endophytic bacteria

Abstract

The development of pharmaceutical raw materials as local Indonesian probiotic concentrates requires attention to strain identity accuracy, safety, and health potential. This is due to the high potential of probiotics in preventing and addressing various health disorders from infancy to old age. Previous research successfully obtained lactic acid bacteria (LAB) isolates as probiotic candidates from tempeh produced in Jember, and preclinical testing showed that administering a consortium of five LAB isolates at a dose of 10^8 CFU/ml to BALB/c mice was an effective treatment for further application. This study aimed to perform molecular identification of the consortium of five LAB isolates from tempeh in Jember, East Java, based on 16S rRNA gene sequences. The consortium includes TA1, TB1, TK1, TK2, and TK4 isolates. This study used both qualitative descriptive and quantitative methods. Identification based on the 16S rRNA gene to determine LAB species strains involved bacterial genomic DNA isolation, amplification of the bacterial 16S rRNA gene, PCR product analysis, and phylogenetic identification and analysis. DNA amplification results showed that the consortium of five LAB isolates produced DNA fragments with bands of approximately 1300 bp. BLAST-N and phylogenetic analysis of the 16S rRNA gene showed that TB1 had 99.05% similarity with Kosakonia cowanii JCM 10956, TA1 had 99.46% similarity with Lactiplantibacillus plantarum JCM 1149, TK1 had 99.38% similarity with Lactiplantibacillus pentosus 124-2, TK2 had 96.86% similarity with Pseudomonas fluvialis ASS-1, and TK4 had 99.38% similarity with Lactiplantibacillus plantarum NBRC 15891. TA1, TB1, and TK4 were identified as non-pathogenic LAB strains, while TK1 and TK2 were not classified as LAB.

Abstract

Hand is the main transmission pathogen entry into the body which is the part of the body that is most often in direct contact with other limbs and surrounding objects. The importance of maintaining hand hygiene by always washing hands with soap is the first step in preventing transmission of COVID-19. However, during a pandemic like this, it will be difficult to find soap available everywhere, and tends to be hard to carry all the time. Therefore, this service aims to invite the people of Pematang Gubernur, Bengkulu City, to use paper soap because it is easier to use its use and flexible to carry anywhere besides being an effort to prevent the COVID-19 outbreak, it can also add to new information and skills for residents in making attractive soaps. The feedback response given by the community service implementation team to the community service participants resulted in a positive response to this activity, who initially did not know about making and use of paper soap and became aware of this.

Abstract

This study aims to inventory the types of food crops growing around tailings ponds and to quantify mercury concentrations in food crop tissues growing in the area around the gold mine. The samples taken are plants consumed by residents. Mercury content testing uses a mercury analyzer. The results of the study showed that from 4 observation locations, a total of 11 types of food crops were found, all of which contained mercury (Hg) with a concentration range of 0.01-0.54 mg/kg. Similarly, the soil as a growing medium contains quite high levels of mercury at location 1, namely 54.36 mg/kg, and the lowest at location 3 at 0.12 mg/kg. The concentration of mercury in vegetable leaves is much higher than in other organs such as tubers, flowers, and fruits. Furthermore, to plant plants for consumption, it is best to pay attention to the condition of the growing medium, as a prevention of heavy metal (mercury) contamination, such as the environment (air, soil, water) that is far from waste retention ponds or waste discharge streams containing heavy metals (mercury). Analisis Konsentrasi Merkuri pada Tanaman Pangan di Sekitar Tambang Emas Kabupaten Lebong ABSTRAK: Penelitian ini bertujuan untuk menginventarisasi jenis tanaman pangan yang tumbuh disekitar kolam penampungan limbah (tailing) dan untuk mengkuantifikasi konsentrasi merkuri dalam jaringan tanaman pangan yang tumbuh di area sekitar tambang emas. Sampel yang diambil merupakan tanaman yang memang dikonsumsi oleh warga setempat. Uji kandungan merkuri menggunakan mercury analyzer. Hasil penelitian menunjukkan dari 4 lokasi pengamatan, ditemukkan Sebanyak 11 jenis tanaman pangan yang semuanya mengandung merkuri (Hg) dengan kisaran konsentrasi 0,01-0,11 mg/kg. Demikian pula, tanah sebagai media tanam mengandung kadar merkuri yang cukup tinggi di lokasi 1, yaitu 54,36 mg/kg, dan paling rendah dilokasi 3 sebesar 0,12 mg/kg. konsentrasi merkuri dalam daun sayuran jauh lebih tinggi dibandingkan organ lain seperti tuber, bunga, dan buah. Selanjutnya, untuk menanam tanaman yang akan dikonsumsi sebaiknya memperhatikan kondisi media tumbuh, sebagai pencegahan dari kontaminasi logam berat (merkuri), seperti lingkungan (udara, tanah,air) yang jauh dari kolam penampungan limbah maupun aliran pembuangan limbah yang mengandung merkuri.

Abstract

The increasing intensification of dragon fruit (Hylocereus sp.) cultivation is followed by an increase in disease attacks that have the potential to reduce plant productivity. This study aims to isolate, identify, and test the pathogenicity of bacteria that cause dragon fruit stem rot in Pematang Donok Village, Kepahiang, Bengkulu. Bacterial isolation was carried out from disease-infected stem tissue, followed by isolation purification. Bacterial identification was carried out based on colony morphology and Gram staining, while pathogenicity and hemolysis tests were carried out on potatoes and blood agar media, respectively. The data were analyzed descriptively qualitatively through observation of colony morphological characteristics and Gram staining, as well as pathogenicity tests on potatoes and hemolysis tests on blood agar. The isolation results obtained two bacterial isolates, namely isolates BBNK1 and BBNK2 with different colony morphology characteristics. Both isolates are Gram-positive bacteria with bacillus and coccus cell shapes. Pathogenicity tests on potatoes showed that both isolates were able to cause soft rot symptoms such as tissue softening, discoloration, and mucus formation compared to the control. Hemolysis tests showed that only isolate BBNK1 showed hemolytic activity with the formation of a clear zone around the isolate. The findings of this study offer foundational data to support future research on molecular identification and the development of more environmentally friendly and sustainable disease management strategies.
