Abstract

Background: In acute coronary syndrome (ACS), antiplatelet therapy is crucial for inhibiting platelet aggregation. Dual antiplatelet therapy (DAPT) commonly employs aspirin along with clopidogrel, prasugrel, or ticagrelor. This is well known that aspirin acts as a cyclooxygenase (COX)-1 inhibitor, while clopidogrel, prasugrel, and ticagrelor act as P2Y12 inhibitors. Despite DAPT's proven efficacy in more effectively reducing cardiovascular events in ACS patients, this is associated with an increased risk of bleeding compared to mono antiplatelet therapy (MAPT). To minimize the cost and side effect that might arise from the use of DAPT, this is necessary to assess the potential of MAPT using a P2Y12 inhibitor drug, to understand whether they are capable of binding to both COX-1 and P2Y12. Hence, this study was conducted to identify P2Y12 inhibitor drugs that have the ability to bind to COX-1, allowing them to be proposed as MAPT.Materials and methods: Molecular docking was employed to assess binding affinity, interaction types, amino acid residues, binding distances, and visualizations in both 3D and 2D formats. The applications utilized were BIOVIA Discovery Studio and AutoDock, while the websites utilized were research collaboratory for structural bioinformatics protein data bank (RCSB PDB) and PubChem. Results: In silico findings reveal differences in binding strength among clopidogrel, prasugrel, and ticagrelor to COX-1 and P2Y12, with ticagrelor emerging as the stronger ligand due to a higher number of bindings and/or closer binding distances. Notably, only prasugrel and ticagrelor demonstrate the ability to bind to the active site of COX-1.Conclusion: Therefore, prasugrel and ticagrelor emerge as potential MAPT agents for ACS patients. Keywords: clopidogrel, prasugrel, ticagrelor, antiplatelet, ACS, in silico, molecular docking

Abstract

The North Coast of Central Java is one of the potential areas for small pelagic fisheries. Estimation of fishing areas can be facilitated by utilizing geographic information technology (GIS) in determining potential fishing areas. This study aims to determine potential areas for small pelagic fishing based on sea surface temperature (SST), chlorophyll-a, and current variables. The study was conducted in the North Coast of Central Java using data throughout 2023. This study uses spatial modeling techniques. Sea surface temperature (SST) and chlorophyll-a were obtained from Aqua MODIS imagery, while current data was obtained from AVISO. Catch data is secondary data obtained from the Pekalongan Nusantara Fisheries Port. The results of the study showed that the average value of sea surface temperature ranged from 28.34-30.73⁰C, the value of chlorophyll-a concentration ranged from 0.33-0.76 mg/m³, and the current speed ranged from 0.14-0.28 m/s indicating that sea surface temperature (SST), chlorophyll-a, and currents affect the determination of small pelagic fish catch areas. The regression correlation analysis showed a positive regression correlation result for sea surface temperature and fish catches, conversely chlorophyll-a and current speed were negatively correlated with small pelagic fish catches. The potential areas for small pelagic fishing in the North Waters of Central Java in 2023 showed a varied distribution in each season. The area with the highest frequency of distribution of small pelagic fishing areas occurred in the transition season I, while the East season showed fewer fishing areas.

Abstract

Pati regency is a coastal area that contributes significantly to Indonesia’s fisheries sector. One if its major fishing ports, PPP Bajomulyo, serves as a hub for fishing activities, with catches ranging from small to large pelagic fish, including Spanish mackerel (Scomberomorus commerson). This study aims to examine to relationship between oceanographic parameters and Spanish mackerel catch volumes at PPP Bajomulyo over the 2021-2023 period. A survey-based approach incorporating satellite image overlay was employed. The analysis of non-linier relationships between variables was conducted using Generalized Additive Model (GAM), implemented in RStudio. Satellite image processing was performed using SeaDAS, while spatial analysis was carried out in ArcGIS. The findings reveal that the interaction between sea surface temperature (SST) and chlorophyll-a concentrations accounted for 11,44% oof catch variability. Elevated SST above 29,5 °C were associated with increased catch rates, whereas extreme temperature (>35 °C or <28 °C) corresponded with significant declines. Chlorophyll-a exerted a relativity weak influence on catch, suggesting that although it serves as a proxy for primary productivity, Spanish mackerel distribution is more strongly governed by thermal conditions and prey availability. The study further identified that optimal fishing grounds were predominantly located in northern Central Java waters, particularly during seasons characterized by favorable oceanographic conditions.
