Abstract

BACKGROUND: Amino acids are important for proliferation and maintenance of tumor cells. Breast cancer patients were found to have significant changes in the number of amino acids, which are assumed to be correlated with the molecular subtypes of breast cancer. Therefore, current study was conducted to analyze plasma amino acids in breast cancer patients with luminal A and B subtypes.METHODS: Breast cancer and control subjects were recruited, and venous blood was collected for the measurement of plasma amino acids. Total 19 plasma amino acids were measured using reverse-phase high-performance liquid chromatography with C18 column. Mean comparison for normally distributed and homogeneous data was further analzyed using independent sample T-test, with p<0.05 was considered as significant.RESULTS: From total 19 amino acids, only 7 amino acids; cysteine, glutamic acid, histidine, ornithine, threonine, tyrosine, valine, were statistically different between the healthy control and breast cancer subjects. Eventhough no amino acids was found to be statistically different between breast cancer subjects with luminal A and B subtypes, but some amino acids were found to be significantly different when correlated to various breast cancer risk factors.CONCLUSION: Amino acid profile of patients with Luminal A and B subtypes of breast cancer differs compared to healthy controls and is also correlated with breast cancer risk factors. Increase in cysteine level in Luminal A subtype patients and decrease of alanine and leucine in Luminal B subtype patients can be used as a biomarker.KEYWORDS: amino acid, plasma, breast cancer, risk factor, biomarker

Abstract

Dental emergencies are potentially life-threatening conditions and immediate treatment is needed to stop ongoing bleeding, reduce pain and also uncontrolled bleeding, cellulitis or bacterial tissue infection causing swelling in the mouth or mouth area and trauma to the bones in the facial area . Delay in treatment of trauma to the teeth can lead to treatment failure. Delay in emergency dental treatment after trauma will increase the risk of complications, non-vital teeth and an unpredictable prognosis.Dental emergencies often occur at the elementary school level and the most common cases are loose or avulsed teeth due to falls and impacts. Teachers in schools not only have a central role as educators, but teachers also act as first aid agents in dealing with health problems in schools. In addition, teachers also play a role and assist health workers in collecting data and also as the first emergency referral before being handled by health workers.SDI Assalaam has a staff of 46 teachers and 250 students. Having students who are actively moving has a risk of accidents such as falling and so on which can cause injury, especially the oral cavity, while knowledge of dental emergencies and their management according to the Principal of SDI Assalaam has never been received by the teachers who teach at SDI Assalaam , so that if there is a dental emergency the teachers don't know what action to take.To overcome the lack of knowledge about dental emergencies and their management. So the teachers of SDI Assalam are given counseling and training about dental emergencies and their handling so that the importance of dental emergencies is socialized so that teachers at SDI Assalaam can increase their knowledge about dental emergencies and their management.Based on the evaluation results of community service activities. There was a significant increase in the level of knowledge of SDI Assalaam teachers regarding dental emergencies and their treatment.

Abstract

BACKGROUND: Currently, laboratory diagnosis of intestinal tuberculosis (ITB) is limited based on clinical manifestations, providing opportunities for alternative laboratory tests to diagnose ITB. At the present time, the role of serum adenosine deaminase (ADA) and hematological tests in ITB patients are not widely known. The objective of this study was to determine the role of ADA and hematological tests in patients suspected with ITB.METHODS: Subjects that were suspected of ITB were classified as ITB group, while subjects with inflammatory bowel disease, hemorrhoid, and intestinal malignancy were classified as non-ITB group. Colonoscopy, histopathological examinations, and hematological test were performed. ADA measurement was also performed with clinical chemistry analyzer based on enzymatic colorimetry principle.RESULTS: Out of 143 subjects, 16 (11.2%) subjects were diagnosed with ITB and 127 (88.8%) subjects were classified as non-ITB group. ADA level and erythrocyte sedimentation rate (ESR) of ITB group were significantly higher than the ones of non-ITB group (p<0.05). Cut-off, sensitivity, and specificity of ADA level were 12.56 IU/L, 75%, and 57%, respectively. Cut-off, sensitivity, and specificity of ESR were 32.5 mm/hour, 81%, and 62%, respectively. Colonoscopy of ITB subjects displayed multiple ulcerations, edema, and hyperemic mucosa. Histopathological examination of ITB subjects exhibited granulomatous inflammation, epitheloid cells, giant cells, and lymphocyte aggregates.CONCLUSION: ADA level and ESR were significantly higher among ITB patients compared with non-ITB patients. Since the sensitivities of ADA and ESR tests were high, the ADA and ESR tests could be considered as a screening test for ITB.KEYWORDS: intestinal tuberculosis, adenosine deaminase, hematological tests

Abstract

BACKGROUND: Emergence of drug resistance due to epidermal growth factor receptor (EGFR) Exon 20 T790M poses a challenge in the effective management of non-small cell lung carcinoma (NSCLC). Significant breakthrough in the management of NSCLC with a specific genetic alteration causes substantial condition improvement in patients whose cancer progressed after first-generation tyrosine kinase inhibitor treatment and who developed tumors with EGFR Exon 20 T790M mutation. The present study analyzed a cohort of NSCLC patients and investigated the incidence of the EGFR Exon 20 T790M status with Osimertinib therapy, along with its impact on survival rates.METHODS: This was a retrospective cohort study on 22 NSCLC subjects who were genetically examined for EGFR status from plasmic cell free total nucleic acid. Subjects with EGFR Exon 20 T790M mutation were treated with/without Osimertinib. Demographic and clinical data were descriptively summarized, and the differences of each variable and correlation between survival rate and EGFR Exon 20 T790M were analyzed.RESULTS: Subjects with (n=13) and without (n=9) EGFR Exon 20 T790M had survival rates of 10.77±2.45 and 4.78±1.48, respectively (p=0.000). Based on 1-year survival status of subjects with EGFR Exon 20 T790M, there were 3 Osimertinib-treated survivors and 2 Osimertinib-treated non-survivors. Eight subjects with EGFR Exon 20 T790M and without Osimertinib treatment did not survive (p=0.001).CONCLUSION: Since the treatment of Osimertinib demonstrated a noteworthy survival rate among NSCLC subjects with EGFR Exon 20 T790M, thus Osimertinib could be suggested as a potential targeted therapy for NSCLC subjects with EGFR Exon 20 T790M.KEYWORDS: non-small cell lung carcinoma, EGFR, Exon 20, T790M, osimertinib

Abstract

Dental emergency is associated with immediate measures that are carried out with the aim to alleviate pain. Dental trauma is one of dental emergencies, which is injury to the teeth and or the supporting tissues of the teeth. Dental injuries are often experienced by school-aged children, in which the majority of traumatic dental injuries occur at home or school, hence it is hoped that parents and school teachers can provide appropriate treatment when dental emergencies occur in children. This community service counseling aims to increase knowledge and skills in the management of dental emergencies. Counseling was carried out by the Trisakti University Faculty of Dentistry Team to parents and teachers of Al-Ikhlas Islamic Elementary School, Cipete, Jakarta Selatan on March 11, 2023. Counseling includes learning materials regarding the management of dental emergencies and dental health education through powerpoint presentation, video, and live demonstration. Final questionnaire shows an increase in knowledge and understanding regarding the management of dental emergencies. Therefore, it can be concluded that after the counseling activity, there is an increase in the level of knowledge among parents and teachers of Al-Ikhlas Islamic Elementary School regarding the management of dental emergencies. Further training is needed to improve the ability to handle dental emergencies better.

Abstract

ABSTRAK Anak – anak usia sekolah dasar, yaitu usia 6-10 tahun adalah populasi resiko karies tinggi yaitu mencapai 54%. Hal ini termasuk pada anak usia sekolah di SDK Jakarta Barat. Sekolah ini memiliki jumlah siswa yang banyak dan terdapat fasilitas UKGS (unit kesehatan gigi sekolah). Akan tetapi, dokter gigi berpraktik hanya 1x seminggu untuk anak – anak SD kelas 1. Hal ini  menyebabkan kurangnya tenaga penyuluhan kesehatan gigi terutama dalam upaya mencegah gigi berlubang dan tatalaksana darurat jika terjadi trauma pada anak - anak. Pengabdian kepada mayarakat ini bertujuan mengedukasi anak-anak SDK Di Jakarta Barat dalam mencegah dan mangatasi permasalahan kesehatan gigi sederhana yang sering terjadi di usia mereka dengan metode interaktif. Kegiatan ini diikuti oleh 137 orang murid kelas 1. Edukasi dilakukan dengan metode interaktif, diawali dengan paparan menggunakan media audio visual dan tanya jawab serta permainan.  Keberhasilan metode ini terlihat dari tingkat antusias peserta anak dalam mengikuti rangkaian kegiatan penyuluhan dengan tertib dan dapat diarahkan serta evaluasi pretest dan post test peserta edukasi. Hasil menunjukkan bahwa ada peningkatan secara signifikan pengetahuan anak-anak dengan metode interaktif. Metode interaktif merupakan metode yang efektif dan menyenangkan untuk edukasi kesehatan gigi pada anak usia 6-7 tahun di SDK Jakarta Barat. Kata Kunci: Metode Interaktif, Edukasi, Kesehatan Gigi Anak  ABSTRACT Children aged 6-10 years have a high caries risk, reaching 54%. This includes school-age children at SDK Jakarta Barat, especially grade 1 children. This school has many students, and even though this school has Usaha Kesehatan Gigi Sekolah (UKGS), the dentist practices only once a week for grade 1 elementary school children. This results in insufficient information needs for a large number of students. In this community, there are no external health workers who provide education about dental health, in this case how to prevent cavities and emergency management if trauma occurs in children.  This community service aims to educate elementary school children in West Jakarta on preventing basic dental health problems that often occur. This activity was attended by 137 grade 1 students. Education was conducted using an interactive method, starting with exposure using audio-visual media, questions and answers, and games. Results are seen from educational participants' pretest and posttest evaluation. The results show a significant increase in children's knowledge with this interactive method. Abstract

Recent advances in cancer treatment have focused on developing alternative therapies with reduced adverse effects. Chemoprevention using natural products derived from plants has gained significant attention. Apium graveolens has demonstrated anticancer properties against various cancer cell types, suggesting its potential efficacy against tongue cancer cells. The aim of this study was to evaluate the cytotoxic effects and mechanisms of action of Apium graveolens leaf ethanolic extract (AGLEE) on the HSC-3 tongue cancer cell line. The leaves were processed and extracted with 70% ethanol to obtain an ethanolic extract. HSC-3 cells were cultured, subjected to starvation, and pre-treated with or without Z-DEVD-FMK, a caspase-3 inhibitor. Subsequently, the cells were treated with or without doxorubicin or varying concentrations of AGLEE. To assess cell viability and apoptosis, MTT and sub-G1 assays were performed. Additionally, treated HSC-3 cells were collected, lysed, and analyzed for levels of cleaved-caspase-3 and cleaved-poly (ADP-ribose) polymerase (cleaved-PARP) using ELISA. The inhibitory concentration (IC50) value of AGLEE for reducing viable HSC-3 cells was determined to be 48.29 μg/mL. AGLEE significantly decreased HSC-3 cell viability and increased the percentage of apoptotic cells. It exhibited a concentration-dependent reduction in cell viability and an increase in apoptosis. Furthermore, the extract elevated the levels of cleaved-caspase-3 and cleaved-PARP in HSC-3 cells. Pre-treatment with Z-DEVD-FMK reduced the levels of cleaved-caspase-3 and cleaved-PARP induced by AGLEE. Taken together, AGLEE could be proposed as a potential natural therapeutic agent by inducing apoptosis through the caspase-3/PARP pathway in tongue cancer cells.

Abstract

Dental caries is a significant health issue in Indonesia, particularly among preschool children, due to a lack of knowledge and habits in maintaining dental hygiene. Schools serve as strategic venues to promote dental health through activities such as tooth-brushing programs. Teachers play a crucial role in setting examples and guiding children in practicing routine oral hygiene. A health education program at Santa Maria Juanda Kindergarten aims to enhance awareness and understanding of oral health among teachers and students. Based on the pretest and posttest results, this results demonstrate an increase in knowledge by 12.69%, and participants are expected to be able to use the knowledge they have acquired in their everyday lives.

Abstract

Background. Regenerative medicine is a new therapeutic modality using cell, stem cell and tissue engineering technologies. Purpose. To describe the regenerative capacity of dental pulp mesenchymal stem cell. Review. In dentistry, stem cell and tissue engineering technologies develop incredibly and attract great interest, due to the capacity to facilitate innovation in dental material and regeneration of dental and oral tissues. Mesenchymal stem cells derived from dental pulp, periodontal ligament and dental follicle, can be isolated, cultured and differentiated into various cells, so that can be useful for regeneration of dental, nerves, periodontal and bone tissues. Tissue engineering is a technology in reconstructive biology, which utilizes mechanical, cellular, or biological mediators to facilitate regeneration or reconstruction of a particular tissue. The multipotency, high proliferation rates and accessibility, make dental pulp as an attractive source of mesenchymal stem cells for tissue regeneration. Revitalized dental pulp and continued root development is the focus of regenerative endodontic while biological techniques that can restore lost alveolar bone, periodontal ligament, and root cementum is the focus of regenerative periodontic. Conclucion. Dentin-derived morphogens such as BMP are known to be involved in the regulation of odontogenesis. The multipotency and angiogenic capacity of DPSCs as the regenerative capacity of human dentin / pulp complex indicated that dental pulp may contain progenitors that are responsible for dentin repair. The human periodontal ligament is a viable alternative source for possible primitive precursors to be used in stem cell therapy.

Abstract

BACKGROUND: Insulin resistance (IR) is considered as the main driver of polycystic ovary syndrome (PCOS) pathogenesis. In PCOS condition, IR is frequently related to glucose, anthropometric profile, lipid profile, and hormone profile parameters. However, not all PCOS phenotype show IR. Therefore, this study was conducted to determine the association the parameters mentioned above in PCOS subjects with and without IR.METHODS: Fifty PCOS women with IR and 26 PCOS women without IR were recruited. All subjects underwent physical examination for measurement of weight, waist circumference (WC), and body mass index (BMI). Ferriman Gallwey Score (FGS) was used to evaluate hirsutism. Blood sample was taken from each subject for measurement of fasting glucose, postprandial glucose, fasting insulin, low-density lipoprotein (LDL), high-density lipoprotein (HDL), total cholesterol, triglyceride (TG), sex hormone binding globulin (SHBG), thyroid-stimulating hormone (TSH), luteinizing hormone (LH), follicle stimulating hormone (FSH), and prolactin. Homeostatic model assessment for IR (HOMA-IR), TG-glucose index (TyGI), and free testosterone index (FTI) were then calculated.RESULTS: From all the parameters examined, only fasting insulin (p<0.001), HOMA-IR (p<0.001), SHBG (p=0.012), TG (p<0.001), and TyGI (p=0.008) that show significant differences between PCOS subjects with and without IR. After multivariate analysis, TyGI was found to have strong association with IR occurrence in PCOS subjects (p=0.005) with an odd ratio of 5.26 (1.65–16.74).CONCLUSION: TyGI appears to have a significant association with the IR occurrence in PCOS subjects. Hence, it can be suggested that TyGI could be an important marker for PCOS women with IR.KEYWORDS: insulin resistance, lipid metabolism, polycystic ovary syndrome, triglyceride-glucose index
