Abstract

The aims of this research was to identify the qualitative and quantitative difference of the phenotypes of native mallard and Muscovy, and Muscovy the genetic diversity between Muscovy and mallard using RFLP method with growth hormone primer (GH) gene. The materials were 30-week old male and female ducks from 5 strains—Magelang, Mojosari and Tegal ducks, white-feathered, and black and white-feathered Muscovy. Muscovy Completely Randomized Design was used to analyze parameters 5 strains of ducks based on gender and with 5 replications. The parameters measured were local and Muscovy duck phenotypes and genetic diversity (number of alleles) based on GH gene, heterozygosity and genetic distance. Qualitative properties were analyzed descriptively and quantitative properties were analyzed using Anova followed by HSD in case of significant differences, whereas RFLP analysis was used to determine the allele frequencies, genotype frequencies, genetic diversity, and genetic distance of local ducks. Result showed phenotypic differences between Anas platyrhynchos and Cairina moschata. The dominant feather colorof Muscovy was black and white while mallard was brown. The bill color Muscovy Muscovy was pink with dark brown but black in mallard. The dominant shank’s color in both Muscovy and mallard was black. Body size of mallard was larger than that of Muscovy Muscovy, and Magelang ducks weigherd more than Tegal and Mojosari Duck. The PCR-RFLP results showed lower heterozygosity of mallard compared to Muscovy based on GH gene, and genetic distance of the Tegal duck was closer to Mojosari and Magelang ducks, whereas Magelang and Mojosari ducks had a considerable genetic distance based on the GH gene.

Abstract

The increasing demand for the utilize services computer networks has increased the traffic on the network. In general, the server provides service round the clock without a break, so that problems such as breakdown of communication in the network is lost that must be avoided. This cases need for a bettersystem with high performance and reliable every time. Current server technology development will be fast but the memory access speed is slower than the increase in the consumption of bandwidth. This is one o f the factors causing the bottleneck experienced of server. With the load balancing can be provide abetter solution. The concept used to provide a server has the same service. Then, the load balancing will check the CPU usage on each server. The server will perform selection based on the value of the smallest CPU Usage o f all existing servers. The application load balancing is placed before the user accessing ofserver with the object of distributing requests from the user to any of the servers that have been selected by the load balancer. Load balancing provides many benefits to systems that use it. The first, low cost, existing resources do not need to be replaced or discarded. Second, don't always need high end equipment. Third, easily developed when needed arose and a drastic decline in the quality o f service. Load balancing is also more secure, when a failure occurs on the server, load balancing application will redirect the user to the server is still available.

Abstract

BOD, COD, phenol and ammonia-free in the effluent of hospitals wastewater often exceed the quality standards. This was due to less optimal biological processes to degrade the pollutants. So we need an efforts to find optimal process conditions through the engineering process and the factors that affect the biodegradation of pollutants. On the other hand, AF2B reactor containing biofilter with bee nest shaped has a large specific surface area so as to maximize the biodegradation process of pollutants by microorganisms. While bacteria consortium consisting of several types of bacteria have a greater capacity than a single bacterium in the degradation of pollutants. The research aims to determine the effect of starter volume and air flowrate to decrease the concentration of pollutants (BOD, COD, phenol and ammonia-free) in AF2B reactor batch using a bacterial consortium. The research was conducted in three phase which are the making of growth curve, acclimatization and biodegradation of pollutants in an AF2B reactor batch using a bacterial consortium. The experiment variables are the starter volume (85%, 75%, 65% in volume), and the air flowrate (2.5; 5; and 7.5 liter/min). Materials used are hospital waste water and bacterial consortium. The experiment begins with setting up the AF2B reactor containing biofilter with bee nest shaped and then filled it with a starter from the acclimatization process on a given volume and followed by supplies of air at a certain flowrate. Then hospital waste water and 5% of nutrients were added till reach 10 liters of total volume in reactor. Samples were taken every 30 minutes for up to 360 minutes of biodegradation process. The samples were then analyzed its pollutants concentration (BOD, COD, phenol and ammonia-free). The BOD concentration was analyzed using Winkler bottles method, COD using open reflux method, while phenol and ammonia-free using UV-Vis spectrophotometry method. The results showed that the starter volume and air flowrate affect the decrease of pollutants concentration till reach quality standard, which at starter volume of 85%, air flowrate of 7.5 liter/min and biodegradation time of 360 min can reduce the BOD, COD, phenol, and ammonia-free to 92%, 86%, 88% and 76% respectively.

Abstract

Periodontitis is multifactorial inflamation process and related to disproportion of cytokine. IL-10 is a dominant noninflammatory cytokines that related to gene polymorphism. Polymorphism G-1082A IL-10 genes has been reported to increase the risk of periodontitis occurs in Italian populations, apart from different result found in Brazilian. The purpose of this research was to determine the polymorphism G-1082A IL-10 in periodontitis patients in Indonesia, especially among Yogyakarta’s Javanese. This is a case-control research with subjects according to the inclusion criteria. DNA was taken by cotton swab from the epithelial cells of buccal mucosa, and was isolated using a PrestoTM (GeneAid) kit. Genotyping analysis by using the PCR RFLP technique and descriptive results were presented. The result showed that A allele frequency is 100% and no G allele was found. AA genotype in case group has lower frequency than in control group and vice versa. From this research, it was concluded that A allele was dominant in Yogyakarta’s Javanese, and AA genotype frequency is lower in individual with periodontitis.

Abstract

Penelitian ini dilakukan karena pelaksanaan layanan bimbingan dan konseling di sekolah tidak berjalan dengan baik terutama dalam pembinaan guru bimbingan dan konseling. Ada tiga tujuan dari penelitian ini. Yang pertama adalah menggambarkan upaya pembinaan kinerja yang dilakukan oleh guru itu sendiri. Yang kedua adalah untuk menggambarkan upaya pembinaan kinerja yang dilakukan oleh kepala sekolah. Yang ketiga adalah untuk menggambarkan upaya pembinaan kinerja yang dilakukan oleh pengawas bimbingan dan konseling. Upaya pembinaan kinerja yang dibahas dalam penelitian ini terkait dengan pelaksanaan layanan bimbingan dan konseling di sekolah serta fasilitas dan dana pendukungnya. Jenis penelitian ini adalah kuantitatif dengan menggunakan metode deskriptif. Sampel penelitian ini adalah lima puluh lima guru bimbingan dan konseling dan dua supervisor dari SMP dan SMA yang dipilih dengan menggunakan teknik total sampling. Instrumen yang digunakan adalah skala model Likert. Temuan penelitian ini antara lain. Pertama, upaya yang dilakukan oleh guru bimbingan dan konseling secara rata-rata masih ?kurang?. Kedua, upaya yang dilakukan oleh kepala sekolah secara rata-rata masih "sangat kurang". Ketiga, upaya yang dilakukan oleh pengawas secara rata-rata masih ?sangat kurang?. Namun, upaya pembinaan perlu ditingkatkan dan dikembangkan baik untuk proses maupun implementasinya. Hasil penelitian ini dapat digunakan sebagai pertimbangan dalam pembinaan kinerja guru bimbingan dan konseling dalam melaksanakan layanan bimbingan dan konseling yang sukses di sekolah.

Abstract

The aim of present study was to investigate myostatin gene polymorphism and its association with weaning weight and body weight in Batur sheep, 30 heads of Batur all lambs were fed by concentrated feed. Body weight measured monthly after weaning to the six months of age. DNA Extraction used 200 ul of whole blood samples. To amplify exon 3 region of MSTN gene a specific primer designed using the Primer3 software. The 25 μl volume contained 25 ng of genomic DNA, 12.5 μl 2x Reaction mix of each primer. The cycling protocol was 5 minutes at 95°C as initial denaturation, 35 cycles of denaturing at 94°C for 45 seconds, annealing at 73.9 for 45 seconds, extending at 72°C for 40 s, with final extension at 72°C for 10 minutes. Eleven polymorphic sites were observed in the in 3rd exon region transversions at c.*121 G instead of A, del-T at c.*129, one individual at c.*139 and one individual at c.*158 positions however, one individual sequence disrupted reading frame in whole MSTN sequenced, also weaning weight and 6-month weight of Batur lambs were 21.13±5.70 and 31.64±7.06 to the homozygous whilst 19.99±4.33 and 30.50±5.44 for heterozygote respectively but there is no significe differences seen above all the studied parameters. Homozygous AA lambs had fairly low weaning weight and body weight compared to the heterozygous AB lambs but might have a higher carcass weight, further investigation is needed into the interaction of MSTN with other genes involved in muscle growth.

Abstract

Learning is characterized by the use of good questioning skills , especially in learning for large groups of children. Skills in question play an important role in student learning activities. The low skill of students asking in the learning process can lead to disruption of an effective learning process and must be handled by the BK teacher appropriately. Questions are one of the stimuli of thinking that is good for teaching students. The research method used is R & D (Research and Development) by following the steps in developing the ADDIE pattern (Analyze, Design, Development, Implementation, Evaluation). The subject of the research trial consisted of experts to test the feasibility of the model, the BK teacher to assess the suitability of the model, and students to test the effectiveness. The study was conducted by collecting initial data as a material for consideration in preparing the initial model which was then tested for its suitability and effectiveness through Focus Group Discussion (FGD) and expert testing. The results of this study show that the students' skills in question are in the high category.

Abstract

Rapid external changes without being followed by internal changes will have a negative impact on tertiary institutions. Internal change begins with the ability of senior lecturers because they are at the forefront in following changes. Organizational commitment is evidenced by the high quality of the lecturers? work ability. This study utilized combined research methods or exploratory mix methods with data taken from forlap dikti and Sinta, and quantitative and qualitative analysis using WAI (Work Ability Index). Results of the study showed that the work ability of senior lecturers is lacking (0.1-1), while organizational commitment is still low as evidenced by the results of scientific journals during the past three years. The innovation of this study is the Emotional Work Community (EWC) as a concept model to improve lecturers' performance abilities that are lacking.

Abstract

This research purposed to describe the results of AUM PTSDL and its use by Guidance and Counseling Teacher or Counselor for Guidance and Counseling services to students. This research used descriptive method with qualitative and quantitative approach. The instruments of research used AUM PTSDL Format 2, The format of BMB3 and guideline for interview. The results of research are analyzed statistically and narrative. This research shows that AUM PTSDL results of Senior High School students in Padang who became samples in form quality score of learning activities still very low or less is average 36.30% from ideal score, and also their learning problems pretty much is average 28,14% from number of learning problems that may exist. Response dynamics of BMB3 student to data of AUM PTSDL results are quite positive, means that students are aware of their flaws and eager to fix it. Guidance and Counseling Teacher or Counselor welcomes positively to awareness and enthusiasm of students accompanied prepare themselves in use AUM PTSDL and utilization results to improve the quality of their learning activities by Guidance and Counseling services, which organized through various types of services and their supporting activities in specific format. The school’s leadership have role in facilitating the implementation of Guidance and Counseling services and supports Guidance and Counseling Teacher or Counselor performance at school.

Abstract

This research is motivated because supervision of Guidance and Counseling (GC) in the senior high school was not optimum. The purpose of this study was to reveal and describe the implementation of the basic tasks related to monitoring, assessment, and coaching by GC Supervisor to GC Teacher or Counselor. This study used quantitative approach with descriptive methods. The samples in this research were 33 people GC Teacher or Counselor at senior high school level in city of Pariaman with total sampling technique. The instrument used is Likert scale models. Finding of this study are as follows: (1) the quality of the implementation of monitoring by the Supervisor GC into GC Teacher or Counselor in senior high school are in the enough category, and the vocational high school are in the enough category,(2 the quality of the implementation of the assessment by GC Supervisor into GC Teacher or Counselor in senior high school are in the enough category, and the vocational high school are in the enough category, (3) and implementation guidance for GC Teacher or Counselor by GC supervisor in senior high school are in the enough category, while the vocational high school also in good category. Monitoring, assessment, and guidance that carried by GC Supervisor for GC Teacher or Counselor still needs to be improved and developed in order to advance the implementation of services GC in senior high school. The implication of this study can be used as a reference and material for consideration on the implementation of the basic tasks of GC Supervisor in senior high school.
