Abstract

This study aims to evaluate the expression of BAX, BCL-2, and BAX/BCL-2 ratio in maturation media of cow oocytes which supplemented with Urea in vitro because BAX and BCL-2 are the main regulators of apoptosis. A total of 263 oocytes from follicle aspirations originating from ovaries taken from slaughterhouses and were saturated with 3 addition of urea which was divided into three groups. The control group (P0) was control group without the addition of urea, P1 group was added with urea 20 mg/dL, while P2 group was added with urea 40 mg/dL. The results of in vitro oocyte maturation were continued with identification using immunocytochemical staining with the addition of BAX and BCL-2 antibodies. Positive results showed a brownish color on the oocyte and its cumulus. The results of this study indicated that there were significant differences (P0.05) in BAX and BCL-2 expression, although both curves were equally increase. The increase in BCL-2 was more significant than BAX, while the BAX BCL-2 ratio did not show a significant difference (P0.05) in whichthe curve of BAX/BCL-2 ratio was decreased. It can be concluded that the addition of urea does not affect the level of apoptosis.

Abstract

This study aimed to determine the occurences of mice brain ventricles dilatation that congenitally infected with Toxoplasma gondii (T. gondii) as a marker of hydrocephalus and cellular changes in the brain. A total of twenty pregnant mice (11.5 days pregnacy) were divided into 2 groups, which were control (P1) group and treatment (P2) group. The mice in the treatment group were infected with 101 tachyzoites of T. gondii. All mice were maintained until delivery. The newborn mice were sacrificed and their brain were removed and fixed in 10% buffered formalin to prepare histology slides with HE staining for observation of ventricular width, TUNEL assay for apoptosis observation, and immunohistochemistry for the expression of transforming growth factor beta (TGF-β) observations. The data were analyzed using t test and linear regression. The results showed that ventricular width and apoptosis index significantly increased (P0.01) in the treatment group compared to control group, but there was no difference in the expression of TGF-β (P0.05) in both groups. Dilatation of ventricle correlated with the apoptotic index of brain cells but did not correlated with the expression of TGF-β.

Abstract

Pigs have potentially to transmit zoonotic gastrointestinal parasite disease both caused by protozoa and worm. The aim of this study was to identify gastrointestinal parasites that were potentially zoonotic in pigs in the province of Bali. A total of 100 fresh feces samples was collected from several pig farms in Bali, from Badung and Tabanan districts, each consisted of 50 samples. Pig feces samples were examined for the presence of eggs worms, cysts and oocysts for protozoa based on the morphology and size. Identification for protozoa and worms used native, sedimentation and sucrose flotation methods. Parameters measured were sex, feed and cage management. The result showed that the characteristic parameters for pigs in both district were generally female. Cage management for raising pigs mostly used group cage. Feed that provided in both district mostly used bran and concentrate. All of 100 pig feces samples that examined positive for parasites. There were 8 types of gastrointestinal parasites that have been identified. Four types of protozoa found were Entamoeba sp. (99%), Balantidium sp. (79%), Eimeria sp. (78%), Blastocystis sp. (69%) and four types of worms were Ascaris sp. (20%), Trichuris sp. (20%), Strongyloides sp. (19%), and Oesophagostomum sp. (8%). All pigs were infected with two or more parasites. The prevalence of parasitic gastrointestinal infections was different for each district, six genera (Entamoeba sp., Balantidium sp., Blastocystis sp., Eimeria sp., Oesophagostomum sp. and Trichuris sp.)  were higher found in Tabanan district and the two genera (Ascaris sp. and Strongyloides sp.) were higher in Badung district. Oesophagostomum sp. was only found to infect pigs in Tabanan district. The conclusion is gastrointestinal parasites that found in pigs at Badung and Tabanan district Bali Province mostly have zoonotic potential.

Abstract

Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri daun sirsak terhadap Staphylococcus aureus. Ekstraksi diproses dengan metode maserasi terlebih dahulu dengan n-heksana kemudian kloroform. Aktivitas antibakteri diukur secara in vitro dengan menggunakan metode difusi agar menggunakan paperdisk. Aktivitas antibakteri diuji dengan analisis variansi (ANOVA) dengan nilai signifikansi 0,5% untuk mengetahui pengobatan mana yang memiliki efek atau berbeda secara signifikan dengan dosis masing-masing 300 mg/ml, 250 mg/ml, 200 mg/ml, 150 mg/ml, 100 mg/ml dan menghasilkan aktivitas antibakteri dengan zona hambat berturut-turut 16,70 mm; 14,05 mm; 11,45 mm; 9,85 mm; 3,00. Hasil pemeriksaan aktivitas antibakteri menunjukkan ekstrak n-heksana dan kloroform dapat menghambat pertumbuhan Staphylococcus aureus pada konsentrasi 250 mg / ml dengan diameter zona hambat 14,05 mm. Ekstrak dengan aktivitas tertinggi ditentukan konsentrasi hambat tumbuh minimum (MIC). Ekstrak kloroform daun sirsak dari bakteri Staphylococcus aureus yang berada pada konsentrasi 100 mg/ml dengan zona hambat 3,00 mm.

Abstract

Biawak air berjenis kelamin betina, berusia tiga bulan dengan berat badan 1.2 kg diambil dari suatu penangkaran, mengalami kelumpuhan selama tiga hari dan didiagnosa menderita metabolic bone disease. Eutanasia dan nekropsi dilakukan untuk mengamati gambar mikroskopis dari jaringan tulang. Sampel tulang diambil dari tujuh tulang ekstremitas yaitu os scapula, humerus, radius, ulna, femur, tibia, dan fibula. Pengamatan mikroskopis menunjukkan aktivitas sel osteoklas yang berlebihan dalam jaringan kompak, jaringan tulang rawan abnormal berada di antara jaringan tulang kompak dewasa, serta beberapa jaringan tulang rawan yang mengandung jaringan lemak. Hal ini menyebabkan tulang kompak kehilangan kepadatan tulang dan membuat struktur tulang biawak air menjadi sulit untuk regenerasi dengan baik sehingga pertumbuhan tulang pada biawak air menjadi terhambat. Hal ini menyebabkan biawak air kesulitan untuk berjalan atau mengalami kelumpuhan. Pemeriksaan kalsium dan fosfor dalam darah ditemukan ketidakseimbangan.

Abstract

The aim of this study was examine the effect of robusta coffee in bali cattle semen. A total of six cattles cement were given four treatments each P0 without adding Robusta coffee extract, P1 was given an additional 5%, P2 10%, and P3 15%. The results are observed by assessing motility, viability, and abnormalities. The data obtained were analyzed by Analysis of Variance (ANOVA) and continued with the duncan. Based on the analysis of variance in obtaining the results of the study, it was shown motility of P0 was significantly different from P1, P2, and P3 (p<0.05). Viability of P0 (control) different significantly from P1 , P2, and P3 (p<0.05). P3 motility different significantly from P0, P1, and P2 (p<0.05). Addition of robusta coffee extract of 10% and 15% decreased spermatozoa quality.

Abstract

Monosodium glutamate (MSG) is a sodium salt from glutamic acid which is currently very popular to be used as a food flavoring ingredient to stimulate appetite. Exessive consumption of MSG can disrupt the balance of antioxidants and ROS, and cause the negative effects of oxidative stress on testes. Coffee has high chlorogenic acid, function for antioxidants and reduce the negative effect on cell damage in the testes. This study aims to analyze the effect of robusta coffee extract on histological of mice testes induced with MSG. This study was completely randomized design (CRD), using 20 male mice for 5 treatment groups: K+ (MSG 0.12 mg and Vitamin C 6 mg), K- (MSG 0.12 mg and CMC Na 0.1 ml), P1 (MSG 0.12 mg and RCE 0.1 mg), P2 (MSG 0.12 mg and RCE 0.2 mg), and P3 (MSG 0.12 mg and RCE 0.4 mg) orally for 42 days. Histophatological scores were analyzed with Saphiro-Wilk Test and Anova. There is no effect of increasing testicular weight. 0,12 mg MSG can cause a decrease in the number of spermatogonia and spermatocytes. 0,2 mg robusta coffee extract can maintain the number of spermatogonia and spermatocytes.

Abstract

The purpose of this study was to determine the risk factors and incidence of Feline Lower Urinary Tract Disease (FLUTD) at the Sahabat Satwa Genteng clinic. A total of 36 samples were evaluated according to research procedures with diagnosis, data collection, urine sampling, dipstick and sediment examination methods, and Chi-square data analysis. The results showed that there was a association between gender, history of castration, and diet on the risk of FLUTD. Meanwhile, there was no association between age and the risk of FLUTD. Clinical signs that often occur in FLUTD disease initiate from stranguria, hematuria, dysuria, periuria, and polykisuria. The results of the dipstick examination founded that 86,1% of cats affected by FLUTD had a normal pH and 13,9% had an alkaline pH. The results of the specific gravity (SG) of cats with FLUTD have values below normal while the protein levels found in all cats suffering from FLUTD have values above normal. Stranguria was found in 69,4% of cats with FLUTD. Leukocytes were also found in all the urine of cats suffering from FLUTD. The most common results of sediment examination were struvite, calcium oxalate, and cystine crystals.
