Abstract

RAPD analysis to determine the diversity, status and genetic close relatioship Batur with Merino, Garut, Thin Tail and Fat Tail sheep genomic DNA was used in 27 Batur, 15 Merino, 17 Garut, 15   Thin Tails and  Fat Tails animals. The process of RAPD-PCR used five primers of 10 nucleotides. PCR results were electrophoresed with 2% agarose gel. Identified similarities and differences between individual RAPD bands one and cross-sample of the population, genetic distance and closeness of genetic relationship. The process 89 sample  sheep RAPD-PCR generated of 4189 band from 100 to 1500 bp which consisted of 91 type. Batur sheep samples produced bands at most (1666 tape) and the lowest Fat Tails (493 bands). The number monomorfism of bands most of the Batur (891 bands) and the lowest Garut (170 bands), but the polymorphism  bands highest  of the Batur (775 bands) and the lowest Fat Tails (262 bands). Between individuals within populations have similar genetic Merino highest and the lowest Thin Tail. Cross-population genetic similarity is highest individuals in the population Batur and Merino, while the lowest Merino and Thin Tail. The closest genetic distance was Batur with Merino population and the most apart distant Merino with Thin Tail or Merino and Fat Tails. Batur sheep population has the closest genetic relationship with the Merino and the most apart distant is with Fat Tail. Abstract

The objective of this research was to know the ability of various acidic solutions on dissolving collagen  chicken feet, with different soaked time.  Each acid 5 percent (v/v), collagen extraction was done by washing chicken feet and then cutted into small pieces and finally grinded.  Every 100 gram treatment was soaked in acetic acid (a1), citric acid (a2), lactic acid (a3) and hydrochloric acid (a4), for 12, 24 and 36 hours.  Precipitated collagen in the filtrate was 5 percent NaOH to reach the neutral pH (pH 7).  Collagen precipitate was separated by filtration usingfilter paper and then  rendement was calculated, HPLC was used to determin amino acid composition, and SDS-PAGE was use determin the type of collagen.  This experiment use factorial completely randomized design (CRD) 4 x 3 and three time replication.   Result showed that lactic acid has highest capability to dissolve collagen, while citric acid the lowest.  Combination of acid solution and soaking time had significant (P<0.01) effect on dissolving collagen of chicken feet.  Extracted collagen in all acid solution, hassame amino acid, composition but different in percentage of amino acid molecules.  Collagen type in treatment combination was the same, but for soaking time of 36 hours revealed some peptide band.  Lactic acid had highest capability of collagen extraction in chicken feet than citric acid, acetic acid and hydrochloric acid with soaking time of 12, 24 and 36 hours.  It was estimated that extracted collagen can be grouped to type I consisted of two chain of a1. Abstract

One of the processes steps for producing rubber sole is vulcanization process. This process is carried out by using pressed moulding  methods in which rubber compound was heated and pressed in the mould of rubber sole at certain time and temperature. The aim of this research is to know the influence of the temperature used for vulcanization for the tensile strength, elongation and tearing strength properties of the rubber sole compound. Temperature vulcanization used for the research was varied at 1400C, 1500C, 1600C and 1700C. The results shows that vulcanization at 1400C, give the highest properties in tensile strength, elongation and tearing strength, those are : 113,70 kg/cm2; 326,60 % and 107,00 kg/cm2respectively. Increasing temperature vulcanization cause decreasing those physical properties.    INTISARI Salah satu tahapan proses pada pembuatan sol karet adalah proses vulkanisasi. Proses ini dilakukan dengan menggunakan metoda cetak tekan dimana kompon karet dipanaskan dan ditekan dalam cetakan sol karet pada temperature dan waktu tertentu. Tujuan dari penelitian ini adalah untuk mengethui pengaruh temperature yang digunakan untuk vulkanisasi pada sifat kuat tarik, kemuluran dan kuat sobek dari kompon sol karet. Temperatur vulkanisasi pada penelitian ini adalah divariasi pada suhu 1400C 1500C, 1600C dan 1700C. Hasil penelitian menunjukkan bahwa vulkanisasi dengan suhu 1400C akan memberikan sifat-sifat kuat tarik, kemuluran dan kuat sobek yang terbaik dari kompon sol karet, yaitu : 113,70 kg/cm2; 326,60 % dan 107,00 kg/cm2. Kenaikan temperature vulkanisasi akan menurunkan sifat-sifat fisika tersebut.

Abstract

Leather tanning industry is one of the industry which potentially causes environmental problem. For processing, a lot of water and chemical are used, generates a lot of liquid waste, which are powerful pollution in water an in soil. There will also be animal residues from such operation as cleaning, fleshing, splitting and trimming. Each of these generates solid waste which must be disposed. It is a big problem for handling the solid waste because of their volume and highly odours when they decompose in their solid form. Untanned  solid waste and liquid waste generated in the tanning process have a high organic content (protein and fats) and their humidity …. are such that they are readily degradated by methane fermentation.   ABSTRAK  Industri penyamak kulit adalah salah satu industri yang sangat potensial menyebabkan problem lingkungan. Dalam prosesnya diperlukan banyak air dan bahan kimia yang akan menjadi limbah cair yang dapat menyebabkan terjadinya pencemaran air atau masuk dalam tanah. Juga sisa-sisa dari proses pencucian, fleshing, splitting dan trimming akan menghasilkan limbah padat. Untuk menangani limbah padat banyak menimbulkan permasalahan karena volume yang cukup besar dan bau yang ditimbulkan. Limbah tak tersamak dan limbah ciar yang diperoleh dalam proses penyamakan mempunyai kandungan bahan organic yang cukup tinggi (protein dan lemak) dan juga kandungan air yang cukup sehingga memungkinkan untuk didegradasi dengan cara fermentasi methane.

Abstract

Enzime is natural protein which has katalistic properties in the chemical reaction. The use of enzymes in leather processing has been well known for many years. One of the enzyme which have big role in the leather processing is protease. It is used in the unhairing and bating processes. The use of protease on the leather processing can reduce the environment pollution especially in unhairing process.   INTISARI Enzima adalah merupakan protein yang terjadi secara alami mempunyai sifat katalis pada suatu reaksi kimia. Penggunaan enzima pada proses penyamakan kulit telah lama dikenal pada industry penyamakan kulit, salah satu jenis enzima yang sangat berperan dalam proses penyamakan kulit adalah enzima protease yang digunakan pada proses bating.  Penggunaan enzima proteolitik dalam penyamakan kulit dapat menekan terjadinya pencemaran lingkungan, terutama penggunaannya pada proses buang bulu.

Abstract

One of the plastic material which is usually used for packaging is polyethylene. That plastic has a good performance for plasticity, chemicals resistence, flexibility and no toxic subtances, therefore, this is suitable for good packaging. One of the use plastic film for packaging is for packing of salted ginger for export purpose. Research in physical properties of the salted ginger packaging shows that plastic film packaging has a weakness in the tearing strength properties and some leaking caused by the unproper sealing during the process. INTISARI Salah satu bahan plastik yang banyak digunakan untuk bahan pengemasan adalah bahan plastik polyethylena. Bahan tersebut mempunyai sifat yang baik dalam keuletan, ketahanan terhadap bahan kimia serta fleksibilitasnya dan tidak mengandung bahan-bahan yang toxic sehingga sangat cocok untuk pengemas makanan. Penggunaan kantong plastik film polyethylena untuk mengemas makanan diantaranya untuk mengemas bahan setengah jadi asinan jahe untuk keperluan ekspor. Dari hasil penelitian sifat fisis kantong plastik untuk kemasan asinan jahe masih dijumpai adanya kelemahan pada bahan plastiknya, yaitu pada kekuatan sobeknya dan kebocoran-kebocoran yang sering terjadi yang disebabkan kurang sempurnanya dalam pembuatan kantong plastik, terutama dalam proses lekat panas.

Abstract

Kinetic of Rotating Biological Contractor have been investigated by manu researchers. In general the model emploued either saturation kinetic (following Monod equation) or first order kinetic to describe substrate removal. In this experiment, saturation kinetic model was used to find out the saturation constant of the RBC unit. The influent fed into the unit was synthetic waste water with glucose as organic carbon sources. The flow was maintained constant at 20.4 1/d and COD influent resulted the saturation constant (Ks) 11.17 mg/I with 20 hours of detention time.

Abstract

The objective of the research was to use of liquid membrane emulsion technique for the extraction of chrom in the nitrate acid solution. The liquid membrane phase as emulsion consists of kerosene as the solvent, sorbitan monooleat (span-80) as the surfactant, trioktil amin, as the carrier, natrium hidroksida as the internal phase. The optimum conditions obtained for making membrane, the liquid membrane phase was consists of organic phase (O), the internal liquid phase (W) with the ratio of O/W = 1, the concentration sorbitan monooleat (span-80) in the membrane phase was 4%(v/v), the optimum time to achieve a emulsion was 5 minutes, the rate of stirring for membrane 8000 rpm. The  optimum conditions obtained for extraction chrom ion as of chrom nitrate  from solution with percentage of liquid membrane extraction used of 97,8% and the percentage of liquid membrane extraction used of 97,8% and the percentage of liquid-liquid extraction of 81,24% were as follow : the concentration of nitric acid in external phase was 3M, the concentration of tri oktil amin in the membrane phase was 5% (v/v), the concentration of natrium hidroksida in internal phase was 1% (v/v), the extraction time was 15 minutes and volume ratio of membrane phase and external phase was 1 : 3. The addition of Fe and Zn ions solution decreased the percent extraction of chrom ion. INTISARI Penelitian ini bertujuan untuk meneliti teknik membran cair dalam ekstraksi krom pada larutan asam nitrat. Fasa membran yang berupa emulsi terdiri dari kerosene sebagai pelarut, sorbitan monooleat (span-80) sebagai surfaktan, tri oktil amin (TOA), sebagai zat pembawa serta natrium hidroksida sebagai fasa internal. Kondisi optimum yang diperoleh untuk pembuatan membrane cair, fasa membran terdiri dari fasa organic (O), fasa internal (W) dengan perbandingan O/W = 1, konsentrasi sorbitan monooleat dalam fasa membran 4%(v/v), waktu optimum pembuatan emulsi 4 menit, kecepatan pengadukan 8000 rpm. Kondisi optimum yang diperoleh untuk mengekstraksi krom dari larutan yang mengandung krom nitrat dengan prosen ekstraksi membran-cair sebesar 97,80% dan proses ekstraksi cair-cair sebesar 81,24% adalah sebagai berikut : konsentrasi asam nitrat dalam fasa eksternal adalah 3M, konsentrasi tri oktil amin dalam fasa membran adalah 5%(v/v), konsentrasi natrium hidroksida, dalam fasa internal 1%(v/v), 1 M dan lamanya waktu ekstraksi ialah 15 menit dan perbandingan  volume fasa membran dan volume fasa eksternal adalah 1 : 3. Penambahan ion ferro dan seng dalam larutan krom akan menurunkan prosen ekstraksi krom

Abstract

An immobilized microbial cell is a physical confinement or localization of intact cell to a certain defined region of space with the preservation of some desired catalytic activity. The immobilization cell process has been increasingly used, one of those is for waste water treatment industry. Microbial entrapping process one of the method for the microbial cell immobilization by using some carrier such as collagen, gelatin, alginate, carragena and cellulose tri acetat at the time being is commonly used. Immobilization cell is effective enough for waste water treatment containing toxic substance such as phenol and by using immobilization cell, secondary sedimentation tanks is no longer used.   INTISARI Immobilisasi mikrobia cell merupakan suatu cara untuk melokalisasi atau menahan secara physis cell lengkap pada tempat tertentu dengan memelihara aktivitas katalitik cell sesuai dengan yang diinginkan. Penggunaan immobilisasi cell saat ini terus meningkat, salah satunya adalah penggunaannya untuk pengolahan limbah cair industri. Proses immobilisasi cell dengan cara menjerat mikrobia dengan berbagai bahan pembaw seperti collagen, gelatin, agar alginate, carrogen, cellulose tri acetat saat ini merupakan metoda yang banyak digunakan. Metoda immobilisasi cell dapat memberikan efektifitas yang cukup tinggi untuk mengolah limbah cair yang mengandung bahan toksik seperti phemol dan tidak lagi diperlukan pengendap sekunder dalam proses pengolahan limbahnya.
