Abstract

ABSTRAK. Hati adalah kelenjar terbesar yang terdapat di dalam tubuh dan dapat dianggap sebagai organ pusat dalam pemeliharaan suplai energi. Penelitian ini bertujuan untuk mengetahui histologi, histomorfometri, dan histokimia sebaran glikogen pada hati ayam buras sebelum dan sesudah menetas. Sampel penelitian dibagi menjadi empat kelompok tingkat umur berbeda dan masing-masing kelompok berjumlah enam hati ayam. Hasil penelitian menunjukkan perbedaan nyata (P0,05) pertumbuhan hati ayam buras secara makroskopis. Pada setiap perkembangan sudah terdapat hepatosit berbentuk poligon yang memiliki inti sel bulat di tengah hepatosit, terdapat juga vena central yang dilapisi sel endotel pipih selapis serta didapati sinusoid yang berada diantara hepatosit, dan juga ditemukan Kupffers cell hanya pada hati ayam setelah menetas hari ke-7. Hasil pengukuran diameter vena central hati ayam buras hari ke-7, hari ke-14, hari ke- 20 sebelum menetas dan hari ke-7 setelah menetas berturut-turut adalah 17,70 1,16 m; 60,32 2,22 m; 49,316 1,93 m; dan 129,11 7,43 m. Hasil histokimia sebaran glikogen pada hati ayam buras hari ke-14, hari ke- 20 sebelum menetas dan hari ke-7 setelah menetas berturut-turut adalah 1,44 0,93; 2,27 0,74; dan 2,77 0,15. Dapat disimpulkan, bahwa diameter vena central dan sebaran glikogen meningkat seiring pertumbuhan ayam buras.(Histological, histomorphometrical, and histochemical of liver on native chicken (Gallus gallus domesticus) during pre and posthatch period) ABSTRACT. The liver is the largest gland in the body and considered as a central organ in maintaining energy supplies. The study aimed to determine the histologic, histomorphometric and histochemical of glycogen distribution in the liver of native chicken during pre and posthatching period.. The study sample were divided into four different age groups and each group consisted of six chicken livers. The results showed a significant difference (P0,05) in native chicken liver growth macroscopically. In every development there are already polygonal hepatocytes that have a round cell nucleus in the middle of the hepatocytes, there is also a central vein that is coated with a layer of flat endothelial cells and also sinusoids that are between the hepatocytes, and kupffer's cells also found in chicken liver 7th day posthatching. The results measurements of the central vein diameter in liver native chickens on the 7th, 14th, 20th pre hatching period and 7th post hatching were 17.70 1.16 m; 60.32 2.22 m; 49.32 1.93 m; and 129.11 7,43 m; The measure results of the glycogen distribution in liver native chickens on the 14th, 20th pre hatching period and 7th post hatching were 1,44 0,93; 2,27 0,74; and 2,77 0,15. It can be concluded, that central vein diameter and glycogen distribution were increased by growth of native chickens.

Abstract

The gain body weight and Spoilage of meat broiler which giving jaloh extract to combine with chromiumABSTRACT. A study was conducted on broiler chickens under heat stress to evaluate the effect of combined jaloh (Salix tetrasperma Roxb) extract with chromium mineral in the body weight, value of feed ratio conversion, and time to meat to become decomposed. Twenty four of 20-day old Cobb female broiler chickens were randomly assigned and divided to 4 treatment groups. Completely randomized design was used in this study. The treatments were as follows: 1) chickens given no heat stress and jaloh extract nor chromium (tCp); 2) chickens under heat stress without given jaloh extract nor chromium (Cp); 3) chickens under heat stress and given jaloh extract in 1.000 mg/lt drinking water (Cp+Ej); and 4) chickens under heat stress, given jaloh extract in 1.000 mg/lt drinking water, and chromium in 1.000 ug/lt drinking water (Cp+Ej+Cr). Heat stress given was 33 1oC of cage temperature during 4 hours per day within 15 days. Jaloh extract and chromium treatments in drinking water were given at 2 hour before cage temperature reaching 33 1oC and were stopped being given after 1 hour, when cage temperature back to room temperature. On the 16th day of the study chickens were measured before slaughtered. Samples obtained were chicken meat that taken from breast part (musculus pectoralis). The result suggested that either giving jaloh extract per se at the dose of 1.000 mg/lt in drinking water or combine it with chromium that given two hours before cage temperature reaching 33 1oC, can prevent chickens from decreasing their body weight, decreasing value of feed ratio convertion, and extending time to meat to become decomposed.

Abstract

Collagen is one of the determinant factors of beef tenderness. This study analyzed the expression and content of type III collagen connective tissue in two muscle types of Aceh cattle through immunohistochemical staining. Meat samples were obtained from the gluteobiceps and psoas major muscles of three Aceh cattle bulls weighing 250-300 kg, with a body condition score of three. Histology preparations were created from samples and subjected to immunohistochemical staining utilizing specific antibodies targeting type III collagen. Qualitative data were analyzed descriptively, while quantitative data were analyzed using IMAGE J software. The results showed that the expression of type III collagen connective tissue was found in the endomysium and perimysium layers of both muscles. The expression of type III collagen connective tissue varied significantly among the muscles (P<0.05). The gluteobiceps muscle had an average percentage of the area containing type III collagen connective tissue of 4.47±2.41%, while the psoas major muscle was lower at 2.51±1.69%. This study concludes that the gluteobiceps muscle of Aceh cattle has a higher amount of type III collagen connective tissue than the psoas major muscle.

Abstract

Accurate determination of estrus status in an animal based on an appropriate estrus detection method can guarantee the cows' pregnancy after mating. This study aims to determine the estrus status of Aceh cows using the van Eerdenburg estrus behavior scoring method. This study was conducted from 22 January to 12 February 2021, at the Technical Implementation Unit for Animals of the Faculty of Veterinary Medicine, Universitas Syiah Kuala, Banda Aceh, Indonesia. The sample used was eleven Aceh cows aged 4-6 years old. Parameters observed were the number of cows exhibiting signs of estrus using the estrus behavior scoring method. The accuracy of estrus detection was confirmed by palpation per rectum to detect the presence of uterine tone. By observing signs of estrus using the method studied, 90.9% (10/11) of the cows were determined to be in estrus. Confirmation by the presence of uterine tone showed a 100% accuracy rate. The most effective sign of estrus was the passivity of the cow when being ridden by another cow or bull. This study concluded that estrus status in local cows could be determined using the estrus behavior scoring of the van Eerdenburg method.

Abstract

The objective of this study was to determine the effect of ethanol extract of neem (Azadirachta indica) leaves on the number of spermatids and androgen receptor expression in the testicular tissue of New Zealand White (NZW) rabbits. Twelve adults male NZW rabbits with body weight (BW) 2-2.5 kg were divided into three treatment groups (n = 4), namely P1 (control group) given 0 mg/kg BW, P2 and P3 were given 200 mg/kg BW and 300 mg/kg BW of neem leaves ethanol extract respectively. The extract was administered orally using a gastric tube for 52 days. At the end of the treatment, the rabbits were terminated and the testes were collected to be processed into histological preparations and stained with Periodic Acid Schiff (PAS) staining to detect the number of spermatid and Immunohistochemical (IHC) staining to detect androgen receptor expression. The results showed that neem leaves extract administered at P3 rabbits significantly decreased the number of spermatids and androgen receptor expression in myoid peritubular cells and connective tissue P0.05), but not significantly different in the Leydig cells and blood vessels (P0.05) of testicular tissue. The conclusion of this study is that the ethanol extract of neem leaves at a dose of 300 mg/kg BW can be used as a contraceptive candidate compared to a dose of 200 mg/kg BW.

Abstract

This study aimed to analyze the muscle microstructure of longissimus dorsi Brahman cross (BX) cattle slaughtered at the slaughterhouse of North Aceh District. The meat samples for the study were taken from three male BX cattle aged 18-30 months with body weights ranging from 400 kg to 500 kg and the average body condition scores (BCS) of 3-4. The muscle used was the longissimus dorsi muscle. The mean (SE) diameter and cross-sectional area (CSA) of muscle fibers and the percentage of fast fibers in the longissimus dorsi BX muscle were 71.691.23 m, 4263.43305.69 m2 , and 48.178.81%, respectively. This study concluded that the longissimus dorsi muscle of the BX has relatively large muscle fibers, some of which in each fascicle are composed of fast muscle (fast fiber/type II).

Abstract

The aim of this research was to find out in vitro antiplasmodium activity of Malacca leaves (Phyllantus emblica) ethanolic extract against Plasmodium falciparum growth. In this study, Plasmodium culture contained 5% parasitemia in ring stage was cultured using candle jar method and antiplasmodial activity test was carried out using microculture. The treatments were divided into 7 groups with four repetitions. K1 as negative control group was given Roswell Park Memorial Institute (RPMI), while K2 as positive control group was given artesdiaquine. Groups K3, K4, K5, K6, and K7 group was added with 100 g/mL, 75 g/mL, 50 g/mL, 25 g/mL, and 5 g/mL of Malacca leaves ethanolic extract, respectively. Antiplasmodial activity was determined by inhibition concentration of 50% parasite growth (IC50). The data were analyzed using ANOVA and followed by Duncan test. The average of parasitemia level in group K1, K2, K3, K4, K5, K6, and K7 were 55.2515.62, 8.502.52, 8.503.00, 9.250.95, 9.002.70, 9.792.06, and 10.752.22, respectively. The average of inhibition percentage in group K1, K2; K3; K4; K5; K6; and K7 were 0.000.00%, 84.624.55%; 84.625.43%; 83.261.73%; 83.714,90%; 82.353,73%; and 80.546.83%, respectively (P0.01). The results showed that the administration of malacca leaves ethanolic extract significantly affect (P0.01) the inhibition of Plasmodium growth as compared to group K1 (negative control). Probit analysis reveals the IC50 value was 3.889 g/mL. In conclusion, all doses of malacca leaves ethanolic extract used in this study was able to inhibit Plasmodium falciparum growth with IC50 value was 3.889 g/mL.

Abstract

The objective of this study was to evaluate the effect of storage time of epididymis at 5 oC on spermatozoa quality of Aceh local goat. This research was conducted at Reproduction Laboratory, FKH Unsyiah from May to June 2009. Nine epididymis from local goat at the age of 1.5-2.0 years were used in this study. The epididymis were collected from abattoir in Banda Aceh. The experimental design used in this study was completely randomized design with 3 treatments and 3 replications. Spermatozoa quality which consist of sperm concentration, motility, amount of life sperm, and sperm abnormality were examined after the collection of epididymis on day 0 (H-0), day 1 (H-1), and day 3 (H-3) post storage at 5 oC. Data were analyzed using one-way analyses of variance (ANOVA). The result showed that no significant difference (P0.05) seen in the spermatozoa quality after different storage time. The average of spermatozoa concentration on H-0, H-1, and H-3 were 318x107/ml, 282x107/ml, and 241x107/ml respectively. On the average, the percentage of spermatozoa motility on H-0, H-1, and H-3 were 82,272,75; 80,252,83; and 78,070,92%, respectively. Life spermatozoa observed on H-0, H-1, and H-3 were 82,292,71; 80,631,87; and 80,093,31%, respectively. Observation on spermatozoa abnormality showed that the average of spermatozoa abnormality on H-0, H-1, and H-3 were 7,230,27; 8,210,55; and 10,753,14%, respectively. It could be concluded that the spermatozoa quality were not affected by the storage time at temperature of 5 oC.

Abstract

This study aimed to verify the effect of ethanol extract of jaloh leaves at various doses on parasitemia percentage of in mice (Mus musculus), wich inoculated Plasmodium berghei. This study used 12 male mice (Mus musculus) strains Balb / C with average body weight of 30.2 grams of 2-month-old. This research using Completely Randomized Design (CRD) with four treatments and three replications. All the mice were infected with Plasmodium berghei prior to treatment on three days post infected the mice in group 2,3 and 4 were given with extract ethanol jaloh leaves at dose 50, 100, 150 mg/kgbw inspectively. The mice in group 1 (K0) were not given anything and consident as control.Observations parasitemia percentage using microscopy on preparations stained thin blood smear Giemsa. The parasitemia percentage data were analyzed using one way ANOVA. On the average (SD) percentage of parasitemia during the three days in four treatments (K0, K1, K2, and K3) on the first day were 24.871.86; 24.331.53; 24.201.06; and 24.931.01. On the second day were 22.672.52; 23.001.00; 22.801.20; and 24.132.01; and on the third day were 24.771.66; 22.973.76; 21.131.90; and 24.271.42. Statistical analysis showed that ethanol extract of the jaloh leaves for KO, KI, KII, and KIV respectively treatment did not affect the percentage of parasitemia in mice (Mus musculus) inoculated Plasmodium berghei.

Abstract

The study was done to investigate the glycogen distribution in the stomach of long tailed macaques (Macaca fascicularis). The study used eight individuals of long tailed macaques consist of 6 fetuses (55, 70, 85, 100, 120, and 150 days pregnancy) and 2 infants (10 and 105 days). The gastrict tract were histologically stained using Hematoxyllin-Eosin (HE), and histochemically stained using Periodic Acid Schiff (PAS), to detect glycogen. The results revealed that the histogenesis pattern of stomach glandullar cells are developed in line with the age of the fetuses and infant. We also found that glycogen existance are increase in line with the age in the glandullar stomach. From the present study we concluded that the glycogen distribution in gastrict glandular cells were influenced by the age in the fetal period, while in the infant was under influence of the digestion activities, and the activities of the gastrict glandular cells was already seen at 55 days gestation.
