Abstract

Abstrak: Diabetes melitus memiliki jumlah kasus yang cukup banyak baik di Dunia maupun di Indonesia. Pola hidup masyarakat yang kurang sehat menjadi faktor utama terjadinya peningkatan kasus diabetes. Program pengabdian kepada masyarakat ini bertujuan untuk meningkatkan pengetahuan dan keterampilan masyarakat dalam penerapan pola hidup sehat dan pemanfaatan herbal untuk pengendalian diabetes. Metode yang digunakan dalam pelaksanaan program antara lain penyuluhan dan pelatihan terhadap warga Kelurahan Kalirungkut Surabaya yang berjumlah 60 orang. Materi penyuluhan dan pelatihan terkait dengan pentingnya pola hidup sehat dan penerapan budidaya tanaman herbal serta pemanfaatannya dalam pengendalian diabetes. Evaluasi yang dilakukan untuk mengetahui peningkatan pemahaman warga terhadap materi adalah metode kuesioner pre-test dan post-test menggunakan 10 buah pertanyaan. Hasil peningkatan pemahaman warga pada kegiatan penyuluhan sebesar 33,95 % dan pelatihan 45,47 %. Tindak lanjut untuk mengoptimalkan peningkatan pemahaman tersebut, perlu dilakukan pendampingan warga melalui kader kesehatan terutama kelompok warga yang memiliki riwayat dan resiko diabetes.Abstract: Diabetes mellitus has a high number of cases worldwide, including in Indonesia. Unhealthy community lifestyles are a major factor in the increase in diabetes cases. This community service program aims to increase community knowledge and skills in implementing healthy lifestyles and utilizing herbs to control diabetes. The methods used in the implementation of the program include counseling and training for 60 residents of Kelurahan Kalirungkut, Surabaya. The counseling and training materials are related to the importance of a healthy lifestyle and the application of herbal plant cultivation and their use in controlling diabetes. Evaluation conducted to determine the increase in residents' understanding of the material is the pre-test and post-test questionnaire method using 10 questions. The results showed an increase in understanding of residents in counseling activities by 33.95% and training by 45.47%. To further optimize the increase in understanding, it is necessary to provide mentoring to residents through health cadres, especially for groups of residents who have a history and risk of diabetes.

Abstract

Background: Colorectal cancer is one of the cancers with high prevalence in Indonesia. Management generally involves surgery and adjuvant chemotherapy. However, chemotherapy often causes side effects that can affect the nutritional status and functional capacity of patients. Nutritional status and Karnofsky index are important indicators in assessing the quality of life of post-chemotherapy patients. Aims: This study aims to analyze the relationship between adjuvant chemotherapy with nutritional status and Karnofsky index in patients with stage III colorectal cancer after surgery. Methods: This study used an analytic observational approach with a cross-sectional design. A total of 65 stage III colorectal cancer patients at Sultan Agung Islamic Hospital Semarang who met the inclusion and exclusion criteria were analyzed using medical record data. Nutritional status was assessed through body mass index (BMI), and Karnofsky index was classified into good and bad categories. Data analysis was performed using the Chi-Square test. Results: Most patients had BMI in the normal category (58.5%) and Karnofsky index in the good category (64.6%). Statistical test results showed that there was no significant relationship between adjuvant chemotherapy and nutritional status (p = 0.356) or Karnofsky index (p = 0.841). Conclusion: Adjuvant chemotherapy was not significantly associated with nutritional status and Karnofsky index in post-surgical stage III colorectal cancer patients. This study emphasizes the importance of a holistic care approach and nutritional support in maintaining patients' quality of life.

Abstract

Inspeksi pemeliharaan gedung secara konvensional memerlukan biaya tinggi, waktu yang lama, dan menimbulkan risiko besar bagi pekerja. Quadcopter berbasis kecerdasan buatan menawarkan solusi yang lebih efisien, akan tetapi kemampuan pemrosesan pada perangkat tersebut terbatas. Penelitian ini merupakan pengembangan dari studi sebelumnya dengan menganalisis dan membandingkan kinerja YOLOv8 dan YOLOv11 dalam mendeteksi keretakan bangunan. Lingkungan simulasi yang realistis dirancang menggunakan Gazebo dan ROS, sementara model YOLOv8n dan YOLOv11n dilatih dan diuji baik pada komputer desktop maupun sistem tertanam Raspberry Pi 5. Evaluasi dilakukan menggunakan metrik Presisi (%) dan waktu inferensi rata-rata (ms). Hasil penelitian menunjukkan adanya trade-off kinerja pada berbagai skenario. Dalam simulasi, YOLOv8 lebih cepat (53,58 ms vs. 56,19 ms) dan unggul pada Presisi Dataset B (100% vs. 95,19%). Pada sistem tertanam, YOLOv8 tetap menunjukkan Presisi tertinggi (hingga 100% pada Dataset B), sedangkan YOLOv11 secara konsisten unggul dalam kecepatan pemrosesan. Rata-rata waktu inferensi YOLOv11 pada Raspberry Pi 5 mencapai 341 ms, lebih cepat dibandingkan YOLOv8 (350 ms), dan pola ini berlanjut pada simulasi langsung. Dapat disimpulkan bahwa pemilihan model bergantung pada kebutuhan aplikasi: YOLOv8 cocok untuk akurasi deteksi maksimal, sementara YOLOv11 lebih sesuai untuk aplikasi yang menuntut kecepatan dan efisiensi komputasi waktu nyata (real-time).

Abstract

Background: Androgenic alopecia (AGA) is characterized by hair follicle miniaturization and chronic inflammation mediated by dihydrotestosterone (DHT). Current therapies primarily target hormonal pathways and do not adequately address inflammatory dysregulation. Exosomes derived from hypoxia-conditioned mesenchymal stem cells (EH-MSCs) exhibit enhanced immunomodulatory properties. However, their effects on key inflammatory mediators in AGA remain unclear. This study evaluated the effects of EH-MSCs on CXCL12 and IL-10 expression in a DHT-induced AGA mouse model.Materials and methods: Male C57BL/6 mice were allocated into five groups: healthy control, DHT-induced alopecia without treatment, DHT-induced alopecia treated with topical minoxidil, and EH-MSCs administered at 100 or 200 µL/kgBW. Alopecia was induced by subcutaneous DHT injection for 17 days. Following model validation, treatments were administered on days 25 and 32. CXCL12 and IL-10 expression in dorsal skin tissue was analyzed using RT-PCR on day 39.Results: DHT induction significantly increased CXCL12 expression and reduced IL-10 levels (p<0.05). EH-MSC administration dose-dependently downregulated CXCL12 (1.73±0.57 and 1.54±0.44 fold-change) and upregulated IL-10 expression (3.10±0.75 and 3.29±0.67 fold-change), demonstrating greater immunomodulatory effects compared with minoxidil.Conclusion: EH-MSCs effectively modulated inflammatory biomarkers by suppressing CXCL12 and enhancing IL-10 expression in a DHT-induced AGA model, suggesting their potential as an immunoregenerative therapeutic strategy for androgenic alopecia.Keywords: androgenic alopecia, mesenchymal stem cells, exosomes, hypoxia, CXCL12, IL-10

Abstract

Background: Ultraviolet-B (UV-B) radiation accelerates photoaging by disrupting extracellular matrix (ECM) homeostasis through dysregulation of mechanistic target of rapamycin complex 1 (mTORC1) and upregulation of matrix metalloproteinase-2 (MMP-2). This study evaluated the effects of exosomes of hypoxia-conditioned mesenchymal stem cells (EH-MSCs) on mTORC1 and MMP-2 expression in a UV-B–induced collagen loss rat model.Materials and Methods: Thirty male Wistar rats were randomized into five groups: healthy control, UV-B + saline, UV-B + hyaluronic acid, UV-B + 200 µL EH-MSCs, and UV-B + 300 µL EH-MSCs. Collagen loss was induced by UV-B irradiation for two weeks (10 sessions, 8 min/session). A single treatment was administered on day 22, and tissue was collected on day 29. Exosomes were isolated from hypoxia-conditioned MSCs and characterized by morphology and surface markers. Gene expression of mTORC1 and MMP-2 was assessed by qRT-PCR and analyzed using one-way ANOVA.Results: UV-B exposure induced collagen loss histologically. EH-MSCs significantly increased mTORC1 expression, highest in the 300 µL group (p < 0.001), and reduced MMP-2 expression, lowest in the 300 µL group.Conclusion: EH-MSCs exert dual regulatory effects by upregulating mTORC1 and suppressing MMP-2 in UV-B–induced collagen loss, suggesting therapeutic potential to mitigate photoaging via anabolic signaling (via mTORC1) and reduced ECM degradation (via MMP-2).Keywords: collagen loss, exosomes, MMP-2, mTORC1, UV-B

Abstract

Ultraviolet-B (UV-B) radiation-induced skin photoaging is marked by collagen degradation and chronic inflammation, with limited effective treatments currently available. Hypoxic-preconditioned mesenchymal stem cells (H-MSCs) offer a novel therapeutic approach due to their immunomodulatory capabilities. This study evaluated the effects of H-MSCs at two doses (2.5×105 and 5×105 cells) on UVB-induced collagen loss, focusing on CD68 expression and TNF-α levels in a rat model. Male Wistar rats were divided into five groups: healthy controls, UV-B-exposed negative controls (saline), positive controls (hyaluronic acid), and two H-MSC-treated groups. After UV-B exposure (160 mJ/cm², five times per week for two weeks), validated H-MSCs were administered subcutaneously. Collagen content was assessed histologically, while CD68 gene expression and TNF-α levels were measured by qRT-PCR and ELISA, respectively. UVB exposure led to significant reductions in collagen and increased levels of inflammatory markers. H-MSC treatment showed dose-dependent anti-inflammatory effects, with the higher dose (5×105 cells) optimally reducing CD68 expression and TNF-α levels, nearly matching healthy controls. These results suggest that H-MSCs, particularly at higher doses, may be a promising therapy for UVB-induced skin damage and collagen loss.

Abstract

Wound healing involves complex biological processes influenced by inflammation and tissue regeneration, where excessive interleukin-1 (IL-1) activity can delay recovery, while platelet-derived growth factor (PDGF) plays a pivotal role in fibroblast proliferation and collagen deposition. This study aimed to evaluate the effect of topical Noni (Morinda citrifolia L.) extract cream on IL-1 levels and PDGF expression in incised skin tissue. A post-test only control group design was conducted using 30 male Wistar rats randomly allocated into five groups: healthy control, negative control with basic cream, positive control with povidone iodine, treatment with 15% Noni cream, and treatment with 30% Noni cream. Incised wounds were created on the dorsal skin and treated once daily for seven days, followed by analysis of IL-1 using ELISA and PDGF expression using RT-qPCR. Statistical analysis showed no significant differences among groups (p=0.088 for IL-1; p=0.056 for PDGF), yet descriptive results revealed lower IL-1 levels in the 30% Noni group and higher PDGF expression in the 15% group, while povidone iodine produced the least favorable outcomes. These findings suggest a dose-dependent dual action of Noni extract cream, with the 30% formulation tending to attenuate inflammation and the 15% formulation enhancing regenerative activity.  These findings descriptively showed the lowest IL-1 levels, although the difference was not statistically significant. Within the limitations of this study, topical Noni extract cream demonstrates potential as a safe and natural adjunctive therapy for wound management, warranting further investigation with larger sample sizes, extended observation periods, and histopathological confirmation
