Abstract

Antibiotic Growth Promoters (AGP/AGPs) have been used to improve feed efficiency in poultry, but are banned in Indonesia due to food safety concerns. This study evaluated the potential of probiotics as an AGP alternative in ISA Brown male layer chickens during the finishing period, focusing on blood glucose and cholesterol levels. A total of 180 chickens were reared for 42 days. Treatment groups received basal feed supplemented with probiotics at 1–5 mL/kg, equivalent to 1.2×10⁹ to 6×10⁹ CFU/kg feed, containing Lactobacillus acidophilus, Lactobacillus plantarum, and Bifidobacterium spp. Blood cholesterol was analysed using an enzymatic colorimetric method at 500 nm, while glucose was measured using the glucose oxidase method at 505 nm. Results showed that probiotic treatments (P2, P3, P5) significantly reduced glucose levels compared to the control (P0) and AGP group (P1). A moderate decrease in cholesterol levels was also observed in probiotic groups. These effects may be linked to improved gut microbial balance and nutrient metabolism. The probiotic combination used in this study shows potential as a safer and more sustainable alternative to AGPs. Further studies are recommended to optimize dosage and assess long-term impacts on poultry health and productivity.

Abstract

Noninvasive genetic sampling is the most reliable approach to collect endangered species which are typically rare and elusive. However, the approach is hampered by poor quality and quantity of DNA amount, and therefore, a pilot study is required. Our current short communication article reports the first noninvasive genetic sampling approach using feces of critically endangered Bawean deer (Axis kuhlii) to test the effect of aging on the amount of DNA extraction. Fecal samples of the Bawean deer were collected from the Bawean Deer Captive Unit in Sangkapura Village, Bawean Island, East Java. The samples were grouped into two different categories, namely: 1) New samples: for feces that had just been expelled for less than a day and 2) Old samples: for feces that were estimated to have been expelled for more than a day. There were 11 New samples and 16 Old samples. The samples were extracted using the QIAGEN Mini Stool Kit. The DNA quantification was carried out by using a fluorometer. The results of the extraction between the two categories were analyzed by the Kruskal Wallis test. The results showed that the mean rank value of the New samples was better (15.27) than the Old samples (13.13). The P value of the Kruskal Wallis test, however, was higher than the Asymp significant value, indicating that there was no meaningful differences (P = 0.49) between the two samples categories. The work described in this short communication article is a preliminary result and an important step in the study of Bawean deer population genetic.

Abstract

Laser cutting is a sheet metal cutting technology using laser media combined with a CNC system. The problems of the laser cutting process are defects in the surface of the cutting product caused by a mismatch of parameter variations. The parameters of laser cutting in this research are cutting speed, focusing distance, and gas pressure. Variations in the cutting speed parameters studied were 1440 m/min, 1620 m/min, and 1800 m/min. The variations in the focusing distance studied were 5.5 mm, 6.0 mm, and 6.5 mm. Variations in the gas pressure parameters used are 0.5 bar, 0.7 bar, and 0.9 bar. The Taguchi method is used to obtain variations with minimum hardness and surface roughness values of the material. Surface roughness was tested using a surface roughness tester; surface hardness was tested using a brine hardness tester. The magnitude of the influence of each parameter is analyzed using the ANOVA method. The results showed that the focusing distance parameter had the most significant effect on surface roughness, with a percentage of 50.385%, and the gas pressure parameter was the most influential parameter on surface hardness, with a percentage of 54.241%. The minimum roughness value can be achieved with a cutting speed of 1800 m/min, focusing of 5.5 mm, and gas pressure of 0.5 bar. The minimum hardness value is achieved with a cutting speed of 1800 m/min, a focusing of 6.5 mm, and a gas pressure of 0.5 bar.

Abstract
