Abstract

Artificial insemination (AI) of dairy cows using sexing semen is the right choice to obtain female calves as replacement stock. The success of AI is influenced by the quality of the spermatozoa used. The purpose of this study was to determine the quality of semen produced by Lembang Artificial Insemination Center. The material used is 30 frozen semen straws of FH cattle produced by Lembang Artificial Insemination Center. This research method uses observational methods. The research variables included individual motility, viability, abnormalities, concentration, total motile spermatozoa, and intact acrosome caps of spermatozoa. The data obtained were analyzed by the Analysis of Variance (ANOVA) test and continued with Duncan's test if significantly different. The results showed that individual motility, concentration, total motile spermatozoa, and intact acrosome caps of spermatozoa had a very significant effect on all treatments (P<0.01), while spermatozoa abnormalities had a significant effect (P<0.05) on all treatments, but on viability, there was no effect (P> 0.05) in all treatments. Motility of individual spermatozoa in non-sexing semen (55.80±4.56)%, sperm X (46.30±5.36)%, sperm Y (46.70±6.40)%. Viability of spermatozoa in non-sexing semen (74.57±4.05)%, sperm X (73.24±2.30)%, sperm Y (72.51±2.68)%. Spermatozoa abnormalities in non-sexing semen (2.73±2.24)%, sperm X (4.94±1.57)%, sperm Y (3.25±2.72)%. Concentration of non-sexing semen (33.88±1.98 million/mini straw), sperm X (25.06±4.29 million/mini straw), sperm Y (25.13±5.74 million/mini straw). The number of sperm motile non-sexing (18.60±1.58 million/mini straw), sperm X (11.68±2.82 million/mini straw), sperm Y (25, 11.64±2.68 million/mini straw). Intact acrosome caps, non-sexing semen (78.40±4.84)%, sperm X (74.08±3.80)%, sperm Y (70.70±4.78)%.

Abstract

This study was carried out to evaluate the seasonal variation on semen characteristics of Murrah buffalo bulls in Indonesia. A total of three Murrah buffalo bulls were included in this study. The semen was collected once a week during the rainy season (from January to April and from October to December) and the dry season (from May to September). Immediately after collection, the semen volume, sperm concentration, total sperm output, individual motility, post-thawing motility, and straw production were evaluated. The results showed that the season did not change (P0.05) the semen volume, individual motility, and post-thawing motility. However, sperm concentration, total sperm output, and straw production were decreased in the dry season (P0.05) as compared to the rainy season. In conclusion, several semen characteristics of Murrah buffalo bulls may vary depending on the season of collection. The semen collection during the rainy season is more profitable than the dry season.

Abstract

The purpose of this research was to analyse the quality and storability of native chicken semen at room temperature after diluted with physiological saline solution (NaCl) supplemented with noni fruit extract. Research methodology used was nested randomized design which variables observed covering the quality of chicken spermatozoa macroscopically and microscopically with the doses of noni fruit extract of 0 % (P0), 10 % (P1), 20 % (P2), and 30 % (P3). Rate of spermatozoa motility at 0, 1, 2, and 3 hours on P0 were 83.70±4.74, 63.30±9.63, 32.90±8.52, and 15.00±6.20; P1 were 85.30±5.44, 72.70±10.06, 51.90±11.75, and 28.50±5.68; P2 were 84.60±6.40, 78.50±8.59, 57.90±10.73, and 33.70±9.06; and P3 were 81.80±7.30, 64.20±7.93, 40.30±11.66, and 19.80±7.47, respectively. Rate of spermatozoa viability at 0, 1, 2, and 3 hours on P0 were 85.30±9.09, 65.10±6.15, 32.10±11.86, and 10.30±9.09; P1 wer 87.40±6.22, 72.70±5.33, 50.80±13.59, and 26.50±10.99; P2 were 68.58±5.30, 77.70±4.79, 56.30±13.76, and 32.70±13.79; P3 were 81.20±8.04, 68.70±10.40, 36.20±16.61, and 16.90±11.93, respectively. Rate of spermatozoa abnormality at 0, 1, 2, and 3 hours on P0 were 8.70±3.40, 12.20±4.42, 17.80±5.67, and 20.30±6.38; P1 were 7.80±2.04, 9.80±2.69, 13.60±4.45, and 16.50±5.19; P2 wer 8.40±4.33, 10.00±2.45, 12.50±5.21, and 15.80±3.71; and P3 were 9.60±3.41, 10.90±2.64, 17.10±5.61, and 21.20±8.16, respectively. It can be concluded that the addition of noni fruit extract at the dose of 10 % and 20 % in physiological saline solutin maintain the native chicken spermatozoa quality up to 2 hours post storage at room temperature.

Abstract

The purpose of this current study was to evaluate the influence of nutritional flushing on body morphometrics of female breeding goats. This study used 32 local female goats or does with an average age of 3.28±1.08 years and an average body weight of 42.47±8.28 kg. The goats were distributed in a completely randomized design using a 2 x 2 factorial pattern with 8 replications. The first factor was the flushing feed (FF) type, consisting of rice bran and concentrate, whereas the second factor was the level of tannin-protected arginine (TPA: 0 and 300 mg/kg). The body morphometrics observed in this study were initial body length (IBL), final body length (FBL), body length change (BLC), initial chest girth (ICG), final chest girth (FCG), chest girth change (CGC), initial wither height (IWH), final wither height (FWH), and wither height change (WHC). The IBL, FBL, BLC, ICG, FCG, IWH, FWH, and WHC of the local does were not significantly different (P0.05) after receiving the FF type. However, FF significantly affected the CGC of the goats (P0.001). The use of concentrate for the local female local goats resulted in a higher CGC (P0.001) compared to the use of rice bran. On the other hand, TPA did not significantly affect all body morphometrics of the goats (P0.05). Thus, it can be concluded that the use of concentrate leads to higher chest girth change, giving better body growth compared to the use of rice bran. The addition of tannin-protected arginine was also found to have no significant contribution to the body morphometrics of the local female goats. _____________

Abstract

Penelitian ini bertujuan mengidentifikasi kandungan gonadotropin releasing hormone (GnRH) ekstrak jaringan otak sapi dewasa pada fase folikuler (ovari berfolikel) dan fase luteal (ovari berkorpus luteum). Pada penelitian ini digunakan 2 buah otak sapi perah peranakan Friesien Holstein (FH) dewasa dengan umur 7-8 tahun yang pada saat disembelih kondisi ovarinya menunjukkan fase folikuler (1 buah otak) dan fase luteal (1 buah otak). Analisis kandungan GnRH dilakukan dengan metode enzyme-linked immunosorbent assay (ELISA) indirect. Hasil penelitian menunjukkan bahwa konsentrasi GnRH bagian otak sapi pada fase folikuler vs fase luteal berturut-turut untuk bagian otak parietal lobe of cerebral hemisphere, corpus callosum, thalamus, hipothalamus, pituitary gland (hipofise), midbrain, cerebellum, pons, dan medulla oblongata masing-masing adalah 260 vs 270, 110 vs 170, 230 vs 320, 1590 vs 1310, 840 vs 1250, 530 vs 810, 200 vs 480, 230 vs 100, dan 220 vs 70 μg/ml. Dari hasil penelitian disimpulkan bahwa semua bagian otak sapi yang diamati mengandung GnRH dengan konsentrasi yang berbeda antara fase folikuler dengan fase luteal.

Abstract

This study investigated the influences of rainy and dry seasons on semen characteristics of fat-tailed and Garut rams under tropical conditions. A total of 4 healthy rams (2 fat-tailed rams and 2 Garut rams) aged 3 to 4 years old were used in this study. The semen was collected from each ram during November 2016 to February 2017 (rainy season) and May to August 2017 (dry season). The measurement of semen characteristics including semen volume (SV), sperm concentration (SpC), total sperm (TSp), individual sperm motility (ISM), post-thawing sperm motility (PTSM), and frozen semen production (FSP). The rainy season resulted higher SV (P= 0.021), TSp (P= 0.005), ISM (P= 0.028), and FSP (P= 0.005) than the dry season. On the other hand, fat-tailed rams had higher (P0.001) SV, TSp, and FSP compared to Garut rams. There was no interaction (P0.05) between season and breed on all semen characteristic parameters found in the present study. It can be concluded that the semen collection during the rainy season had a beneficial impact to produce high-quality semen for artificial insemination program than in the dry season. Fat-tailed rams have better semen characteristics compared to Garut rams.

Abstract

Penelitian bertujuan mengetahui kualitas spermatozoa ayam kampung dalam pengencer NaCl fisiologis dengan lama simpan yang berbeda pada suhu kamar. Materi yang digunakan dalam penelitian ini adalah semen ayam kampung berumur 1-1,5 tahun. Lama simpan pada suhu kamar adalah 0, 30, 60, 90, dan 120 menit dengan 10 kali ulangan. Rasio pengenceran semen dengan pengencer 1:10. Variabel yang diamati yaitu motilitas dan viabilitas spermatozoa. Metode yang digunakan adalah metode percobaan menggunakan rancangan acak lengkap (RAL). Hasil analisis statistik menunjukkan bahwa lama simpan semen ayam kampung pada suhu kamar dengan pengencer NaCl fisiologis selama 0, 30, 60, 90, dan 120 menit berpengaruh sangat nyata (P0,01) terhadap motilitas individu dan viabilitas. Rata-rata motilitas individu pada penyimpanan selama 0, 30, 60, 90, dan 120 menit masing-masing adalah 68,5+5,5; 58,5+5,79; 49,5+7,25; 39,5+8,9; dan 26+11,7% sedangkan rata-rata viabilitas masing-masing adalah 83,1+3,63; 77,6+3,47; 67,8+4,4; 57,7+7,7; dan 45,6+7,19%. Dapat disimpulkan bahwa motilitas dan viabilitas spermatozoa ayam kampung mengalami penurunan secara bertahap seiring dengan lama simpan pada suhu kamar.

Abstract

This study aims to determine the effect of the substitution of bovine serum albumin (BSA) with cattle blood serum and egg white in the diluent of Cauda epididymal Plasma 2 (CEP-2) on sperm quality of Limousin cattle during cooling at 3-5 C. The research material used was rejected Limousin bull sperm (motility of 50-60%) from Artificial Insemination Centre Singosari, Malang. This research was a laboratory experiment using a randomized block design which was composed of six treatments with 10 replications, those were T0 as controls ((90% CEP-2 with BSA + 10% egg yolk); T1 (83.84% CEP-2 + 6.16% cattle blood serum + 10% egg yolk); T2 (81.84% CEP-2 + 8.16% cattle blood serum + 10% egg yolk); dan T3 (90% CEP-2 + 0,4% egg white + 10% egg yolk); T4 (90% CEP-2 + 0.8% egg white + 10% egg yolk); and T5 (90% CEP-2 without BSA + 10% egg yolk). Parameters measured were the percentage of motility, viability, and abnormality of sperms. Results of research after 48 hours of storage showed that the percentage of sperm motility in T0, T1, T2, T3, T4, and T5 were 40.50±5.90, 36±36.16, 34.00±6.58, 40.50±3.69, 38.50±3.37, and 38.50±4.12, respectively, while the percentage of sperms viability were 75.16±4.30, 70.50±2.88, 73.80±2.80, 74.80±3.30, 75.13±3.13, and 74.03±4.13, respectively, and the percentage of sperms abnormality were 10.14±2.34, 10.62±1.34, 11.33±2.00, 10.94±2.82, 10.02±1.95, and 10.78±1.96, respectively. Abstract

This research was conducted to determine the effect of glucose supplementation in liquid semen diluent on the quality of Kampung Unggul Balitbangtan (KUB) spermatozoa.The chicken used is KUB chicken as many as 8 tails.The research procedure consists of; the manufacture of diluent, collection of semen, dilution of semen, storage, and evaluation of semen.Evaluation of semen was carried out on motility, viability, abnormalities, and plasma membrane integrity at 0 and 3 days of storage.Data were analyzed using the T-test to compare the quality of spermatozoa with added glucose into the liquid semen diluent and without the addition of glucose.The addition of glucose into the diluent was able to maintain spermatozoa motility at 35.00 ± 11.50, spermatozoa viability at 41.42 ± 10.15, and spermatozoa plasma membrane integrity at 82.94 ± 7.42 at 5oC storage for three days.This study concluded that glucose supplementation into liquid semen diluent was able to maintain plasma membrane motility, viability, and integrity at 5oC storage for three days, spermatozoa quality of KUB chickens was able to survive at 5oC storage for three days, and glucose can be added to the liquid semen diluent at a dose of 10 mM.

Abstract

There is lack of evidence on the black soldier fly (Hermetia Illucens L.) and its use as a biological control and waste recycle agent. Unknown information related with mating and ovipositional activities. The aim of this research is to determine the reproductive profile and its manure of the Black Soldier Fly (Hermetia Illucens L.) through different rearing substrate as ruminant feed. A total of 4 treatments and 4 replicates were used in this experiment. The treatment diet was formulated as follows:    T0:  dry fermented cow waste, T1: 21% of crude protein, T2: 22% of protein, T3: 23% of protein. The data was analyzed using ODA SAS on an academic general linear model (GLM).  The result showed using different rearing substrates gave insignificant differences (p > 0.05) on the reproductive profile, nutritional content, and manure nutritional content. In summary, the use of different leves of protein can be used as the rearing substrate without any adverse effects.
