Abstract

BACKGROUND: Coal dust is known to trigger hypersensitivity and inflammation of the respiratory tract as it increases oxidative stress leading to asthma. To date, the relationship of coal dust exposure in the pathomechanism of asthma remains unclear. This study was aimed to examine the effect of coal dust exposure on the superoxide dismutase (SOD) activity and the oxidative DNA damage indicated by increased serum 8-hydroxy-2' -deoxyguanosine (8-OHdG) in asthmatic mice.METHODS: Twenty-four female balb/c mice were divided into four groups. The first group was the control group. The second group was the negative control group which composed of mice exposed to coal dust particles. The third group was composed of ovalbumin (OVA)-sensitized mice. The fourth group was composed of OVA-sensitized mice and exposed to coal dust particles. The inflammatory process was identified by serum interleukin (IL)-13 concentration using Enzyme-linked Immunosorbent Assay (ELISA) method. Meanwhile, the oxidative stress was examined by measuring the SOD activity using the Nitro Blue Tetrazolium (NBT) method, and the 8-OHdG concentration was quantified by ELISA method.RESULTS: There was an increasing IL-13 in OVAtreated coal dust exposed group along with the increment of 8-OHdG (statistically not significant). SOD activity measured in serum was decreased in all groups (p>0.05). Combination of OVA and coal dust showed the worst effect on IL-13, 8-OHdG and SOD activity.CONCLUSION: Coal dust exposure for four weeks does not adequately induce the oxidative DNA damage in asthmatic mice.KEYWORDS: asthma, coal dust, IL-13, SOD, 8-OHdG

Abstract

Latar belakang: autisme merupakan gangguan perkembangan berat dalam halberkomunikasi, interaksi sosial, dan perilaku yang tampak sebelum anak berusia 3 tahun.Gangguan perkembangan ini dihubungkan dengan faktor psikologi, kelainanneurobiologi, genetik, gangguan metabolik, pencernaan, keracunan logam berat danbiokimia.Tujuan: mengetahui kelainan biokimia yaitu senyawa oksigen reaktif (SOR) dan nitrogenoksida (NO) pada autisme. Peningkatan SOR dan NO diduga menyebabkan stresoksidatif yang berperan dalam neuropatologi pada autisme.Metoda: penelitian terhadap 39 anak autisme sebagai kelompok risiko dan 10 anaknormal sebagai kontrol, melalui studi observasional analitik dengan metode crosssectional.Akumulasi SOR ditentukan dengan mengukur aktivitas katalase dan kadarsenyawa karbonil secara spektrofotometrik. Aktivitas NO ditentukan secara tidaklangsung, yakni mengukur kadar methemoglobin dalam plasma.Hasil: rerata aktifitas katalase pada autisme 0,0061±0,0013 s-1, sedangkan kontrol adalah0,0086±0,0042 s-1. Rerata hasil pengukuran methemoglobin pada autisme adalah30,38±15,44% sedangkan kelompok kontrol 21,73±9,66%. Kadar senyawa karbonilpada autisme adalah 0,411±337693 ìM ml-1/gr dan kontrol 0,424±265861 ìM ml-1/gr.Uji-t untuk dua rata-rata pada a=5% menyimpulkan aktivitas katalase dan kadarmethemoglobin berbeda secara bermakna dibandingkan kontrol (P<0,05), sedangkankadar senyawa karbonil kelompok risiko dan kontrol tidak berbeda (P>0,05)Kesimpulan: aktivitas katalase pada anak autisme lebih rendah dan kadar methemoglobinlebih tinggi secara bermakna dibandingkan anak normal. Meskipun demikian, keadaantersebut belum sampai mempengaruhi kerusakan oksidatif protein. Perlu dilakukanpenelitian lebih lanjut untuk mengetahui aktivitas antioksidan endogen lainnya sepertisuperoksida dismutase dan glutation peroksidase serta apakah kadar senyawa antioksidanberhubungan dengan derajat gejala klinis anak autisme.

Abstract

Background: Stenochlaena palustris, also known as kelakai or lemidi, is frequently linked to anti-inflammatory, anti-bacterial, anti-fungal, and antioxidant properties. S. palustris phytosterols are suggested to suppress the progression of breast cancer. Objective: The objective of this study is to evaluate the potential of phytosterols found in S. palustris to act as estrogen receptor (ER) inhibitors. Methods: Phytosterols (alpha-tocopherol, beta-sitosterol, campesterol, stigmasterol, fucosterol) were docked to estrogen receptor (PDB ID: 7KBS). Molecular docking parameters included Gibb's free energy and interactions between ligand and protein. ADMET properties were analyzed using pkCSM and SwissADME. Results: Alpha-tocopherol showed the highest interaction with the estrogen receptor with ΔG value -8. 9254 kcal/mol (the native ligand, raloxifene, had a G value of -12.052 kcal/mol). Leu387 (hydrogen bond); Phe404 (Phi-phi-T shaped), Leu391, Leu346, Trp383, Leu354, Ala350, Leu525, Leu349 (Alkyl) were among the residues by which a-tocopherol interacted with ER. Alpha-tocopherol has no hepatotoxicity and no skin sensitization. Conclusion: By suppressing ERa, phytosterols from S. palustris may have potential anti-breast cancer activity and may be used to prevent estrogen-dependent human cancers like breast cancer.

Abstract

Cadmium (Cd) and Mercury (Hg) is a heavy metal with high cytotoxicity, implicated as causes of chronic inflammation, oxidative stress, obesity, hyperglycemia, and even diabetes. The long-term exposure of Cd and Hg can affect enzymes involved in carbohydrate metabolism, on the process of glycolysis and glycogenesis. by changing the structure and enzymatic activity of proteins which can cause toxic effects on enzymes involved therein including glycogen synthase (GS), which can cause glycogen content depletion and has potential to limit the glycolysis process in liver and muscles by decreasing the phosphofructokinase (PFK) enzyme activity. There are not many studies that explain the interactions between Cd and Hg on GS and PFK enzymes. For this reason, this research was carried out using in silico. The structure of the enzymes was obtained from the RCSB Protein Data Bank (http://www.rcsb.org) with the following code, GS (PDB ID: 1RZV) and PFK (PDB ID: 4WLO). The interactions between Cd and Hg with these enzymes were used by MIB: Metal Ion-Binding site prediction and docking server (http://bioinfo.cmu.edu.tw/MIB/). The interactions between Cd and amino acids of targeted protein were visualized on UCSF Chimera 1.14. The results showed that Hg interacted with amino acid residues at the active site of the glycogen synthase enzyme at the end of the C-terminal domain, namely at 3 cysteine ??residues Cys 295, cys 366, and cys 390. Meanwhile, Cd and Hg did not interact with amino acid residues. on the active site of the phosphofructokinase enzyme, but interacts with the protein structure of the enzyme.

Abstract

Potensi air gambut di Kalimantan sebagai air  baku yang berlimpah tidak secara langsung dapat digunakan untuk keperluan sehari-hari seperti air minum, mandi dan mencuci. Kecamatan Gambut dan Kabupaten Hulu Sungai Selatan memiliki kualitas air gambut yang ekstrim yaitu derajat keasaman pH antara 3,7 – 4,3, kandungan zat organik tinggi dengan nilai KMnO4 antara 38-280 mg/l, serta kadar logam terlarut  Fe antara 0,45-5,96 mg/l. Kandungan logam besi dan mangan terlarut yang tinggi dapat mempengaruhi kehidupan biota, sumber daya dan kenyamanan ekosistem perairan. Tujuan Penelitian ini untuk menganalisis kemampuan rancang media penyaring campuran pasir dan ampas tahu kering dalam berbagai variasi berat dan waktu kontak untuk menurunkan kadar logam besi dan mangan pada air gambut. Penelitian ini menggunakan metode quasi eksperimental ( tidak dilakukan randomisasi) dengan post test group design tanpa kelompok kontrol. Media filter terdiri atas pasir dan ampas tahu kering dengan variabel bebasnya yaitu waktu kontak dan berat penjerap. Data dianalisis menggunakan uji Anova dua arah, kemudian dilanjutkan dengan uji Post Hoc. Uji Anova dua arah menunjukkan hubungan yang signifikan dan terdapat interaksi pada variabel waktu kontak dan berat penjerap dalam menurunkan kadar besi dan logam di dalam air gambut (p<0,05). Kesimpulan penelitian ini yaitu terdapat hubungan signifikan dalam menurunkan kadar besi dan mangan dilihat dari waktu kontak dan berat penjerap.

Abstract

Sectio caesarea merupakan persalinan buatan dimana janin dilahirkan melalui suatu insisi pada dinding perut dan dinding rahim serta berat diatas 500 gr. World Health Organization (WHO), menetapkan standar rata-rata sectio caesarea disebuah negara sekitar 5-15% per 1000 kelahiran. Kementerian Kesehatan RI mematok persalinan secara sectio caesarea sebanyak 20% dari total persalinan di Indonesia. Berdasarkan data yang didapatkan di RSUD Dr. Moch. Ansari Saleh pada tahun 2019 persalinan dengan tindakan sectio caesarea sebanyak 586 kasus (24%). Tujuan penelitian ini untuk untuk menganalisis hubungan usia ibu, paritas, komplikasi kehamilan, penyulit persalinan dan jaminan kesehatan dengan tindakan sectio caesarea. Metode penelitian analitik observasional dengan pendekatan kasus kontrol retrospektif. Populasi penelitian adalah seluruh ibu bersalin di RSUD Dr. H.Moch. Ansari Saleh Banjarmasin pada tahun 2019 dengan jumlah 2495 orang. Analisis data secara deksriptif dan statistik dengan menggunakan uji chi-square dan uji regresi logistik berganda. Hasil uji chi-square menunjukkan terdapat hubungan antara usia ibu (0,000), paritas (0,002), komplikasi kehamilan (0,000), penyulit persalinan (0,000) dan jaminan kesehatan (0,035) dengan tindakan sectio caesarea. Hasil analisa regresi logistik menunjukkan variabel usia ibu memiliki hubungan yang paling dominan dengan tindakan sectio caesarea (p=0,000, exp. B= 9,000).

Abstract

Cadmium and its compounds are used for various industrial interests. However, the accumulation of cadmium in liver will lead to liver damage. This research aims to analyze the effect of the long exposure of cadmium to wards the levels of H2O2, MDA and MG in white-mouse livers. The number of white-mouse livers used in this research were 24 male mouses which were divided into four treatments of cadmium exposure for zero, two, four and six weeks. After the surgeries, the levels of H2O2, MDA and MG of the white-mouse livers were measured using spectrophotometer UV – VIS. The results showed that there were significant differences in the levels of H2O2 between treatment without exposure to Cd and that exposure. The levels of H2O2 increased respectively 3.164 mmol, 11.076 mmol, 16.292 mmol and 31.588 mmol at the weeks of 0, 2, 4, and 6. The results of the Kruskal-Wallis statistical test (p = 0.000; p <0.05), stated that the long exposure of Cd increased significantly the level of H2O2 in the white-mouse livers. The level of MDA of the white-mouse livers also increased consecutively in the week of 0, 2, 4, and 6 as much as 211.5 μM, 230.6 μM, 269.2 μM and 533 μM. The results of the Kruskal-Wallis statistical test (p = 0.000; p <0.05) showed that the Cd exposure could significantly increase the MDA levels in white-mouse livers. This happened because the Cd exposure activated the phagocytic cells to perform respiratory burst resulting in the accumulation of peroxide compounds. The increased Peroxide with Fe metal that existed in the cytoplasm triggered the lipid peroxidation in the membrane producing the MDA compounds. In addition, the results also showed the increased levels of liver MG consecutively in week 0, 2, 4, and 6 as much as 20.039 %, 24.055 %, 27.985 % and 33.87 % . The results of the Kruskal - Wallis statistical test (p = 0.001 ; p < 0.05), showed that the Cd exposure increased significantly the level of MG which was caused by the metal Cd increased the glycation reaction, i.e., the reaction between glucose and proteins to form Amadori product. At the time of the Amadory formation, the formation of 2,3- enediol which was are easily oxidized by the presence of Cd and oxygen to produce MG occurred. It can be concluded that the exposure of heavy metal cadmium (Cd) for 6 weeks may increase the levels of H2O2, MDA and MG

Abstract

Mechanism process and work activity  tyre unit in PT. Rahman Abdijaya workshop using machines and working equipments. Noise with high intensity of those machines and working equipments will raise noise exposure and will increase the risk of the worker.  This study aims to analysis affect of noise to hearing function and blood pressure at tyre worker in workshop PT. Rahman Abdijaya   in  Tabalong district.  It is an observational study with cross-sectional design. Sample size was 32 worker. Data collection by audiometric  measurement, tension before and after working measurement also noise in working environment measurement. Univariat analysis, bivariat analysis. With chi-square test are used to analyzed the data. The results of the study showed that there was not significant affect of noise to hearing function (sig.α = 0,401 > 0,05), but there was significant affect of noise to sistole blood pressure (sig.α = 0,039 < 0,05) and diastole blood pressure (sig.α = 0,019 < 0,05). Suggestion for the company are necessary noise measurement in all working company area, maintenance activity must be done for periodically and all of broken equipment must be replaced, audiometric measurement every ones in a year, blood pressure measurement must be controlled, Safety sign compatible as safety and health risk, prepared APD like ear muff, training  and socialized about noise affect to the health and how to control them.

Abstract

The purpose of this study was to analyze the influence of meteorological factors directly or indirectly through the concentration of particulate (PM10) on the incidence of Acute Respiratory Infections (ARI) in the District of South Banjarbaru, Banjarbaru. The method used in this research is cross-sectional study, where data meteorological factors, the concentration of particulate matter (PM10) and the incidence of ARI are collected simultaneously. Data meteorological factors and the concentration of particulate matter (PM10) derived from Banjarbaru Climatological Station, while data came from health ARI Banjarbaru and Sei Besar which is located in the district of South Banjarbaru. While the analysis used in this study were Path Analysis (path analysis) was an analysis of the relationship between the independent variables, intermediate variables, and the dependent variable was presented in the form of a diagram. The results showed the meteorological factors that had a direct impact on the incidence of ARI was the largest factor relative air humidity of by 18.7%, followed by a factor of 7.1% of air temperature, wind speed factor and its influence on the intensity of rainfall was below 1%. While the indirect influence of meteorological factors on the concentration of particulate matter (PM10) on the incidence of ARI in the District of South Banjarbaru effect was below 1%. It can be concluded that the direct effect of meteorological factors and the concentration of particulate matter (PM10) on the incidence of ARI in the District of South Banjarbaru significant factor was the relative air humidity and air temperature. While the indirect influence of meteorological factors against ARI through PM10, the effect was not significant.

Abstract

Cassia spectabilis is one of the Indonesian medicinal plants traditionally used to treat different diseases, including malaria. Quality of a drug derived from plants is also influenced by the quality of its raw materials. Thus, in order to assure the quality of products made from plants, it is necessary to standardize the raw materials and following with the antimalarial activity test. The aim of this study was to standardize the quality of C. spectabilis leaves and to evaluate its antimalarial activities of ethyl acetate extract. The fresh material of C. spectabilis leaves were observed its specific and non-specific parameters. In vitro test was done by using Plasmodium falciparum 3D7. In vivo test was done using 4-day suppressive test method against mice infected with P. berghei for four consecutive days. Heme detoxification inhibitory activity test was carried out using the modified Basilico method. The leaves of C. spectabilis meet the quality requirement for raw material of traditional medicine. The ethyl acetate extract showed in vitro antiplasmodial activity against P. falciparum 3D7 and in vivo antimalarial activity against P. berghei infection with IC50 value of 27.28 μg/mL and ED50 value of 1.74 mg/kg, respectively. The extract also showed heme detoxification inhibitory activity with IC50 value of 0.33±0.01 mg/mL. The leaves of C. spectabilis leaves meet the quality requirement and the ethyl acetate extract from standardized C. spectabilis leaves possessed a potential an antimalarial activity which deserves to be further developed.
