Abstract

Acathina fulica is a terrestrial mollusk that often invades terrestrial vegetation that has tropical and subtropical climates. Acathina fulica has been widely reported in environments as diverse as urban areas, forests, capoeiras, caatings, plantations, decomposing materials, savannahs and vegetable gardens. Soil is source of food and a defense strategy for A. fulica, therefore it has an important role for them. Therefore, A. fulica may be host several species of nematodes. Microscopic examination of snails A. fulica fecal samples in Cemorokandang Village, Malang City using the native method aimed to detect Rhabditis spp. The results showed that 14 (12.5%) out of 112 samples contained Rhabditis spp. The genus rhabditis belongs to the Rhabditidae family and is one of the free-living nematodes. This species has long been known and of concern in the veterinary world. The genus rhabditis has been reported to have infected humans and have been isolated from urine samples, vaginal swab, and human stool. The research was conducted in the dry season with low rainfall. Nematode of the genus Rhabditis spp. can be naturally found in humid places and decaying organic matter.

Abstract

Toxoplasmosis is one of the zoonotic diseases caused by the protozoa Toxoplasma gondii. The disease infects warm-blooded animals including humans. The purpose of this study was to determine the contamination of Toxoplasma oocysts in water sources in goat-sheep farms in Sidoarjo Regency. A total of 63 water samples were used for the study. Water samples were taken from 5 sub-districts that have many goat-sheep farms, each sub-district has 2 villages, with each village having 6-7 samples. The examination used the floating method and was examined under a microscope with a magnification of 400x. The results of the examination found 21 samples of Toxoplasma gondii oocysts or 33.33% (21/63). The morphology of T.gondii oocysts is round, ovoid or ellipsoid with sizes varying between 8.02 - 12.72 micrometers. Preventive measures are needed by treating water before giving it to livestock.

Abstract

The COVID-19 pandemic caused by SARS-CoV-2 requires effective vaccines to be developed. This study aimed to assess the impact of a SARS-CoV-2 inactivated vaccine candidate in k18-hACE2 mice by monitoring their body weight, immune activation, and inflammatory cytokines including IL-4, IL-6, TNF-α, and IFN-γ. The study utilized k18-hACE2 mice expressing the human angiotensin-converting enzyme 2 (hACE2) receptor. The mice were administered the inactivated vaccine candidate compared with sham and vehicle. Body weight was monitored, and serum samples were collected to measure IL-4, IL-6, TNF-α, and IFN-γ levels using ELISA. Data were evaluated using SPSS statistical analysis software. The administration of the SARS-CoV-2 inactivated vaccine candidate in k18-hACE2 mice did not result in significant changes in body weight compared to the control group. Furthermore, the levels of IL-4, IL-6, TNF-α, and IFN-γ were significantly reduced in the vaccinated mice compared to the control group, suggesting a dampening effect on the inflammatory response. This study demonstrates that the SARS-CoV-2 inactivated vaccine candidate has a minimal impact on the body weight of k18-hACE2 mice. Nevertheless, it successfully regulates the levels of IL-4, IL-6, TNF-α, and IFN-γ, suggesting its safety and beneficial impact. These findings contribute to understanding the vaccine's efficacy and safety profile in vaccine development.

Abstract

Lamongan Regency, located in East Java, Indonesia, is a significant center for beef cattle production. Despite its prominence, studies on the identification and differentiation of Eimeria spp. parasites in this region are notably lacking. This research aims to address this gap by evaluating the prevalence of Eimeria spp. and identifying two pathogenic species: Eimeria bovis and Eimeria zuernii. The study involved the collection of 250 fecal samples from beef cattle raised on smallholder farms across Lamongan. Sampling was conducted during the rainy season to optimize the detection of Eimeria infections. Using the sugar flotation method, Eimeria oocysts were isolated from 5–10 g of fecal matter per sample. Molecular identification employed polymerase chain reaction (PCR) assays targeting ribosomal RNA’s internal transcribed spacer 1 (ITS-1) region to detect the pathogenic species. Fecal examination using the Whitlock test revealed a prevalence of Eimeria spp. at 44.45% (111/250). PCR analysis further identified E. bovis with a 238 bp amplicon in Solokuro District and E. zuernii with a 344 bp amplicon in Tikung District, highlighting the presence of these two pathogenic species in distinct geographic areas. The findings underline the need for further research that includes expanded sampling from different regions and cattle breeds and the application of varied diagnostic methods. Such studies will contribute to a more comprehensive understanding of the genetic diversity, distribution, and epidemiology of E. bovis and E. zuernii in Indonesia, supporting improved cattle health and management practices in the region.

Abstract

Hymenolepiasis is one of the neglected zoonotic diseases in humans, caused by the cestodes Hymenolepis nana and Hymenolepis diminuta. Its definitive hosts are rodents (mostly rats). This study aims to identify the morphology both microscopically and macroscopically. This research is a laboratory exploratory research and the sampling method uses purposive sampling technique. The samples used in this study amounted to 100 samples of wild rats that had been trapped and then necropsied, worms obtained from the digestive tract were stained with dark red stain and observed with a microscope. The results showed that microscopic morphological examination showed that the anterior part of the H. nana worm had hooks and crowns. The overall prevalence of H. nana worms was found to be 18% (18/100). The prevalence of H. nana in wild rats in Surabaya City in Rattus tanezumi was 16% (12/77). Rattus novergicus in Surabaya area with positive samples was 26% (6/23). Morphology in H. nana worms has a distinctive difference. The anterior part of H. nana has hooks that surround the rostellum, the anterior part has a ‘crown of hooks’.

Abstract

Salah satu permasalahan yang sering dihadapi para peternak kerbau adalah penyakit Fascioliasis yang disebabkan oleh cacing hati atau Fasciola spp., dimana penyakit tersebut adalah salah satu penyakit yang bersifat zoonosis. Oleh sebab itu penelitian ini bertujuan untuk mengetahui prevalensi Fasciola spp. pada kerbau di Kabupaten Agam, Sumatra Barat. Sampel berupa feses 105 ekor kerbau dari tiga kecamatan lalu diperiksa dengan menggunakan metode natif (sederhana) dan sedimentasi sederhana. Hasil penelitian diperoleh 25 ekor dari 105 ekor kerbau dinyatakan positif terinfeksi Fasciola spp. Prevalensi yang diperoleh secara keseluruhan adalah 23,81% (25/105), dan hasil ini dapat dikembangkan dalam penelitian lebih lanjut.

Abstract

Cryptosporidiosis is a waterborne zoonosis caused by Cryptosporidium, which is a parasite that causes infection of the small intestine and leads to acute diarrhea in both humans and animals. Wild rats (Rattus spp.) are known to carry many zoonotic pathogens including Cryptosporidium. Given their close proximity to humans in urban environments, the likelihood of disease transmission is high. This study aims to detect the Cryptosporidium spp. infection in wild rats in Surabaya City, East Java Province. Intestinal fecal samples were collected using necropsy. The flotation test and Ziehl-Neelsen stain were used to observe the presence of Cryptosporidium under the microscope. The results showed that 69 samples were positive for Cryptosporidium spp. in the oocyst stage, which appeared dark pink with a clear cavity and the size of the oocysts ranged from 2-6 μm. The high incidence of cryptosporidiosis in wild rats is a potential health threat to both animals and humans.

Abstract

Subclinical mastitis is an economically important disease in dairy goat farming. Lumajang is a center for dairy goats in East Java. However, routine examination of subclinical mastitis for screening tests has no data. This study aimed to provide prevalence data and identification factors that influenced subclinical mastitis. Therefore, this study was carried out as part of the national surveillance program. The research design is a cross-sectional study with simple random sampling. Samples came from 19 farms with a total individual sample of 224 dairy goats. Detection of subclinical mastitis using an undirect test with the California Mastitis Test (CMT). Identification of management factors and characteristics that can influence subclinical mastitis in dairy goat farms using a structured questionnaire. The results showed that the prevalence of subclinical mastitis in Lumajang District was 36.2%. There was a significant difference in subclinical mastitis in each village (p<0.05). The highest prevalence of subclinical mastitis was in Kandangtepus village. A farmer's characteristic factor that significantly influences subclinical mastitis is the farmer's experience. The aspect of livestock management that significantly affects subclinical mastitis is structured work management. Subclinical mastitis was prevalent in dairy goats in Lumajang District and requires prevention efforts. Prevention approach through education for farmers to increase knowledge, attitude, and socialization to improve livestock management.

Abstract

The aim of this research is to know about the staging of spermatogenesis and leydig cells in mice that injected by Toxoplasma gondii after giving honey as a preventive substance. This research used 25 mice in age 2-4 month with 20-30 weight. Mice were divided into 5 groups (P0, P1, P2, P3 dan P4). P0 as a control teratment were given aquades in peroral, P1 was a group given honey in doses I (0.08ml/0.2ml/day) during 14 days in peroral, P2 was a group that given infection by T.gondii in doses 103 in peritonial, P3 was given honey in doses I 0.08ml/0.2ml/day) then injected by T.gondii, and P4 was given honey in doses II (0.12ml/0.2ml/day) then injected by T.gondii. This research conducted in 18 days, in day 14th mice were given honey and the day 15th–18th mice injected by T.gondii doses 103. The data result was analyzed with Analysis of Variant (ANOVA) and cotinued with Duncan. Comparision statistical showed with SPSS statistic 20 for Windows. The result of analysis, P0 and P1 were significantly different (p<0.05), with the increase number of spermatogenic and leydig cells. P0 and P2 also significantly different, with the decrease number of spermatogenic and leydig cells. In P2 and P3 group were not significantly different (P>0.05), also in P4 group were not significantly different with P0 and P1. The results of this research showed that honey in doses II can maintain the number of sprematogenic and leydig cells as a preventive substance against T.gondii.

Abstract

This study aimed to determine the occurences of mice brain ventricles dilatation that congenitally infected with Toxoplasma gondii (T. gondii) as a marker of hydrocephalus and cellular changes in the brain. A total of twenty pregnant mice (11.5 days pregnacy) were divided into 2 groups, which were control (P1) group and treatment (P2) group. The mice in the treatment group were infected with 101 tachyzoites of T. gondii. All mice were maintained until delivery. The newborn mice were sacrificed and their brain were removed and fixed in 10% buffered formalin to prepare histology slides with HE staining for observation of ventricular width, TUNEL assay for apoptosis observation, and immunohistochemistry for the expression of transforming growth factor beta (TGF-) observations. The data were analyzed using t test and linear regression. The results showed that ventricular width and apoptosis index significantly increased (P0.01) in the treatment group compared to control group, but there was no difference in the expression of TGF- (P0.05) in both groups. Dilatation of ventricle correlated with the apoptotic index of brain cells but did not correlated with the expression of TGF-.
