Abstract

Hypertension in pregnancy is one of the causes of morbidity and mortality in mothers and babies. PlGF has been shown to be associated with the diagnosis of hypertensive disorders in pregnancy. Pregnant women with hypertension are characterized by reduced circulating PlGF levels, even before clinical signs and symptoms. A decrease in the amount of PlGF is positively correlated with the severity of disease progression This study aimed to analyze the relationship between PlGF levels and blood pressure in pregnant women with hypertension. Observational analytic with cross sectional study design.  Sampling was total sampling. The number of subject were 34 peoples. The results showed that there was no significant correlation between PlGF levels and blood pressure in pregnant women with hypertension. PlGF levels have no relationship with blood pressure.

Abstract

Prolonged labor, also known as failure to progress, occurs when the total duration of labor is more than 24 hours and if not handled properly will cause various complications for the mother and baby and even lead to death. This study aimed to determine the effectiveness of labor dance on the duration of the first active phase of labor in nulliparous women. This study was a true experimental randomized controlled trial (single blind) with a posttest only with control group design. This research was conducted from 1 February to 31 March 2023 at the Central General Hospital Prof. dr.I.G.N.G Ngoerah Denpasar and Regional General Hospital Wangaya Denpasar. 42 primigravida mothers were randomly selected according to arrival, with 21 people in the intervention and control groups. Data were analyzed using the Mann Whitney test. The average length of the first stage of labor in the experimental group was 252,62 minutes and in the control group was 355,95 minutes (p-value=0,000); effect size of 1,61 (strong effect) with a percentage of 94,5%. There was a statistically significant difference in the duration of the active phase I stage of labor between the experimental and control groups. Abstract

Metallothionein is an important protein for detoxifying heavy metal ions. This research was conducted to isolate and clone cDNA of gene encoding for metallothionein type 2 from Melastoma affine. Total RNA was isolated from young leaves. Total cDNA was synthesized from the total RNA by reverse transcription. The MaMt2 cDNA was successfully isolated by PCR technique. The MaMt2 cDNA was inserted into pGEM-T Easy and the recombinant plasmid was successfully introduced into Escherichia coli DH5α. DNA sequencing analysis showed that this cDNA is full length consisting of 246 pb encoding 81 amino acid residues. This cDNA is identical to mRNA of AtMt2 from Arabidopsis thaliana. It does not contain any restriction sites found in the cloning sites of pGEM-T easy. The deduced protein of MaMT2 contains 14 cysteine residues distributed in the Cys-Cys, Cys-X-Cys, and Cys-X-X-Cys motifs.   Key words: cDNA, metallothionein, Melastoma affine, cloning, cysteine

Abstract

Lactobacillus casei WSP-derived an aspartic protease was sequentially purified by using chromatography gel filtration sephadex G-50. It resulted in a 22.81-fold increase of specific activity (51.5 U/mg) with a final yield of 1.9%. The estimated molecular weight of the purified enzyme was 37 kDa and showed gelatinolytic activity in zymogram assay. The enzyme exhibited optimum activity at 40ºC and pH 6 with casein as the substrate. Enzyme activity was significantly inhibited by pepstatin A (0.5 mM and 1 mM), confirming that this enzyme is a group of aspartic proteases, while other inhibitors such as EDTA, PMSF and iodoacetic acid showed no inhibition effect on the activity of enzyme. The addition of metal ion to the enzyme decreased enzyme activity, indicating the proteolytic enzyme was metal ion- dependent. Denaturant such as DDT tended to increase caseinolytic activity. Furthermore, this enzyme was capable of generating the new peptides from skimmed milk with the size 8 kDa, 10 kDa and 15 kDa. These peptides have potential as antibacterial and antioxidant agents.

Abstract

The embryogenesis (SE) has important role for genetic engineering of cassava (Manihot esculenta Crantz). However, the success of SE induction depend on plant growth regulator s (PGR)s and treatment enriched in induction media. This experiment tried to induce callus formation of cassava from several in vitro explants: immature leaf, apical bud, and internode; and to develop somatic embryogenesis of cassava in several media enriched with tyrosine and copper sulphate (CuSO4) added into media enrich with picloram as treatment. Different response of explants source to callus induction treatment from those three varieties in callus induction as well as friable callus formation were found in this experiment. The best medium to induce varied with variety; MS media supplemented 12 mg/L picloram + 0.5 mg/L CuSO4 was the best for “Adira 4” and half MS and half GD media supplemented 12 mg/L picloram + 100 mg/L tyrosine for “Malang 6”. All treatments resulted somatic embryo which developed indirectly and in morphologically normal somatic embryos

Abstract

The synthetic gene of CalBsyn was previously constructed to encode Candida antarctica lipase B (CALB). Lipase of CalBsyn gene is slightly different from wild type CALB (CALB-wt) where it has three amino acids substitutions at different positions, i.e. V210I, A281E, and V221D, in order to improve its thermostability and catalytic efficiency. The CalBsyn gene was isolated from pJ912-CalBsyn vector by digestion using XhoI restriction enzyme. The 1136 bp fragment of CalBsyn gene was then ligated to pGAPZα expression vector and transformed into Escherichia coli TOP10F’ to obtain recombinant vector pGAPZα-CalBsyn. The result show that pGAPZα-CalBsyn recombinant vector was successfully transformed into E. coli TOP10F’ with transformation efficiency of 4.11x103 cfu/μg plasmid DNA. The pGAPZα-CalBsyn recombinant plasmid was successfully introduced into Pichia pastoris SMD1168H using electroporation method with transformation efficiency of 1.01x102 cfu/μg DNA. Recombinant protein expression was analyzed in several selected P. pastoris recombinant strains. Qualitative lipase activity assays results show that transformed P. pastoris-produced extracellular recombinant lipase (CALB) showing lipolytic activity; while results of quantitative lipase activity assays show that this Pichia-derived lipase achieved an activity of 6.35 Units/mL within 48 hours. SDS-PAGE analysis confirms the succesfull expression of CALB protein with molecular size was approximately 45 kDa.

Abstract

This research aims to find out the strategy for developing Mangrove ecotourism in the coastal area of Kertomulyo village, Trangkil sub-district, Pati Regency. This research is classified as a type of field research, with a qualitative-descriptive method. The data collection techniques use observation, interviews and documentation. And data analysis using SWOT theory. The results of this research show that the strengths possessed by the organization in carrying out the strategy that has been determined are the availability of applicable laws and regulations, the availability of human resources both in quality and quantity, sufficient availability of supporting facilities and infrastructure, and awareness from the public. The weaknesses that an organization has in implementing the strategy that has been determined can be an obstacle to the success of the strategy, such as: Lack of professionalism of the employee apparatus. Public awareness regarding the implementation of development has not yet accumulated. Accurate data regarding the development that will be implemented is not yet available. There has not been a culture of transparency in budget management and good management. Opportunities or opportunities that an organization has in carrying out a predetermined strategic plan can be supporting factors that will have a positive impact on the success of the strategy, including: The existence of business opportunities. There is development in the business world. The occurrence is cross-sectoral. Increased mobility of goods and services. There is coordination from related agencies. Threats definitely occur in every organization which can be a trigger to increase an organization's efforts and creativity in carrying out planned strategies. Some of the threats in tourism strategies are: High business competition, lack of development of facilities and infrastructure, lack of government role, and lack of attractiveness and uniqueness of tourist attractions.

Abstract

Students need the ability to explore, think critically and analyze a problem for a logical and appropriate solution. Problem solving abilities accompanied with a belief in their abilities (self efficacy) will maximize student abilities in and get maximum grades. This research aims to analyze the correlation between self efficacy and students' problem-solving abilities in ecosystem material. The method used is a quantitative method with a correlational approach (relationship study). The data collection technique was in the form of a problem-solving ability test in the form of an essay about ecosystem and a self efficacy questionnaire. The data analysis technique uses the Product Moment correlation test. Based on the results of data analysis and hypothesis testing, a correlation coefficient value of 0.675 was obtained, which means that the correlation coefficient level is strong. It can be concluded that there is a correlation between self efficacy and problem-solving abilities of students in ecosystem material with a strong correlation coefficient.

Abstract

Cottonii seaweed (Kappaphycus alvarezii Doty) is one of the most important commercial sources of carrageenan, which is widely used in the pharmaceutical and food industries. A major problem in the cultivation of this seaweed is ice-ice disease, which is caused by extreme changes in environmental conditions such as temperature and seawater salinity. Gene transformation to produce transgenic Kappaphycus lines that are tolerant to environmental stress is a potential solution to this problem. The Gα gene, which encodes the heterotrimeric G-protein alpha subunit, plays an important role in tolerance to biotic and abiotic environmental stress. This study aimed to: (a) introduce the Gα gene into the callus cells of K. alvarezii and regenerate transformed callus cells into transgenic plantlets; and (b) determine the appropriate concentration of acetosyringone and Agrobacterium tumefaciens strain for successful gene transfer into the callus of K. alvarezii. The callus cells of K. alvarezii were transformed using Agrobacterium tumefaciens strains LBA4404 and EHA105 carrying the expression vector pGWB502-Gα under the control of the CaMV-35S promoter. The calli and A. tumefaciens were co-cultivated in different concentrations of acetosyringone (20, 40, and 60 mg/L). The regeneration of transformed callus cells into transgenic plantlets was successfully achieved using the somatic embryogenesis technique. The results showed that the highest percentage of putative transgenic micropropagule formation occurred at acetosyringone concentrations of 20–40 mg/L. Polymerase chain reaction (PCR) analysis of twenty regenerated plantlets indicated that the Gα gene was successfully introduced into the genomic DNA of all samples. The highest transformation efficiency was obtained from the 20–40 mg/L acetosyringone co-cultivation treatment (22–28%). The transformation efficiency produced by Agrobacterium tumefaciens EHA105 (23%) was not significantly different from that produced using strain LBA4404 (15%).
