Abstract

Tujuan dari penelitian ini adalah mengidentifikasi suatu protein dengan berat molekul 46 kDa yang diduga sebagai epidermal growth factor(EGF) yang diisolasi dari oosit sapi yang telah dimaturasi secara in vitro dengan metode elektroforesis. Ovarium sapi yang berasal dari rumah potong hewan, diaspirasi pada folikel dengan diameter permukaan 5 mm menggunakan spuit dan jarum. Oosit dimaturasi dalam tissue culture medium (TCM) 199 selama 22 jam pada suhu 38,5 C di dalam inkubator CO2. Preparasi protein dengan berat molekul 46 kDa menggunakansodium dodecyl sulphate polyacrilamide gel elektroforesis (SDS PAGE). Berdasarkan perhitungan jumlah regresi dari protein marker, didapatkan 12 fraksi protein yaitu BM 172,7; 153,09; 118,24; 102,43; 89,27; 59,75; 46,41; 43,82; 40,2; 36,06; 23,45; dan 18,42 kDa. Protein dengan berat molekul 46,41 kDa yang tampak pada pita protein dapat diidentifikasi sebagai protein yang diduga EGF yang berperan dalam proses maturasi oosit.

Abstract

Penelitian ini bertujuan mengindentifikasi GDF-9 di dalam oosit yang berasal dari preantral folikel sapi yang dimatangkan secara in vitro. Penelitian ini menggunakan metode imunositokimia ikatan antara molekul avidin dan biotin yang terkandung dalam antibodi sekunder bentuk kompleks avidin-biotin. Ovarium sapi diambil dari rumah potong hewan (RPH) Pegirian Surabaya. Penelitian ini menggunakan 105 oosit matang yang berasal dari 62 ovarium. Hasil imunositokimia menunjukkan bahwa 77,14% GDF-9 teridentifikasi.

Abstract

This study aims to explores blood urea nitrogen (BUN) levels in Wistar rats exposed to synthetic and organic MSG to identify potential renoprotective effects. The study design involved a control group (K) that was fed a normal diet only, and six treatment groups (P1–P6) that received doses of organic or synthetic MSG at 60, 120, or 240 mg/kg BW for 19 days. Each group consisted of six rats. MSG was administered orally and BUN levels were measured using the colorimetric method. The results showed no significant difference in BUN levels between the control group and the treatment groups (p > 0.05). The highest BUN levels were found in the control group, while the lowest levels were found in the group that received the highest dose of synthetic MSG (240 mg/kg BW). Additionally, BUN levels were lower with organic MSG at 120 and 240 mg/kg BW than with the 60 mg/kg BW dose, suggesting that bioactive compounds in tempeh and pineapple bromelain may have renoprotective effects. While the lowest BUN levels were observed in the group receiving the highest dose of synthetic MSG (240 mg/kg BW), this may be due to reduced protein intake or other metabolic adaptations rather than safety factors. Synthetic MSG at a dose of 240 mg/kg BW produced the lowest BUN levels, which may indicate reduced protein intake or other metabolic adaptations, rather than safety factors.

Abstract

Intelligence is one of the basic capital in advancing a country. Efforts that can be done during pregnancy is to provide adequate nutrition and stimulation, stimulation that is easily accepted by the fetus is sound and music is the most harmonious combination of sounds. Analyzing the differences in the number of glia cerebrum cells and the newly born Rattus norvegicus cerebellum from mothers exposed to Mozart music, traditional Javanese gamelan music, Sundanese gamelan, Balinese gamelan and those who were not exposed to music during pregnancy. Experimental research with randomized post-test only control group design. Used by experimental animals Rattus norvegicus as a model of music stimulation. Cutting was done with 10 IU PMSG injection, 10 IU HCG and monomating. randomized into 5 groups. The treatment was given from 10-19 days of pregnancy, for 1 hour in the soundproof room, intensity of 65 Db. Children of Rattus norvegicus were decapitated and dissected by the brain then counted the number of glia cells using Hematoksilin - Eosin and analyzed by statistics. There were significant differences in the number of newly born Rattus norvegicus glia cerebrum cells between groups. Mozart group (25.16 ± 1.72; Astrocytes), (26.16 ± 1.50; Oligodendrocytes) and (21.40 ± 4.27; Microglia) were the highest compared to the control group, Javanese gamelan music, Sundanese, and Bali. Average Number of newborn Rattus norvegicus glia cerebellum cells in the Mozart music group (24.48 ± 3.66; Astrocytes), (19.92 ± 3.77; Oligodendrocytes) and (19.92 ± 3.86; Microglia) the highest compared to control groups, Javanese gamelan music, Sundanese gamelan, and Balinese gamelan. The Mozart exposure group had more glia cerebrum and cerebellum cells than those exposed to Javanese, Sundanese, Balinese gamelan and those who were not exposed to music.

Abstract

Stimulation classical music has been shown to influence the increase in brain function and the development of child intelligence. Mozart's classical music has a frequency that can stimulate the growth of synaptic tissue. This research was analyze differences in the expression of synapsin 1 cerebrum and cerebellum Rattus norvegicus was born from mothers exposed to Mozart music, traditional Javanese gamelan music, Sundanese, Balinese and who were not exposed to music during pregnancy. This research was using methods experimental with post test only control group design. The treatment was given to Rattus norvegicus animals from pregnancy day to 10-19, for 1 hour in a soundproof room, intensity of 65 dB. On the 20th day of pregnancy, Rattus norvegicus was sacrificed and 3 of the heaviest, medium and lowest Rattus norvegicus children were selected and the heads of Rattus norvegicus children were decapitated and brain dissection and synapsin 1 expression was calculated using an immunohistochemical method and analyzed with appropriate statistics. Result There were significant differences in the expression of newborn synapsin 1 Rattus norvegicus cerebrum between groups with a value of p = 0.042 (mean in the control group IRS: 2.04 ± 0.26, in the Mozart group IRS: 3.84 ± 1.66, Javanese gamelan group IRS: 2.64 ± 0.51, Sundanese gamelan group IRS: 2.92 ± 0.41, and in the Balinese gamelan group IRS: 2.52 ± 1.18) and so in the cerebellum with a value of p = 0.001 (mean in the control group IRS: 2.02 ± 0.30, in the Mozart group IRS: 4.72 ± 0.54, the Javanese gamelan group was IRS: 3.20 ± 0.82, the Sundanese gamelan group was IRS: 3.12 ± 0.83, and in the Bali gamelan group IRS: 2.08 ± 0.30). Conclusion There are differences in the expression of synapsin 1 of the Rattus norvegicus offspring exposed to Mozart music, gamelan traditional music Javanese, Sundanese and Balinese in the cerebellum but there are no differences in the cerebrum during pregnancy. The highest synapsin expression was in the Mozart group.

Abstract

Tanaman sambiloto sering digunakan digunakan sebagai diuretika. Ekstrak sambiloto yang memiliki kandungan salah satunya diantaranya yaitu Diterpe lactone dimana dapat merangsang pelepasan insulin dan menghambat absorbsi glukosa melalui penghambatan enzim alfaglukosidase dan alfa-amilase yang dapat menurunkan kadar gula darah serta meningkatkan sensitivitas insulin melalui peningkatkan reseptor GLUT-4 sehingga menurunkan kadar insulin. Tujuan penelitian ini untuk mempelajari pengaruh kadar insulin pada rattus norvegicus model sopk dengan resistensi insulin yang diberi ekstrak sambiloto, yang diamati kadar insulin. Penelitian ini eksperimen laboratories, jenis rancangan acak lengkap dengan kelompok kontrol dan kelompok perlakuan sebanyak 25 ekor tikus putih (Ratus norvegicus) dibagi secara acak menjadi 5 kelompok masing- masing terdiri 5 ekor tikus putih. Kelompok kontrol (K) ada 2 yaitu K- tidak mendapat perlakuan, K+ dibuat model SOPK- resistensi insulin dengan pemberian testosterone propionate 28 hari. Kelompok perlakuan ekstrak sambiloto dosis 18 mg/ 100g bb /hari, kelompok perlakuan ekstrak sambiloto dosis 36 mg/ 100g bb /hari, kelompok perlakuan ekstrak sambiloto dosis 72 mg/ 100g bb /hari. Pemberian perlakuan diberikan satu kali sehari, dilaksanakan selama 4 siklus birahi tikus putih atau 15 hari. Hasil penelitian menunjukkan bahwa rerata dari hasil dari uji Manova diperoleh nilai signifikansi 0.554 (p>0,05) artinya tidak adanya perubahan yang nyata antara kadar insulin kelompok kontrol dan kelompok perlakuan. Kadar insulin yang tidak ada perubahan bermakna, mengindikasikan kemungkinan ekstrak sambiloto belum dapat memperbaiki keadaan insulin dengan memiliki jumlah reseptor insulin kurang, mengakibatkan penurunan sensitivitas insulin sehingga menyebabkan terjadinya keadaan hiperinsulinemia. Dengan asumsi ekstrak sambiloto belum mampu memperbaiki keadaaan kadar insulin, kemungkinan dipengaruhi oleh beberapa faktor diantaranya jumlah dosis pemberian

Abstract

Seiring bertambah banyaknya kasus kejadian SOPK- resistensi insulin yang terjadi angka kejadiaan 69% (Samsulhadi, 2003), yang mengakibatkan kesulitan untuk mendapatkan anak khususnya pasangan usia subur (infertilitas). Tujuan penelitian ini untuk mengetahui mekanisme perbaikan perkembangan folikel pada rattus norvegicus model sopk dengan resistensi insulin yang diberi ekstrak sambiloto. Penelitian ini eksperimen laboratories, jenis rancangan acak lengkap dengan kelompok kontrol dan kelompok perlakuan sebanyak 25 ekor tikus putih (Ratus norvegicus) dibagi secara acak menjadi 5 kelompok masing- masing terdiri 5 ekor tikus putih. Kelompok kontrol (K) ada 2 yaitu K- tidak mendapat perlakuan, K+ dibuat model SOPK- resistensi insulin dengan pemberian testosterone propionate 28 hari. Kelompok perlakuan ekstrak sambiloto dosis 18 mg/ 100g bb /hari, kelompok perlakuan ekstrak sambiloto dosis 36 mg/ 100g bb /hari, kelompok perlakuan ekstrak sambiloto dosis 72 mg/ 100g bb /hari. Pemberian perlakuan diberikan satu kali sehari, dilaksanakan selama 4 siklus birahi tikus putih atau 15 hari. Hasil penelitian bahwa rerata dari uji Anova yang terbesar dengan nilai 8.0000 adalah folikel primer pada kelompok K- dan diikuti kelompok P1 dan P3 ekstrak sambiloto dosis 18 mg dan 36 mg dengan nilai retara sebesar 6.8000. Hasil uji normalitas dengan oneway sample kolmogorov smirnov test bahwa variabel folikel primer, folikel sekunder, folikel tersier, folikel de Graff mempunyai nilai p > 0.05 disimpulkan data berdistribusi normal. Hasil uji LSD terdapat perbedaan yang bermakna antara perkembangan folikel primer kelompok kontrol dengan kelompok perlakuan dengan nilai p sebesar 0.031, dan perkembangan folikel de Graff kelompok control dan kelompok perlakuan dengan nilai p sebesar 0.002, maka dengan ini disimpulkan terdapat hubungan yang bermakna dengan penggunaan ekstrak sambiloto. Perlunya penggunaan dosis yang tepat dan waktu yang lebih lama dari ekstrak sambiloto dalam memperbaiki perkembangan folikel

Abstract

Infertility is a problem experienced by some women and men around the world. Most infertility problems in women is caused by impaired reproductive organs or disrupted ovulation.. One factor that causes impaired oocyte maturation is wireless fidelity (Wi-Fi) radiation which has a radio frequency field of 2.45 GHz. The radiation may increase the activity of free radical cells through the fenton reaction pathways that cause infertility because of the disrupted oocyte development. This study aimed to determine the effect of Wi-Fi radiation on the number of oocytes. This was an experimental study using control group design. The subjects were 32 female mice selected through complete random sampling and divided into two groups: control (R0) and exposure (R1). Each group consisted of 16 mice. The Wi-Fi radio frequency used was 2.5 GHz. Mice (R1) were placed closer to Wi-Fi source (± 15cm), and there were two types of laptop PCs and 3G mobile phones connected to internet placed next to mice for 15 hours/28 days. The mice underwent a simultaneous cycle with intraperitoneal injection of PMSG and HCG. Furthermore, the mice were mated with vasectomized male monomatingly to induce ovulation. The fertilization pouch in both oviducts were observed for oocyte collection. The number of oocytes was calculated using an inverted microscope. There was a difference in the number of oocytes between control and exposure group. Statistical tests were analyzed using Mann Whitney U and resulted in significant values (p value = 0.00). No oocytes count in exposure group. In other words, the group underwent anovulation. In conclusion, Wi-Fi radiation affected the number of oocyte stimulation in mice. Therefore, it was important to minimize the risk factors that trigger electromagnetic radiation on reproductive health.

Abstract

This study aimed to determine the effects of red fruit (Pandanus conoideus Lam) oil on MDA levels and spermatozoa quality in mice (Mus musculus) exposed to MSG. The quality includes motility, viability, concentration, and morphology of spermatozoa. This experimental study used randomized post-test only control group design. The subjects of this study were 25 mice (Mus musculus), divided into 5 groups (5 mice per group). K- group received distilled water for 35 days. K+ group received 4 mg/g BW MSG for 21 days. P1, P2, and P3 treatment groups received 4 mg/g BW MSG for 21 days and 0.02; 0.04; 0.08 ml/g BW red fruit oil, respectively, from day 22 to 35. The results showed that mean spermatozoa morphology in K-, K+, P1, P2, P3 groups were as follows: 0.86; 0.56; 0.67; 0.61; and 0.87 (%). The spermatozoa concentrations were sequentially as follows: 21; 10; 15; 32,8,19 (107 cells/ml). The spermatozoa's vitalities were as follows: 0,64; 0,14; 0,24; P2: 0.36; 0.68 (%). MDA levels were respectively: 0.29; 0.60; 0.35; 0.23; and 0.19 (nm). As a conclusion, testicular MDA levels in mice exposed to MSG and given with red fruit oil were lower than those in mice exposed to MSG without receiving red fruit oil. The quality of spermatozoa in mice exposed to MSG and receiving red fruit oil was higher than that of mice exposed to MSG without being given with red fruit oil.

Abstract

Folic acid contains 5-methyltetrahydrofolate (5-MTHF), one of antioxidants that can be used to inhibit reaction caused by cigarette smoke exposure. Folic acid is able to inhibit the levels of Reactive Oxygen Species (ROS) so as to reduce the occurrence of neuronal apoptosis. The objectives of this study was to determine the effect of folic acid intake during pregnancy whose mother was exposed to cigarette smoke on brain neurons apoptosis index and quantity of mice (Mus musculus) offspring. This study was an experimental laboratory study with posttest-only control group design. The subjects were 24 female mice divided into 4 groups consisting of one control group and three treatment groups. Sampling used simple random sampling, each group consisting of 6 animals. Control group 1 was not given treatment during pregnancy, group 2 was given with folic acid orally on days 0-17 of gestational age as much as 0.06 mg/day/mice. Group 3 was given with cigarette smoke exposure on days 7-17 of gestational age, amounting to 2 cigarettes/day. Group 4 was given with cigarette smoke exposure on days 7-17 of gestational age, amounting to 2 cigarettes/day and folic acid orally on days 0-17 of gestational age as much as 0.06 mg/day/mice. The result showed significant differences in all variables. In neurons apoptosis index, there were significant differences between groups 1 and 3, groups 2 and 3, groups 3 and 4, significant differences in the quantity of neurons between groups 1 and 3, groups 2 and 3, groups 2 and 4. The study concluded that there were significant difference in neural apoptosis index between all groups, and there were also significant differences in the quantity of neurons between all groups.
