Abstract

Booklet is one of the learning media prepared for training or counseling for the community. The fruit craftsman community needs a booklet on "Nata de jambu, rich fiber fermented food " to help study the nata from Guava Darsono production. The booklets were prepared based on the experimental research result on "The Effect of Guava Darsono and Varieties and Sugar sorts on Nata Quality Based on the Thickness, Weight and Nata Fiber. The purpose of this study is to produce a booklet about nata from Guava Darsono production. This booklet development model uses the Hannafin & Peck development model. The booklet validator consists of: media expert validator, material expert validator, and 15 person fruit processing craftsman for legibility test respondent. Data collection technique by questionnaire. The research results is a booklet with the title "Nata de jambu, Rich Fiber Fermented Food " has been made. The validator's assessment stated that this booklet is worthy to be used as a learning media for fruit craftsmen.Booklet adalah salah satu media belajar yang disusun untuk keperluan pelatihan atau penyuluhan bagi masyarakat. Masyarakat pengrajin olahan buah membutuhkan booklet tentang “Nata de jambu, makanan fermentasi kaya serat” untuk membantu mempelajari pembuatan nata dari jambu Darsono. Booklet disusun berdasarkan hasil penelitian eksperimen tentang “Pengaruh Varietas Jambu Darsono dan Macam Gula terhadap Kualitas Nata Berdasarkan Tebal, Berat dan Kadar Serat Nata. Tujuan penelitian ini yaitu untuk menghasilkan booklet tentang pembuatan nata dari jambu Darsono. Model pengembangan booklet ini menggunakan model pengembangan Hannafin & Peck. Validator booklet terdiri atas validator ahli media, ahli materi, dan responden yaitu pengrajin olahan buah berjumlah 15 orang untuk uji keterbacaan. Teknik Pengumpulan data dilakukan melalui penyebaran angket. Hasil penelitian ini adalah Booklet dengan judul “Nata de jambu, Makanan Fermentasi Kaya Serat” telah tersusun. Hasil penilaian para validator menyatakan booklet ini layak digunakan sebagai media pembelajaran bagi para pengrajin olahan buah.

Abstract

Abstract: Learning is expected to provide learning experiences to students in order to understand learning material. The contextuality of learning resources and learning media must be improved. Biotechnology Handout "Utilization of Jackfruit Dami as the Baseline for Making Nata" is used to guide students to use learning media. This study aims to determine the effect of handouts on the ability to think creatively. This research is a quasi-experimental design with the pretest-posttest control group design method, using one control class and one treatment class as the research sample. Data analysis uses gain score and independent t-test. Based on the t-test, the Sig value is obtained. 2-tailed (0,000) <0.05. The results of the study prove that the use of handouts can improve students' creative thinking skills.Abstrak: Suatu pembelajaran diharapkan memberikan pengalaman belajar kepada siswa agar dapat memahami materi pembelajaran. Konstekstualitas sumber belajar dan media pembelajaran harus ditingkatkan. Handout Bioteknologi “Pemanfaatan Dami Nangka sebagai Bahan Dasar Pembuatan Nata” digunakan untuk menuntun siswa menggunakan media pembelajaran. Tujuan penelitian ini yaitu mengetahui pengaruh handout terhadap kemampuan berpikir kreatif. Penelitian ini merupakan penelitian Quasi-eksperimental design dengan metode pretest-postest control group design menggunakan satu kelas kontrol dan satu kelas perlakuan sebagai sampel penelitian. Analisis data menggunakan gain score dan uji-t independen Berdasarkan uji-t diperoleh nilai Sig. 2-tailed (0,000) < 0,05. Hasil penelitian membuktikan bahwa penggunaan handout dapat meningkatkan kemampuan berpikir kreatif siswa.

Abstract

Padi (Oryza sativa L.) adalah tanaman pangan utama masyarakat Indonesia. Masalah kekeringan dalam upaya penanaman padi adalah hal yang biasa di kalangan petani. Penelitian tahan cekaman kekeringan pada padi perlu dilakukan untuk mengatasi masalah ketersediaan pangan. Sifat tahan cekaman kekeringan pada tanaman salah satunya dikendalikan oleh gen DREB2A. Varietas padi lokal Indonesia untuk tahan cekaman kekeringan belum banyak diteliti. Penelitian ini bertujuan untuk mempelajari varietas padi lokal Jawa Timur yang tahan terhadap cekaman kekeringan menggunakan gen DREB2A, yang diisolasi menggunakan primer forward 5’-CCT CAT TGG GTC AGG AAG AA-3’ dan primer reverse 5’-GGA TCT CAG CCA CCC ACT TA-3’. Penelitian ini menggunakan tiga varietas padi lokal, yaitu Berlian dan SOJ A3, keduanya dari Banyuwangi, dan varietas Jawa dari Malang. Isolasi DNA total dilakukan mengikuti protokol Kit Macherey-Nagel. Amplifikasi gen DREB2A dilakukan menggunakan metode Polymerase Chain Reaction (PCR). Fragmen gen DREB2A berhasil diamplifikasi sepanjang 239 bp (varietas Berlian), 239 bp (varietas SOJ A3), dan 240 bp (varietas Jawa). Analisis sampel gen DREB2A varietas padi lokal Jawa Timur menunjukkan hilangnya kemampuan gen DREB2A dari varietas Jawa untuk mengikat cis-regulator elemen promotor gen target.

Abstract

Abstract - Petunia has been used as research object on flavonoids studies especially anthocyanin biosynthesis. Hf1 gene encode F3'5'H enzyme which plays a role in the synthesis of color in Petunia. Research on the regulation of Petunia color is potential for the development of ornamental plant breeding in Indonesia, hence the information regarding the gene analysis is necessary. Recently, there are many reagent kits for the isolation and purification of plant DNA available, each has different protocols regarding the special character of plant tissue with various secondary metabolites accumulation. Secondary metabolites in plants have different chemical contents, for that reason its needs to develop a proper protocol resulting in pure DNA with high concentrations. This study aims to obtain the effective and efficient DNA isolation and purification protocol for Petunia hybrida. Optimization results showed that samples immersed in absolute ethanol for two days then dried and stored in the freezer until used have given the optimal results for further analysis. Abstract

The White-bellied Sea Eagle (Haliaeetus leucogaster) is one of the top predators living in the coastal area of Indonesia. This species has the range of distribution in all across Indonesian archipelago. However, this raptor population decreases particularly in the Java southern seas due to illegal hunts and the decreasing quality of their natural habitat. Meanwhile, this bird is still poorly studied in Indonesia. This research focuses on the identification of Haliaeetus leucogaster and the phylogenetic study of this species by means of morphometrical analysis and DNA barcode cytochrome-c oxcidace sub unit I (COI) in regards to the conservation of thisspecies. The method used in this research is by measuring the morphological characteristics and mitochondrial DNA isolation using Forward primer BirdF1 5’- TTC TCC AAC CAC AAA GAC ATT GGC AC-3’ and Reverse primer BirdR2 5’ ACT ACA TGT GAG ATG ATT CCG AAT-3’. The phylogenetic analysis using MEGA 6 with Maximum Likelihood method shows that Haliaeetus leucogaster in this study is closely related to Haliaeetus albicilla, Haliaeetus leucocephalus dan Haliaeetus pelagicus.  Keywords: phylogenetic study, Heliaeetus leucogaster, morphological chacarteristics, DNA barcoding, Cytochrome-c Oxcidace Sub Unit I (COI)

Abstract

Abdopus aculeatus has good camouflage capabilities in nature. The objective of this study is to obtain information on the camouflage behavior of Abdopus aculeatus in nature (in situ). We used an explorative description to observe the camouflage behavior of 24 octopus samples at a number of points on the Panjang Island, Buton Regency. Abdopus aculeatus was identified molecularly using a mtDNA COI blast to determine the target species of observation. Any of the camouflage behavior found was documented and analyzed descriptively. Generally, there are four forms of observations for camouflage behavior; matching body color to wide array of natural background, immersing itself in the sand, resembling a specific object, and being part of the objects surrounding. The various camouflages patterns obtained show a process of defense evolution in minimizing the detection of predators or octopus prey.

Abstract

The purpose of this study is to know what kinds of sexual abuse that have been done to the women figures in the novels “Perempuan di Titik Nol” by Nawal El Saadawi and “The Color Purple” by Alice Walker. Besides, the purpose of the study is to know the impact of sexual abuse that happened to the women. To achieve the objectives above, the study used qualitative design and feminism approach. The re- sult of the studyshows that sexual abuse happened to the leading women figures in the novels from before, during, and after theirmarriage. The kinds of sexual abuse which happened to the leading women figures are gender harrassment,  seduction behaviour, sexual bribery, sexual coercion, dan sexual imposition

Abstract

SARS-CoV-2 is a virus that has caused COVID-19 pandemic. This virus is a new variant of the SARS-CoV virus and also closely related to MERS-CoV, which caused similar acute respiratory infections. All these viruses recognize target cells by binding to the Receptor Binding Domain (RBD) on Spike protein with receptors. This study aimed to determine the SARS-CoV-2, MERS-CoV, and SARS-CoV genome structure, Spike protein sequence differences, and variations of RBDs Receptor Binding Motif (RBM). This research was using data mining approach. Genome sequences were downloaded from NCBI, except for Indonesian samples were downloaded from GISAID. Genomic structures, Spike sequence, and RBD structure were analyzed using Bioedit, followed by protein modelling using SwissModel and PyMol applications. The result showed that the SARS-CoV-2, MERS-CoV, and SARS-CoV genome shared the same genes yet in different numbers and length. SARS-CoV-2 Spike protein sequence was quite similar to SARS-CoV Spike protein, but very different to the Spike protein of MERS-CoV. There were variations of RBDs RBM structure due to the mutations occurred among these viruses. It is suggested that these differences may increase the affinity between SARS-CoV-2 Spike protein to its hACE2 receptor which caused SARS-CoV-2 becomes more infective and spread wider than SARS-CoV and MERS-CoV, in turn. This result expected to be basic information for the development of SARS-CoV-2 introduction inhibition agent and spreading prevention.

Abstract

This study aimed to investigate students' understanding of the nature of science (NOS) in project-based learning combined with mind mapping (PjBL-MM), students' NOS understanding across gender, and PjBL-MM and gender interaction's effect on students' NOS understanding of conservation education. It employed a pretest-post-test non-equivalent group design. The research population consisted of first-year students at the Biology Department, Faculty of Mathematics and Natural Sciences, Universitas Negeri Semarang, Central Java, Indonesia. Thus, the study consisted of 98 students (40 males and 58 females) selected randomly from three different classes. The students' NOS understanding was assessed using the views of NOS type B questionnaire. The learning models' effectiveness was tested using ANCOVA. The results showed a significant difference in students' NOS understanding, PjBL-MM group reported the highest NOS score among all treatment groups in the Conservation education course. There is a significant difference NOS understanding between male and female students. Females outperformed males in NOS understanding. However, PjBL-MM and gender interaction did not affect students' NOS understanding. This study is expected to encourage the implementation of PjBL-MM to improve the students' NOS understanding. The educators are also expected to empower NOS understanding through students' active participation in science by implementing project-based learning combined with mind mapping techniques.
