Abstract

This study is about the analysis of language anxiety factors affecting Technical department 51 Batch 4th semester cadets‟ speaking performance. The aim of this study is to list the language anxiety factors that affect cadets‟ speaking performance and to explain how the language anxiety can affect cadets‟ speaking performance. The data of this study were taken from questionnaire, open-ended interview, and students‟ speaking performance score. There were 60 cadets from two classes on class F and G of Indonesia Merchant Marine of Semarang. The questionnaire data were analyzed based on Horwitz‟s theory of Language Anxiety. Then for the language anxiety factors, each statement was calculated to know five chosen statement. The relation between language anxiety and students‟ speaking performance were analyzed based on students‟ interview answer, speaking score, and anxiety level. There were five factors of language anxiety affects students‟ speaking performance: 1) lack of preparation during language class, 2) peer-pressure in language as a whole, 3) sudden-activity in class, 4) peer-pressure in English speaking, and 5) students‟ negative attitudes toward English language. There were no solid correlation between students‟ speaking performance and language anxiety, what made their speaking performance differ from one to another was their linguistic competence and how they cope with their anxiety during speaking class.

Abstract

The caudal fin of Zebra fish as a model for studies regeneration both in terms of process and study of molecular analysis. This research is a descriptive exploratory. A sample of zebra fish (Danio rerio) 4-6 months of age as much as 30 individuals. The regeneration caudal fin of zebrafish observed by microscope with cutting vertically. The parameters observed were: 1) the time required for the regeneration process, 2) The regeneration process in details covering and, 3) Type of regeneration. Data in the form of the regeneration process ranging from wound healing to the original length obtained are collected in the form of digital photos. The results showed that the time required to regenerate the caudal fin to recover to normal at a temperature of 22-31° C was 22.25 ± 1.63 days for a model of vertical cuts. The regeneration process begins with the events of wound healing (H1) followed by the formation of blastema and will radius (H2) and continued differentiation characterized by the process of angiogenesis (H2). Regenerative development begins with the formation of the fingers and the growth of the peripheral areas tail fins faster than the medial region (H3-H8). End process regeneration in caudal fin of zebrafish are included in the development is the formation of regenerative segment radius starting from the proximal to the distal portion regenerat regenerat (H9 - ± H21). From the results of this research note that caudal fin of zebrafish to regenerate in epimorphosis.

Abstract

Gatul fish in included in the family Poecilidae which is a small fresh water, male is smaller and more colorfull than female. This fish is easy to be taken care in laboratory and has a short reproduction cycle, thus allowing these fish to be one animal model development in the field of biology. Yet, in Indonesia the biological study upon Gatul fish is very limited. The aim of this study is to understand the process of caudal fin regeneration of Gatul fish are malformation (abnormal growth). Caudal fin of Gatul fish were vertically cut in a half of its length. Observation was done using 400x magnification. Picture with taken by camera with digital zooming. The subject of this study with a body length of 2-3 cm. The result shows that regeneration of Gatul caudal fin is started with wound healing in day-1 (D1= 24 hours after amputation) and blastema formation, then stump regeneration begin to grow in D2. Regenerative outgrowth begin in D3 characterized by the formation Blood island and fin rays. Differentiation begin in D6 and extended up to D18. From this research we can conclude that the process is going step by step through wound healing, stump growth, and tissue differentiation, and respectively

Abstract

Padi (Oryza sativa L.) adalah tanaman pangan utama masyarakat Indonesia. Masalah kekeringan dalam upaya penanaman padi adalah hal yang biasa di kalangan petani. Penelitian tahan cekaman kekeringan pada padi perlu dilakukan untuk mengatasi masalah ketersediaan pangan. Sifat tahan cekaman kekeringan pada tanaman salah satunya dikendalikan oleh gen DREB2A. Varietas padi lokal Indonesia untuk tahan cekaman kekeringan belum banyak diteliti. Penelitian ini bertujuan untuk mempelajari varietas padi lokal Jawa Timur yang tahan terhadap cekaman kekeringan menggunakan gen DREB2A, yang diisolasi menggunakan primer forward 5’-CCT CAT TGG GTC AGG AAG AA-3’ dan primer reverse 5’-GGA TCT CAG CCA CCC ACT TA-3’. Penelitian ini menggunakan tiga varietas padi lokal, yaitu Berlian dan SOJ A3, keduanya dari Banyuwangi, dan varietas Jawa dari Malang. Isolasi DNA total dilakukan mengikuti protokol Kit Macherey-Nagel. Amplifikasi gen DREB2A dilakukan menggunakan metode Polymerase Chain Reaction (PCR). Fragmen gen DREB2A berhasil diamplifikasi sepanjang 239 bp (varietas Berlian), 239 bp (varietas SOJ A3), dan 240 bp (varietas Jawa). Analisis sampel gen DREB2A varietas padi lokal Jawa Timur menunjukkan hilangnya kemampuan gen DREB2A dari varietas Jawa untuk mengikat cis-regulator elemen promotor gen target.

Abstract

Padi lokal merupakan padi liar yang telah dibudidayakan oleh petani Indonesia. Potensi padi lokal dapat digunakan sebagai plasma nutfah yang memiliki berbagai gen dalam mengendalikan sifat tertentu. Beberapa beras di Indonesia memiliki potensi sebagai beras aromatik. Berbagai varietas padi aromatik berpotensi meningkatkan kualitas beras di Indonesia. Aroma beras dikendalikan oleh gen betain aldehid dehidrogenase (BADH). Gen ini terdiri dari dua alel yaitu BADH1 dan BADH2, mengkodekan enzim yang bertanggung jawab untuk mengkatalisis sintesis senyawa 2-acetyl-1-pyrroline (2-AP). Penelitian ini bertujuan untuk mengidentifikasi potensi aroma varietas Berlian. Gen BADH2 diisolasi dari daun pada ketiga varietas tersebut dan diamplifikasi menggunakan primer forward 5'-TTGTTTGGCTCTTGCTGATG -3' dan primer reverse 5'CATAGGAGCAGCTGAAATATATACC-3'. Fragmen gen BADH2 berhasil diamplifikasi dengan ukuran 258 bp (varietas Berlian). Sekuen BADH2 varietas Berlian menunjukkan kemiripan 100% dengan sekuen BADH2 padi tidak beraroma dan tidak memiliki kemiripan dengan sekuen BADH2 varietas padi beraroma. Analisis potensi padi beraroma menunjukkan bahwa varietas Berlian tidak berpotensi sebagai padi beraroma.

Abstract

This research was a study which attempted to explain how effective the use of Instagram in improving students’ skill of writing procedure texts is, and to explain the factors that make Instagram improves students’ skill of writing procedure texts. This study used quasi-experimental. The subjects of the study were the 53 batch students of 4th semester of Technical Department of Politeknik Bumi Akpelni. There were two groups in this study, TB as the control group and TC as the experimental group, in each group there was 24 students. The control group was taught by using conventional method, while the experimental group was taught by using Instagram as a medium. The resultanalysis of the quantitative data shows that the mean scores of the experimental group increases from 64.13 to 82.45. The result indicates that the working hypothesis is accepted. It means that the there is significant difference between students who were taught by using Instagram and those who were taught by using conventional method. In addition, the analysis of the questionnaire shows that 51.61% of the students agree that the implementation of Instagram assists their writing. Therefore, the analysis reveals that Instagram is effective in teaching writing procedure text.

Abstract

Chili pepper is widely used and cultivated by Indonesian people. There are three species of chili pepper, i.e.: Capsicum annuum L., Capsicum frutescens L., and Capsicum violaceum HBK. Capsicum frutescens L. has a higher economic value due to its pungency and carotenoid content. C. frutescens has several cultivars, one of those is Capsicum frutescens cv. Cakra Hijau. This cultivar is resistant against pest and disease and has very high pungency. This special character of chili pepper is born by its secondary metabolic, Capsaicin. Moreover, capsaicin also serves as defense mechanism, antiarthritis, analgesic, and anticancer. This study aimed to isolate Acyltransferase (AT3) gene which encoding Capsaicin Synthase (CS) enzyme. AT3 gene was isolated through PCR using forward primer 5-ATG GCT TTT GCA TTA CCA TCA-3 and reverse primer 5-CCT TCA CAA TTA TTC GCC CA-3. Data were analyzed using DNA Baser, BLAST, and ClustalX. This study has successfully isolated 404 bp fragments of AT3 gene. This fragments located at 1918-1434 bp referred to AT3 gene from Capsicum frutescens cv. Shuanla. Isolation of upstream and downstream fragments of AT3 gene from Capsicum frutescens cv. Cakra Hijau is undergoing.

Abstract

Poeciliidae family consists of small live-bearer fishes with sexual dimorphic. This fishes are widely distributed across Indonesian archipelago, but biological studies on this fish are very limited. This research is aimed to identify the species of Peciliidae fishes which lives in Sari Lake Pasuruan based on gonopodium structures and morphometric analysis. A specific characteristic of the Poeciliidae family is its gonopodium structures which is a modification of 3rd, 4th, and 5th rays of the males anal fin. Each species represented 50 samples male and female from each type we found and measured 26 characters (in mm). Cladogram based on morphometric measurements showed that sample 1 and 2 had 100% similarity, sample 3 had 94% similarity with sample 1 and 2. Sample 4 had 54% similarity with sample 1, 2, and 3. Based on gonopodium structures sample 1 and sample 2 included Xiphophorus hellerii Heckel possess large claw on the terminus of ray 5 hook is small crescent-moon shaped on 3rd ray and lacking of gonopodium palp, sample 3 possesses distal platform with 11 retrosse serrae,and membranous hook so we were able to identify the sample 3 as Poecilia mexicana Steindachner, whilst sample 4 was Poecilia reticulata Peter had a long-pointed comb-like spines on ray 3rd and and has a small hook on the tip of 5p ray.

Abstract

Frog is an amphibian which widely distributed around the world. Indonesia houses 450 species which represent 11% of frog species in the world. In Java Island living 42 species of frogs and toads. Frogs can be used as an environment indicator due to the presence of frog in a particular place indicates that the place is stay natural and unpolluted. State University of Malang Campus #1 which is located in the heart of Malang District has been developing rapidly, currently. Thus, it requires for the construction of new various facilities to support it widen activities. Extensive construction can be destructive even damaging to the habitat of frog, which potentially threat the frog life, if it does not take a consideration on the environmental impact, carefully. This study aimed to identify the species of frog which is survives in State University of Malang with consideration of the frog species found in 1995. Species identification conducted by observing the morphologicalcharacter. This study found four species with three species which are survive and as found in 1995; those are Duttaphrynus melanostictus, Polypedates leucomystax, and Kaloula baleta; and one new species that is Rana chalconota. This study also reveals there are four species which are extinct; those are Fejervarya cancrivora, Fejervarya limnocharis, Ingerophrynus biporcatus, and Occidoziga lima. This situation shows the decreasing amount of species from 7 to 4 within 17 years. This result indicates that there is a serious environmental degradation which caused the losing of frog habitats. Further research is needed to study the ecological condition changing in order to save the frog species.

Abstract

Tree frogs live in natural places which are unpolluted. Regarding their role as an ecological indicator, the decrease of frogs population in a particular habitat indicates the danger of environment quality decrease. Moreover, this condition can harm the frogs themselves. All kinds of frogs breed in aqueous environment such as ponds, marshes, and farming fields. One of the tree frogs, Polypedates leucomystax, which belongs to Familia Rachophoridae, is widely spread in Indonesia. This frog has yellowish brown skin with black spots or six lines extending from head to the posterior tip of body. A breeding couple of the frog produces foam nests on the water or plants around water body, where they will nest their fertilized eggs. This species produces over a hundred embryos in one spawning season. These embryos require appropriate conditions to develop normally in the nature. Frog embryo development may becomes a reference to understand how the frog population survives. This study focused on P. leucomystax with regards to its decrease in number due to the drying up of the environment and a lot lost of trees in Campus of State University of Malang. The development of P. leucomystax embryos in the reproduction foam was observed until it reached a tadpole stage. The result showed that the embryos developed in the foam until they hatched then they move out of the foam into the water under which they would continue their development. Considering that water body is a critical requirement for the development of P. leucomystax embryos, it is our responsibility to make any efforts to conserve not only the trees but also any type of water bodies including ponds, marshes, and farming fields as well.
