Abstract

Milk consumption during adolescence is considered an early means of preventing osteoporosis in adults. Osteoporosis is a systemic skeletal disease characterised by low bone density and microarchitectural deterioration of bone tissues, with a consequent increase in bone fragility and suspectibility to fracture. Augmenting bone mass during adolescence has been suggested as a strategy to prevent osteoporosis, because adolescents may represent the final opportunity for substantially increasing bone mass before skeletal consolidation. The purpose of this study was to determine effects of fresh and high calcium milk on blood calcium concentration and bone density. Variables measured in this study were blood calcium concentration, bone density of spine and bone density of whole body. The study using 55 adolescent boys that had 17 to 19 years old. The design of this study is nested randomized design with two factors are kind of mil (fresh milk, high calcium milk) and volume of each kind of milk (250 ml, 500 ml, 750 ml). Results of the study indicated that fresh and high calcium milk in this research not significant effects (P>0.05) on blood calcium concentration and bone density of whole body. But, high calcium milk consumption was able to increase bone density. It was found that high calcium consumption showed very highly significant effect (P<0.01) on bone density of spine with 1.79% contributions. Based on general linear model equation, bone density of trunk can be estimated by high calcium milk consumption. Keywords : milk consumption, adolescence, blood calcium concentration, bone density

Abstract

Background: Uncontrolled diabetes can lead to macrovascular and microvascular complications in various organs. Organ fibrosis is one of the microvascular complications due to diabetes. Hyperglycemia triggers ROS formation thereby causing oxidative stress. Oxidative stress enhances pro-inflammatory and pro-fibrotic activities. The lungs have a lot of vascularization and connective tissue, making them susceptible to diabetes complications. Moringa is called 'The Miracle Tree' because it has many properties. Moringa leaves are the most studied part, for their efficacy as antioxidants. This study examines the effect of moringa leaf extract (MLE) in inhibiting lung fibrosis in diabetic Wistar rats. Methods: A total of 27 male Wistar rats were randomized into three groups, K0, K1 and P. Group K0 was given saline (ip) and saline (orally); K1 was given Streptozotocin (STZ) (ip) and saline (orally); P was given STZ (ip) and MLE (orally). MLE was administered daily at 1000 mg/Kg BW dose for four weeks. Lung fibrosis assessment was carried out using a modified Ashcroft scale. Results: Lung fibrosis assessment showed that the K1 group had the highest mean of lung fibrosis and K0 had the lowest mean of lung fibrosis. Kruskal-Wallis test showed that there was a significant difference in the data comparison between the three groups. The post-hoc test showed that there was a significant difference in the data comparison between groups. Comparison of the K0-K1 group data showed the smallest significance value and comparison of the K0-P group data showed the highest significance value. Conclusion: MLE can inhibit lung fibrosis in diabetic Wistar rats.

Abstract

Cardiac fibrosis remains as the leading cause of death worldwide and is often associated with elevated levels of transforming growth factor-β 1 (TGF-β1) and galectin-3, making them potential therapeutic targets. Recent studies revealed that quercetin, myricetin, and kaempferol have the biological effect for several cardiovascular diseases. However, the investigation into this topic through molecular models and analysis remain unexplored. The aim of this study was to evaluate the potential effect of quercetin, myricetin, and kaempferol which targeted TGF-β1 and galectin-3. In this study, quercetin, myricetin, and kaempferol roled as the tested ligands. Subsequently, colchicine and native ligand acted as control ligands that were screened through molecular docking against TGF-β1 and galectin-3 using AutoDock tools to identify the potential inhibitor. The stability of ligand-receptor complexes was assessed through molecular dynamic (MD) simulations using NMAD. Absorption, Distribution, Metabolism, Excretion and toxicity (ADMET) prediction were also performed using ADMETlab 2.0. Molecular docking analysis revealed that quercetin, myricetin, and kaempferol exhibited strong binding affinity which are -8.9 kcal/mol, -8.5 kcal/mol, -7.6 kcal/mol respectively with TGF-β1, and -7.5 kcal/mol, -7.0 kcal/mol, -5.7 kcal/mol respectively with galetcin-3; low inhibition constant (Ki); and stable interaction with the active sites of TGF-β1 and galectin-3. MD simulations confirmed the stability and compactness of the ligand-receptor complexes. ADMET analysis also showed high Plasma Protein Binding (PPB) values (quercetin: 95%, myricetin: 92%, and kaempferol: 97%) and moderate clearance values (quercetin: 8.284%, myricetin, and 7.716%, kaempferol: 6.868%) for the tested compounds. In conclusion, the in silico analyses suggested that quercetin, myricetin, and kaempferol are promising for cardiac fibrosis therapies by inhibiting TGF-β1 and galectin-3.

Abstract

Background: In acute coronary syndrome (ACS), antiplatelet therapy is crucial for inhibiting platelet aggregation. Dual antiplatelet therapy (DAPT) commonly employs aspirin along with clopidogrel, prasugrel, or ticagrelor. This is well known that aspirin acts as a cyclooxygenase (COX)-1 inhibitor, while clopidogrel, prasugrel, and ticagrelor act as P2Y12 inhibitors. Despite DAPT's proven efficacy in more effectively reducing cardiovascular events in ACS patients, this is associated with an increased risk of bleeding compared to mono antiplatelet therapy (MAPT). To minimize the cost and side effect that might arise from the use of DAPT, this is necessary to assess the potential of MAPT using a P2Y12 inhibitor drug, to understand whether they are capable of binding to both COX-1 and P2Y12. Hence, this study was conducted to identify P2Y12 inhibitor drugs that have the ability to bind to COX-1, allowing them to be proposed as MAPT.Materials and methods: Molecular docking was employed to assess binding affinity, interaction types, amino acid residues, binding distances, and visualizations in both 3D and 2D formats. The applications utilized were BIOVIA Discovery Studio and AutoDock, while the websites utilized were research collaboratory for structural bioinformatics protein data bank (RCSB PDB) and PubChem. Results: In silico findings reveal differences in binding strength among clopidogrel, prasugrel, and ticagrelor to COX-1 and P2Y12, with ticagrelor emerging as the stronger ligand due to a higher number of bindings and/or closer binding distances. Notably, only prasugrel and ticagrelor demonstrate the ability to bind to the active site of COX-1.Conclusion: Therefore, prasugrel and ticagrelor emerge as potential MAPT agents for ACS patients. Keywords: clopidogrel, prasugrel, ticagrelor, antiplatelet, ACS, in silico, molecular docking

Abstract

This study aims to examine the implementation of character building at MTsN 6 Sragen, as well as the supporting and inhibiting factors in the process. The background of this research stems from the importance of character education in shaping students with integrity, honesty, discipline, and responsibility. In an era of globalization filled with moral challenges, character education becomes a vital aspect that must be implemented in schools. This research employed a qualitative method with a descriptive approach. Data were collected through interviews, observations, and documentation involving teachers, students, and school staff at MTsN 6 Sragen. The research findings indicate that the implementation of character building at MTsN 6 Sragen has been effective, although some challenges remain, such as limited teaching time and minimal parental involvement. Other findings show that students participating in this program tend to exhibit better discipline and responsibility. The main supporting factors include the commitment of teachers in running the program and a conducive school environment. However, social media influence and lack of family engagement present the primary challenges. The study implies that the success of character education in schools requires strong collaboration between schools, families, and communities.
