Abstract

This study aims to evaluate the expression of BAX, BCL-2, and BAX/BCL-2 ratio in maturation media of cow oocytes which supplemented with Urea in vitro because BAX and BCL-2 are the main regulators of apoptosis.A total of 263 oocytes from follicle aspirations originating from ovaries taken from slaughterhouses and were saturated with 3 addition of urea which was divided into three groups. The control group (P0) was control group without the addition of urea, P1 group was added with urea 20 mg/dL, while P2 group was added with urea 40 mg/dL.The results of in vitro oocyte maturation were continued with identification using immunocytochemical staining with the addition of BAX and BCL-2 antibodies.Positive results showed a brownish color on the oocyte and its cumulus. The results of this study indicated that there were significant differences (P0.05) in BAX and BCL-2 expression, although both curves were equally increase. The increase in BCL-2 was more significant than BAX, while the BAX BCL-2 ratio did not show a significant difference (P0.05) in whichthe curve of BAX/BCL-2 ratio was decreased. It can be concluded that the addition of urea does not affect the level of apoptosis.

Abstract

This study aims to explores blood urea nitrogen (BUN) levels in Wistar rats exposed to synthetic and organic MSG to identify potential renoprotective effects. The study design involved a control group (K) that was fed a normal diet only, and six treatment groups (P1–P6) that received doses of organic or synthetic MSG at 60, 120, or 240 mg/kg BW for 19 days. Each group consisted of six rats. MSG was administered orally and BUN levels were measured using the colorimetric method. The results showed no significant difference in BUN levels between the control group and the treatment groups (p > 0.05). The highest BUN levels were found in the control group, while the lowest levels were found in the group that received the highest dose of synthetic MSG (240 mg/kg BW). Additionally, BUN levels were lower with organic MSG at 120 and 240 mg/kg BW than with the 60 mg/kg BW dose, suggesting that bioactive compounds in tempeh and pineapple bromelain may have renoprotective effects. While the lowest BUN levels were observed in the group receiving the highest dose of synthetic MSG (240 mg/kg BW), this may be due to reduced protein intake or other metabolic adaptations rather than safety factors. Synthetic MSG at a dose of 240 mg/kg BW produced the lowest BUN levels, which may indicate reduced protein intake or other metabolic adaptations, rather than safety factors.

Abstract

Infertility is a problem experienced by some women and men around the world. Most infertility problems in women is caused by impaired reproductive organs or disrupted ovulation.. One factor that causes impaired oocyte maturation is wireless fidelity (Wi-Fi) radiation which has a radio frequency field of 2.45 GHz. The radiation may increase the activity of free radical cells through the fenton reaction pathways that cause infertility because of the disrupted oocyte development. This study aimed to determine the effect of Wi-Fi radiation on the number of oocytes. This was an experimental study using control group design. The subjects were 32 female mice selected through complete random sampling and divided into two groups: control (R0) and exposure (R1). Each group consisted of 16 mice. The Wi-Fi radio frequency used was 2.5 GHz. Mice (R1) were placed closer to Wi-Fi source (± 15cm), and there were two types of laptop PCs and 3G mobile phones connected to internet placed next to mice for 15 hours/28 days. The mice underwent a simultaneous cycle with intraperitoneal injection of PMSG and HCG. Furthermore, the mice were mated with vasectomized male monomatingly to induce ovulation. The fertilization pouch in both oviducts were observed for oocyte collection. The number of oocytes was calculated using an inverted microscope. There was a difference in the number of oocytes between control and exposure group. Statistical tests were analyzed using Mann Whitney U and resulted in significant values (p value = 0.00). No oocytes count in exposure group. In other words, the group underwent anovulation. In conclusion, Wi-Fi radiation affected the number of oocyte stimulation in mice. Therefore, it was important to minimize the risk factors that trigger electromagnetic radiation on reproductive health.

Abstract

BACKGROUND: Endometriosis impairs female reproductive function through oxidative stress and apoptosis, reducing oocyte quality and causing infertility. Current therapies are limited by suboptimal efficacy and side effects, including ovulation suppression. Curcumin offers antioxidant and anti-inflammatory benefits but has low bioavailability and poor solubility, which can be improved through nanoparticle formulation. Although nanocurcumin is suggested to act through multiple pathways, its mechanisms remain unclear. This study was conducted to evaluate the antioxidant and anti-apoptotic effects of nanocurcumin in a mouse model of endometriosis.METHODS: Thirty-five mice were allocated into five groups and induced to develop endometriosis using cyclosporine A, ethinyl estradiol, and human endometrial tissue. Nanocurcumin was formulated at three particle sizes (3.71; 3.98; and 25.60 nm) and administered orally at doses of 50, 100, or 200 mg/kg/day for 14 days. After treatment, the mice were euthanized, and ovarian tissues were collected for immunohistochemical analysis of glutathione peroxidase (GPx) and B-cell lymphoma-2 (Bcl-2) expression.RESULTS: The highest GPx expression was observed in the group receiving 50 mg/kg/day nanocurcumin (mean±SD= 6.31±1.97; p=0.042). The lowest expression of Bcl-2 was observed in control group with no treatment (mean±SD=4.15±2.48; p=0.582). Nanocurcumin administration significantly increased GPx expression in a dose-dependent manner compared with the untreated group, while no significant differences were found in Bcl-2 expression.CONCLUSION: Nanocurcumin increases GPx expression, particularly at 50 mg/kg/day, indicating its potential as an antioxidant in reducing oxidative damage associated with endometriosis. However, nanocurcumin did not significantly influence Bcl-2 expression. These findings support nanocurcumin’s role as an effective antioxidant agent in protecting ovarian granulosa cells in endometriosis.KEYWORDS: nanocurcumin, GPx, Bcl-2, endometriosis, granulosa cells

Abstract

The integration of 21st-century skills into economics education is essential to prepare students for complex global challenges. This quasi-experimental study examined the effectiveness of role-playing pedagogy in enhancing critical thinking, collaboration, and communication skills among senior high school students in Jember, Indonesia. A total of 160 eleventh-grade students participated, divided into an experimental group (n = 80) receiving role-playing instruction and a control group (n = 80) taught through traditional lectures. Data were collected using validated rubrics and analyzed with SPSS 26.0, employing independent samples t-tests and ANCOVA with pretest scores as covariates. Results revealed that the experimental group significantly outperformed the control group across all measured domains, with large effect sizes (Cohen’s d > 1.0). Specifically, role-playing was found to foster deeper cognitive engagement, collaborative interactions, and effective communication within simulated economic contexts. These findings confirm the pedagogical potential of role-playing as a transformative instructional strategy that aligns with experiential and constructivist learning theories. The study contributes to the growing evidence base advocating for active learning approaches in secondary education and highlights the importance of embedding authentic, simulation-based activities into economics curricula. Implications for practice suggest that integrating role-playing can enhance both academic outcomes and transversal competencies, equipping students with critical skills necessary for future academic and professional success
