Abstract

The aim of this research was to show the influence of seminiferous tubules histopathology of mice (Mus musculus) caused Fusarium graminearumexposure. This research was done in April-May 2017 in Microbilogy Laboratory Faculty of Science and Technology, Animal Laboratory, and Microbilogy Laboratory Faculty of Veterinary Medicine UniversitasAirlangga. This research used 20 male mice (Mus musculus) aged 6 weeks with 18-30 gram body weight. The mice devided into four groups: P0 given 0,25 ml Sodium chloride without Fusarium graminearum exposure by oral; P1 given 0,25 ml Fusarium graminearum exposure with dilution 102 by oral; P2 given 0,25 ml Fusarium graminearum exposure with dilution 103 by oral; and P3 given 0,25 ml Fusarium graminearum exposure with dilution 104 by oral. This treatment carried out for 21 days. Each milliliter dilution containing 228x106 spore for P1, 228x107 spore for P2, and 228x108 spore for P3. Then do the surgery and harvesting the testes then performed histopathological examination by scoring of seminiferous tubules. For data analyzing used non parametric difference Kruskall-Wallis and continued with Mann-Whitney. The result of this research was showed that decreased the spermatogenic cells in the process of spermatogenesis significant.

Abstract

The purpose of this study was to prove that the provision of pare leaf extract can reduce the process of follicular atrophy in the ovaries caused by hyperglycemic conditions. The total of this study was completed within 20 days, including the induction of Streptozotocin for 5 days, treatment for 14 days, and 1 day for ovarian collection. The K- group as a negative control without being induced by Streptozotocin and received no treatment. The K+ group as a positive control was only induced by Streptozotocin at a dose of 55 mg / Kg BW and D10 without treatment. The T1 group was induced by Streptozotocin at doses of 55 mg / Kg BW and D10 and treated with pare leaves extract (Momordica charantia) at a dose of 1.26 ml / Kg BW. T2 group was induced by Streptozotocin with dose of 55 mg / Kg BW and D10 and was treated with pare leaves extract (Momordica charantia) at a dose of 2.52 ml / Kg BW. Induction of Streptozotocin intraperitoneally and once-daily peroral treatment. The result showed that treatment group could make a significant difference with the positive control. Showing that amount of gap could make Momordica charantia to be alternative treatment for hyperglycemic condition.

Abstract

This study investigated the preventive effect of virgin coconut oil (VCO) on the epithelium thickness and the diameter of the seminiferous tubules induced by ethanol in mice (Mus musculus). This study used 20 male mice as the experimental animal which were divided into five groups with four mice in each group. Negative control (C-) was given 2% Tween and aquadest, while positive control (C+) was given 2% Tween and 33% ethanol. T1, T2, and T3 were respectively given 0.09, 0.19, and 0.37 mL/kg bw VCO and 33% ethanol (0.2 mL/kg bw). VCO was given orally for 39 days, and ethanol was given orally seven days later for 32 days. Ethanol was administered two hours after the VCO administration. Analysis of variance followed by Tukey's range test on the epithelium thickness and the diameter of the seminiferous tubules showed significant differences (p <0.05) of C+ group from the other groups. Whereas, there was no significant difference (p >0.05) was found among C-, T1, T2 and T3 group. The result concluded that VCO could protect the testis of mice from the damage caused by ethanol.

Abstract

This research aims to determine the effect of Moringa oleifera leaf extract (MLE) on the diameter and epithelium thickness of the seminiferous tubules of rats induced with gentamicin. This study used 25 male rats (Rattus norvegicus) aged 8-12 weeks with 150-200 grams body weight. Negative control group (C-) was injected with aquadest and given 0.5% Na-CMC orally, positive control group (C+) was induced with gentamicin injection of 5 mg/kg BW/day and given 0.5% Na-CMC orally, while groups T1, T2, and T3 were induced with gentamicin at the same dose and treated orally with MLE of 200, 316, 500 mg/kg BW/day. Analysis of variance showed significant differences and further analysis using Duncan Multiple Range Test showed difference in each group. The results showed that treatment with MLE of 500 mg/kg BW/day improved the diameter and epithelium thickness of the seminiferous tubules of rats injected with gentamicin. It could be concluded that MLE restored the diameter and epithelium thickness of the seminiferous tubules of rats induced with gentamicin.

Abstract

This study aimed to determine the effect of taurine on the enhancement of the spermatogenetic process in male mice (Mus musculus) induced by paraquat (PQ). Twenty-five male mice (Mus musculus) aged 2-3 months with a bodyweight of around 35 grams were divided randomly into five groups. The K + and the treatment group (P1, P2, and P3) mice were induced using PQ. PQ was given intraperitoneally (IP) twice a week for 21 consecutive days at a dose of 30 mg/kg BW. Two hours after the administration of PQ, P1, P2, and P3 groups were given taurine at a dose of 250, 500, and 1000 mg/kg BW/day for three weeks (Heidari et al., 2019). K- group was given distilled water (IP) only. On day-29, mice were sacrificed for testicles histopathological preparations with hematoxylin-eosin staining. Results showed that the mice exposed to PQ only (the K+ group) had a reduced spermatogenesis score compared to those of the K- group (p <0.05).  Taurine treatment on PQ-exposed mice was followed by an increase spermatogenesis score. The optimal curative dose of taurine was 500 mg/kg (P2 group). However, a higher dose (1000 mg/kg BW) of taurine resulted in a decline in the spermatogenesis score than those of at the 500 mg/kg. It could be concluded that treatment with taurine could enhance the spermatogenetic process of male mice (Mus musculus) induced by PQ.

Abstract

Predicting the turtle's reproductive status and sexual maturity is necessary for ex-situ breeding and conservation program. The histology of the seminiferous tubules of the Indonesian box turtle (Cuora amboinensis couro) at various ages has not been studied much. This study aims to develop equations to predict the sexual maturity of Indonesian box turtles based on straight carapace length (SCL). Six turtles with SCL sizes 10-21 cm were sacrificed and their testes were taken for morphometric measurements and histological preparations for Hematoxylin Eosin staining. Pearson's correlation of SCL with testicular maturity parameters was analyzed, followed by Anova Regression. The results showed that there were differences in the morphometry and topographic anatomy of the Indonesian box turtle testes between SCL below 15 cm and above 15 cm. Six turtles with SCL size of 10-21 cm were sacrificed and their testes were taken for morphometric measurements and histological preparations for Hematoxylin Eosin staining. Pearson's correlation of SCL with testicular maturity parameters was analyzed, followed by Regression Anova. The results showed that there were differences in the morphometry and topographic anatomy of the Indonesian box turtle testes between SCL below 15 cm and above 15 cm.

Abstract

This study aims to evaluate the expression of BAX, BCL-2, and BAX/BCL-2 ratio in maturation media of cow oocytes which supplemented with Urea in vitro because BAX and BCL-2 are the main regulators of apoptosis.A total of 263 oocytes from follicle aspirations originating from ovaries taken from slaughterhouses and were saturated with 3 addition of urea which was divided into three groups. The control group (P0) was control group without the addition of urea, P1 group was added with urea 20 mg/dL, while P2 group was added with urea 40 mg/dL.The results of in vitro oocyte maturation were continued with identification using immunocytochemical staining with the addition of BAX and BCL-2 antibodies.Positive results showed a brownish color on the oocyte and its cumulus.The results of this study indicated that there were significant differences (P0.05) in BAX and BCL-2 expression, although both curves were equally increase. The increase in BCL-2 was more significant than BAX, while the BAX BCL-2 ratio did not show a significant difference (P0.05) in whichthe curve of BAX/BCL-2 ratio was decreased.It can be concluded that the addition of urea does not affect the level of apoptosis.

Abstract

This study aimed to determine the occurences of mice brain ventricles dilatation that congenitally infected with Toxoplasma gondii (T. gondii) as a marker of hydrocephalus and cellular changes in the brain. A total of twenty pregnant mice (11.5 days pregnacy) were divided into 2 groups, which were control (P1) group and treatment (P2) group. The mice in the treatment group were infected with 101 tachyzoites of T. gondii. All mice were maintained until delivery. The newborn mice were sacrificed and their brain were removed and fixed in 10% buffered formalin to prepare histology slides with HE staining for observation of ventricular width, TUNEL assay for apoptosis observation, and immunohistochemistry for the expression of transforming growth factor beta (TGF-) observations. The data were analyzed using t test and linear regression. The results showed that ventricular width and apoptosis index significantly increased (P0.01) in the treatment group compared to control group, but there was no difference in the expression of TGF- (P0.05) in both groups. Dilatation of ventricle correlated with the apoptotic index of brain cells but did not correlated with the expression of TGF-.

Abstract

Background: Cigarettes are tobacco products processed with or without additives and are known to contain addictive substances that pose serious health risks to individuals and the public. Inhaled cigarette smoke directly stimulates the activation of macrophage cells in the respiratory tract. Among the affected cells, type II alveolar cells which produce pulmonary surfactant to reduce alveolar surface tension are particularly vulnerable to smoke-induced damage. Purpose:  This study aimed to compare the effects of conventional cigarette smoke and e-cigarette vapor exposure on the number of alveolar macrophages and type II alveolar cells in male mice (Mus musculus). Method: A total of 28 male mice were randomly divided into seven groups. The control group (C) was exposed to ambient air only. Groups A1, A2, and A3 were exposed to 1, 2, and 3 conventional cigarettes per day, respectively, while groups B1, B2, and B3 were exposed to 0.7 mL, 1.3 mL, and 2 mL of e-liquid vapor per day, respectively. All exposures were conducted for 36 consecutive days. Results: A significant difference (p<0.05) was observed in the number of alveolar macrophages among groups C, A1, A2 , A3 , B2 , and B3 , whereas no significant difference was found between A1 and B1. The number of type II alveolar cells significantly differed (p < 0.05) across all groups: C, A1, A2, A3, B1, B2, and B3. Conclusion: Both conventional cigarette smoke and e-cigarette vapor increase alveolar macrophage counts and reduce type II alveolar cell numbers compared to controls. Although both exposures pose health risks, e-cigarette vapor appears to cause milder pulmonary effects than conventional cigarette smoke.
