Abstract

Frog is an amphibian which widely distributed around the world. Indonesia houses 450 species which represent 11% of frog species in the world. In Java Island living 42 species of frogs and toads. Frogs can be used as an environment indicator due to the presence of frog in a particular place indicates that the place is stay natural and unpolluted. State University of Malang Campus #1 which is located in the heart of Malang District has been developing rapidly, currently. Thus, it requires for the construction of new various facilities to support it widen activities. Extensive construction can be destructive even damaging to the habitat of frog, which potentially threat the frog life, if it does not take a consideration on the environmental impact, carefully. This study aimed to identify the species of frog which is survives in State University of Malang with consideration of the frog species found in 1995. Species identification conducted by observing the morphologicalcharacter. This study found four species with three species which are survive and as found in 1995; those are Duttaphrynus melanostictus, Polypedates leucomystax, and Kaloula baleta; and one new species that is Rana chalconota. This study also reveals there are four species which are extinct; those are Fejervarya cancrivora, Fejervarya limnocharis, Ingerophrynus biporcatus, and Occidoziga lima. This situation shows the decreasing amount of species from 7 to 4 within 17 years. This result indicates that there is a serious environmental degradation which caused the losing of frog habitats. Further research is needed to study the ecological condition changing in order to save the frog species.

Abstract

Chili pepper is widely used and cultivated by Indonesian people. There are three species of chili pepper, i.e.: Capsicum annuum L., Capsicum frutescens L., and Capsicum violaceum HBK. Capsicum frutescens L. has a higher economic value due to its pungency and carotenoid content. C. frutescens has several cultivars, one of those is Capsicum frutescens cv. Cakra Hijau. This cultivar is resistant against pest and disease and has very high pungency. This special character of chili pepper is born by its secondary metabolic, Capsaicin. Moreover, capsaicin also serves as defense mechanism, antiarthritis, analgesic, and anticancer. This study aimed to isolate Acyltransferase (AT3) gene which encoding Capsaicin Synthase (CS) enzyme. AT3 gene was isolated through PCR using forward primer 5-ATG GCT TTT GCA TTA CCA TCA-3 and reverse primer 5-CCT TCA CAA TTA TTC GCC CA-3. Data were analyzed using DNA Baser, BLAST, and ClustalX. This study has successfully isolated 404 bp fragments of AT3 gene. This fragments located at 1918-1434 bp referred to AT3 gene from Capsicum frutescens cv. Shuanla. Isolation of upstream and downstream fragments of AT3 gene from Capsicum frutescens cv. Cakra Hijau is undergoing.

Abstract

Poeciliidae family consists of small live-bearer fishes with sexual dimorphic. This fishes are widely distributed across Indonesian archipelago, but biological studies on this fish are very limited. This research is aimed to identify the species of Peciliidae fishes which lives in Sari Lake Pasuruan based on gonopodium structures and morphometric analysis. A specific characteristic of the Poeciliidae family is its gonopodium structures which is a modification of 3rd, 4th, and 5th rays of the males anal fin. Each species represented 50 samples male and female from each type we found and measured 26 characters (in mm). Cladogram based on morphometric measurements showed that sample 1 and 2 had 100% similarity, sample 3 had 94% similarity with sample 1 and 2. Sample 4 had 54% similarity with sample 1, 2, and 3. Based on gonopodium structures sample 1 and sample 2 included Xiphophorus hellerii Heckel possess large claw on the terminus of ray 5 hook is small crescent-moon shaped on 3rd ray and lacking of gonopodium palp, sample 3 possesses distal platform with 11 retrosse serrae,and membranous hook so we were able to identify the sample 3 as Poecilia mexicana Steindachner, whilst sample 4 was Poecilia reticulata Peter had a long-pointed comb-like spines on ray 3rd and and has a small hook on the tip of 5p ray.

Abstract

Hylarana chalconota is a cryptic species of frog endemic to Java Island . This species is small with long legs, and brown skin. The Snout-Vent Length (SVL) ranges between 30-40 mm for male and 45-65 mm for female. Reports the existence of this species in State University of Malang, which was not found in 1995. To reveal the haplotype diversity of COI gene in this species, we analyzed Cytochrome-c oxidase subunit-1 (COI) sequences of both samples. The both samples had 604 bp and 574 bp fragment length, respectively. These fragments showed polymorphism; with mutation position in sites 104, 105, and 124. Based on this result, we suggest that the two samples share a different haplotypes, proposed as UM1 and UM2

Abstract

The Incidence of Indonesia breast cancer case in 2018 was reported at 20.7% or 160,653 in number. The factors that caused breast cancer is TP53 gene mutation and Single Nucleotide Polymorphism (SNP). This study aimed to determine the mutation and Single Nucleotide Polymorphism (SNP) of TP53 gene in breast cancer. Samples of this study were 9 people chosen based on a purposive technique. The methods include total DNA isolation, DNA quantification, PCR, and sequencing. The results of the sequencing were then analyzed using alignment and blast. The SNP is browsed by the SNP finder on NCBI both followed by protein modeling. The results of this study indicate the existence of mutation in the exon and intron regions. Substitution of Guanine (G) to base Adenine (A) is occur in codon 496 which is a coding region. SNP was also obtained in exon of sample 12. The conclusion of this study is breast cancer can be caused by mutations that occur in the exon and altered the structur of protein structure, so then affect the binding affinity between p53 and its target domain. Tp53 gene mutation is not the only cause of breast cancer staging development, there are other genes that also contribute to the development of cancer stage. In addition, Single Nucleotide Polymorphism in the TP53 gene can be used as a predisposition marker of breast cancer that has potential to be inherited

Abstract

Untuk membina karier kepangkatan dan profesionalisme, guru harus membuat artikel yang dimuat di jurnal ilmiah. Berdasarkan hasil wawancara, kesulitan guru SMPN 15 Malang dalam menulis artikel ilmiah adalah motivasi, waktu, teknik penulisan, kurang melek teknologi, hingga sumber referensi. Oleh karena itu, guru perlu didampingi dalam menulis artikel ilmiah di jurnal terindeks. Agar lebih mudah, kegiatan penulisan dilakukan dengan aplikasi reference manager, Zotero. Metode pelaksanaan kegiatan pendampingan penulisan artikel ilmiah berbasis manajer referensi dilaksanakan dengan tahapan berikut: (a) persiapan strategi kegiatan observasi di SMPN 15 Malang, (b) pendampingan penulisan artikel ilmiah berbasis manajer referensi untuk guru SMPN 15 Malang, dan (c) refleksi kegiatan dengan  rincian sebagai berikut. Kegiatan dilakukan melalui sinergi lima dosen dengan dua mahasiswa untuk 49 guru SMPN 15 Malang. Hasil kegiatan yakni berupa draf pendampingan penulisan artikel jurnal terindeks dengan aplikasi Zotero yang sengajar dipilih berdasarkan kebutuhan guru dan kesederhanaan fitur aplikasi. Kegiatan ini rencananya akan ditindaklanjuti secara luring dan daring untuk menyempurnakan draf artikel yang selanjutnya akan dikirim ke jurnal terindeks yang berbasis Online Journal System (OJS).

Abstract

The scientific literacy framework in PISA 2015 includes context, competence, attitudes and knowledge. Knowledge is closely related to students' cognitive abilities. Analysis of needs in the class of 2020 Biology at the State University of Malang shows that 93.8% of students' knowledge about amylolytic and cellulolytic bacteria in vegetable waste disposal soil is relatively low. One factor is that there are no teaching materials related to this material. The research aims to determine the effectiveness and practicality of handouts in improving students' cognitive abilities. This research method is quasi-experimental with a nonrandomized pretest-posttest control group design—implementation in the control and ex-perimental classes by giving a pretest and posttest. The increase in posttest scores compared to the learning outcome scores for both classes was analyzed using the T-test to compare learning outcomes based on the N-gain score. The results of the unpaired t-test analysis stated that the cognitive abilities of students in the experimental class using handout were higher than those in the control class using PowerPoint—comparison value p-value (0.046) < α (0.05). The conclusion is that the handouts are practical and effective in improving students' cognitive abilities. Students can find out the existence of bacteria that play a role in the degradation process of starch and cellulose so that students can protect the environment and not throw away rubbish, which causes an imbalance in the soil ecosystem.Abstrak Kerangka kerja literasi sains dalam PISA 2015 meliputi konteks, kompetensi, sikap, dan pengetahuan. Pengetahuan erat hubungannya dengan kemampuan kogintif mahasiswa. Analisis kebutuhan di biologi angkatan 2020 Universitas Negeri Malang menunjukkan pengetahuan 93,8% mahasiswa tentang bakteri amilolitik dan bakteri selulolitik pada tanah pembuangan sampah sayur tergolong rendah. Salah satu faktornya adalah tidak ada bahan ajar terkait materi tersebut. Tujuan dari penelitian untuk mengetahui keefektifan dan kepraktisan handout dalam meningkatkan kemampuan kognitif mahasiswa. Metode penelitian ini kuasi eksperimen dengan desain nonrandomized pretest-posttest control group. Implementasi pada kelas kontrol dan kelas eksperimenn dengan diberikan pretest dan posttest. Peningkatan nilai posttest dibandingkan untuk nilai hasil belajar kedua kelas dan dianalisis dengan uji T untuk membandingkan hasil belajar berdasarkan N-gain skor. Hasil analisis uji-t tak berpasangan menyatakan kemampuan kognitif mahasiswa kelas eksperimen menggunakan handout lebih tinggi dibandingkan kelas kontrol menggunakan Power Point. Nilai perbandingan p-value (0,046) < α (0,05). Kesimpulannya adalah handout praktis dan efektif meningkatkan kemampuan kognitif mahasiswa. Mahasiswa dapat mengetahui keberadaan bakteri yang berperan dalam proses degradasi amilum dan selulosa, sehingga mahasiswa dapat menjaga lingkungan, tidak membuang sampah yang menyebabkan ketidakseimbangan ekosistem tanah.

Abstract

Penelitian ini bertujuan untuk mengetahui manifestasi ideologi sosial dalam cerita pendek majalah Horison tahun 1998─1988 melalui tindakan rasional instrumental, tindakan rasionalitas nilai, tindakan afektif, dan tindakan tradisional. Jenis penelitian yang dilakukan adalah deskriptif kualitatif. Data penelitian ini berupa unit teks yang berwujud kalimat dan paragraf cerita dalam cerita pendek. Sumber data dalam penelitian ini adalah cerita pendek majalah Horison tahun 1988─1998. Pengumpulan data dalam penelitian ini dilakukan dengan menyusun panduan dokumentasi studi. Teknik analisis data yang digunakan adalah reduksi data, penyajian data, serta menarik kesimpulan. Manifestasi ideologi sosial yang ditemukan dalam cerita pendek majalah Horison tahun 1988-1998 terbagi menjadi empat, meliputi perwujudan ideologi sosial melalui (1) tindakan rasional instrumental dalam kehidupan bermasyarakat, kehidupan keluarga, dan kehidupan beragama. (2) tindakan rasional nilai yang ditunjukkan dalam bekerja, mengambil keputusan, dan pendidikan. (3) tindakan afektif yang ditunjukkan melalui hubungan seksual, rumah tangga, dan lingkungan masyarakat. (4) tindakan tradisional yang ditunjukkan terhadap anak, orang tua, dan masyarakat.

Abstract

Etika lingkungan hidup dapat diartikan sebagai disiplin ilmu yang membahas nilai, norma, dan prinsip moral yang mengatur tingkah laku manusia terkait dengan lingkungan, serta nilai dan prinsip moral yang mendasari sikap manusia dalam interaksinya dengan lingkungan. Etika lingkungan hidup memperhitungkan keberadaan moral semua makhluk, tidak hanya manusia, dalam lingkup perhatian moral manusia. Novel Kekal karya Jalu Kancana menceritakan kegiatan eksploitasi yang dilakukan oleh perusahaan tambang panas bumi di Kawasan Cagar Alam Kamojang. Penelitian ini membahas paradigma etika lingkungan. Penelitian ini bertujuan untuk mendeskripsikan paradigma etika lingkungan aposentrisme, ekosentrisme,dan bosentrisme. Metode analisis data yang digunakan dalam penelitian ini adalah analisis isi. Sumber data penelitian ini ialah novel dengan judul Kekal karya Jalu Kancana yang diterbitkan oleh Buku Mojok di Sleman Yogyakarta tahun 2019. Hasil analisis menunjukkan sudut pandang antroposentrisme yang memandang manusia sebagai pusat peradaban, sudut pandang ekosentrisme ditandai dengan hubungan antara sesama makhluk hidup dan ekologi yang mana ekologi lebih penting daripada ekonomi, dan sudut pandang biosentrisme dapat dilihat dari hubungan makhluk hidup dengan makhluk hidup lain yang memiliki hak untuk hidup dan diperlakukan setara.

Abstract

The Rambut Monte lake in Krisik Village, Blitar, is a historical and cultural heritage tourism destination that is continuously preserved and promoted. The symbolic potential behind the story of the Rambut Monte lake is what attracts tourists to visit it. The artifacts in Rambut Monte as a relic of the Majapahit kingdom have become historical tours that need to be explored for their attractiveness. Apart from that, the myth of the Rambut Monte lake is a community belief that is continuously maintained to protect the cosmos. The promotion of Rambut Monte tourism using a cultural mechanism of production, distribution, and consumption can make the Rambut Monte lake known by the wider community.
