Abstract

Background: Cytokines and growth factors were reported to play an important role in stimulating fibroblast proliferation. In vitro culture, fibroblast is mostly culture in medium containing fetal bovine serum (FBS).  Human umbilical cord blood (hUCB) has been reported to have low immunogenic property and potential in wound healing, so therefore hUCB serum (hUCBS) could be potential and were investigated in current study.Materials and Methods: Five hUCBs were collected from healthy volunteers with normal delivering procedure. hUCB was ex utero immediately collected from umbilical vein in vacutainers and processed. NIH3T3 cells were cultured in DMEM with 10% FBS or 5-20% hUCBS for 48 hours. Cells were then quantified using MTT assay. Protein concentration of FBS and hUCBS were quantified using Bradford assay.Results: NIH3T3 cells density grown in DMEM with 10% FBS was the lowest. NIH3T3 cells densities were increased along with the increment of hUCBS concentrations. MTT results showed that average number of NIH3T3 cells grown in DMEM with 10% FBS was 6,185±1,243. Meanwhile average numbers of NIH3T3 cells grown in DMEM with 5%, 10% and 20% hUCBS were 8,126±628, 9,685±313 and 12,200±304, respectively. Average numbers of NIH3T3 cells grown in DMEM with 5% hUCBS were significantly higher than the ones with 10% FBS (p=0.000). Bradford results showed that concentration of hUCBS was significantly higher than the one of FBS (p=0.000).Conclusion: hUCBS could induce higher proliferation rate of NIH3T3 cells than FBS. Hence hUCBS could be suggested as an alternate of FBS in inducing fibroblast.Keywords: NIH3T3, fibroblast, UCB, serum, FBS, proliferation

Abstract

Background: Caffeic acid has been reported that when it is combined with all-trans retinoic acid, it can inhibit proliferation activity of SaOS-2 or OSA-01 cells. In addition, caffeic acid merely could reduce cell viability of SaOS-2 cells. However, there is not any study in caffeic acids possible effect to induce apoptosis in osteosarcoma cell.Materials and Methods: MG-63 cells were cultured in Dulbecco’s Modified Eagle Medium containing 10% fetal bovine serum. Cells were treated with various concentrations of caffeic acid. Apoptosis were analyzed with Sub-G1 assay and activation of caspase-8, -9, and -3 were analyzed with immunoblotting. Caffeic acid-induced percentage of apoptotic cells and cleaved-8, -9, -3 were then statistically analyzed.Results: Sub-G1 results showed that caffeic acid significantly induced apoptosis in MG-63 osteosarcoma cells in concentration dependent manner. Immunoblotting results showed that caffeic acid induced cleavage of caspase-8, -9 and -3. Cleaved-caspase-8 and -9 were increased at 1-hour treatment of caffeic acid, while cleaved-caspase 3 was increased markedly at 6-hours treatment of caffeic acid.Conclusions: Caffeic acid induces apoptosis significantly in concentration dependent manner through caspase-dependent intrinsic apoptotic pathway.Keywords: caffeic acid, osteosarcoma, MG-63, apoptosis, caspase

Abstract

Background: Interaction between advanced glycation end product (AGE) and receptor for AGE (RAGE) triggers the escalation of inflammatory cytokine expressions. High-sensitivity C-reactive protein (hsCRP), an important inflammatory marker, has been reported to be modulated by soluble RAGE (sRAGE). However, the relationship between hsCRP and sRAGE in diabetes was not clearly described. Therefore present study was conducted to determine the relationship between sRAGE with hsCRP in men with central obese diabetic and non-diabetic. Materials and Methods: Adult men aged 25-60 years with central obese diabetes and non-diabetes, were recruited. Patient’s profiles were collected before the physical and blood examination. Physical examinations were conducted by measuring waist/abdomen, blood pressure, height, and weight. The routine blood test was performed to obtain concentrations of fasting blood glucose, HbA1c, hsCRP and sRAGE level.Results: Fifty-seven subjects with central obese and waist size ≥90 cm were selected. It was found that hsCRP values were significantly different (p=0.000) in HbA1c <6.5% dan HbA1c ≥6.5% groups. There was an inverse relationship between hsCRP and sRAGE levels for both in HbA1c <6.5% (r=-0.073) and HbA1c≥6.5% (r=-0.022) groups. In HbA1c ≥6.5% group, sRAGE showed strong positive correlation with 1 mg/dL ≤ hsCRP <3 mg/dL group (r>0.5).Conclusions: In the early stages of diabetes with hsCRP <1 mg/dL, the protective function was demonstrated with greater sRAGE levels. However, in further phase with 3 ≤ hsCRP < 10 mg/dL, the level of sRAGE was low, which is assumed to be associated with complications. Hence, sRAGE could be suggested as a complementary marker for hsCRP to evaluate diabetic men with central obesity.Keywords: sRAGE, hsCRP, diabetes, HbA1c, central obesity

Abstract

Secara  in vitro,  Embryonic Stem Cell  (ESC) dapat diarahkan perkembangannya menjadi sel neuron dan sel glia. Conditionedmedium  dari kultur primer sel syaraf mengandung sejumlah faktor pertumbuhan antara lain  nerve growth factor (NGF), glial derived-neurotrophic factor (GDNF), nestin, dan glial fibrillary acidic protein (GFAP). Dengan melakukan purifikasi protein yang terkandung  di dalam CM, maka diharapkan spektrum protein yang ada menjadi lebih sempit sehingga protein target dapat terdeteksi. Penelitian ini mempelajari kultur primer sel syaraf yang berasal dari hemisfer Mus musculus. Tujuan penelitian adalah untuk mendapatkan CM dari kultur primer sel syaraf  Mus musculus. Medium yang digunakan adalah Dulbecco?s Modified Eagle?s Medium (DMEM) highglucose FBS 10%. Penggantian medium kultur dilakukan setiap 2 hari sekali. Kepadatan sel sekitar 32x103 sel/2 cm2. Setelah hari ke-4 terlihat adanya pertumbuhan neuron bipolar dan neural progenitor cell (NPC). Sel-sel astrosit akan teramati ketika periode kultur diperpanjang. Sel mengalami konfluensi setelah 12 hari kultur. Sel-sel yang tumbuh berguna untuk penjelasan neurogenesis. Kultur primer sel syaraf secara monolayer yang berasal dari hemisfer neonatus mampu mendukung  pertumbuhan sel yang tergolong sebagai neurogenic dan nonneurogenic.  Kata kunci: kultur primer, sel syaraf, conditioned medium, neural progenitor cell, neurogenesis.

Abstract

Abstract: The escalation of COVID-19 cases both in morbidity and mortality rates in Daerah Khusus Ibukota (DKI) Jakarta and its surrounding areas (JABODETABEK) continues to occur. Adherence in implementing health protocols depends on the knowledge and attitudes of the individuals. This study aimed to assess the knowledge and attitude regarding Covid-19 prevention in an informal community, namely music community. This study was conducted online with 44 participants obtained through convenience sampling method. Participants filled out the questionnaires covering individual characteristics, general knowledge regarding Covid-19, and attitudes towards protective measures against Covid-19. The Mann-Whitney test was used to analyze the differences in the mean score of knowledge based on selected variables. There were 52.4% of participants that had low level of knowledge. Meanwhile, 86.4% of participants had positive attitude towards protective measures against Covid-19. Significant differences were found in the mean scores of knowledge based on age (p=0.001), place of residence (p=0.004), and occupational status (p=0.032). In conclusion, most participants of the music community in Jakarta and surrounding areas have low level of knowledge, however, they have positive attitude towards protective measures against Covid-19.Keywords: Covid-19; knowledge; attitude; music community Abstrak: Peningkatan kasus Covid-19 baik dalam angka morbiditas maupun mortalitas di wilayah Daerah Khusus Ibukota (DKI) Jakarta dan sekitarnya (JABODETABEK) masih berlangsung. Kepatuhan dalam menerapkan protokol kesehatan bergantung pada pengetahuan dan sikap individu. Penelitian ini bertujuan untuk mengetahui gambaran pengetahuan dan sikap mengenai pencegahan Covid-19 pada salah satu komunitas informal di masyarakat, yaitu komunitas musik. Penelitian ini dilakukan secara daring dengan 44 peserta yang didapatkan melalui metode convenience sampling. Peserta mengisi kuesioner survei yang berisi data karateristik individu, pengetahuan umum, dan sikap terkait Covid-19. Uji Mann-Whitney digunakan untuk melihat perbedaan rerata nilai pengetahuan berdasarkan setiap variabel. Hasil penelitian mendapatkan sebanyak 52,4% responden memiliki tingkat pengetahuan rendah namun 86,4% responden memiliki sikap positif terhadap upaya pencegahan penularan Covid-19. Perbedaan rerata nilai pengetahuan secara bermakna ditemukan berdasarkan usia (p=0,001), wilayah tinggal (p=0,004), dan status pekerjaan (p=0,032). Simpulan penelitian ini ialah sebagian besar komunitas musik di DKI Jakarta dan sekitarnya memiliki tingkat pengetahuan rendah namun memiliki sikap positif terhadap upaya pencegahan penularan Covid -19. Kata kunci: Covid-19; pengetahuan; sikap; komunitas musik

Abstract

Background: Empirically, Erythrina fusca has been used as traditional herb for its antibacterial and antiinflammation properties. Periodontal disease is one of the most oral infectious diseases with microorganism predominated as the contributing factors. Porphyromonas gingivalis (P. gingivalis) is one of the main bacteria pathogen found in periodontal diseases. Purpose: The purpose of this study was to examine the bactericidal effect of Erythrina fusca Leaves Aquadest Extract (EFLAE) at various concentrations on P. gingivalis and cytotoxic effect on fibroblast. Methods: Pure P. gingivalis was cultured in Brain Heart Infusion (BHI) medium for 24 hours with or without various concentrations of treatment of EFLAE. Calculation and statistical analysis of remaining bacteria were performed by inhibitory zone method to evaluate the EFLAE bactericidal effect and compared to chlorhexidine as positive control. To evaluate the cytotoxic effect, NIH 3T3 cells were cultured in Dulbecco’s Modification of Eagle’s Medium (DMEM) containing of 10% fetal bovine serum (FBS) and 1% penicillin-streptomycin, pH 7.2, in 5% CO2, and stored in humidified incubator under temperature 370 C. Cells were treated with/without various concentrations of EFLAE for 48 hours. The viable cells were then counted using 3-(4,5- Dimethylthiazol-2-yl)-2,5 diphenyl tetrazodium bromide (MTT) method. Results: EFLAE have bactericidal effect on P. gingivalis in a concentration dependent manner starting from 78%. The concentration of 90% EFLAE had stronger bactericidal effect (35.004 ± 1.546) than those of chlorhexidine as positive control (32.313 ± 1.619). One-way ANOVA showed significant bactericidal effect differences among concentrations of EFLAE and chlorhexidine (p<0.05) while Tuckey HSD test showed significant difference only between lower concentration of EFLAE (78%, 79%) and chlorhexidine. With the highest concentration of EFLAE (100%) applied in the bactericidal test, no cytotoxic effect of EFLAE on NIH 3T3 cells was detected. Conclusion: EFLAE could inhibit the growth of P. gingivalis in a concentration dependent manner, starting from 78%. There was no evidence of EFLAE’s cytotoxic effect on fibroblast.Latar belakang: Erythrina fusca telah digunakan secara empiris sebagai tanaman obat tradisional untuk khasiat antibakteri dan antiradang. Penyakit periodontal merupakan salah satu penyakit infeksi mulut terbanyak dengan mikroorganisme sebagai faktor kontributor utama. Porphyromonas gingivalis (P. gingivalis) merupakan salah satu bakteri patogen utama yang ditemukan pada penyakit periodontal. Tujuan: Tujuan penelitian ini untuk mengamati efek bakterisid terhadap P. gingivalis dan efek sitotoksik terhadap sel fibroblast dari beberapa konsentrasi ekstrak akuades daun Erythrina fusca (EFLAE). Metode: P. gingivalis murni dikultur pada medium Brain Heart Infusion (BHI) selama 24 jam dengan atau tanpa pemberian beberapa konsentrasi EFLAE. Perhitungan dan analisis statistik terhadap bakteri yang masih hidup dilakukan dengan metode zona hambat untuk mengevaluasi efek bakterisid EFLAE dibandingkan dengan chlorhexidine sebagai kontrol positif. Untuk mengevaluasi efek sitotoksik, digunakan kultur sel NIH 3T3 pada medium Dulbecco’s Modification of Eagle’s Medium (DMEM) yang berisi fetal bovine serum (FBS) 10% dan penicillin-streptomycin 1%, pH 7.2, dalam CO2 5%, dan diinkubasi pada suhu 37° C. Sel diberi perlakuan dengan atau tanpa beberapa konsentrasi EFLAE selama 48 jam, kemudian sel yang masih hidup dihitung menggunakan metode 3-(4,5-Dimethylthiazol-2-yl)-2,5 diphenyl tetrazodium bromide (MTT). Hasil: EFLAE mempunyai efek bakterisid terhadap P. gingivalis mengikuti kenaikan konsentrasinya dimulai dari 78%. Pada konsentrasi 90%, EFLAE menunjukkan efek bakterisid lebih kuat (35.004 ± 1.546) dibandingkan dengan chlorhexidine (32.313 ± 1.619) sebagai kontrol positif ANOVA-1 jalan menunjukkan perbedaan efek bakterisid yang bermakna di antara beberapa konsentrasi EFLAE dan chlorhexidine (p<0.05) sedangkan uji Tuckey HSD menunjukkan perbedaan bermakna hanya ditemukan antara konsentrasi EFLAE yang lebih rendah (78%, 79%) dengan chlorhexidine. Efek sitotoksik terhadap sel NIH 3T3 tidak terdeteksi pada pemberian konsentrasi tertinggi EFLAE (100%) yang telah diaplikasikan pada uji bakterisid. Kesimpulan: EFLAE dapat menghambat pertumbuhan P. gingivalis sesuai dengan konsentrasinya dimulai dari 78%. Tidak ada efek sitotoksik EFLAE terhadap sel fibroblast.

Abstract

Background: In Jakarta, oral squamous cell carcinoma (OSCC) usually detected in late stage with very low survival rate ofabout 1.1 years. OSCC may be preceded by premalignant lesion, so that early detection of the lesion may decrease the mortality rate due to oral malignancy. CD133 is a hematopoietic stem cell that play role in tissue regeneration, inflammation and tumor. Upregulated of CD133 was reported on tumor progression. Purpose: The aim of study is to determine circulating CD133 expression on premalignant (PML) and proliferative (PL) lesion. Method: Observational research was carried out on patients who seek treatment of PML and PL at Oral Medicine clinic. CD133 was taken from peripheral blood serum, examined using PCR. Data was analyzed by Chi square test. Result: 15 subjects (each of five subjects for PML, PL and control) consist of 40% male and 60% female. Age group of above 41 years old was most affected PML and PL (66.7%). Tongue is common site for oral lesion (40%). There is a significant different of circulating CD133 rate among all groups lesion (p=0.039). Conclusion: CD133 express differently in premalignant and proliferative lesions.

Abstract

BACKGROUND: Our previous report showed that phosphorylated-survivin at Ser81 induces survivin back loop to activate protein kinase A (PKA) in the cytoprotection mechanism. Activated PKA could possibly induce the cytoprotection via Phosphatydilinositol 3-kinase (PI3K). Therefore our current study was conducted to investigate the possibility of survivin-PKA-PI3K signaling pathway.METHODS: Viral productions by BOSC23 cells of Survivin, Antisense Survivin (Survivin-AS) and Ser81Ala mutant (Survivin-S81A) in pMSCV-IRES-GFP vector with cytomegalovirus (CMV) promoter were conducted. L929 cells were pretreated with/without PKI 6-22 amide and infected with viral particle of Survivin, Survivin-AS, Survivin-S81A or vector only. Cells were harvested, lysed and immunoprecipitated with anti-PI3K (p85) antibody and immunoblotted to detect PI3K (p85) and phospho-(Tyr) p85 PI3K. To confirm PI3K activation, PI3K Activity Assay was conducted by using phosphoinositide fraction containing PtdIns(4,5)P2 and [32P]ATP.RESULTS: Immunoblot and PI3K activity results showed similar results. Upon infection of virus with survivin, a markedly increased level of tyrosine phosphorylation of p85 PI3K or PI3K activity in L929 cells was seen. Low levels of tyrosine phosphorylation of p85 PI3K or PI3K activity were observed for Survivin-AS and Survivin-S81A-viral-infected L929 cells. With higher concentrations of Survivin-viral-infection, levels of tyrosine phosphorylation of p85 PI3K or PI3K activity in L929 cells were gradually increased. However, when L929 cells were pretreated with PKI 6-22 amide, prior to Survivin-viral-infection, level of tyrosine phosphorylation level of p85 PI3K or PI3K activity was detected much lower.CONCLUSION: Our result suggest that Ser81 Survivin play role in inducing PI3K activation and the Survivin-PI3K signaling pathway was PKA-dependent.KEYWORDS: Ser81, Survivin, PKA, PI3K, L929

Abstract

BACKGROUND: Acute myocardial infarction (AMI) is one of cardiovascular diseases with high morbidity and mortality rates. Novel biomarkers that can detect accurately acute coronary syndrome (ACS) at early stage, are necessary to improve current strategies and/or to identify subjects who are at risk. Fibrinogen and high-sensitivity C-reactive protein (hs-CRP) roles in inflammation process could be potential for ACS early detection. This study was conducted to evaluate measurements of fibrinogen and hs-CRP on ACS.METHODS: An analytic observational study with cross sectional approach was conducted on patients with Troponin I positive. After signing informed consent, anamnesis and complete blood count were conducted. Besides that, liver function, renal function, and blood glucose tests were conducted as well. Samples of selected subjects were quantified with enzyme-linked immunosorbent assay (ELISA) for Troponin I, fibrinogen and hs-CRP. Then statistical analyses were performed.RESULTS: There were 76 subjects in each ACS and non-ACS groups. ACS group showed significant higher levels of both fibrinogen and hs-CRP compared to Non-ACS group (p=0.000). Among evaluated risk factors, diabetes mellitus (DM) (p=0.003) and hypertension (p=0.000) were significantly higher in ACS group than in non-ACS group. Among evaluated clinical factors, blood glucose (p=0.001) and age (p=0.000) were significantly higher in ACS group than in non-ACS group. Combination of fibrinogen and hs-CRP measurements showed the highest sensitivity (75.00%), specificity (80.26%), accuracy (77.63%), positive predictive value (79.19%) and negative predictive value (76.25%).CONCLUSION: Since fibrinogen and hs-CRP were increased in ACS group and combination of fibrinogen and hs-CRP measurements showed the highest sensitivity, specificity, accuracy, positive predictive value and negative predictive value, we suggest that combination of fibrinogen and hs-CRP measurements could give added value to identify ACS.KEYWORDS: fibrinogen, hs-CRP, biomarker, ACS, acute coronary syndrome, atherosclerosis, inflammation

Abstract

BACKGROUND: Secreted factors contained in conditioned media (CM) of human umbilical cord blood mesenchymal stem cells (hUCB-MSCs) known as secretome, was suspected to have important roles in regulating cells. This study was conducted to investigate the role of CM-hUCB-MSCs-derived secretome in apoptosis and growth of HeLa cells.METHODS: HeLa cells were treated with secretome in various concentrations (0, 0.2, 2 and 20%) for 24 and 48 hours. Trypan blue exclusion assay was performed to detect cell viability. Meanwhile sub-G1 apoptotic assay was performed to detect apoptotic cells. The transition of mitochondrial transmembrane potential (TMP), which occurs in the apoptotic process, was analyzed by mitochondrial membrane potential (ΔΨM) assay. Both sub-G1 and ΔΨM assays were performed using FACSCanto flow cytometer. Statistical analyses were conducted using IBM SPSS Statistics to detect significance level at p<0.05.RESULTS: Secretome significantly induced cell death starting at concentration of 0.2% within a 24-hour period (p<0.05). Secretome significantly induced cell death in concentration and time dependent manner (p<0.05). The cell death was then confirmed as apoptosis through sub-G1 analysis. Due to the underlying apoptotic mechanism, we found distinct decrease of TMP, indicating an increase in mitochondrial membrane permeability of HeLa cells. In addition, we found that HeLa cell growth was inhibited partially by secretome.CONCLUSION: Taken together, we conclude that CMhUCB-MSCs-derived secretome significantly induced apoptosis of HeLa cells in a concentration and time dependent manner through mitochondrial apoptotic pathway.
