Abstract

The quail’s egg have value of nutrient which similar to the chicken’s egg and the duck’s egg. Various of the programme of the light to improve repair of the quantity and the quality of the quail egg. The objectives of this research were to applied monochromatic light to repair the quality and quantity of the quail egg. Two hundred and seventy female quails (DOQ) were divided into nine treatments of light, with ten replications and three quails in each replication, the treatments were without light, controls with 15 and 25 W, red, green, and blue lights with intensities of 15 and 25 lux. Control treatment used incandescent bulb. The red, green, and blue lights were provided by light emitting diodes (LED). All lights treatment were given for 14 h daily, started from 17.00 to 07.00. Parameters measured were amount of the egg weekly, henday egg production, and quality of the egg consist of the weights of the egg, protein and fat of the egg. The result showed that quails exposed to blue light had higher of the amount of the egg, henday production, and the weights of the egg, while the quality of the egg, such as protein and fat in the egg were not change (P>0,05). Blue light could be used to improve repairing quantities of the quail egg.

Abstract

Light plays an important role in the avian life. The present study was designed to evaluate the effects of monochromatic light on serum estrogen concentrations and profile the oviduct, that were the length and weight of the oviduct in the quail. Two hundred and seventy quails were divided into nine treatments of light, with ten replications and three quails in each replication. The treatments were without light,  controls  with 15 and 25 W, red, green, and blue lights with intensities of  15 and 25 lux. Control  treatment used incandescent bulb.  The red,  green,  and  blue lights  were  provided  by   light emitting   diodes  (LED). All lights   treatment   were   given   for   14 h  daily,   started from 17.00 to 07.00. Parameters   measured  were  serum estrogen  concentrations, and  weights  and  lengths  of   the  oviduct  on  weeks 5,  7,  and  9.  Quails   exposed  to  monochromatic   light had  higher  serum  estrogen  concentrations (P<0,05). Quails  exposed to monochromatic light had  better  weights  and  lenghts of  the  oviduct  development. Blue light could be used to increase serum estrogen concentrations and stimulate shell gland development.

Abstract

Indera pendengaran tikus sawah memiliki dua puncak tanggap akustik yaitu pada kisaran suara terdengar (frekuensi 20 Hz â?? 20 KHz) dan ultrasonik (>20 KHz). Kemampuan indera tersebut penting dalam menunjang aktivitas kehidupan tikus sawah sebagai hewan nokturnal. Penelitian eksploratif dilakukan untuk mengumpulkan dan menginventarisasi vokalisasi alami tikus sawah pada rentang suara terdengar dalam kondisi alami di lapangan sepanjang musim tanam padi. Vokalisasi yang diperoleh dimurnikan dan dikarakterisasi menggunakan perangkat lunak Cool Edit Pro 2.1, selanjutnya dibuat databasenya. Hasil penelitian menunjukkan bahwa tikus sawah pada kondisi alami di lapangan tidak setiap saat melantangkan vokalisasi bioakustik sepanjang musim tanam padi. Eksplorasi sepanjang musim tanam padi diperoleh 6 pola vokalisasi bioakustik yang dilantangkan tikus sawah pada saat pengolahan lahan, padi stadia anakan maksimum, bunting, dan berbunga, serta seminggu pascapanen. Vokalisasi bioakustik berdurasi singkat, rata-rata 12,41 detik (0,5-25,1 detik) dengan frekuensi dominan 1-2 kHz yang disertai frekuensi 5-9 kHz selama pelantangan. Taraf intensitas menunjukkan tingkat kebisingan suara berkisar 6,94-93,90 desibel (rata-rata 43,91 dB). Penelitian lanjutan diperlukan untuk mengetahui respon perilaku tikus sawah apabila dipaparkan vokalisasi tersebut.

Abstract

Tanaman kipahit merupakan salah satu tanaman yang berpotensi sebagai sediaan alami antipiretik. Penelitian bertujuan mengetahui dosis efektif ekstrak etanol daun kipahit (Tithonia diversifolia) sebagai antipiretik. Sebanyak 24 ekor tikus putih jantan galur Sprague?dawley dengan bobot badan 150?200 g dibagi menjadi 6 perlakuan dan 4 ulangan. Perlakuan tersebut ialah tikus percobaan tidak diinduksi demam dan tanpa pemberian sediaan antipiretik (kontrol 0), tikus percobaan diinduksi demam tanpa mendapatkan sediaan antipiretik (kontrol ?), tikus percobaan diinduksi demam dan diberi sediaan ekstrak etanol daun kipahit (EEDK) dosis 100 mg/kg BB (perlakuan 1), 200 mg/kg BB (perlakuan 2), 300 mg/kg BB (perlakuan 3), dan 400 mg/kg BB (perlakuan 4). Induksi demam dilakukan dengan menyuntikkan vaksin DTP?HB?Hib dosis 0.2 mL/200 g BB IM. Pemberian EEDK dilakukan secara per oral pada 90 menit pascainduksi demam. Pengamatan dilakukan dengan mengukur suhu rektal tikus menggunakan termometer digital (tingkat ketelitian 0,1ºC) pada menit ke?0 (sebelum injeksi DTP?HB?Hib atau suhu normal), 90, 120, 150, dan 180 pascainduksi demam. Hasil penelitian menunjukkan bahwa pemberian EEDK mampu menurunkan suhu rektal tikus dibanding dengan kontrol negatif. Pemberian EEDK terbaik dalam menurunkan suhu rektal tikus percobaan terdapat pada dosis 100 dan 200 mg/kg BB. Simpulan, penelitian ini  menunjukkan bahwa daun kipahit memiliki aktivitas antipiretik. [MKB. 2017;49(3):139?44]Kata kunci: Daun, demam, ekstrak etanol, kipahit Exploration of Kipahit Leaf Ethanol Extracts as Natural AntipyreticsKipahit plant is one of the plants that have the potential to be used as natural antipyretics. This study was conducted to explore  the effective doses of ethanol extract of kipahit leaf (Tithonia diversifolia) as an antipyretics. Twenty four male sprague?dawley white rats weighed150?200 g were divided into 6 groups and replicated 4 times. The experiment included  experimental rats without fever induction and without administration of antipyretic substance (control 0), experimental rats with induced fever without administration of antipyretic substance (negative control), experimental rats with induced fever and  ethanol extract of kipahit leaf (EEDK) with various doses: 100 mg/kg BW (treatment 1), 200 mg/kg BW (treatment 2), 300 mg/kg BW (treatment 3), and 400 mg/kg BW (treatment 4). Fever was induced by injecting DTP?HB?Hib vaccines intramuscularly at a dose of 0.2 mL/200 g BW. Administration of EEDK was conducted orally at 90 minutes pos-injection of DPT?HB?Hib vaccines. The antipyretic effects of EEDK were observed by measuring rectal temperature by using digital thermometer (correction factor 0.1ºC) in 0 minute (before the injection of DTP?HB?Hib or normal temperature), 90, 120, 150, and 180 minutes post?fever induction. The result showed that the administration of EEDK decreased rectal temperature as compared to negative control. The optimum doses of EEDK administration that decreased rectal temperature were 100 and 200 mg/kg BW. It is concluded that the EEDK has an effect. Abstract

Serat pangan telah banyak digunakan sebagai pangan fungsional dan direkomendasikan untuk menurunkan kadar lipid darah untuk mencegah hiperkolesterolemia. Bekatul merupakan bahan pangan yang mengandung serat cukup tinggi. Tujuan penelitian untuk mempelajari pengaruh penambahan bekatul pada parameter lipid darah mencit jantan hiperkolesterolemia. Penelitian dilakukan di kandang hewan Jurusan Pendidikan Biologi Universitas Pendidikan Indonesia dan Laboratorium Fisiologi Fakultas Kedokteran Hewan Institut Pertanian Bogor periode Agustus 2011?Maret 2012. Desain penelitian menggunakan rancangan acak lengkap dengan lima kelompok perlakuan, yaitu kontrol negatif mencit normokolesterolemia yang diberi pakan standar tanpa suplementasi bekatul dan kontrol positif mencit hiperkolesterolemia tanpa suplementasi bekatul, selanjutnya kelompok mencit hiperkolesterolemia diberi suplementasi bekatul 16%, 38%, dan 57%. Parameter yang diukur ialah bobot badan, konsentrasi kolesterol total serum, hati, dan feses, kadar trigliserida, high density lipoprotein (HDL-c), low density lipoprotein (LDL-c), dan glukosa darah. Hasil penelitian menunjukkan bahwa suplementasi bekatul dalam diet menurunkan bobot badan, konsentrasi kolesterol serum total dan hati, trigliserida dan LDL-c, serta menaikkan konsentrasi HDL-c dan kolesterol feses, tanpa mengubah konsentrasi glukosa darah. Suplementasi bekatul sebesar 57% menurunkan bobot badan sebesar 10,31%, kadar total kolesterol 17,28%, trigliserida 28,63%, dan LDL-c 79,35%, serta meningkatkan HDL-c sebesar 24,41%. Suplementasi bekatul menurunkan kolesterol hati 57,76% dan meningkatkan pembuangan kolesterol melalui feses sebesar 39,86%. Simpulan, bekatul sebagai suplemen makanan memperbaiki parameter lipida darah mencit jantan hiperkolesterolemia dengan meningkatkan pembuangan kolesterol melalui feses dan menurunkan bobot badan tanpa mengubah kadar glukosa darah [MKB. 2013;45(1):1?9]Kata kunci: Bekatul, hiperkolesterolemia, lipid darah, mencit, serat pangan Improvement of Blood Lipid Parameters of Hypercholesterolemic Mice by Supplementation of Rice BranDietary fiber is widely used as a functional food and recommended to reduce blood lipid level to prevent hypercholesterolemia. An experiment was conducted on the effects of rice bran that has high dietary fiber content on blood lipid parameters of hypercholesterolemic male mice, which was conducted at the Biology Education Department animal cages, Indonesia University of Education and the Veterinary Medicine Faculty Laboratory, Bogor Agricultural Institute from August 2011 to March 2012. The mice were randomly assigned into groups with 5 different treatments: negative control group i.e. normocholesterol mice with a standard diet without rice bran; positive control group i.e. hypercholesterolemic mice with a standard diet without rice bran; and groups with hypercholesterolemic mice with 16%, 38%, and 75% rice bran supplements. The parameters were body weight as well as blood serum, liver and feces cholesterol, triglyceride, high density lipoprotein (HDL-c), low density lipoprotein (LDL-c) and glucose levels. Body weight, blood serum and liver cholesterol as well as triglyceride and LDL concentrations decreased while serum HDL-c and feces cholesterol increased without any significant effect on blood glucose concentration with rice bran supplement. Supplementation of rice bran by 57% (14.30% of nonsoluble dietary fiber content) decreased body weight, total serum cholesterol, triglyceride, and LDL-c of 10.31%, 17.28%, 28.63%, and 79.35%, respectively, and increased HDL-c by 24.41%. Rice bran supplement reduced liver cholesterol levels by 57.76% and increased fecal cholesterol excretion by 39.68%. In conclusion, 57% rice bran supplementation is effective to improve blood lipid parameters of hypercholesterolemic male mice by increasing feces excretion and decreasing body weight with no change in blood glucose level. Abstract

Asam valproat memengaruhi aktivitas histone deacetylase yang berperan dalam ekspresi gen selama organogenesis. Insulin berperan dalam proliferasi dan diferensiasi sel-sel saraf dentate gyrus hipokampus. Penelitian ini bertujuan mempelajari pengaruh pemaparan asam valproat pada induk bunting terhadap ekspresi gen insulin pada dentate gyrus. Penelitian dilakukan di UPHL IPB pada bulan Mei 2015 hingga Desember 2016 dengan  84 ekor anak tikus yang dilahirkan oleh induk tikus kontrol yang diberi asam valproat 250 mg pada umur kebuntingan 10, 13, dan 16 hari digunakan untuk pengamatan kadar glukosa, insulin, DNA, RNA, dan rasio RNA/DNA serta pengamatan mikroskopis otak. Pengamatan dilakukan selang waktu empat minggu, dimulai dari umur 4 sampai 32 minggu. Anak tikus yang dilahirkan oleh induk tikus yang diberi asam valproat selama kebuntingan mempunyai kadar glukosa otak yang lebih tinggi (p<0,01) dan insulin yang lebih rendah (p<0,05).  Selama periode pertumbuhan, anak tikus yang dilahirkan oleh induk tikus yang diberi asam valproat mengalami peningkatan kadar glukosa dan penurunan kadar insulin (p<0.05). Pengamatan mikroskopis sel-sel dentate gyrus menunjukkan degenerasi sel dan tidak terlihat reaksi imunoreaktif terhadap insulin, namun terjadi penurunan konsentrasi DNA, RNA, serta rasio RNA/DNA (p<0,05). Pemberian asam valproat pada induk tikus pada umur kebuntingan 10, 13, dan 16 hari memengaruhi organogenesis otak anak tikus sehingga menyebabkan kerusakan sel-sel saraf penghasil insulin otak yang ditunjukkan oleh  penurunan sekresi dan kadar insulin. Insulin plays a role in regulating the proliferation and differentiation of the dentate gyrus. This experiment was designed to study the effects of valproic acid administration on the expression of insulin gene in the dentate gyrus of pregnant rats.  This study was carried out at the Laboratory Animal Management Unit (UPHL) of Bogor Agricultural University  from May 2015 to December 2016, with eighty four newborn rats from control rats and VA-administered rats at the age of 10, 13, and 16 days of pregnancy. The measurements were done for glucose, insulin, DNA, and RNA concentrations in dentate gyrus and to observe the microscopic condition of the brain at the ages of 4 to 32 weeks with 4 weeks interval.  Rats born form mothers administered by 250 mg valproic acid during pregnancy had higher glucose concentrations (p<0.01) and lower insulin concentrations (p<0.05) in the dentate gyrus. During the post-natal growth phase, rats born from mothers administered with valproic acid had increased glucose concentrations and decreased insulin concentrations (p<0.05). Microscopic observations of the dentate gyrus showed degeneration of nerve cells with no immunoreaction to insulin was seen in dentate gyrus cells. However, the concentrations of DNA, RNA, and the ratio of RNA/DNA in the dentate gyrus cells decreased (p<0.05). The administration of valproic acid to pregnant rats at the age of 10, 13, and 16 days of pregnancy destructs  brain organogenesis of the offsprings which eventually disturbs insulin production in the dentate gyrus indicated by decreased insulin secretion and concentrations.  [MKB. 2017;49(3):156–64]Key words: Dentate gyrus, insulin, organogenesis, valproic acid

Abstract

The  aim  of  this  research  is  to  study  the  effects  of the  methanol  extract and  its  derived  fraction  (n-hexane,  chloroform,  ethyl  acetate,  and  methanolwater)  on  liver  function  of  male  rats.  The  treatment  groups  were  administered 500  mg/kg body weight of metanol extract and its derived fractions (n-hexane, chloroform,  ethyl  acetate,  n-butanol,  methanol-water)  of  E.  longifolia for  7 consecutive  days.  Positive  control  group  received  25  mg  silymarin/kg  body weight and negative control group received 2 mL aquadest/kg body weight daily for  7  consecutive  days.  Liver  function  was  monitored  by  measuring  alanine transaminase  (ALT),  aspartate  transaminase  (AST),  total  protein,  alkaline phosphatase  (ALP),  total  bilirubin,  direct  bilirubin,  and  indirect  bilirubin consentrations  in  the  serum.  Compared  with  control,  oral  administration  of methanol  extract  and  derived  fractions  of  methanol extract  of  E.  longifolia root had  no  significant  effects  on  liver  function  (p>0.05).  Methanol-water fraction gave similar results to silymarin.Key words: Eurycoma longfolia Jack.,liver function, biochemical blood Histopathologi

Abstract

Forty eight gilts with average body weight of 107.83 + 5.08 kg were used in experiments to study the use of pregnant mare serum gonadotrophin (PMSG) and human chorionic gonadotrophin (hCG) as superovulation agent in gilts to increase piglet production. Four groups of twelve gilts were injected with PMSG dan hCG dose levels of 0, 600, 1200, and 1800 IU/gilt. Injections were conducted three days before estrus. During gestation, gilts were placed in colony pigpens. On days 15, 35, and 70 of gestation, gilts were slaughtered in order to measure the number of corpus luteum, growth and development of the uterus and placenta. Blood samples were collected to determine progesterone and estradiol concentrations. The resuts showed that superovulation dose levels of 600 to 1200 IU/gilt increased progesterone and estradiol secretions, growth and development of the uterus and placenta in gestation ages of 15, 35, and 70 days. It is concluded that superovulation with dose of 600 to 1200 IU can improve the gilts reproduction. Key words: gilts, superovulation, gestation, progesterone, estradiol, corpus luteum

Abstract

The limiting factor which is a weakness of sea water pearl production are high costs, the risk of major business failures and a long coating time. From the issue of freshwater pearls appear to have prospects of alternative substitution for sea water pearl. This present study aimed to evaluate effect of loads (the number and diameter nucleus) on freshwater pearl coating process and the number and size of the appropriate nucleus diameter, to produce the optimum coating thickness of half-round pearls. The research consists of experimental implantation of 2, 4, and 6 nucleus number per individual mussel was maintained by the method stocked in hapa in bottom waters. Observation method and factorial randomized block design used in the study of the influence of the load to the successfulness of  pearl coating and the pearl layer thickness. The results showed that A. woodiana can be utilized as a producer of freshwater pearls. In addition, the number of optimum nucleus that can be attached to the mussel A. woodiana was 2 grains/individuals with a diameter of 10 mm. Shells implanted with the optimum nucleus diameter and number of pearls produced the highest layer thickness of 17 mm after 9 months cultivation. This result was good enough compared with the layer thickness of sea water pearl production after the same cultivation time.

Abstract

An experiment was conducted to study the effect of male and female host oysters on the pearl sac formation in Pinctada maxima oyster. One hundred sixty oysters were used in a completely randomized design with 2 x 4 factorial arrangement and 20 replications. The 1st factor was that sex of host oyster consisted of two levels that is males and females. The 2nd factor was week after nucleus implantation with four levels that is 1, 2, 3, and 4 weeks. The parameters observed were the percentage of successful oysters to form the pearl sac, the speed of pearl sac formation, the percentage of nucleus coverage by the pearl sac, histology of the pearl sac growth and development, and haemolymph glucose, calcium and phosphorus concentrations. Our results showed that the percentages of host oysters that succeeded in forming a pearl sac were 80% and 75% in female and male host oysters, respectively. There was no statistical difference in nucleus rejection and mortality in male and female host oysters but the results indicated that male host oysters showed a numerically higher nucleus rejection. The speed of pearl sac growth and the percentage of nucleus coverage by the pearl sac in female host oysters were better than those in male host oysters. Haemolymph calcium, phosphorus and glucose concentrations, oxygen consumption, and histological development of the pearl sac were not different between male and female host oysters. Pearl sac formation in the female host oysters was better than that in male host oysters.
