Abstract

Introduction: To investigate Brain-Derived Neurotrophic Factor levels, Mammalian Target of Rapamycin Complex 1 levels, and Dendritic Density in the cerebrum and cerebellum of Rattus norvegicus offspring following prenatal Mozart compositions exposure. Material and Methods: An experimental study with a randomized post-test-only control group design was conducted at the Animal Laboratory of the Faculty of Veterinary Medicine, Universitas Airlangga, following ethical clearance. A 13 healthy pregnant female Rattus norvegicus were randomly chosen and exposed to 14 Mozart compositions, beginning on gestation day 10 until delivery, while another 13 served as the control group. At day 20, the rats were sacrificed, 2 offsprings' brains (the heaviest) were extracted, and BDNF levels were analyzed using the ab203573 immunohistochemistry kit, mTORC1 levels using the pSer2448 kit, and Dendritic Density using silver impregnation. Statistical analysis was performed accordingly with a significance level of p < 0.05. Results: BDNF levels and Dendritic density in the cerebrum of the treatment group were higher (2.27 ± 1.41 vs. tr 7.44 ± 2.09, p < 0.05) and (4.42 ± 2.04 vs. 6.67 ± 1.23, p < 0.05). In the cerebellum, BDNF levels and Dendritic density in the treatment group were higher (1 vs. 8, p < 0.05) and (3.34 ± 1.94 and 7.91 ± 2.45, p < 0.05). mTORC1 levels in the treatment group were lower but not statistically significant in both areas. Conclusion: Prenatal 14 Mozart compositions exposure during pregnancy increased Rattus norvegicus offsprings' cerebrum and cerebellum BDNF levels and Dendritic Density. A correlation was found between BDNF levels and dendritic density.

Abstract

This research aimed to assess the efficacy of ethylene glycol-sucrose as an alternative cryoprotectant compared to commercial cryoprotectants by focusing on the expression of insulin-like growth factor-1 (IGF-1) and bone morphogenetic protein-15 (BMP-15) in goat oocyte cumulus complexes after warming with identification using the immunocytochemical analysis. Maturated oocytes were divided into 3 groups, i.e., (C) the control group followed by an immunocytochemical examination, and two treatment groups, i.e., (P1) was exposed to a commercial cryoprotectant for 15 minutes, then placed in a hemistraw and dipped in liquid nitrogen, and (P2) was exposed to 30% ethylene glycol and 1 M sucrose for 15 minutes, then placed in a hemistraw and dipped in liquid nitrogen. The results revealed divergent responses in IGF-1 (CG = 9.00 ± 3.00; P1 = 9.50 ± 3.20; P2 = 4.67 ± 0.94) and BMP-15 (CG = 10.50 ± 3.35; P1 = 9.50 ± 2.69; P2 = 5.50 ± 3.64) expression patterns, possibly influenced by the two cryoprotectant abilities and oocytes performing their permeabilities to cryoprotectant solution. We concluded that each cryoprotectant is necessary to mitigate the adverse impact on gene expression in oocytes so that we can use it for reproductive technologies.

Abstract

The aim of this study is to analyze the IGF-1 levels decrease in mice modeled by Polycystic Ovary Syndrome Insulin Resistance post carbohydrate high in protein. True Experimental Design with a Posttest Control Group Design research using 36 tails, female Wistar Rattus Norvegicus Strains, 3 months, 100-200 grams and randomly divided into 3 groups, namely K-(n =12, negative control group), K+(n=12, positive control group) and KP(n=12, treatment group). SOPK-IR modeling is done by mice in the injection of the hormone testosterone provionate at a dose of 100 mg/kg BW for 28 days, on the last day after injection a vaginal swab is performed to identify the condition of diestrus. The measurement of IGF-1 levels used the Enzyme-Linked Immunosorbent Assay method. Data analysis techniques used ANOVA test and LSD post hoc test. The results obtained mean levels of IGF-1 at K- (234.126±32.568) ng/mL, K+ (169.662±30.150) ng/mL, KP (146.528±29.562) ng/mL (p=0.000)

Abstract

This study is a true experiment with a posttest-only control group design. 30 pregnant Rattus norvegicus aged 2-3 months were randomly divided into 3 groups, namely the control group (K), the mackerel oil treatment group (P1), and the omega-3 supplements group (P2). The experimental animals were acclimatized and superovulated. The treatment was given at day 1-17 of pregnancy. 3 newborn Rattus norvegicus with the criteria of the largest, medium, and lightest body weight were taken from each parent to be made preparations on the 18th day. Examination of preparations carried out by immunohistochemistry. The mean ± SD of the apoptosis index in the cerebrum were 4.93 ± 0.87 (K), 2.53 ± 0.45 (P1), and 3.72 ± 0.54 (P2). The results of statistical tests showed that there was a significant effect on the apoptotic index in the cerebrum of newborn Rattus norvegicus between groups with p-value <0.05.

Abstract

Latar Belakang : Pada stres prenatal akan memicu peningkatan kadar CRH dan kortisol maternal. ini akan meningkatkan jumlah CRH dan kortisol pada janin dan menurunkan ekspresi 11β-HSD2 di plasenta. Meningkatnya kadar CRH akan meningkatkan aktifitas glukokortikoid yang akan menurunkan Growth Factors,juga akan mempengaruhi ekspresimTORC1 sebagai mekanisme pertahanan sel. GH, dan IGF-1 sedangkan menurunnya ekspresi 11β-HSD2 pada akhirnya akan menyebabkan perkembangan organ yang tak seimbang. Tujuan.Untuk mengetahui pengaruh frekuensi paparan stresor cahaya selama kebuntingan terhadap jumlah dendrit dan ekspresi mTORC1 anak mencit (Mus musculus). Metode : Penelitian ini merupakan penelitian eksperimen dengan post test only group design. Jumlah dendrit diukur dengan Golgy Cox hasil rerata jumlah dendrit dalam 5x lapangan pandang. Ekspresi mTORC1diukur dengan indeks skala Remelle . Analisis data menggunakan uji Post Hoc dengan tingkat kemaknaan 95% (0,05). Hasil : Hasil penelitian menunjukkan adanya perbedaan rerata jumlahdendrit yang signifikan antara kelompok kontrol (22.30) dan kelompok perlakuan 2 (7.55 ) lebih rendah. Hal yang serupa juga didapatkan padaekspresimTORC1. Terdapat perbedaan yang berarti padaekspresimTORC1. kelompok kontrol (1.14) dan kelompok perlakuan 2 (4.28) lebih tinggi. Selain itu, didapatkan juga hubungan (p < 0,05) antara jumlah sel dendrit danekspresimTORC1 otak anak mencit baru lahir dengan menggunakan korelasiPearson. Hasil uji korelasi Pearson didapatkan nilai signifikansi 0,068 dengan p < 0,05 dan nilai korelasi 0,338 dengan arah korelasi negative Kesimpulan : Jumlah dendrit pada kelompok perlakuan1 dan 2 lebih rendah dibandingkan kelompok kontrol, sedangkan ekspresi mTORC1 pada kelompok perlakuan1 dan 2 lebih tinggi dibandingkan kelompok kontrol.

Abstract

The purpose of the research was to know  the effect of Insulin-Like Growth Factor-I (IGFI) derived from pregnant crossbreed mare serum (PMS) on endometrium thickness of mice (Mus musculus). The subject of this research were 35 female mice. The research was arranged by Completely Randomized Design (CRD) with seven treatment and five replications. The treatment were P0 = 10 ng/ml of physiological NaCl, P11 = 10 ng/ml of IGF-I PMS, P12 = 20 ng/ml of IGF-I PMS, P13 = 40 ng/ml of IGF-I PMS, P21 = 10 ng/ml of IGF-I recombinant mouse, P22 = 20 ng/ml of IGF-I recombinant mouse, and P23 = 40 ng/ml of IGF-I recombinant mouse. Observed variables include histopatological endometrium thickness of mice. The data were analyzed by Analysis of Variance (ANOVA), followed by HSD (Honestly Significant Difference) test. The data was also be analyzed using General Linear Model Univarieted to see the comparison between IGF-I PMS and recombinant mouse. The result showed that the addition of IGF-I PMS did not significantly affect (p>0,05) on endometrium thickness of mice . It showed that did not significantly difference (p>0,05) between the effect of IGF-I PMS and IGF-I recombinant mouse against the endometrium thickness of mice.

Abstract

This study was aimed to determine the effect of urea in maturation medium on in vitro oocyte maturation rate. The medium used was TCM-199 added with Hepes, NaHCO3, Kanamycin 0.15 IU/mL, PMSG, 0.15 IU/mL hCG, and 10% FBS. Cumulus oocyte complexes (COCs) of cows derived from follicle aspiration were divided into three groups. In control group (P0), the COCs were matured in vitro in a maturation medium without urea addition, meanwhile in the P1 and P2 groups, the medium was added with urea 20 and 40 mg/dL, respectively. Each petri dish contained three drops of maturation medium (300 µl/drops) according to the groups. Microdrops were coated with mineral oil and then incubated in a 5% CO2 incubator, at 39 ËšC with maximum humidity. Aceto-orcein staining was conducted to evaluate the maturation of oocytes based on the achievement of metaphase II phase that is indicated by the presence of metaphase plate and/or first polar body. The result showed that the oocyte maturation rates of P0, P1, and P2 were 51.25, 52.43 (p >0.05), and 46.88 % (p <0.05) respectively. It could be concluded that the presence of urea at 40 mg/dL in maturation medium reduced the percentage of bovine oocyte maturation in vitro.

Abstract

 This research aimed to determine the effect of α-tocopherol on the count of spermatogenic and Leydig cells in rats (Rattus norvegicus) exposed to 2,3,7,8 tetrachlorodibenzo-p-dioxin (dioxin). Dioxin is an endocrine-disrupting agent that adversely affects reproductive health, while α-tocopherol maintains fertility. This research used 25 rats aged 10-12 weeks weighing 150-200 grams. Rats were divided into five groups (K, P0, P1, P2 and P3). The K (control group) was administered with corn oil 1 ml/day. P0 was exposed to 700 ng/kg/day dioxin. P1, P2 and P3 was exposed to dioxin at a dose of 700 ng/kg/day and administered with α-tocopherol at a dose of 77, 140 and 259 mg/kg/day respectively. Dioxin exposure, corn oil and α-tocopherol administration were conducted orally for 20 days. On day-21, all rats were sacrificed for histological slides preparation of testicles with hematoxylin-eosin staining. Data were analyzed with analysis of variance and continued with the Duncan test. The results indicated that exposure to dioxin caused a decrease in the number of spermatogenic and Leydig cells. The administration of α-tocopherol at a dose of 140 mg/kg/day eliminated the effect of reducing the number of spermatogenic and Leydig cells caused by exposure to dioxin. The conclusion was the administration of α-tocopherol at 140 mg/kg/day was effective in maintaining the number of spermatogenic and Leydig cells in rats (Rattus norvegicus) exposed to 2,3,7,8 tetrachlorodibenzo-p-dioxin.

Abstract

This study aims to evaluate the expression of BAX, BCL-2, and BAX/BCL-2 ratio in maturation media of cow oocytes which supplemented with Urea in vitro because BAX and BCL-2 are the main regulators of apoptosis.A total of 263 oocytes from follicle aspirations originating from ovaries taken from slaughterhouses and were saturated with 3 addition of urea which was divided into three groups. The control group (P0) was control group without the addition of urea, P1 group was added with urea 20 mg/dL, while P2 group was added with urea 40 mg/dL.The results of in vitro oocyte maturation were continued with identification using immunocytochemical staining with the addition of BAX and BCL-2 antibodies.Positive results showed a brownish color on the oocyte and its cumulus.The results of this study indicated that there were significant differences (P0.05) in BAX and BCL-2 expression, although both curves were equally increase. The increase in BCL-2 was more significant than BAX, while the BAX BCL-2 ratio did not show a significant difference (P0.05) in whichthe curve of BAX/BCL-2 ratio was decreased.It can be concluded that the addition of urea does not affect the level of apoptosis.

Abstract

Tujuan penelitian ini adalah membuktikan potensi dan efektivitas rat bone marrow mesenchymal stem cell (rbmmsc) sebagai terapi pada kasus teratogenik selama kebuntingan dengan melihat ekspresi tumor necrosis factor (TNF-), Bax, dan Bcl-2. Dua puluh empat ekor tikus bunting model teratogenik dibagi menjadi empat kelompok perlakuan yang berbeda yaitu P1-Kontrol (dipapar carbon black dosis 532 mg/m3 selama 4 jam pada umur kebuntingan ke-6 s/d ke-11 + Injeksi MEM 0,1 ml), P1-Terapi (dipapar carbon black dosis 532 mg/m3 selama 4 jam pada umur kebuntingan ke-6 s/d ke-11 + rbmmsc dengan dosis 1x106 sel/0,1 ml), P2-Kontrol (dipapar carbon black dosis 532 mg/m3 selama 4 jam pada umur kebuntingan ke-6 s/d ke-17 + Injeksi MEM 0,1 ml), P2-Terapi (dipapar carbon black dosis 532 mg/m3 selama 4 jam pada umurkebuntingan ke-6 s/d ke-11 + rbmmsc dengan dosis 1x106 sel/0,1 ml). Data yang didapatkan kemudian dianalisis menggunakan uji MannWhitney. Hasil penelitian ini menunjukkan bahwa pemberian terapi rat bone marrow mesenchymal stem cell pada tikus model teratogenikparticulate matter tidak berpengaruh dalam menurunkan parameter ekspresi TNF- dan Bax, serta meningkatkan ekspresi Bcl-2 pada plasenta.
