Abstract

Objective: To create an animal model of vesicovaginal fistula as a modality to develop therapy.Material and Methods: This study was an preliminary experimental study to create new fistula vesicovaginal formation in a rabbit model. Six female New Zealand white rabbits of Stem Cell Research and Development Center of Universitas Airlangga were used in this study and were divided into 3 groups in which there were 2 rabbits in each group. Vesicovaginal fistula formation was performed through laparotomy surgery. After the abdomen is opened, the Nasogastric tube is inserted vaginally into the anterior vaginal wall. The incision is made on the anterior vaginal wall and the posterior wall of the bladder. The Nasogastric tube that comes out of the vagina is inserted into the bladder incision to maintain the formation of vesicovaginal fistula. Both mucosal walls are sutured and tested to determine if there is any leak. Evaluation the model of vesicovaginal fistula was carried out 3 weeks postoperatively according the group.Result: Four out of six rabbits have formed vesicovaginal fistulas, which are from groups two and three. There are no complications during the treatment period until terminationConclusion: Rabbits have the potential to become experimental animals for the vesicovaginal fistula model. The best time to release a nasogastric tube to maintain a fistula is 2 weeks.

Abstract

HIGHLIGHTS 1. Endometriosis treatment remains controversial, whether it should be managed with hormonal modulation and/or surgical removal of the lesion.2. NF-kB becomes potential therapeutic target as it has constitutive activation in peritoneal endometriosis.3. It has been recognized that NF-kB expression decreases when red fruit extract is given.4. Effect of red fruit extract, compared to leuprolide, a widely used treatment for endometriosis, on NF-kB expression was examined.5. NF-kB was significantly lower in endometriotic group receiving red fruit extract than in group receiving leuprolide.   ABSTRACT Objectives: This study aimed to prove the effect of different administration of red fruit (Pandanus conoideus Lam) extract and leuprolide on the expression of NF-kB and endometrial implant width in mouse models of endometriosis. Materials and Methods: This study used completely randomized design with pre-post separated sample. The sample size was 36 female mice (Mus muculus), which were divided into 3 groups randomly. They were given with cyclosporine A, estrogen and endometrial tissue to form endometriosis model. Results: The mean expression of NF-kBin in treatment group receiving leuprolide (4.13 + 0.70) and red fruit (2.70 + 1.08) were significantly lower than that in control group (6.94 + 1.007). Whereas, in treatment group receiving red fruit (2.70 + 1.08), it was significantly lower than in those receiving leuprolide (4.13 + 0.70). Mean endometrial implant width in treatment group receiving leuprolide (13.62 + 3.21) and red fruit (8.93 + 2.74) were significantly lower than that in control group (28.89 + 8.28). Whereas, in treatment group receiving leuprolide (13.62 + 3.21), it was not significantly higher than in those receiving red fruit (8.93 + 2.74). Conclusion: The expression of NF-kB was significantly lower after the administration of red fruit than leuprolide. The width of endometrial implant was not significantly lower after the administration of red fruit than after the administration of leuprolide.

Abstract

Background: Increased oxidative stress causes inflammation and increases angiogenesis. It presumed to promote the proliferation of endometriosis tissue. Kebar grass (Biophytum petersianum) and other herbs such as green tea and Cucumis melo, which contain high antioxidants, are expected to decrease oxidative stress, inflammation, angiogenesis, and reduced endometriosis implants. Objective: To investigate the effects of Kebar grass, green tea, and Cucumis melo to malondialdehyde serum, tumor necrosis factor alpha, and vascular endothelial growth factor expression, and the area of the endometriotic implants. Methods: Twenty-eight mice were divided into four groups, i.e., the first group of endometriosis mice was given Kebar grass extract; the second group was assigned green tea extract, the third group was given the combination of Cucumis melo extract–gliadin, and the last containing the untreated endometriosis mice as the control. Each treatment was given for 14 days. The data of MDA serum level, the area of the endometriotic implants, TNF-α, and VEGF expression were collected and analyzed. Results: The MDA serum levels of the groups treated with Kebar grass extract, green tea extract, and Cucumis melo extract – gliadin were significantly lower (p=0.001) than the control group. TNF-α expression of the groups provided with each treatment also lower than the control groups (p=0.002). However, only the administration of the Cucumis melo extract–gliadin resulted in lower VEGF expression compare with the control (p=0.017). Finally, the area of the endometriotic implants of the mice models administered with each treatment was smaller than the control group (p=0.003).Conclusion: Kebar grass as well as green tea and Cucumis melo–gliadin inhibits endometriotic implants extension by decreasing MDA serum and TNF-α expression.

Abstract

Background: Endometriosis is a gynaecological disorder characterised by the presence of endometrial tissue outside the uterine cavity. Apoptosis, regulated by the balance of Bcl-2/Bax, plays an important role in the endometrial improvement. Metformin, an insulin sensitizer that is known to have beneficial effect in the endometriosis treatment, is expected to lower Bcl-2/Bax expression ratio and reduce endometrial implants. Objective: To explore the effect of metformin on the Bcl-2/Bax expression ratio and endometrial implant area of endometriosis-induced mice. Methods: This experimental study used 3-month old 33 BALB/c mice of endometriosis that were randomly and equally divided into three groups (P0, P1, and P2). On the 15th day, the P0 group was first terminated for Bcl-2/Bax examination and the size of endometrial implants. The P1 group was given aquabidest, whereas the P2 group was given metformin 4 mg/day for 14 days. The immunohistochemistry of Bcl-2/Bax expression were performed from cavum abdomen and pelvis peritoneal tissues of the mice and measured by the Remmele Scale Index, whereas the extracted mice’ endometrial implants were analysed with computer tracing method. All data normality test was calculated with Shapiro-Wilk test. The mean difference test of all groups was analysed using the one-way ANOVA test, whereas the mean difference test between groups was completed using the Unpaired T-test (LSD/Least Significance Difference). Results: The Bcl-2/Bax expression ratio and endometrial implant area in the P2 group were significantly lower compared to P0 and P1 (p<0.001). There were no significant differences in the Bcl-2/Bax expression ratio or endometrial implant area in P0 and P1 (p>0.05)Conclusion: Metformin may be a potential effective drug treatment for endometriosis by decreasing Bcl-2/Bax expression ratio and endometrial implants.

Abstract

BACKGROUNDIndonesia has the third largest tobacco consumption in the world after China and India. Nicotine as the main component of cigarette smoke has negative effects on the reproductive system, such as oocyte maturation, ovulation, and fertilization, and increasing the diploidy of oocytes. The goal of this researchwas to evaluate the effect of nicotine on oocyte maturation in Rattus norvegicus.METHODSThis was an experimental study with post test only control group design. The subjects were 40 rats selected homogenously and randomly. They were divided into a control group (receiving carboxy-methyl-cellulose sodium and 3 treatment groups (I-III) receiving nicotine subcutaneously for 7 days at dosages of 21 mg/ kgBW, 41 kg/kgBW and 84/kgBW, respectively. The observations comprised oocyte maturation stage, viz. germinal vesicle (GV), germinal vesicle breakdown (GVBD), metaphase I and metaphase II. Data were analyzed by one-way Anova with á=0.05, followed by Tukey’s HSD test.RESULTSOne-way Anova showed significant differences in oocyte  maturation in all groups. Tukey’s HSD test showed that for GV, the differing groups were control and I, control and II, I and III. For GVBD, the differing groups were control and I, I andII, I and III. For metaphase I, the differing groups were control with I, II, and III, I and II, I and III. For metaphase II, the differing groups were control versus I, II, and III, I and II, I and III.CONCLUSIONLow dose of nicotine is capable of affecting oocyte maturation in Rattus norvegicus.
