Abstract

ABSTRAK. Pelestarian isolat mikroba sangat penting untuk penelitian, diagnostik, dan bioteknologi. Metode konvensional seperti stok gliserol atau liofilisasi efektif namun seringkali mahal dan rentan terhadap risiko kontaminasi. Strategi alternatif penyimpanan berbasis butiran, memungkinkan pengambilan mikroorganisme yang masih hidup dengan mudah sambil meminimalkan siklus pembekuan-pencairan berulang. Studi ini bertujuan untuk mengevaluasi kinerja awal sistem GAMICBANK, metode penyimpanan mikroba berbasis butiran menggunakan butiran mikrobank plastik yang dimasukkan dalam larutan susu skim atau Skim Milk–Tryptone–Glucose–Glycerol (STGG), untuk menyimpan bakteri dan ragi pada suhu –80 °C. Isolat Gram-positif, Gram-negatif, dan ragi yang representatif (Staphylococcus aureus, Streptococcus spp., Listeria monocytogenes, Salmonella typhi, Shigella flexneri, Escherichia coli, Neisseria gonorrhoeae, Chromobacterium violaceum, dan Candida albicans) disimpan dalam butiran dengan susu skim atau STGG. Butiran tersebut diambil kembali setelah 1 dan 3 bulan, ditanam pada media selektif, dan dievaluasi pertumbuhan koloni dan viabilitasnya. Setelah 1 bulan, semua isolat tetap memiliki viabilitas yang sangat baik di kedua media. Pada 3 bulan, sebagian besar spesies tetap sangat viabel (>99%) di kedua media. Namun, Listeria monocytogenes dan Neisseria gonorrhoeae menunjukkan penurunan viabilitas yang signifikan dalam susu skim (30%) tetapi tetap stabil dalam STGG (99%). Demikian pula, Candida albicans menunjukkan penurunan viabilitas dalam susu skim (50%) tetapi mempertahankan 99% kelangsungan hidup dalam STGG. Sistem bead GAMICBANK secara efektif melestarikan mikroorganisme yang tangguh dan sensitif pada suhu –80 °C. STGG memberikan perlindungan beku yang lebih unggul dibandingkan susu skim, terutama untuk bakteri sensitif dan ragi. Kesimpulannya bahwa sistem GAMICBANK dapat dijadikan sebagai metode alternatif untuk penyimpanan isolat bakteri dan ragi.ABSTRACK.The preservation of microbial isolates is crucial for research, diagnostics, and biotechnology. Conventional methods, such as glycerol stocks or lyophilization, are effective but frequently expensive and susceptible to contamination risks. An alternative strategy, bead-based storage, facilitates the easy retrieval of viable microorganisms while minimizing repeated freeze-thaw cycles. This study aims to evaluate the initial performance of the GAMICBANK system, a bead-based microbial storage method utilizing plastic microbank beads submerged in either skim milk or Skim Milk–Tryptone–Glucose–Glycerol (STGG) solutions, for the cryopreservation of bacteria and yeast at –80 °C. Representative Gram-positive, Gram-negative, and yeast isolates (Staphylococcus aureus, Streptococcus spp., Listeria monocytogenes, Salmonella typhi, Shigella flexneri, Escherichia coli, Neisseria gonorrhoeae, Chromobacterium violaceum, and Candida albicans) were preserved in beads using either skim milk or STGG. The beads were retrieved after 1 and 3 months of storage, cultured on selective media, and evaluated for colony growth and viability. After 1 month, all isolates maintained excellent viability in both preservation media. At 3 months, the majority of the species remained highly viable (>99%) in both media. However, Listeria monocytogenes and Neisseria gonorrhoeae exhibited a significant decline in viability when stored in skim milk (30%) but remained stable in STGG (99%). Similarly, Candida albicans demonstrated a reduction in viability in skim milk (50%) while maintaining a 99% survival rate in STGG. The GAMICBANK bead system effectively preserves both robust and sensitive microorganisms at –80 °C. STGG provides superior cryoprotection compared to skim milk, particularly for sensitive bacteria and yeast. In conclusion, the GAMICBANK system serves as a viable alternative method for the preservation of bacterial and yeast isolates.

Abstract

Currently, Indonesia is implementing a new paradigm curriculum, namely the PSP (Program Sekolah Penggerak) curriculum (PSP) in junior high schools, particularly among English teachers, amid Indonesia’s ongoing curriculum reform efforts. Despite its implementation, the PSP curriculum has generated considerable concern, particularly regarding teachers’ capacity to adapt to its demands. However, in implementing this curriculum, there is a lot of attention from various parties, especially teachers. The purpose of the study is to analyze the difficulties faced by English teachers in implementing the PSP (Program Sekolah Penggerak) curriculum. The research only focused on junior high school teachers’ difficulties in implementing the PSP (Program Sekolah Penggerak) curriculum. The researcher limits the data of the study to teachers. This study employed the qualitative method as it was a more comprehensive account of the phenomenon under study would be presented. The research used a case study design. The participant consisted of two English teachers participants as the object of research. The instrument for collecting data was an interview. The type of interview used in this research is the semi-structured interview. To analyze the result of interviews this study used data reduction, data display, and conclusion. The first was the teacher’s understanding of the PSP curriculum. The second was the difficulties faced by English teachers. First, the difficulties faced were in terms of providing materials and teaching methods. Second, the teacher had difficulty managing time. Third, the difficulties faced by English teachers were the teaching devices. Fourth, the PSP curriculum was not implemented effectively and teachers had to be ready. The third was the advantages and disadvantages. The fourth was the differences between the PSP curriculum and the 2013 curriculum. The result of this study indicated that the teachers faced difficulties in providing materials and teaching methods, and had difficulty managing time, the teaching devices, effectiveness, and readiness.

Abstract

This article discusses the development of literary appreciation learning models in higher education, emphasizing systematic, creative, and collaborative approaches. The aim is to identify strategies that enhance students’ interest, comprehension, and engagement in appreciating literary works. The ADDIE model serves as the primary framework, combined with creative-innovative and collaborative methods. Findings suggest that integrating theory, practice, and digital technology into literary appreciation learning significantly improves students’ cognitive, affective, and evaluative abilities.

Abstract

Purpose of the study: This research aims to find out about the Suro one night tradition, especially the earth almsgiving and its influence on the socio-religious life of the community. Methodology: This research adopts a quantitative approach with a phenomenological focus. In this research, the data collection techniques used involved interview and observation instruments. In detailing the findings from data collection, this research applies Milles and Huberman data analysis techniques. Main Findings: The results of observations show that the Suro first night tradition, especially in the context of earth almsgiving, has a significant impact on the social and religious life of the community. From the results of interviews with participants, it was revealed that the Suro first night tradition, especially the earth almsgiving, has a deep meaning for the community. Novelty/Originality of this study: One Suro Night Tradition, deeply rooted in local culture, serves as a unique catalyst in shaping the social and religious fabric of communities. Its profound influence extends beyond traditional practices, resonating within madrasahs, islamic boarding school, and public schools, fostering a harmonious convergence of cultural heritage and modern education.

Abstract

Pemanfaatan limbah pertanian seperti daun pala menjadi produk bernilai tambah merupakan strategi krusial dalam mendukung konsep green economy, namun pelaku UMKM sering kali terhambat oleh lemahnya manajemen keuangan. Penelitian melalui kegiatan pengabdian ini berfokus pada perumusan model akuntansi biaya pada UMKM Penyulingan Daun Pala Feryandi yang menghadapi kendala pencatatan manual dan ketidakakuratan penetapan harga jual. Tahapan penelitian dilaksanakan menggunakan pendekatan partisipatif yang meliputi survei awal, persiapan modul edukatif, serta pendampingan intensif mengenai perhitungan Harga Pokok Produksi (HPP) dan analisis Break Even Point (BEP). Hasil pengabdian menunjukkan peningkatan signifikan pada kapasitas manajerial mitra, di mana pelaku usaha kini mampu memisahkan keuangan pribadi dari bisnis serta mengintegrasikan biaya bahan baku, tenaga kerja, dan overhead secara menyeluruh. Temuan lapangan mengindikasikan bahwa pemahaman terhadap HPP dan strategi pemasaran digital secara efektif membantu UMKM mengoptimalkan margin keuntungan sekaligus mengurangi dampak lingkungan melalui pengolahan limbah menjadi minyak atsiri. Simpulan utama penelitian menegaskan bahwa integrasi akuntansi biaya dengan prinsip ekonomi hijau menciptakan model pengelolaan usaha yang sistematis dan berkelanjutan. Implementasi model ini tidak hanya meningkatkan efisiensi operasional dan akurasi pengambilan keputusan strategis, tetapi juga memperkuat daya saing produk lokal di pasar digital dalam jangka panjang demi mewujudkan pembangunan ekonomi berkelanjutan yang ramah lingkungan bagi masyarakat secara berkesinambungan. ABSTRACT Utilizing agricultural waste such as nutmeg leaves into value-added products is a crucial strategy in supporting the green economy concept, but MSMEs are often hampered by weak financial management. This community service research focused on formulating a cost accounting model for the Feryandi Nutmeg Leaf Distillery MSME, which faces obstacles such as manual recording and inaccurate selling prices. The research phase was implemented using a participatory approach, including an initial survey, preparation of educational modules, and intensive mentoring on calculating the Cost of Goods Sold (COGS) and analyzing the Break Even Point (BEP). The community service results showed a significant increase in the partners' managerial capacity, with entrepreneurs now able to separate personal finances from business finances and comprehensively integrate raw material, labor, and overhead costs. Field findings indicate that understanding COGS and digital marketing strategies effectively helps MSMEs optimize profit margins while reducing environmental impacts through processing waste into essential oils. The main conclusion of the research confirms that integrating cost accounting with green economy principles creates a systematic and sustainable business management model. Implementing this model not only improves operational efficiency and the accuracy of strategic decision-making, but also strengthens the competitiveness of local products in the digital market in the long term to realize sustainable economic development that is environmentally friendly for the community in a sustainable manner.
